Claims (4)
- 동물 연골의 주성분을 이루는 콜라겐(collagen), 히알루로닉산(hyaluronic acid), 피브린(fibrin)의 기질(matrix)을 주사기의 선단 믹싱팁을 이용하여 상호 혼합하면서 연골손상부위에 주입하여 이식함을 특징으로 하는 주입형 연골세포치료제의 이식방법.
- 바이알 1(Red)의 세포배양액 중 1ml을 멸균된 1mL 주사기 안으로 집어 넣은 후, 바이알 2(Blue)의 백색 또는 미황색의 동결건조분말(Fibrinogen)을 수직으로 한 상태에서 주사기의 바늘을 삽입하여 주입하여 10∼200mg/ml의 농도로 만드는 단계;상기 바이알 1(Red)의 세포배양액 중 1ml을 주사기를 이용하여 백색 동결 건조 분말(Thrombin)이 들어있는 바이알 3(Red, small)에 수직으로 한 상태에서 주사기의 바늘을 삽입하여 서서히 붉은색 액체를 주입하는 단계;1mL 주사기를 이용하여 바이알 3(Red, small)의 내용물 1mL중 0.1cc를 채취하여 바이알 4(yellow)의 바닥에 주입하여 1∼200IU/ml의 농도로 만드는 단계;연골세포현탁액 바이알 2개를 멸균된 1mL 주사기를 이용하여 전량을 바이알 4(yellow)에 넣고 주사기를 이용하여 2~3회 혼합하는 단계;상기 바이알 2(Blue)의 내용물이 전부 용해된 것을 확인한 후, 1mL 주사기에 전량을 주입하는 단계;상기 바이알 4(Yellow)의 잘 혼합된 세포 치료제를 1mL 주사기에 주입하는 단계; 및상기 바이알 2(Blue)와 바이알 4(Yellow)의 내용물이 충진된 2개의 1mL 주사기를 거치대에 장착한 후 장착된 주사기에 스크류 팁(screw tip)을 장착하고, 이어서 주사기의 선단 믹싱팁을 이용하여 상호 혼합하면서 동물의 연골손상부위에 주입하여 이식하는 단계;가 포함됨을 특징으로 하는 주입형 연골세포치료제의 이식방법.
- 제 2 청구항에 있어서,상기 주사기의 바늘을 삽입하여 주입하는 단계는, 바늘 끝이 동결 건조물에 닿지 않도록 하고, 바이알의 내벽을 따라 주입하지 말고 동결 건조물 위에 직접 주입함을 특징으로 하는 주입형 연골세포치료제의 이식방법.
- 제 2 청구항에 있어서,상기 바이알 1(Red)의 세포배양액에는 0.01∼2%(W/V)의 히알루로닉산(hyaluronic acid)과 100∼20,000KIU/ml의 아프로티닌(aprotinin), 50ug/ml - 1mg/ml의 제 2형 콜라겐(Collagen type 2) 중에서 선택되는 어느 하나 이상이 첨가되는 것을 특징으로 하는 주입형 연골세포치료제의 이식방법.
