Abstract
본 발명은 시험 물질의 미백 효과를 스크리닝하는 방법에 관한 것으로서, 보다 구체적으로는 장수 유전자인 FoxO3a(Forkhead box O3A)를 이용하여 시험 물질이 FoxO3a 를 활성화 시키거나 발현을 촉진시키는지 여부를 확인하여 시험 물질의 미백 효능을 스크리닝하는 방법에 관한 것이다.
Description
본 발명은 시험 물질의 미백 효과를 스크리닝하는 방법에 관한 것으로서, 보다 구체적으로는 장수 유전자인 FoxO3(Forkhead box O3) 유전자에 의해 인코딩되는 단백질인 FoxO3a(Forkhead box O3A)를 이용하여 시험 물질이 FoxO3a 를 활성화 시키거나 발현을 촉진시키는지 여부를 확인하여 시험 물질의 미백 효능을 스크리닝하는 방법에 관한 것이다.
멜라닌은 사람의 피부색을 결정하는 색소로 이 멜라닌의 양과 분포에 의해 피부색이 결정된다. 멜라닌을 만드는 세포는 피부 표피 밑에 멜라닌 형성 세포(Melanocyte)라고 불리는 세포에서 만들어져서 피부의 신진대사로 각질 표면으로 이동하여 떨어져 나간다. 피부색에 관계없이 멜라닌 형성 세포의 수는 거의 동일하다. 단지 멜라닌이 생성되는 양과 종류, 분포가 다르기 때문에 피부색이 차이가 나는 것이다. 피부에서 티로신(Tyrosine)이 티로시나아제(Tyrosinase)라는 인체 효소에 의해 도파(DOPA)로 전환되고, 계속되는 일련의 산화과정을 통해 최종적으로 흑갈색의 중합체인 멜라닌이 생성된다.
멜라닌은 생체 내에서 분해되지 않으며, 각질형성세포가 표피에서 떨어져나갈 때 멜라닌과 함께 피부에서 떨어져나가는 것에 의해 제거된다. 이러한 멜라닌이 과도하게 생성되는 경우 기미나 주근깨, 점 등과 같은 과색소침착증을 유발하여 미용상으로 좋지 않은 결과를 가져오게 되며, 운동 등의 야외 활동에 따른 자외선에 의한 멜라닌 색소 침착을 막고자 하는 요구가 더욱 늘어나는 추세이다. 이에 과도한 멜라닌 생성을 막는 미백제의 개발의 요구가 커지는 실정이다.
따라서 이러한 멜라닌 생성을 저해하여 미백 효능을 갖는 물질을 찾는 것이 중요한 과제로 대두되고 있으며, 관능 평가나 육안에 의한 평가가 아닌 객관적인 기준을 통해 미백 효능을 판단할 수 있는 방법에 대한 필요가 요구된다.
이에 본 발명자들은 장수 유전자인 FoxO3(Forkhead box O3) 유전자에 의해 인코딩되는 단백질인 FoxO3a(Forkhead box O3A)를 이용하여 미백 효과 여부를 확인할 수 있음에 착안하여 본 발명을 완성하게 되었다.
따라서 본 발명은 FoxO3a(Forkhead box O3A)를 이용하여 시험 물질의 미백 효과를 스크리닝하는 방법을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명은 FoxO3a(Forkhead box O3A)의 활성화 또는 발현 촉진 여부를 확인하는 것을 포함하는 시험 물질의 미백 효능 스크리닝 방법을 제공한다.
본 발명에 따른 FoxO3a(Forkhead box O3A)는 장수 유전자로 알려진 FoxO3에 의해 인코딩되는 단백질이며, 인슐린 신호전달경로 (IIS, Insulin/IGF-like signaling)에 위치하는 전사인자로서 Mn-SOD, Catalase와 같은 효소의 발현에 작용하는 단백질이다. FoxO3a가 활성화되면 생체 내 방어기작의 활성화 등을 통해 항노화 효능을 구현한다. 상기 FoxO3a의 활성화는 FoxO3a의 핵으로의 이행에 의해 판단할 수 있다. 즉 세포질에 존재하던 FoxO3가 핵으로 이행하여 효소들의 발현을 개시하는 것이다.
본 발명자들은 FoxO3a의 활성화에 따른 핵 이행 정도와 멜라닌 생성 감소 정도가 비례하는 것을 확인하였다. 따라서 피부 세포에 미백 효능의 후보 물질을 처리하여 FoxO3a의 활성화 또는 발현 촉진 여부를 확인함으로써 상기 물질이 멜라닌 생성을 감소시켜 미백 효능을 나타내는 물질인지 여부를 간편하고 객관적으로 확인할 수 있다.
따라서 본 발명은 시험 물질의 미백 효과를 스크리닝하는 방법으로서,
상기 시험 물질이 FoxO3a(Forkhead box O3A)를 활성화 또는 발현을 촉진시키는지 여부를 확인하는 것을 특징으로 하는 미백 효과 스크리닝 방법을 제공한다.
본 발명의 일실시예에서 상기 시험 물질이 FoxO3a를 활성화시키는지 여부의 확인은
a) 피부 세포에 시험 물질을 처리하는 단계; 및
b) 상기 a) 단계에서 시험 물질을 처리한 피부 세포에서의 FoxO3a의 이동을 관찰하는 단계를 포함할 수 있다.
상기 스크리닝 방법은 c) 상기 피부 세포에서 FoxO3a가 핵으로 이행하는 경우 미백 효능 물질로 판정하는 단계를 더 포함할 수 있으며, 이는 FoxO3a가 핵으로 이행하는 것을 통하여 FoxO3a가 활성화 되는 것을 확인할 수 있기 때문에다. 또한 상기 스크리닝 방법에 의해 판정된 미백 효능 물질은 FoxO3a에 의해 항산화 효소의 발현을 개시시킬 수 있기 때문에 항노화 및 항산화와 동시에 미백 효능을 나타내는 물질로서 판정할 수 있다.
상기 피부 세포는 각질형성세포, 섬유아세포 또는 멜라닌형성세포일 수 있다.
또한 본 발명의 일실시예에서 상기 시험 물질이 FoxO3a의 발현을 촉진시키는지 여부의 확인은 웨스턴블랏(Western blot), ELISA 또는 RT-PCR를 이용하여 수행할 수 있다.
상기 확인 결과를 통하여 시험물질이 FoxO3a 발현을 촉진시키는 경우 미백 효능 물질로 판정할 수 있다. 또한 상기 스크리닝 방법에 의해 판정된 미백 효능 물질은 장수 유전자인 FoxO3의 특성상 FoxO3a에 의해 항산화 효소의 발현을 개시시킬 수 있기 때문에 본 발명에서 발견한 미백 효능 뿐 아니라 항노화 및 항산화 효과를 동시에 나타내는 물질로서 판정할 수 있다.
본 발명에 따른 미백 효능 물질 스크리닝 방법은 실제 멜라닌 형성 감소와 FoxO3a의 활성화 정도가 비례하기 때문에 객관적인 기준을 통하여 시험 물질의 미백 효능 여부를 확인할 수 있어, 임상실험이나 관능 평가와 같은 방법에 의하지 않고 미백 효능 물질을 신속하고 간편하며 정확하게 판단할 수 있다. 또한 상기 스크리닝 방법에 의해 판정된 미백 효능 물질은 FoxO3a에 의해 항산화 효소의 발현을 개시시킬 수 있기 때문에 항노화 및 항산화 효과와 미백 효능을 동시에 나타내는 물질을 스크리닝할 수 있다.

이하, 본 발명의 내용을 실시예 및 시험예를 통하여 보다 구체적으로 설명한다. 이들 실시예는 본 발명의 내용을 이해하기 위해 제시되는 것일 뿐 본 발명의 권리범위가 이들 실시예 및 시험예로 한정되는 것은 아니고, 당업계에서 통상적으로 주지된 변형, 치환 및 삽입 등을 수행할 수 있으며, 이에 대한 것도 본 발명의 범위에 포함된다.
[시험예 1] FoxO3a 활성화와 멜라닌 생성 감소 실험
MNT-1 세포(Human melanoma cell line, Lonza, SWISS) 1X106 세포를 MEM 배지(10% DMEM)에 첨가하고, 각각 야생형(WT) FoxO3a 플라스미드(Plasmid 1787: HA-FOXO3a WT, Addgene사), NLS 돌연변이 FoxO3a 플라스미드(nuclear localization sequence mutant plasmid; Plasmid 8361: FLAG-FOXO3a TM, Addegene사)를 lipofectamin 2000(Invitrogen)을 이용하여 MNT-1 세포에 트랜스펙션하여 37℃에서 48시간동안 배양하였다. 상기 MNT-1 세포를 500㎕의 LIPA buffer(25 mM Tris-HCl, pH 7.4, 0.1% SDS, 0.1% Triton X-100, 1% sodium deoxycholate, 150 mM NaCl, 1 mM, EDTA, 1 mM Na3VO4, 1 mM PMSF, 10 mg/mL aprotinin, 5 mg/mL leupeptin)로 용해(lysis)시킨 후, 세포 용해질을 원심분리(eppendorf, centrifuge 5415R, Germany)하여 그 펠렛을 분리하여 색상을 관찰하였으며, 1N 수산화나트륨 200㎕로 상기 펠렛 속의 내용물을 용해시켜 사진을 도 1a에 나내었다. 대조군으로는 플라스미드를 트랜스펙션하지 않은 MNT-1세포를 사용하였다.
상기 용해액 20㎍을 로딩하여 웨스턴블랏으로 FoxO3a의 단백질 발현량을 확인하였고 세포질 성분의 마커 단백질로 Gapdh, 핵 성분의 마커 단백질로 Lamin B를 사용하여 결과를 도 1b에 나타내었다.
도 1a의 결과에서, 야생형 FoxO3a 플라스미드를 트랜스펙션한 경우 Foxo3a의 과발현에 의해 멜라닌 생성이 현저히 감소함을 확인할 수 있으며, 핵위치서열에 돌연변이가 있는 NLS 돌연변이 FoxO3a 플라스미드를 트랜스펙션한 경우에는 FoxO3a가 핵으로 이행하지 않기 때문에 멜라닌 생성 감소가 나타나지 않음을 확인할 수 있다. 또한, 도 1b의 결과에서 야생형 FoxO3a 플라스미드를 트랜스펙션한 경우 FoxO3a가 활성화되어 핵으로 이행됨이 나타났음을 확인할 수 있다. 따라서 FoxO3a의 활성화가 일어나지 않으면 멜라닌 생성에 영향을 미치지 않으며, FoxO3a의 핵으로의 이행이 멜라닌 감소에 작용함을 확인할 수 있다. 즉 FoxO3a의 활성화가 피부세포에서 멜라닌 생성을 효과적으로 저해할 수 있는 중요한 인자라는 점을 알 수 있다.
[시험예 2] 미백물질 처리에 따른 멜라닌 생성 감소 실험
MNT-1 세포(Human melanoma cell line, Lonza, SWISS) 1X106 세포에 공지의 미백물질인 비타민 C, N-아세틸시스테인(N-acetyl cysteine; NAC), 트롤록스(Trolox)를 각각 0, 1nM, 10nM, 100nM, 1μM, 10μM, 100μM의 농도로 처리한 후, 37℃에서 48시간 동안 배양하였다. 상기 배양된 세포를 500㎕의 LIPA buffer로 용해(lysis)시킨 후, 상기 세포 용해질 중 500㎕를 원심분리(eppendorf, centrifuge 5415R, Germany)하여 그 펠렛을 분리하여 색상을 관찰하였으며, 수산화나트륨으로 상기 펠렛 속의 내용물을 용해시켰다. 상기 용해액을 멜라닌 색소에 특이적인 OD490에서 흡광도를 측정하고, BCA protein assay kit (Pierce)를 이용하여 총 단백질양을 측정한 후 멜라닌 양을 (흡광도)/(총 단백질 양)으로 보정하여 멜라닌/㎍ 단백질로 나타내었으며, 각 미백물질 실험군의 비처리군(0nM)의 멜라닌 양을 1로 보았을 때의 실험군의 멜라닌 양으로 계산하여 도 2에 나타내었다.
[시험예 3] 미백물질 처리에 따른 티로시나아제 활성 억제 평가
상기 시험예 2에서의 세포 용해질 100㎕에 10 mM DOPA(dihydroxyphenylalanine) 100㎕를 첨가하여 37℃에서 1시간 동안 반응시킨 후, 티로시나아제의 작용으로 생성된 도파크롬(dopachrome)의 양을 OD490에서 흡광도를 측정하여 비처리군(0nM)의 값을 1로 보았을 때의 각 실험군의 흡광도로 나타내어 티로시나아제 활성 억제능을 계산하여 결과를 도 2에 나타내었다.

MNT-1 세포(Human melanoma cell line, Lonza, SWISS) 1X106 세포에 공지의 미백물질인 비타민 C, N-아세틸시스테인(N-acetyl cysteine; NAC), 트롤록스(Trolox)를 각각 100nM, 100μM의 농도로 처리한 후, 37℃에서 48시간동안 배양하였다. 대조군으로는 비처리군을 사용하였다. 배양된 세포를 각각 DAPI (핵 염색제-blue), FoxO3a IF antibody (red)로 염색하고, 형광현미경(EVOSfl digital fluorescence microscope, Advanced Microscopy Group)으로 관찰하여 결과를 도 3에 나타내었다.
도 3의 결과에서, 비타민 C, N-아세틸시스테인(N-acetyl cysteine; NAC), 트롤록스(Trolox) 처리시 FoxO3a의 핵 이행이 관찰됨을 알 수 있으며, 각 경우에서 100μM에서는 핵이행이 관찰되지 않는 반면, 100nM에서 극명하게 관찰되는 것을 알 수 있다.
상기 시험예 2~4의 결과로부터, FoxO3a의 핵이행이 관찰된 물질 농도에서 미백 효능을 나타내며, FoxO3a의 활성화를 매개하는 물질은 항노화, 항산화 효능 및 미백 효능을 동시에 구현하는 항노화-미백 물질임을 확인할 수 있다. 따라서 본 발명에 따른 스크리닝 방법에 의하여 시험 물질의 항노화·항산화 효과와 동시에 미백 효과를 객관적으로 정확하게 스크리닝할 수 있음을 알 수 있다.
Claims (8)
- 시험 물질의 미백 효과를 스크리닝하는 방법으로서,
상기 시험 물질이 FoxO3a(Forkhead box O3A)를 활성화 또는 발현을 촉진시키는지 여부를 확인하는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제1항에 있어서,
상기 시험 물질이 FoxO3a를 활성화시키는지 여부의 확인은
a) 피부 세포에 시험 물질을 처리하는 단계; 및
b) 상기 a) 단계에서 시험 물질을 처리한 피부 세포에서의 FoxO3a의 이동을 관찰하는 단계를 포함하는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제2항에 있어서,
상기 스크리닝 방법은
c) 상기 피부 세포에서 FoxO3a가 핵으로 이행하는 경우 미백 효능 물질로 판정하는 단계를 더 포함하는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제2항 또는 제3항에 있어서,
상기 피부 세포는 각질형성세포, 섬유아세포 또는 멜라닌형성세포인 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제1항 내지 제3항 중 어느 한 항에 있어서,
상기 스크리닝 방법은 미백 효과와 항노화 효과를 동시에 스크리닝할 수 있는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제1항에 있어서,
상기 시험 물질이 FoxO3a의 발현을 촉진시키는지 여부의 확인은 웨스턴블랏(Western blot), ELISA 또는 RT-PCR를 이용하여 수행하는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제6항에 있어서,
상기 시험물질이 FoxO3a 발현을 촉진시키는 경우 미백 효능 물질로 판정하는 것을 특징으로 하는 미백 효과 스크리닝 방법. - 제6항 또는 제7항에 있어서,
상기 스크리닝 방법은 미백 효과와 항노화 효과를 동시에 스크리닝할 수 있는 것을 특징으로 하는 미백 효과 스크리닝 방법.
