Claims (3)
- 오리지널 기체 및 하나 이상의 레플리카 기체 상에 cDNA 라이브러리를 고정화시키는 방법으로서, 하기의 단계를 포함하는 방법:(a) 오리지널 기체의 표면 상에 아미노기를 고정화시키는 단계;(b) 오리지널 기체에 올리고 (dT)n 를 접촉시켜 오리지널 기체 상에 올리고 (dT)n 을 고정화시키는 단계(여기서 n 은 15 내지 30 임);(c) 오리지널 기체를 전체 RNA, 역전사 효소, 및 뉴클레오티드를 포함하는 반응액으로 처리하여 mRNA 로부터 cDNA 를 작제하는 단계;(d) 오리지널 기체를 20 ℃ 의 반응액에 놓아둠으로써 오리지널 기체 상의 올리고 (dT)n 와 전체 RNA 용액내의 mRNA 를 혼성화시키는 단계;(e) 오리지널 기체를 역전사 효소용액으로 처리하고, 오리지널 기체를 역전사 효소와 접촉시켜둠으로써 오리지널 기체 상에 고정화된 cDNA 라이브러리를 작제하는 단계;(f) 오리지널 기체를 세척하고 가열하여 오리지널 기체로부터 mRNA 를 탈혼성화시키는 단계; 및(g) 오리지널 기체를 대신하여 레플리카 기체를 사용하여 상기 (b) 내지 (f) 단계를 반복함으로써 기체 상에 고정화된 레플리카 cDNA 라이브러리를 작제하는 단계.
- 제 1 항에 의해 작제된 DNA 라이브러리를 고정화한 기체로서, 상기 기체가 판상, 구체, 입방체 또는 입자상이며, 플라스틱, 유리, 실리콘, 금속, 다이아몬드 및 탄소원자로 만들어진 기체.
- 단일 가닥 DNA 라이브러리가 고정화된 기체로서, 상기 기체가 판상, 구체, 입방체 또는 입자상이며, 플라스틱, 유리, 실리콘, 금속, 다이아몬드 및 탄소원자로 만들어진 기체.
