Abstract

Description
본 발명은 아쥬반트(adjuvant)를 사용하여 면역계를 자극하는 개선된 기술에 관한 것이다. 본 발명은 특히 아쥬반트를 복수개의 디스플레이 방식으로 나타내는 신규한 아쥬반트-함유 생성물, 이들 생성물을 포함하는 조성물 및 면역화에서의 이들 생성물의 용도에 관한 것이다.

운반을 개선시킬 목적으로 합성 아쥬반트를 제제에 혼입하기 위한 노력이 있어왔다. 예를 들어, 미국 제2005/0233105호는 낮은 분자량의 합성 아쥬반트를 포함하는 제제를 기술한다. 그러나 이들 제제는 바이러스 백신과의 아쥬반트의 단순한 혼합물이고 합성 아쥬반트의 고유 활성을 개선시키는 수단을 제공하지 못한다.
비슷하게, WO 2007/078879는 자가 조립 리포솜, 중합체 복합체 및 유화된 지질을 포함하는 조성물을 기술한다. 이들 조성물은 더 자연적인 방식으로 아쥬반트가 존재하도록 의도된 것이지만, 소수성 코어 내에 포획되어 있어서 제제화하기 힘들고 아쥬반트 물질의 대부분은 접근하기 어렵다. 이들 제제는 특정 조건 하에 엉기고 그들의 불안정성으로 인해 그들은 보통 투여 직전에 제조되어야 한다.
그러므로 면역계의 증진된 자극을 제공하는 합성 아쥬반트의 설계에 대한 새로운 접근의 필요가 있다.
본 발명은 그러므로 3개 이상의 아쥬반트에 공유결합으로 연결된 중합체 골격을 포함하는 아쥬반트-중합체 구조물(본원에서 중합체-아쥬반트 구조물로도 지칭됨)을 제공하며, 여기서 3개 이상의 아쥬반트는 각각 펜던트 측쇄에 존재하고, 아쥬반트는 중합체 골격에 직접 또는 스페이서기를 통해 연결된다.
본 발명은 아쥬반트가 다원자가 디스플레이 방식으로 존재하도록 몇몇 작은 합성 아쥬반트를 중합체로 연결하는 합성 아쥬반트 단독의 사용과 비교할 때 면역 자극을 증가시킬 수 있음을 밝혔다. 병원체-관련 분자 패턴(PAMP)를 연상시키는 방법으로의 복수개의 아쥬반트의 형태는 수용체 결합성을 증진시키고 톨-유사 수용체 및 패턴 인식 수용체에게 더 자연적인 형태를 제공하는 것으로 생각된다. 추가로, 아쥬반트의 다원자가 디스플레이는 수용체를 가교 및 신호전달을 하도록 돕는다. 이들 인자는 모두 증가된 면역 자극을 야기하고 그로써 아쥬반트의 더 낮은 용량이 사용될 수 있고 부작용은 감소된다.

본 발명의 바람직한 실시양태에서, 중합체 골격 자체는 친수성이어서 전형적으로 친지성인 아쥬반트의 용해를 돕는다. 이는 아쥬반트의 운반을 촉진시키고 더욱 단순한 제제가 사용될 수 있게 한다. 추가 장점은 수용체와 상호작용 할 수 있는 분자의 수를 증가시키는 것이고, 이 또한 더 낮은 용량이 사용될 수 있게 한다.
본 발명의 아쥬반트-중합체 구조물은 전형적으로 백신과 함께 투여된다. 본 발명은 그러므로 또한 백신에 결합한 본 발명의 아쥬반트-중합체 구조물을 포함하는 백신 포합체를 제공한다. 또한 제공되는 것은 본 발명의 아쥬반트-중합체 구조물 또는 백신 포합체 및 제약상 허용가능한 운반체 또는 희석제를 포함하는 조성물이다.

중합체 골격은 아쥬반트를 포함하는 3개 이상의 펜던트 측쇄를 갖는 선형 중합체일 수 있다. 별법으로, 중합체 골격 자체는 분지된 구조일 수 있다. 예를 들어, 수지상(dendritic) 및 빗살 모양(comb) 중합체가 예상된다.

전형적으로 개시제가 공중합 반응에, 바람직하게는 AIBN에 사용된다. 반응은 일반적으로 유기 용매, 전형적으로 DMSO에서 일어난다. 반응은 일반적으로 50 내지 70℃, 바람직하게는 약 60℃의 온도로 가열된다. 반응은 일반적으로 상기-명시한 온도에서 4 내지 8시간, 바람직하게는 5 내지 7시간, 더욱 바람직하게는 약 6시간 동안 가열된다. 그로써 수득된 중합체는 전형적으로 아세톤-디에틸 에테르(3:1) 혼합물에서 침강되고, 여과되며, 아세톤 및 디에틸 에테르로 세척되며, 진공에서 건조된다. 그로써 수득된 중합체는 추가로 세파덱스-LH(Sephadex-LH) 20 컬럼에서 메탄올을 사용하여 정제될 수 있다.

나노겔 코어 입자를, 이전에 문헌[Blackburn et al., Colloid Polym Sci. 2008; 286(5): 563-569]에서 보고된 것과 같이, 자유-라디칼 침강 중합으로 합성했다.
중합체를 분리하는 열상(thermally phase)의 사용은 매우 단순분산된 나노겔 합성을 위한 침강 중합의 사용을 가능케 한다. 몰 조성은 전체 단량체 농도가 140 mM로, 98%의 N-이소프로필메트아크릴아미드(NIPMAm), 2%의 N,N'-메틸렌비스(아크릴아미드)(BIS)였다. 용액은 또한 공초점 현미경을 통한 시각화를 위해 나노겔에 형광을 주기 위해 소량의(약 0.1 mM) 아크릴아미도플루오레세인(acrylamidofluorescein)(AFA)를 함유했다. 통상적인 합성에서, NIPMAm, BIS, 및 도데실 황산나트륨(SDS, 8 mM 전체 농도)의 여과된 수용액의 100 ml를 반응 플라스크에 첨가했고 그 후 70℃로 가열했다. 용액을 온도가 계속 안정할 때까지 N2 가스로 퍼징했고 격렬하게 교반했다. U937-luc 세포를 100 ng/ml의 LPS, 100 ng/ml의 Pam3Cys, 100 ng/ml의 pHPMA 또는 100 ng/ml pHPMA-Pam3Cys로의 노출 전에 ml 당 1e6의 밀도로 키웠다. 8시간 후에, 세포를 펠렛화하고, 분해하고, 루시퍼라아제 발현에 대해 평가했다(도 4). 이 연구에서, 사용된 HPMA 공중합체는 대략 20 kDa의 평균 분자량을 가졌고 5.22 중량%의 Pam3Cys(GCMS)를 함유했다.
