Abstract
Description
도 2는 각종 비생물학적 스트레스와 ABA 호르몬 처리시 At1g10560 및 At1g60190 유전자의 발현 양상을 분석한 결과를 나타낸 그림이다. 상술한 바 대로, At1g10560과 At1g60190은 식물의 스트레스 반응에 중요한 작용을 하는 식물호르몬인 ABA(abscisic acid)에 의해서 유도될 것이라고 예상되었다. ABA를 처리시 At1g10560 mRNA와 At1g60190 mRNA 발현이 증가하는 것을 확인할 수 있다(도 2a). 또한 mRNA의 발현 정도는 At1g60190 유전자가 At1g10560 유전자보다 상대적으로 높은 것을 확인할 수 있다. 건조, 저온 및 염 처리를 함으로써 이러한 각종 비생물학적 스트레스에 대한 상기 유전자들의 발현 정도를 분석하였다(도 2b). 그 결과 건조, 저온 및 염에 의해서 두 유전자 모두 정도의 차이는 있으나 mRNA 발현량이 증가하는 것을 확인할 수 있었다. 그리고 ABA처리에 의한 mRNA 발현 패턴과 동일하게 At1g60190 유전자가 At1g10560 유전자보다 더 강한 유도성(inducibility)을 가지는 것을 확인할 수 있었다. 이는 두 유전자가 만일 상호 보완적인 기능을 가지고 있다면 아마도 At1g60190 유전자가 At1g10560 유전자보다 더 주된 인자로 작용할 가능성이 있음을 시사한다.
도 3은 조직염색화학 GUS 분석을 통한 At1g10560 유전자와 At1g60190 유전자의 프로모터 활성 분석 결과를 나타낸 그림이다. ABA를 처리하거나 건조 스트레스를 주었을 경우 At1g10560 유전자와 At1g60190 유전자의 프로모터 활성이 모두 증가하는 것을 확인할 수 있다. 건조 스트레스의 경우에는 새로 나는 어린 잎에서의 활성이 높은 것을 확인할 수 있는 반면 ABA를 처리한 경우 상대적으로 잎 전반에 걸쳐 발현이 되는 차이점이 있으나, 두 처리구에서 공통적으로 프로모터 활성이 관찰되었다. 이는 At1g10560 유전자와 At1g60190 유전자가 건조 스트레스 상황에서 공변세포의 조절을 통해 식물체내 수분의 조절로 스트레스 저항성과 같은 반응에 연관될 가능성을 강하게 뒷받침해준다.
도 4는 At1g10560 유전자와 At1g60190 유전자의 이중 녹-아웃(double Knock-out) 돌연변이체와 각각의 과다발현 식물체의 유전자 발현양상을 나타낸 그림이다.
도 5는 At1g10560 유전자와 At1g60190 유전자의 녹-아웃 돌연변이체와 과다발현 식물체 및 야생종에 건조 스트레스를 준 후 그 표현형의 차이를 관찰한 결과를 나타낸 그림이다. 도 5a는 애기장대 야생종과 At1g10560 유전자 및 At1g60190 유전자의 녹-아웃 돌연변이체, 그리고 이를 교배하여 얻어진 이중 녹-아웃 돌연변이체를 약 3주간 키운 후 12일 동안 물을 주지 않고 건조 스트레스를 주고 다시 물을 준 후 3일 뒤의 사진이며, 3일 후와 10일 후의 생존 비율을 나타낸 그래프이다. 이 실험의 결과 각각의 유전자가 기능을 상실할 경우 건조 저항성이 증가하였으며, 두 유전자의 기능이 모두 사라진 이중 녹-아웃 돌연변이체에서 저항성이 더욱 크게 증가하였다.
도 5b는 At1g10560 유전자와 At1g60190 유전자의 발현이 이중 녹-아웃 돌연변이체와 과다발현 식물체 야생종에서, 뿌리를 지상부와 지상부로의 시간이 지나면서 수분의 손실에 따른 질량의 감소를 그래프로 나타낸 것이다. 도 5a의 결과와 동일하게 이중 녹-아웃 돌연변이체는 야생종에 비해 상대적으로 수분 손실에 따른 질량의 감소 폭이 적으나 과다발현체의 경우 그 폭이 큰 것을 알 수 있다.
도 6은 At1g10560 유전자와 At1g60190 유전자의 녹-아웃 돌연변이체와 과다발현체의 ABA 처리 하에서의 공변세포 표현형 분석 결과를 나타낸 그림이다.
녹-아웃 돌연변이체의 경우 야생종에 비해서 기공이 더 많이 닫히는 것을 알 수 있으며 과다발현체의 경우 야생종에 비하여 기공의 닫히는 정도가 덜한 것을 알 수 있다.
도 7은 At1g10560 유전자와 At1g60190 유전자의 녹-아웃 돌연변이체와 과다발현체의 발아율을 분석한 결과를 나타낸 그림이다. 상기 유전자의 녹-아웃 돌연변이체는 그 발아율이 ABA에 의해서 야생종에 비해 상당히 낮아지게 되며, 과다발현 식물체는 야생종보다 그 발아 비율이 높아지는 것을 확인할 수 있다(도 7a). 도 7b는 At1g10560 유전자와 At1g60190 유전자의 과다발현 식물체를 ABA가 함유되지 않은 기본 발아 배지에서 심은 후 2일 그리고 3일 후의 사진이다. 돌연변이체의 경우는 야생종과 큰 차이가 없지만 상대적으로 과다발현체의 경우는 기본 배지에서도 그 발아율과 초기 생장이 빨라지는 것을 확인할 수 있다.
도 8은 At1g10560 유전자와 At1g60190 유전자의 녹-아웃 돌연변이체를 이용한 고염 스트레스 하에서의 표현형 분석결과를 나타낸 그림이다. 관찰 결과 기본배지에서의 생장은 별 차이가 없으나, 120 mM의 NaCl이 포함된 경우 야생종보다 돌연변이 식물체들의 뿌리 생장이 약간 증가한 것을 확인할 수 있다. 또한 120 mM과 140mM NaCl이 각각 포함된 배지에서 야생종의 백화현상이 각각의 유전자가 기능을 상실한 돌연변이체보다 상대적으로 빠르게 나타난 것을 확인할 수 있다.
도 9는 At1g10560 유전자와 At1g60190 유전자의 녹-아웃 돌연변이체를 이용한 저온 스트레스 하에서의 표현형 분석결과를 나타낸 그림이다. 각각의 유전자가 기능을 상실한 돌연변이체의 경우 뿌리와 지상부의 성장이 야생종에 비해 증가한 것을 통해 저온 저항성이 증가하였음을 확인할 수 있다.

- 제 1 항에 있어서, 상기 서열목록 제4서열의 아미노산 서열을 코딩하는 뉴클레오타이드 서열은 서열목록 제2서열의 뉴클레오타이드 서열로 이루어진 것을 특징으로 하는 식물체의 비생물학적 스트레스 내성 증진용 조성물.
- 제 1 항에 있어서, 상기 비생물학적 스트레스는 건조 스트레스, 저온 스트레스 및 염 스트레스로 구성된 군으로부터 선택되는 것을 특징으로 하는 식물체의 비생물학적 스트레스 내성 증진용 조성물.
- 제 1 항에 있어서, 상기 핵산 분자는 T-DNA, siRNA, shRNA, miRNA, 리보자임(ribozyme), PNA(peptide nucleic acids) 또는 안티센스 올리고뉴클레오타이드인 것을 특징으로 하는 조성물.
- 제 5 항에 있어서, 상기 핵산 분자는 T-DNA인 것을 특징으로 하는 조성물.
- 상기 제 1 항 내지 제 3 항 중 어느 한 항의 조성물로 형질전환된 비생물학적 스트레스 내성이 증진된 식물세포.
- 상기 제 1 항 내지 제 3 항 중 어느 한 항의 조성물로 형질전환된 비생물학적 스트레스 내성이 증진된 식물체.
- 다음의 단계를 포함하는 비생물학적 스트레스 내성이 증진된 형질전환 식물체를 제조하는 방법:
(a) 상기 제 1 항 내지 제 3 항 중 어느 한 항의 조성물을 식물 세포에 도입시키는 단계; 및
(b) 상기 식물세포로부터 비생물학적 스트레스 내성이 증진된 형질전환 식물체를 수득하는 단계.
- 제 9 항에 있어서, 상기 비생물학적 스트레스는 건조 스트레스, 저온 스트레스 및 염 스트레스로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 9 항에 있어서, 상기 식물체는 벼, 밀, 보리, 옥수수, 콩, 감자, 밀, 팥, 귀리 및 수수를 포함하는 식량 작물류; 애기장대, 배추, 무, 고추, 딸기, 토마토, 수박, 오이, 양배추, 참외, 호박, 파, 양파 및 당근을 포함하는 채소 작물류; 인삼, 담배, 목화, 참깨, 사탕수수, 사탕무우, 들깨, 땅콩 및 유채를 포함하는 특용작물류; 사과나무, 배나무, 대추나무, 복숭아, 양다래, 포도, 감귤, 감, 자두, 살구 및 바나나를 포함하는 과수류; 장미, 글라디올러스, 거베라, 카네이션, 국화, 백합 및 튤립을 포함하는 화훼류; 및 라이그라스, 레드클로버, 오차드그라스, 알파알파, 톨페스큐 및 페레니얼라이그라스를 포함하는 사료작물류로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 서열목록 제3서열, 제4서열 또는 이들의 조합으로 구성된 군으로부터 선택된 아미노산 서열을 코딩하는 뉴클레오타이드 서열을 포함하는 식물체의 발아 촉진용 조성물.
- 제 12 항에 있어서, 상기 서열목록 제3서열의 아미노산 서열을 코딩하는 뉴클레오타이드 서열은 서열목록 제1서열의 뉴클레오타이드 서열로 이루어진 것을 특징으로 하는 식물체의 발아 촉진용 조성물.
- 제 12 항에 있어서, 상기 서열목록 제4서열의 아미노산 서열을 코딩하는 뉴클레오타이드 서열은 서열목록 제2서열의 뉴클레오타이드 서열로 이루어진 것을 특징으로 하는 식물체의 발아 촉진용 조성물.
- (a) 상기 제 12 항 내지 제 14 항 중 어느 한 항의 뉴클레오타이드 서열; (b) 상기 뉴클레오타이드 서열에 작동적으로 결합(operatively linked)되어있고 식물세포에서 RNA 분자를 형성시키는 프로모터; 및 (c) 식물세포에서 작용하여 RNA 분자의 3'-말단의 폴리아데닐화를 야기시키는 폴리 A 시그널 서열을 포함하는 식물발현용 재조합 벡터를 포함하는 식물체의 발아 촉진용 조성물.
