Abstract

Description
만성 염증은, 알츠하이머 질병, 당뇨병, 죽상 동맥 경화증 및 암을 포함하는 다양한 질병에 관련되어 있다. 또한, 자가 면역 질병, 예를 들면, 골관절염 및 크론병도 만성 염증에 관련되어 있다. 만성 염증은, 병변 부위 근방에서 기준선보다 높은 수준에 있지만 급성 염증에서 발견되는 것보다 몇 배 낮은 수준으로 존재하는 전염증성 인자(pro-inflammatory factor)를 특징으로 할 수 있다. 이러한 인자의 예로는, TNF, IL-1α, IL-1β, IL-5, IL-6, IL-8, IL-12, IL-23, CD2, CD3, CD20, CD22, CD52, CD80, CD86, C5 보체 단백질, BAFF, APRIL, IgE, α4β1 인테그린 및 α4β7 인테그린를 포함한다. 만성 염증에 관련된 질병의 치료는, 예를 들면, 인자의 수용체에 결합하거나 인자에 결합함으로써 전염증성 인자의 작용을 방해하는 치료를 포함한다.

정의
용어 "최종당화산물" 또는 "AGE-개질 단백질"(또한, 최종당화산물)은, 단백질의 측쇄와 당의 반응 결과로 형성되고 추가로 재배열하여 비가역적 가교를 형성하는 개질된 단백질을 지칭한다. 이러한 공정은, 환원 당과 아미노기 사이의 가역적 반응을 시작해서 시프 염기를 형성하고, 공유 결합된 아미도리 재배열 산물을 형성한다. 아미도리 산물이 형성되면, 추가로 재배열하여 AGE를 생성한다. ("Clearance of p16Ink4a-positive senescent cell delays ageing-associated disorders", Nature, vol. 479, pp. 232-236, (2011))에 의한 최근의 연구에서는, 마우스 내에서 노화 세포의 클리어런스 효과를 조사했다. 그러나, 염증 및 전염증성 인자에 대한 효과는 기재되어 있지 않았다. 본 출원 이전에, 염증 및 전염증성 인자에 대한 노화 세포의 제거 또는 사멸의 효과는 공지되어 있지 않았다.

실시예
실시예 1: 안티 최종당화산물 항체 투여의 생체내 연구
안티 최종당화산물 항체의 효과를 조사하기 위해서, 항체는 노화된 CD1(ICR) 마우스(Charles River Laboratories)에, 정맥내 주사에 의해 1일 2회, 1주 1회, 3주간( 1일, 8 일 및 15일) 투여된 후, 10 주간 치료를 하지 않았다. 시험 항체는 키홀 림펫 헤모시아닌과 컨쥬게이팅 된, 카르복시 메틸리신, 일반적인 AGE 에피토프에 대해서 발생되는 시판되는 마우스 안티 최종당화산물 항체이었다. 대조 동물에는 생리식염수 대조구 기준을 사용하였다.
"젊음"으로 지칭된 마우스는 8 주령이었고, "늙음"으로 지칭된 마우스는 88주령(±2일)이었다. 항체의 투여로부터 부작용은 발견되지 않았다. 연구에 사용된 동물의 상이한 그룹은 표 1에 표시된다.
노화 세포의 마커인 P16INK4a mRNA은, Real Time-qPCR에 의해서 그룹의 지방 조직 내에서 정량화되었다. 결과는 표 2에 표시된다. 표에서, ΔΔCt = ΔCt 평균 대조 그룹(2) - ΔCt 평균 실험 그룹 (1 또는 3 또는 5):발현 배수 = 2 - ΔΔCt.
상기 표에서는, 치료되지 않은 늙은 마우스(대조 그룹 2)가 예상대로 치료되지 않은 젊은 마우스 (대조 그룹 1)보다 2.55 배 많은 p16Ink4a mRNA가 발현되는 것이 표시된다. 이는, 치료 22일에 안락사시킨 그룹 1의 치료되지 않은 젊은 마우스에 비해 회복 85일에 안락사시킨 그룹 2의 치료되지 않은 늙은 마우스를 비교하는 경우 관찰되었다. 그룹 2의 치료되지 않은 늙은 마우스의 결과는, (85일에 안락사시킨)그룹 3의 치료된 늙은 마우스의 결과와 비교하는 경우, p16Ink4a mRNA는 그룹 3에서보다 그룹 2에서 1.23 배 높은 것이 관찰되었다. 따라서, p16Ink4a mRNA 발현의 수준은, 늙은 마우스가 항체의 2.5 μg/gram/BID/주에 치료된 경우보다 낮았다.
그룹 2(대조군)의 치료되지 않은 늙은 마우스의 결과는 그룹 5(5 ug/gram) 치료된 늙은 마우스 (22일에 안락사)의 결과와 비교하는 경우, p16Ink4a mRNA 는 그룹 5(5 ug/gram)에서보다 그룹 2(대조군)에서 3.03 배 높은 것이 관찰되었다. 이러한 비교는, 5.0 μg/gram/BID/주로 치료된 경우 p16Ink4a mRNA 발현의 낮은 수준을 갖는 것을 나타내고, 젊은 치료되지 않은 마우스(즉, 그룹 1)의 것에 필적하는 p16Ink4a mRNA 발현 수준을 제공한다. 회복 85일에 안락사시킨 그룹 3((2.5 ug/gram) 마우스와 달리, 그룹 5 마우스는 치료 22일에 안락사되었다.

실시예 2: 시험 항체의 친화성 및 운동성
실시예 1에서 사용되는 시험 항체의 친화성 및 운동성은 세포의 AGE-개질 단백질에 대한 모델 기질로서 Nα,Nα-비스(카르복시메틸)-L-리신 트리플루오로아세테이트 염 (Sigma-Aldrich, St. Louis, MO)을 사용해서 분석하였다. 라벨-부재 상호작용 분석은 Series S 센서 칩 CM5 (GE Healthcare, Pittsburgh, PA)을 사용하여 BIACORETM T200 (GE Healthcare, Pittsburgh, PA) 상에서 수행하고, Fc1은 블랭크로서 세팅되고 Fc2는 시험 항체로 고정된다(분자량 150,000 Da). 러닝 완충액은 25℃ 온도에서 HBS-EP 완충액 (10 mM HEPES, 150 mM NaCl, 3 mM EDTA 및 0.05% P-20, pH of 7.4)이었다. 소프트웨어는 BIACORETM T200 평가 소프트웨어, 버전 2.0이었다. 이중 기준(Fc2-1 및 완충 주사액)은 분석에 사용되고, 데이터는 랑뮤어 1:1 결합 모델에 피팅되었다.

실시예 3(prophetic): 류마티스 관절염의 항원 유도 관절염(AIA) 마우스 모델 내에서 안티 최종당화산물 항체 및 마우스 항-마우스 TNF 항체의 투여의 생체내 연구
안티 최종당화산물 항체 및 마우스 항-마우스 TNF 항체의 류마티스 관절염에 대한 효과를 조사하기 위해서(자가 면역 질환인 기존의 염증성 질환), 2개의 항체가 CD1(ICR) 마우스에 동시에 투여되고, 먼저 전신 주사 이어서 관절 내 주사에 의해서 메틸화 소 혈청 알부민으로 사전에 치료되어 AIA 마우스를 형성한다. 안티 최종당화산물 항체 및 항-TNF 항체의 조합 투여는 정맥내 주사에 의해서 1일 2회, 1주 1회 3주간이다(1일, 8일, 15일). 안티 최종당화산물 항체는 키홀 림펫 헤마시아닌에 컨쥬게이팅 된 일반적인 AGE 에피토프로서 카르복시메틸 리신에 대해 발생되는 시판되는 마우스 안티 최종당화산물 항체이다. 생리식염수의 대조 기준은 제1대조 동물 내에서 사용되고, 제2실험 그룹은 안티 최종당화산물 항체만 투여되고, 항-TNF 항체의 제2대조 기준이 제2대조 동물 내에 사용된다. 각 항체의 5 μg/gm/BID/주의 용량 수준이 사용된다.
연구 과정 중에 동물을 관찰하고, TNF의 수준을 결정하기 위해서 혈액을 취한다. 연구 종료시에, 동물은 안락사하고, 관절 조직은 류마티스 관절염과 관련된 손상의 징후에 대해서 조사한다. 이러한 결과에 따르면, 제2실험 그룹 및 제2대조 그룹은 제1대조 그룹보다 TNF 수준이 낮고 관절 손상이 덜한 것을 나타낸다. 또한, 이러한 제1 실험 그룹은 모든 연구 그룹의 TNF의 가장 낮은 수준 및 적은 관절 손상뿐 아니라, 관절 손상 및 TNF 수준의 감소가 제2실험 그룹 및 제2대조 그룹에 기초해서 예상된 것보다 크다. 이러한 결과는, 안티 최종당화산물 항체 및 항-염증 항체를 사용해서 시너지 효과뿐 아니라 안티 최종당화산물 항체의 항-염증 효과를 나타낸다.

상기 세포 상의 AGE-개질 단백질에 결합하는 항체는 인간 항체이고, 상기 항-염증 항체는 인간 항체인, 조성물.
- 세포 상의 AGE-개질 단백질에 결합하는 항체를 투여하는 단계를 포함하는, 염증 또는 자가 면역 장애의 치료 방법.
- 제9항에 있어서,
항-염증 항체를 투여하는 단계를 더 포함하는, 방법.
- 전염증성 인자(pro-inflammatory factor) 의 활성을 방해하는 단계, 및
노화 세포를 죽이는 단계, 를 포함하는, 염증 또는 자가 면역 장애의 치료 방법.
- 제11항에 있어서,
노화 세포를 죽이는 단계는, 세포 상의 AGE-개질 단백질에 결합하는 항체를 투여하는 단계를 포함하는 방법.
