Abstract
Description
도 2는 선별과정 후 선별된 ssDNA를 확인하기 위해, 12.5% 요소(urea) PAGE 분석법을 수행한 결과를 나타낸 그림이다.
도 3은 반복되는 선별 과정에서 각 단계마다 선별되는 앱타머(aptamer)의 결합력을 나타낸 그래프이다.
도 4는 SELEX를 통해 선별된 DNA 앱타머를 염기서열과 구조적 유사성으로 분리해 놓은 그림이다.
도 5는 형광물질이 표지된 DNA 앱타머를 이용하여 카나마이신(kanamycin)과의 결합정도를 나타낸 그래프이다.

Claims (7)
- 서열번호 1 내지 서열번호 13 중 어느 하나의 염기서열을 갖는 것을 특징으로 하는, 카나마이신(kanamycin)에 특이적으로 결합하는 DNA 앱타머(aptamer).
- 1) 카나마이신(kanamycin)을 비드(bead)에 고정시키는 단계;
2) 카나마이신이 고정된 비드와, 양말단에 보존된 PCR용 프라이머 영역을 가지고 중앙에 30 - 50개의 임의의 염기를 가지는 ssDNA 라이브러리를 반응시켜 결합을 유도하는 단계;
3) 카나마이신이 고정화된 비드에 결합된 ssDNA를 분리하는 단계;
4) 분리된 카나마이신이 고정된 비드에 결합된 ssDNA로부터 ssDNA를 분리하는 단계; 및
5) 상기 PCR용 프라이머 영역에 상보적인 프라이머 쌍을 이용하여 PCR을 수행함으로써 카나마이신에 특이적으로 결합하는 ssDNA를 증폭하는 단계,
를 포함하는 제 1항의 카나마이신에 특이적으로 결합하는 DNA 앱타머의 제조방법.
- 제 2항에 있어서, 상기 비드는 세파로드비드(sepharose bead) 또는 자성비드(magnetic bead)인 것을 특징으로 하는 카나마이신에 특이적으로 결합하는 DNA 압타머의 제조방법.
- 제 2항에 있어서, 단계 2)의 PCR용 프라이머 영역은 N-말단에 서열번호 14로 기재되는 아미노산 서열, 및 C-말단에 서열번호 15로 기재되는 아미노산 서열을 갖는 것을 특징으로 하는 카나마이신에 특이적으로 결합하는 DNA 앱타머의 제조방법.
- 제 2항에 있어서, 단계 2) 내지 단계 5)의 단계를 추가적으로 8회 이상 반복하여 실시하는 단계를 포함하는 카나마이신에 특이적으로 결합하는 DNA 앱타머의 제조방법.
- 서열번호 1 내지 서열번호 13 중 어느 하나의 염기서열을 갖는 것을 특징으로 하는 카나마이신에 특이적으로 결합하는 DNA 앱타머를 포함하는 카나마이신 검출용 조성물.
- 서열번호 1 내지 서열번호 13 중 어느 하나의 염기서열을 갖는 것을 특징으로 하는 카나마이신에 특이적으로 결합하는 DNA 앱타머를 포함하는 카나마이신 검출용 키트.
