Abstract

본 발명은 세프티오푸르나트륨을 활성성분으로 하고 소정의 부형제를 포함하는 주사액 조성물로서, 분산매가 식물성 지방산에스테르임을 특징으로 하는 현탁 주사액 조성물을 개시한다.
상기 구성에 의하면 점도가 개선되고, 재분산성 및 생체이용율이 우수한 세프티오푸르나트륨을 활성성분으로 함유하는 현탁 주사제의 제조가 가능하다.

Description

세프티오푸르나트륨을 활성성분으로 함유하는 현탁주사액 조성물{Injectable Composition Comprising Ceftiofur Sodium as Active Ingredient}
본 발명은 세프티오푸르나트륨을 활성성분으로 하고 소정의 부형제를 포함하는 주사제 조성물로서 보다 상세하게는 점도가 낮고, 재분산성과 생체이용율이 우수한 세프티오푸르나트륨을 활성성분으로 함유하는 현탁주사액 조성물에 관한 것이다.
세프티오푸르([6R-[6α,7β(Z)]]7-[[(2-아미노-4-치아졸릴)(메톡시이미노)아세칠]아미노]-3-[[(2-푸라닐카보닐)치오]메칠]-8-옥소-5-치아-1-아자시클로[4.2.0]옥트-2-엔-2-카르복실릭산)는 베타-락타마제 저항성을 가진 제 3세대 광범위 세팔로스포린계 항생제로서 그람양성 및 그람음성의 병원성 세균주에 대한 폭넓은 항균작용을 나타내며, 수의학적으로 널리 사용되고 있다(Brown, S. A., Comparison of plasma pharmacokinetics and bioavailability of ceftiofur sodium and ceftiofur hydrochloride in pigs after a single intramuscular injection. J. Vet. Pharmacol. Therap. 22, 35-40, 1999; R. J. Yancey et al., In vivo and in vitro antibacterial activity and β-lactamase stability: Am. J. Vet. Res. 48, 1050). 세프티오푸르는 난용성으로 일반적으로 염산염 또는 나트륨염의 형태로 제제화되고 있으나, 수분에 대한 불안정성으로 현탁주사제 개발에 있어서 제한 요소로 작용하고 있다.
현탁주사제에 관한 선행기술로는 세프티오푸르의 용해도 향상을 위해 니코틴아미드를 용해보조제로 하는 수성조성물이 대한민국특허 96-0008236에 개시되어 있다. 하지만 상기 수성조성물은 제제의 불안정이 여전히 문제점으로 지적되고 있다. 이와 같은 문제점을 극복하기 위해 비수성 현탁주사제로서 식물유에 활성성분을 분산시킨 지속성 제제화 기술이 국제공개특허 WO 94/20505에 개시되어 있고, 레시친과 계면활성제를 포함하며 입자크기가 약 10㎛ 이하가 되도록 만들어 0.3∼0.5%의 물을 넣어 분산성을 좋게 한 것을 특징으로 하는 세프티오푸르 염산염의 오일 현탁액이 국제공개특허 WO 98/25621에 개시되어 있다.
하지만 이들은 제제의 장기간 보관 중 주성분이 침강되어 응집괴를 형성하여 재분산이 어려워지는 단점이 있고, 분산매로 사용되는 면실유 등의 점도가 높아 사용하기에 불편한 단점이 있다. 한편 시판되는 세프티오푸르나트륨 제제는 활성성분의 안정화를 위하여 무균상태에서 동결건조하여 분말화한 제제로서 사용할 때 물에 녹여 사용해야 하는 불편함이 있고, 제조 공정이 복잡한 문제를 가지고 있다.
본 발명자는 상기 종래 기술이 가지는 문제점을 극복하기 위한 방안을 연구하여 오던 중 세프티오푸르나트륨의 분산매로서 식물성 지방산에스테르를 적용하는 경우 활성성분의 재분산성이 우수하며, 생체이용율이 우수함을 확인하고서 본 발명을 완성하게 되었다.
이와 같이 본 발명의 목적은 점도가 개선되고, 재분산성 및 생체이용율이 우수한 세프티오푸르나트륨을 활성성분으로 함유하는 현탁주사액 조성물을 제공하는데 있다.
도 1은 본 발명의 현탁주사액(실시예 1)과 대조구인 시판제제(엑세넬)를 웅성 랫트에 근육 주사한 후 일정시간 간격으로 측정한 혈중 활성성분의 농도를 나타낸 그래프.
본 발명은 세프티오푸르나트륨을 활성성분으로 하고 소정의 부형제를 포함하는 주사액 조성물로서, 특히 분산매가 식물성 지방산에스테르인 현탁주사액 조성물을 포함한다.
상기에서 식물성 지방산에스테르라고 함은 식물성 오일 등에 존재하는 공지의 지방산과 공지의 1가 내지 3가 알콜과의 사이에 축합반응을 거쳐 얻어지는 에스테르 화합물로서 단일 성분 또는 2종 이상의 혼합성분으로 구성되는 화합물을 말한다.
바람직하기로는 상기 지방산에스테르에는 모노글리세리드, 디글리세리드, 트리글리세리드, 벤질알콜, 안식향산벤젠, 올레인산에칠, 에틸리놀레이트, 이소프로필팔미테이트, 이소프로필미리스테이트가 포함된다.
상기 지방산 에스테르의 함량은 종류에 따라 상이할 수 있으며, 특별한 한정을 요하는 것은 아니지만 바람직하기로는 세프티오푸르나트륨의 중량 대비 1:0.1∼10, 보다 바람직하기로는 1:0.5∼3으로 함이 좋다. 이는 0.1 미만으로 하는 경우 주성분의 분산성이 문제가 될 수 있고, 10을 초과하는 경우 농도희석으로 인하여 사용상 불편한 문제가 우려되나 위 범위를 다소 벗어나더라도 본 발명을 실시하지 못하는 것은 아니라 볼 것이다. 따라서 상기 범위는 어디까지나 예시적인 것일 뿐 반드시 이에 한정될 필요는 없는 것으로 해석된다.
본 발명의 현탁주사액 조성물은 바람직하기로는 재분산성을 개선하고, 점도를 조절하기 위한 점도조절제로서 적량의 에탄올(또는 무수 에탄올) 등이 추가로 첨가될 수 있다. 에탄올을 첨가하는 경우 함량은 특별한 한정을 요하지 않으며, 당업자에 의해 원하는 점도 및 분산성에 따라 적량 선택하여 첨가될 수 있다.
또한 본 발명의 현탁주사액 조성물은 필요에 따라 의학적으로 허용되는 각종 첨가제를 포함할 수 있다. 이러한 첨가제의 예로는 계면활성제(소르비탄모노올레이트, 폴리옥시에칠렌20올레이트, 폴리옥실20세틸에테르 등), 항산화제(부틸파라히드록시안식향산, 부틸히드로퀴논, 토코페롤 등), 점도조절제(알긴산나트륨, 카르복시메칠셀룰로오스, 메칠셀룰로오스, 트라가칸타, 한천 , 폴리비닐피롤리돈 등) 등이 있다.
이하 본 발명의 내용을 실시예에 의해 보다 상세하게 설명하기로 한다. 다만 이들 실시예는 본 발명의 내용을 이해하기 위해 제시되는 것일 뿐 본 발명의 권리범위가 이들 실시예에 한정되어지는 것으로 해석되어져서는 아니된다.
<실시예 1∼8> 현탁주사제의 제조
하기 표 1의 성분 및 배합비에 따라 세프티오푸르나트륨을 함유하는 주사제를 제조하였으며, 제조 결과 모두 재분산성이 우수하였고, 점도가 낮게 나타났다.

<시험예> 약물학적 효과측정
상기 실시예 1에 의해 제조한 주사제를 시판되는 주사제인 엑세넬(제조사:파마시아업죤)을 대조군으로 하여 근육주사(세프티오프르 45 ㎎/㎏)에 따른 흡수실험을 수행하였다. 실험동물로는 스프라그-도올리계 랫트(웅성, 체중 300±20G)가 사용되었으며, 3마리를 한 그룹으로 하여 절식상태에서 상대적인 생체내 이용률을 측정하였다. 약제 투여 전에 공시험용 혈액을 채취하였고, 약제를 투여한 후 정해진시간에 따라 경정맥에서 혈액 0.2㎖씩 채혈하였으며, 혈중 세프티오푸르의 분석은 고속액체크로마토그래피법을 이용하였다.
분석된 혈중 세프티오푸르 농도의 측정결과는 도 1에 도시된 바와 같으며, 본 발명에 의한 실험군에서 생체이용율이 매우 우수한 것으로 확인되었다.
본 발명에 의하면 점도가 개선되고, 재분산성 및 생체이용율이 우수한 세프티오푸르나트륨을 활성성분으로 함유하는 현탁 주사제의 제조가 가능하다.

Claims (7)

  1. 세프티오푸르나트륨을 활성성분으로 하고 소정의 부형제를 포함하는 주사액 조성물에 있어서,
    분산매가 식물성 지방산에스테르임을 특징으로 하는 현탁주사액 조성물
  2. 제 1항에 있어서,
    에탄올이 추가로 포함됨을 특징으로 하는 현탁주사액 조성물
  3. 제 2항에 있어서,
    에탄올은 무수에탄올인 현탁주사액 조성물
  4. 제 1항에 있어서,
    지방산에스테르는 모노글리세리드, 디글리세리드, 트리글리세리드, , 벤질알콜, 안식향산벤젠, 올레인산에칠, 에틸리놀레이트, 이소프로필팔미테이트, 이소프로필미리스테이트의 군으로부터 선택되는 단독 또는 2종 이상의 혼합물을 포함하는 현탁주사액 조성물
  5. 제 1항에 있어서,
    지방산에스테르는 세프티오푸르나트륨의 중량 대비 1:0.1∼10인 현탁주사액조성물
  6. 제 1항에 있어서,
    지방산에스테르는 세프티오푸르나트륨의 중량 대비 1:0.5∼3인 현탁주사액 조성물
  7. 제 1항에 있어서,
