추가의 상세한 설명없이, 본 분야의 기술자는 상기 설명을 사용함으로써 본 발명을 완전히 사용할 수 있다고 여겨진다. 따라서, 하기의 바람직한 특정의 실시태양은 단지 설명하고자 하는 것이며, 어느 방식으로든, 무엇으로든 본 개시를 제한하고자 함이 아닌 것으로 해석되어야 한다.

침윤성 유방암을 앓는 여성 환자의 우측 유방에
SF64
를 정맥내(i.v.) 주사한 후의 광학 유방조영술
0.9%의 생리 식염수(normal saline)로 동결건조물을 희석시킨 후, SF64를 0.1 mg/kg체중의 투여량으로 유치 전완부 캐뉼러(indwelling forearm canula)를 경유하여 정맥내로 주사하였다. 엎드린 자세로, 공기로 둘러싸인 영상화 챔버내로 자유롭게 그의 유방을 걸쳐놓고 광학 컴퓨터 단층 촬영 레이저 유방조영술 장치를 사용하여 환자를 영상화하였다.
SF64를 정맥내로 주사한 후 1시간 10분째, 및 형광 모드를 사용하여 영상 재구성을 실시하였을 때 거의 오로지 SF64의 형광 신호만이 나타났다. 조영 염료는 침윤성 유방암에 축적되었는데, 이는 휘점에 의한 영상으로 표시된다. 이것이 영상에서 SF64의 표지된 형광 신호를 가져왔다.

Capan
-1 종양을 갖는, 누드 마우스에서의 미세전이암 영상화
배양액중에서 컨플루언스 이하(subconfluence)로 성장시킨 Capan-1 종양 세포를, 트립신으로 처리하여 원심분리하고, PBS중에 재현탁시켰다. 트립판 블루로 염색시키고, 세포 농도를 산출한 후, 세포 현탁액을 3×107/㎖의 농도로 설정하였다. 세포 사용시까지 세포 현탁액을 얼음상에서 냉각시켰다. 암컷 누드 마우스 (NMRI-누드, 체중 24-25g) 3마리를 마취시키고, 복강 절개후 각각의 동물의 췌장 피막하에 30 ㎕ (1×106 세포/동물)의 세포 현탁액을 접종하였다. 도 2에 도시한 구조를 갖는 시아닌 염료를 포함하고, EB-DF 항체 AP39에 접합된 (문헌 [Marty C, et al. (2001) Protein Expr. Purif. 21:156-64] 참조) 물질을 0.05 μmol/㎏체중 (체중 1 kg당 0.04 mg의 염료)으로 각각의 동물에 투여하였다. 뚜렷한 종양 성장이 감지될 수 있는 시점에 (종양 세포 이식후 약 12 내지 14주) 이 물질을 정맥내 투여하였다. 물질 투여후 6시간째 동물을 죽이고, 강화된 CCD 카메라를 사용한 형광 신호에 대하여 미세전이암을 포함하는 창자간막을 생체외에서 영상화하였다. 레이저 다이오드 (0.5W 출력)로 생성된 740 ㎚ 파장의 근적외선광을 사용한 창자간막 조사에 의하여 물질의 형광을 여기시켰다. 형광 영상을 디지탈로 저장하였다. 그 후, 미세전이암의 크기를 저 배율 현미경 (Stemi 2000-C, Fa. Carl Zeis)을 사용하여 측정하였다. 직경이 0.5 내지 2.0 mm 범위인 미세전이암 및 더 큰 장간막 전이암으로부터 형광 신호을 얻었으며, 이는 미시적 평가와 일치하였다. 염료 접합체의 효능은 도 2, 우측 패널에 도시한다.

영상화
2마리의 NMRI-누드 마우스에서 피부의 자발성 다중 미세-병변을 관찰하였다. 0.05 μmol/㎏체중 (0.04 mg 염료 kg b.w.)의 도 2에 도시한 염료-AP39 접합체를 각각의 마우스에 정맥내로 투여하였다. 물질 투여후 6시간째 마취된 마우스에서 영상화를 실시하였다. 마취제 이소플루란(이소플루란 쿠라메드(Isofluran Curamed), 독일 칼스류헤에 소재하는 큐라메드 파마 게엠베하(Curamed Pharma GmbH))을 사용하여 단시간 마취를 유발시켰다. 물질의 형광은 다이오드-레이저 (742 nm의 여기 파장, 레이저 다이오드, 0.5W 출력)로 여기시키고, 강화된 CCD-카메라를 사용하여 검출하였다. 형광 영상을 디지탈로 저장하였다. 그 후, 미소-병변의 크기를 저 배율 현미경 (Stemi 2000-C, Fa. Carl Zeis)을 사용하여 측정하였다. 형광 신호는 < 1 mm 보다 작은 미소-병변으로부터 얻었다. 염료 접합체의 효능은 대표예에 기초하여 도 3에 도시한다.
Claims (15)
- 제1항에 있어서, 화합물이 표적 화합물에 결합하는 것인 용도.
- 제2항에 있어서, 표적 화합물이 폴리펩티드, 핵산, 소분자 또는 당으로 구성된 표적 화합물들의 군으로부터 선택되는 것인 용도.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 폴리펩티드가 수용체 리간드, 항체, 단일쇄 항체, 또는 항체 또는 단일쇄 항체의 결합 단편으로 구성된 군으로부터 선택되는 것인 용도.
- 제1항 내지 제4항중 어느 한 항에 있어서, 증식성 질환이 종양, 전암 증상, 이형성증, 화생(metaplasia), 건선, 건선성 관절염, 류마티스성 관절염, 자궁내막증, 및/또는 안 질환으로 구성된 군으로부터 선택되는 것인 용도.
- 제5항에 있어서, 종양이 원발성 종양 또는 전이암(metastasis)인 용도.
- 제6항에 있어서, 종양이 위장 또는 결장직장관, 간, 췌장, 신장, 방광, 갑상선, 전립선, 자궁내막, 난소, 고환의 악성 종양, 흑색종, 이형성증성 구강 점막, 침습성 구강 암, 소세포 또는 비-소세포 폐 암종; 호르몬-의존성 유방암, 호르몬 비의존성 유방암을 비롯한 유방 종양; 이행 및 편평 세포 암; 신경모세포종, 신경아교종, 성상세포종, 골육종, 수막종을 비롯한 신경계 암; 연조직 육종; 뇌하수체 선종, 크롬친화세포종(pheochromocytoma), 부신경절종을 비롯한 혈관종 및 내분비성 종양, 림프종 및 백혈병을 비롯한 혈액암, 또는 상기 언급한 종양들 중 하나로부터 유래하는 전이암인 용도.
- 제6항에 있어서, 진단용 화합물이 종양 선별 도중에, 또는 수술 이전에, 수술 도중에 또는 수술 이후에 투여되는 용도.
- 제5항에 있어서, 전암 증상이 피부의 전암 증상, 특히 광선 각화증, 피부 각질, 광선 구순염, 타르 각화증, 비소 각화증, x-선 각화증, 보웬(Bowen's) 질환, 보웬양 구진증, 악성 흑점, 경화성 태선 및 태선 고무 점막; 소화관의 전암 증상, 특히 홍색판, 백반, 바레트 식도, 플러머-빈슨(Plummer-Vinson) 증후군, 다리 궤양, 거대 비후성 위질환, 경계 암종, 종양 장 폴립, 직장 폴립, 석회화 담낭; 부인과적 전암 증상, 특히 유관 상피내 암종 (CDIS), 경부 상피내 종양 (CIN), 백반, 자궁내막 증식증 (III 급), 외음부 이상증, 외음부 상피내 종양 (VIN), 포상기태; 비뇨기과 전암 증상, 특히 방광 유두종증, 퀴이라 홍색 비후증(Queyrat's erythroplasia), 고환 상피내 종양 (TIN), 백반; 상피내 암종 (CIS); 만성 염증에 의하여 유발된 전암 증상, 특히 화농 피부증, 골수염, 응괴성 여드름, 심상 루푸스, 및 누공으로 구성된 군으로부터 선택되는 것인 용도.

- 제1항 내지 제13항 중 어느 한 항에 있어서, 화합물이 0.1 mg/kg체중 이하의 양으로 진단용 조성물에 포함되는 용도.
- 진단 적용시마다 0.5 미만 내지 0.001 mg/kg체중 이상의 화합물을 투여하기 위한 진단용 조성물을 제조하기 위한 양으로 제1항의 화학식 I의 화합물 또는 그의 제약학상 허용가능한 염, 및 임의로 제약학상 허용가능한 염, 담체, 부형제 및/또는 완충제를 포함하는 진단용 키트.
