Abstract
치료용 유전자 발현 염기서열 및 유전자 치료용 약제가 개시된다. 편평상피세포에 치료용 유전자를 특이적으로 발현시키는, 하기 (a) ∼ (d) 중 어느 하나의 치료용 유전자 발현 염기서열이 개시된다.

(c) 서열번호 17 로 표시되는 염기서열
(d) 서열번호 16 로 표시되는 염기서열.
또한 상기 어느 하나의 염기서열의 하류영역에 치료용 유전자를 갖고, 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제가 개시된다.
Description
유전자 치료는 질병의 치료을 목적으로 하여 유전자 또는 유전자를 도입한 세포를 사람내에 투여하는 치료법이며, 현재의 치료법을 대신하는 새로운 치료법으로서 주목받고 있다. 1990 년에 미국에서 아데노신 디아미나아제 (ADA) 결손에 의한 선청성 중증복합형 면역부전증인 두 아이에 대한 유전자 치료가 실시된 이래, 1996 년말까지 세계에서 2100 건 이상의 유전자 치료가 실시되고 있다. 일본에서도 ADA 결손환자에 대해 1995 년 8 월, 일본 최초의 유전자 치료가 개시되었다.
유전자 치료연구에서, 유전자 치료용 약제의 안전성을 높이는 연구는 필요불가결한 기술이며, 예컨대 치료의 대상인 세포에만 치료용 유전자를 도입한다는 타깃팅 수법은 투여한 유전자 치료용 벡터의 쓸데없는 확산을 방지함으로써 유효성을 높일 뿐만 아니고, 부작용의 원인이 될 수 있는 외래 유전자의 바람직하지 않은 장기, 조직에서의 발현을 저감한다는 의미에서도 필요불가결한 기술이다.
예컨대 사람 면역부전 바이러스 (HIV) 를 기초로 제작된 HIV 벡터는 그 표면에 존재하는 외피 단백질 (Envelope proteins) 이 CD4 단백질에 대하여 특이적으로 결합하는 성질을 이용하여 CD4 양성 세포에만 유전자 도입이 가능한 것 (Shimada T., et al., J. Clin. Invest., 88, 1043, 1991), 몰로니마우스 백혈병 바이러스 (Molony murine leukemia virus) 의 외피를 헤레글린 (heregulin) 으로 수식한 재조합 바이러스 벡터는 상피세포 성장인자 (Epidermal growth factor receptor : EGFR) 를 과잉 발현하고 있는 폐암세포 등의 세포에만 유전자 도입이 가능한 것 (Han X. L., et al., Proc. Natl. Acad. Sci. USA, 92, 21, 9747, 1995), 그리고 암세포에 엽산수용체가 과잉 발현하고 있는 것을 이용하여 암세포에 대한 친화성을 높인 엽산결합 폴리-L-리신 (Gottschalk S., et al., Gene Ther., 1, 3, 185, 1994, Lee R. J., Huang L., J. Biol. Chem., 271, 14, 8481, 1996) 등의 기술이 개시되어 있다.
한편, 치료용 유전자가 치료의 대상이 되는 조직에 대하여 특이적으로 발현되는 시스템 (조직 특이적 발현 시스템) 의 개발도 진행되고 있다. 조직 특이적 발현 시스템은 특정 장기, 조직에서만 발현되는 단백질에 주목하고, 이 단백질을 코드하는 유전자의 프로모터 및/또는 인핸서 (Enhancer) 등의 유전자 발현을 제어하는 유전자 서열을 이용하는 기술이다.
이들 기술은 이미 공지되어 있는 총설 (Gabi U. Dachs, etal., Oncology Res., 9, 313, 1997) 등에 의해 개시되어 있다. 조직 특이적인 프로모터/인핸서로서는 간세포암에서 특이적으로 발현이 발견되는 α- 페토프로테인 (Alpha-Fetoprotein : AFP) 의 프로모터/인핸서, 전립선암에서 발현이 증가하는 전립선암 특이항원 (Prostate specific antigen : PSA) 의 프로모터영역, 내피세포에서 특이적인 폰 빌레브란드 (von Willebrand) 인자 (vWf) 의 프로모터 및 tie-2/tek 의 프로모터, 유방암에서 발현증가가 발견되는 DF3 의 프로모터, 간장 특이적으로 발현되는 알부민의 인핸서, 멜라노머 (Melanoma) 세포에서의 특이적 발현이 가능한 티로시나아제 (Tyrosinase) 의 프로모터, 글리오머 (Glioma) 세포에 특이적인 미엘린 염기성 단백질 (Myelin Basic Protein : MBP) 의 프로모터, 골육종세포 (Osteosarcoma) 특이적인 오스테오칼신 (Osteocalcin) 의 프로모터 등이 알려져 있다.
질환 특이적인 것으로는, 다양한 암에서 발현성의 증가가 발견되는 암태아성 항원 (Carcinoembryonic Antigen : CEA) 의 프로모터, 유방암 또는 췌장암에서 발현성의 상승이 발견되는 HER/neu 의 프로모터, 세포의 증식, 분화, apo토시스 (Apoptosis) 에 관련한 Myc 단백질 패밀리/Max 단백질 복합체가 작용하여 전사를 촉진하는 Myc-Max 반응요소 (response element) 를 들 수 있다. 또 다양한 조건에 의해 발현이 조절되는 것으로서, 방사선을 조사했을 때의 초기 성장 반응-1 유전자 (Early Growth Response-1 Gene: Egr-1) 의 프로모터 또는 조직형 플라스미노겐 활성화제 (Tissue type plasminogen activator : t-PA), 글루코오스의 고갈 또는 산소결핍상태 등의 종양 특이적 조건에서 유도되는 GRP78/BiP 단백질의 프로모터, 저산소상태에서 특이적으로 발현이 발견되는 저산소증 반응요소 (Hypoxia Response Element : HRE) 등이 보고되고 있다.
그러나, 전체 암의 약 60 % 에 이른다고 하는 자궁경부암, 피부암, 두경부암, 식도암, 폐암 등의 편평상피암에 대해서는, 특이적으로 유전자를 발현시키는 시스템이 존재하지 않아 이들 편평상피암에 대해서 유효한 유전자 치료를 실시하는 시스템이 없었다.
본 발명은 치료용 유전자를 편평 상피세포에 특이적으로 발현시키는 염기서열 및 이 염기서열을 사용한, 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제에 관한 것이다.

(발명의 개시)
본 발명은 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 염기서열을 제공하는 것을 목적으로 한다. 또 이 염기서열을 사용하는 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제를 제공하는 것을 목적으로 한다.
본 발명자들은 상기 과제에 대하여 예의 검토한 결과, 편평상피세포에만 특이적으로 치료용 유전자를 발현시키는 것이 가능한 신규 염기서열을 발견하였다. 또한 이러한 신규 염기서열은 정상 편평상피세포와 비교하여 SKGⅢa 등의 편평상피암세포에서 유전자 발현능력이 더욱 증강되는 것을 발견하고 본 발명의 완성에 이른 것이다.
즉, 본 발명은 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 치료용 유전자 발현 염기서열에 관한 것이다.
보다 상세하게는 편평상피세포에 치료용 유전자를 특이적으로 발현시키는, 하기 (a) ∼ (d) 중 어느 하나의 치료용 유전자 발현 염기서열에 관한 것이다.

또, 본 발명은 하기 (a) ∼ (d) 중 어느 하나로 표시되는 염기서열에서, 1 또는 수개의 염기가 결실, 치환 또는 부가된 염기서열로서, 편평상피세포에 치료용 유전자를 특이적으로 발현시키는 치료용 유전자 발현 염기서열에 관한 것이다.

또한, 본 발명은 상기 치료용 유전자 발현 염기서열의 하류영역에 치료용 유전자를 갖고, 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제에 관한 것이다. 또 이 약제를 사용하여 특정 편평상피세포에 발현시키고자 하는 치료용 유전자를 발현시키는 방법도 포함하는 것이다.
(발명을 실시하기 위한 최선의 형태)
이하, 본 발명을 발명의 실시형태에 입각하여 상세하게 설명한다.
(염기서열)
본 발명의 편평상피세포에 특이적으로 치료용 유전자를 발현시키는 염기서열 (이하, 본 발명의 염기서열이라고 함) 은 편평상피세포암 항원 1 [Squamous Cell Carcinoma Antigen l (이하, SCC-A1 이라고 약기)] 의 아미노산 서열을 규정하는 구조유전자보다도 상류영역의 연속염기서열이다. 여기서 SCC-A 는 자궁경부암 등의 편평상피암의 종양 메이커로서 널리 사용되고 있는 단백질이며, 유전자 서열에 약 95 % 의 상동성을 갖는 SCC-A1 및 SCC-A2 가 존재하고, 이들은 함께 염색체상의 18q21. 3 에 존재한다 (S. S. Schneider, et al., Proc. Natl. Acad. Sci. USA, 92, 3147, 1995). Sakaguchi 등은 SCC-A2 프로모터를 해석하고, 편평상피암세포인 SKGⅢa 의 세포에서의 유전자의 발현은, 전사개시점으로부터 500 bp 상류까지의 길이의 DNA 단편을 사용함으로써 가장 강한 전사활성이 얻어진다는 것을 보고하고 있다 (Sakaguchi Y., Biochemica et Biophysica Acta, 1444, 111-116, 1999). 또 Shirato 등은 방사선치료를 받은 편평상피세포암 환자의 혈청중의 SCC-A 단백질 농도를 측정한 결과, 자궁경부암 환자에서 SCC-A 단백질 농도와 예후가 상관된다는 것을 발견하고, 치료후의 모니터링에 유용하다고 보고하고 있다 (Shirato H., et al., Acta Oncologica 32, 6, 663, 1993). SCC-A 단백질의 아미노산 서열 및 그것을 코드하는 유전자 서열은 일본 공개특허공보 평 4-200387 호에 의해 개시되어 있다.
본 발명의 염기서열은 서열번호 1 에 나타나는 4324 염기의 연속염기서열의 전체 또는 그 일부의 염기서열을 포함하고 또한 편평상피세포에 특이적으로 치료용 유전자를 발현시키는 염기서열이다. 또한 서열번호 18 에 나타나는 1000 염기의 연속염기서열의 전체 또는 그 일부의 염기서열을 포함하고 또한 편평상피세포에 특이적으로 치료용 유전자를 발현시키는 염기서열이다. 또 서열번호 17 에 나타나는 500 염기의 연속염기서열의 전체 또는 그 일부의 염기서열을 포함하고 또한 편평상피세포에 특이적으로 치료용 유전자를 발현시키는 염기서열이다. 그리고 서열번호 16 에 나타나는 250 염기의 연속염기서열의 전체 또는 그 일부의 염기서열을 포함하고 또한 편평상피세포에 특이적으로 치료용 유전자를 발현시키는 염기서열이다.
또, 편평상피세포에만 특이적으로 치료용 유전자를 발현시키는 기능을 갖는 것이면, 상기 어느 하나의 서열에서 1 또는 수개의 염기가 결실, 치환 또는 부가, 혹은 수식되어 있는 염기서열도 본 발명의 염기서열에 포함된다.
본 발명의 염기서열을 얻는 방법으로서는 특별히 한정되지 않는다. 본 명세서에 개시된 염기서열에 기초하여 종래 공지의 방법을 바람직하게 적용할 수 있다. 예컨대 적절한 농도로 조정한 마그네슘이온 등의 염류와 dATP, dGTP, dCTP, dTTP 등을 함유하는 완충액중에서, SCC-A1 유전자 상류게놈영역을 주형으로 하고, 주형 DNA 서열의 목적으로 하는 영역을 증폭가능하도록 디자인한 프라이머 및 내열성 DNA 폴리머라제를 사용하여 통상 시행되는 PCR 법에 의해 조제할 수 있다.
이 때, 사용하는 내열성 DNA 폴리머라제로는, 테무스 아쿠아티쿠스 (Themus aquaticus) 유래의 Taq DNA 폴리머라제, 피로코쿠스 푸리오수스 (Pyrococcus furiosus) 유래의 Pfu DNA 폴리머라제 또는 이들의 대장균에 의한 재조합형 내열성 DNA 폴리머라제 등을 들 수 있다. 또 PCR 법은 시판되고 있는 PCR 용 장치를 사용하여 시행할 수 있다.
본 발명의 염기서열은 서열번호 1, 18, 17 또는 16 에 나타나는 염기서열의 일부로 구성되어 있어도 되므로 100 하기 염기수이어도 되고, 이 경우는 핵산화합물로서 본 발명의 염기서열을 합성하는 것도 가능하다.
핵산화합물로서는, 천연형 올리고데옥시리보누클레오티드, 포스포로티오에이트형 올리고데옥시리보누클레오티드, 메틸포스포네이트형 올리고데옥시리보누클레오티드, 포스포로아미데이트형 올리고데옥시리보누클레오티드, H-포스포네이트형 올리고데옥시리보누클레오티드, 트리에스테르형 올리고데옥시리보누클레오티드 등을 들 수 있다.
또한, 핵산합성하는 경우에는 상기 방법외의 DNA/RNA 합성기를 사용하여 포스파아미다이트법에 의해서도 합성할 수 있다.
포스파아미다이트법이란, 수식 데옥시리보누클레오시드의 3' 말단에 시아노에틸기 등으로 보호한 포스파아미다이트를 결합한 시약을 사용하여 별도의 수식 데옥시리보누클레오시드, 올리고수식 데옥시리보누클레오시드 등의 5' 말단 등에 축합하는 것을 기본체로 하는 합성법이며, 최종 사이클에서 5' 말단의 당수산기의 보호기가 결합한 상태에서 합성을 종료한다. 실온하에서 합성한 올리고머를 서포트로부터 절단한 후, 염기부분 및 인산부분의 탈보호를 실시하여 조(粗)정제물을 얻는다. 조정제물은 통상의 정제방법, 예컨대 에탄올 침전법을 사용하거나, 역상 크로마토그래피, 이온교환 크로마토그래피 및 겔여과 크로마토그래피의 원리에 기초한 고속액체 크로마토그래피, 초임계 크로마토그래피 등 다양한 방법으로 정제된다.
(치료용 유전자)
본 발명의 염기서열은 편평상피세포에 치료용 유전자를 특이적으로 발현시키기 위한 프로모터로서 유용하다.
구체적으로는 편평상피세포에 특이적으로 발현시키고자 하는 치료용 유전자를 본 발명의 염기서열의 하류에 위치시키고, 예컨대 바이러스 벡터의 바이러스 게놈서열의 일부에 삽입함으로써 사용하는 것이 가능하다.

또한, 치료용 유전자는 치료효과가 기대되는 유전자면 사람유래의 유전자일필요는 없고, 이와 같은 예로서, 예컨대 사람 단순 헤르페스 바이러스 유래 티미딘키나아제 유전자, 사람 델타형 간염 바이러스 등의 바이러스·바이로이드 또는 테트라히메나 등의 원생동물 섬모충류 유래의 리보자임 등을 들 수 있다.
상기중, 편평상피세포에 특히 치료효과가 기대되는 유전자로서는, 예컨대 색소성 건피증에서는 누클레오티드 제거수복기구에 관한 XPA 로부터 XPG 중 어느 하나의 유전자를, 표피수포증에서는 케라틴 유전자 또는 Ⅶ 형 콜라겐 유전자를, 티로시나아제 음성형 백피증에서는 티로시나아제 유전자를, 신경섬유종증 1 형에서는 NF1 유전자를 일례로서 들 수 있다.
또, 편평상피암세포에 특히 치료효과가 기대되는 유전자로서는, 예컨대 사람단순 헤르페스 바이러스유래 티미딘키나아제 유전자 등의 자살 유전자, 뇌 글리코겐 포스포릴라아제 유전자 등의 암특이적 항원을 코드하는 유전자, fas 유전자 등의 아폽토시스(apoptosis)를 촉진하는 유전자, 사람 백혈구 항원 유전자 등의 암세포의 면역원성을 높이기 위한 유전자, p53 유전자 등의 암억제 유전자, 암 유전자에 대한 안티센스 또는 리보자임 등의 편평상피암세포에 대하여 길항적으로 작용하는 것을 일례로서 들 수 있다.
또한, 본 발명은 주름의 생성억제, 보습성의 향상, 기미·주근깨의 생성억제, 지성·여드름의 억제 등의 미용을 목적으로 한 치료에도 적용가능하다. 예컨대 주름의 생성억제, 보습성의 향상을 위해 케라틴 유전자 또는 콜라겐 유전자를 1 종 또는 2 종 이상 조합하여 사용가능하다. 또 기미, 주근깨의 생성억제를 위해 티로시나아제 유전자 등의 멜라닌 생성에 관한 일련의 효소 유전자에 대한 안티센스 또는 리보자임 등을 1 종 또는 2 종 이상 조합하여 사용가능하다.
또, 본 발명의 치료용 유전자에는 마커 유전자도 포함된다. 마커 유전자로서는 CAT, GUS, β-gal, GFP, EGFP 등이 사용가능하다.
발현된 치료용 유전자의 세포내에서의 안정성을 제어하기 위해 폴리 A 시그널을 치료용 유전자의 하류에 더 삽입하는 것도 가능하다.
(치료용 유전자의 도입방법)
본 발명의 염기서열의 하류에 치료용 유전자를 도입하고, 이러한 치료용 유전자를 정상 편평상피세포 또는 편평상피암세포 등으로 송달하는 수법으로는 특별히 한정되지 않지만, 예컨대 바이러스 벡터를 사용하는 경우, 또는 합성 폴리아미노산, 양이온성 지질 등의 비바이러스 벡터의 담체를 사용하는 경우가 있다.
바이러스 벡터를 사용하는 경우는, 레트로 바이러스 벡터, 아데노 바이러스 벡터, 아데노 수반 바이러스 벡터, 재조합 HIV 벡터, 단순 헤르페스 바이러스 벡터 등의 바이러스유래의 벡터를 사용할 수 있다. 바이러스 벡터에서는 바이러스 게놈서열의 일부에 본 발명의 염기서열을 삽입하여 사용한다. 예컨대 레트로 바이러스 벡터중, 백혈병 바이러스 벡터의 경우는 바이러스게놈의 5' 측부터 순차적으로, 백혈병 바이러스유래의 LTR (Long Terminal Repeat), 본 발명의 염기서열, 치료용 유전자, 폴리 A 시그널, LTR 과 동일하게 디자인된 재조합 바이러스 게놈을 구축하면 된다. 작성한 바이러스 벡터는 공지의 방법에 의해 바이러스 입자내에 패키징하면 된다.
합성 폴리아미노산, 양이온성 지질 등의 비바이러스 벡터의 담체를 사용하는경우에는, 예컨대 합성 폴리아미노산의 경우, 폴리리신 또는 세린 등의 아미노산을 주체로 하는 담체를 사용할 수 있다. 또 예컨대 양이온성 지질에서는 리포솜 등을 사용할 수 있다. 비바이러스 벡터의 경우는 플라스미드에 본 발명의 염기서열과 그 하류에 치료용 유전자 및 폴리 A 시그널을 삽입하여 사용할 수 있다.
(유전자 치료용 약제)
본 발명의 유전자 치료용 약제는 앞에서 설명한 본 발명의 염기서열을 사용하고 있으며, 특정 치료용 유전자를 편평상피세포에만 특이적으로 발현시킬 수 있다. 이 유전자 치료용 약제는 본 발명의 염기서열을 프로모터로 하고, 그 하류에 치료용 유전자를 도입하여 얻을 수 있다.
이 유전자 치료용 약제를 상기 수법에 의해 세포에 송달하였을 때, 본 발명의 염기서열은 치료용 유전자를 편평상피세포에만 특이적으로 발현시킬 수 있다. 또한 본 발명에 따르면, 치료용 유전자를 특히 암화된 편평상피세포에 강하게 발현시킬 수도 있다.
이러한 효과는 또한 서열번호 1 에 나타내는 4324 염기의 연속염기서열로 확인되지만, 이 서열중, 서열번호 18 에 나타내는 1000 염기의 연속염기서열, 나아가서는 서열번호 17 에 나타내는 500 염기의 연속염기서열, 더불어 서열번호 16 에 나타내는 250 염기의 연속염기서열로 강하게 확인된다.
본 발명의 유전자 치료용 약제에 의한 치료효과는 피부, 폐, 식도, 자궁 등의 편평상피세포를 포함하는 전 조직, 장기의 유전성 질환에서 유용하다. 예컨대 색소성 건피증, 표피수포증, 어린선(魚鱗癬), 백피증, 티로시나아제 음성형 백피증, 신경섬유종증 1 형 등의 유전성 피부질환의 치료에도 유용하다.
또, 피부의 주름의 생성억제, 보습성의 향상, 기미·주근깨의 생성억제, 지성·여드름의 억제 등의 미용을 목적으로 한 치료에도 유용하다.
또한, 본 발명의 유전자 치료용 약제는 편평상피세포에 대한 질환뿐만 아니고, 편평상피암세포의 유전자 치료에도 유용하여 자궁경부암, 피부암, 두경부암, 식도암, 폐암 등에서 특히 유효하다.
본 발명의 유전자 치료용 약제를 투여하는 방법으로는 직접 목적으로 하는 장기, 조직에 투여하는 방법외, 경구 또는 비경구로 투여하는 방법도 사용된다. 경구투여에는 혀아래 투여가 포함된다. 비경구투여에는 예컨대 피하, 근육, 정맥, 동맥, 점적을 투여경로로 하는 주사, 또한 좌제, 도포약, 첩부약이 포함된다.
또, 발명의 유전자 치료용 약제의 투여량은 연령, 투여경로, 투여횟수에 의해 달라지며 적당히 변경할 수 있다. 이 경우, 본 발명의 유전자 치료용 약제의 유효량과, 적절한 희석제 및 약리학적으로 사용할 수 있는 담체의 조성물로서 통상 투여되지만, 그 유효량은 1 ∼ 100,000 ㎍/㎏ 체중/일 이며, 연속적으로 또는 1 일 1 회에서 수회에 걸쳐서 또는 수일마다 1 회 투여된다.
본 발명의 유전자 치료용 약제를 경구투여하는 경우는 정제, 과립제, 세립제, 산제, 캡슐제 등이 적용된다. 이들 조성물중에 통상 함유되는 결합제, 포함제, 부형제, 붕괴제 등을 포함해도 되고, 내용수제, 현탁제, 유제, 시럽제 등 중 어느 형태이어도 된다. 또 비경구투여하는 경우에는 이를 위한 조성물이 안정화제, 완충제, 보존제, 등장화제 등을 포함해도 되고, 통상 단위투여량 앰플 혹은 다투여량 용기 또는 튜브의 상태로 제공된다.
이하, 실시예를 나타내고, 이 발명의 실시형태를 구체적으로 설명하지만, 본 발명은 하기 예에 한정되는 것은 아니다.
(실시예)
(실시예 1 ; 스크리닝용 멤브레인의 조제)
1 차 스크리닝용으로서, 사람게놈 DNA 라이브러리 (EMBL3 SP6/T7, Clontech #HL2067j, Sau3A I partial digest ; Clontech사) 를, 파지의 플라크수가 100 ㎕ 당 1 ×104pfu (plaques forming unit) 가 되도록, 파지희석완충용액 [100 mM NaCl (와코쥰야쿠사), 10 mM MgSO4(와코쥰야쿠사) 를 포함하는 50 mM Tris-HCl (GIBCO사), pH 7.5, 0.01 % 젤라틴] 로 희석하여 플레이트 전면에 구석구석 플라크가 형성되도록 조제하고, 합계 1.5 ×105개의 플라크의 스크리닝을 실시하였다. 2 차 스크리닝용으로서, 1 차 스크리닝에 의해 얻어진 양성 파지클론을 함유하는 파지액을, 파지의 플라크수가 100 ㎕ 당 3000 pfu 가 되도록 파지희석완충용액로 희석하여 플레이트에 단리된 플라크가 형성되도록 조제하였다.
대장균 K802 주를 파종하기 전날부터 5 ㎖ 의 LB 배지 (DIFCO사) 에서 배양하고, 1 ㎖ 를 0.2 % 말토오스, 10 mM MgSO4를 포함하는 5 ㎖ 의 LB 배지에 식균한 후, OD600 nm 의 값이 0.6 이 될 때까지 배양함으로써 파지를 감염시키기 위한 대장균을 조제하였다.
상기 대장균 조제액 300 ㎕ 와 파지희석액을 혼합하여 37 ℃ 에서 15 분간 인큐베이트하였다. 이것을 오토클레이브후, 48 ℃ 에서 보온한 6 ㎖ 의 0.75 % Bacto-Agar (DIFCO사) 를 포함하는 LB 배지에 첨가하고, LB 한천배지에 전체량을 첨가하였다. 약 20 분간 실온에 방치한 후, 37 ℃ 에서 하룻밤 인큐베이트하였다.
플레이트를 4 ℃ 에 1 시간 두고, Hybond-N+(아마샴사) 를 LB 한천배지상에 밀착시켜 실온에서 1 분간 방치하였다. 멤브레인을 바람에 건조하여 변성 (Denature) 완충용액 (0.4 M NaOH 용액) 에 5 분간, 중화 (Neutralization) 완충용액 (1 M NaCl 를 포함하는 0.5 M Tris-HCl 용액, pH 7.5) 에 5 분간, 2 ×SSC 에 5 분간 순차적으로 침지시켜 UV 크로스 링커 (Cross Linker) (UV 스트러터 링커 (Strata Linker) ; 스트러터 진사) 처리하고, 하이브리백 (코스모바이오사) 에 보존하였다.
(실시예 2 ; DNA 프로브의 합성)
스크리닝에 사용한 SCC-A2 유전자 유래의 DNA 프로브 (서열번호 2) 는 하기와 같이 하여 조제하였다. 즉, 사람 자궁경암세포주 HT-Ⅲ (American Type Cell Collection사 : ATCC사) 의 mRNA 로부터 M-MLV 역전사효소 (GIBCO사) 에 의해 cDNA 를 제조하고, PCR primer (서열번호 3,4) 를 사용하여 PCR (GeneAmp PCR System 2400 : Perkin Elmer사) 에 의해 165 bp 의 PCR 산물을 합성하였다. 정제는 QIAquick PCR purification kit (QIAGEN사) 를 사용하여 상기 DNA 프로브를 얻었다.
(실시예 3 ;32P 표지-DNA 프로브의 제작)
사 에서 조제한 DNA 프로브의 라디오아이소토프 표식은 랜덤 프라이머 DNA 표지화 키트 (Random primer DNA labeling kit) (다카라슈조사) 를 사용하여32P 라벨링을 하였다. 정제는 QIAquick nucleotide removal kit (QIAGEN사) 를 사용하여 실시하였다.
(실시예 4 ; 플라크 하이브리다이제이션에 의한 클리닝)
실시예 1 에서 조제한 멤브레인에 예비융합화 용액 (prehybridization solution: GIBCO사) 을 1 매당 20 ㎖ 를 첨가하여 42 ℃ 에서 2 시간 인큐베이트하였다. 예비 융합화 용액을 버린 후, 실시예 3 에서 조제한 DNA 프로브 (1 ×107cpm/㎖) 를 함유하는 예비 융합화 용액 (GIBCO사) 을 20 ㎖ 첨가하여 42 ℃ 에서 하룻밤 인큐베이트하였다. 멤브레인을 1L 의 0.1 % SDS 를 함유하는 6 ×SSC 에 의해 42 ℃ 에서 30 분간의 세정을 3 회 실시하고, 다시 1L 의 0.1 % SDS 를 포함하는 2 ×SSC 에서 65 ℃, 30 분간의 세정을 2 회 실시하였다. 멤브레인을 하이브리백 (코스모바이오사) 에 싸서 -80 ℃ 에서 3 일간, X 선 필름 (후지필름사) 에 감광, 현상하였다. 현상한 필름과 원래의 플레이트를 중첩함으로써 양성 플라크의 위치를 확인하였다. 양성 플라크의 위치를 한천마다 잘라내 회수하고, 500 ㎕ 의 파지희석완충용액와 5 ㎕ 의 클로로포름 (와코쥰야쿠사) 을 첨가하였다. 진탕기(vortex)에 의한 교반 및 원심에 의해 겔편을 가라앉힌 후, 양성 파지입자를 포함하는 상청을 회수하였다. 또한, 양성파지를 단리하기 위해, 실시예 1, 3 및본예의 방법을 사용하여 2 차 스크리닝하고 단리한 플라크중에서 2 종의 양성 파지클론 (B612, C618) 을 단리하였다.
(실시예 5 ; 파지 DNA 의 대량조제)
대장균 K802 주를 파종하기 전날부터 OD 600 nm 의 값이 0.6 이 될 때까지 배양하였다.
실시예 4 에서 얻은 파지액을 파지희석완충용액로 희석하여 B612, C618 을 각각 1010pfu/㎖ 로 조제하였다. 37 ℃ 에서 배양을 실시한 후, 클로로포름과 NaCl 을 첨가하여 37 ℃ 에서 30 분간 진탕하였다. 5000 ×g 에서 30 분간 원심을 실시하여 파지를 포함하는 상청을 회수하였다. 파지 DNA 의 추출은 다음과 같이 하여 실시하였다. 폴리에틸렌글리콜 (와코쥰야쿠사) 100 g 을 첨가하여 4 ℃ 에서 하룻밤동안 침전한 후, 0.75 g/㎖ 의 CsCl (GIBCO사) 로 35000 rpm 에서 18 시간 초원심하였다. 그 후, SM 완충용액 (50 mM Tris-Cl (pH 7.5), 100 mM NaCl, 10 mM MgSO4, 0.01 % 젤라틴) 로 하룻밤 투석하였다. 이것을 0.02 % SDS 함유 페놀용액에서 1 시간 진탕처리한 후, TE 완충용액 를 사용한 투석에 의해 정제하였다. 그 결과, B612 가 800 ㎍, C618 이 910 ㎍ 의 파지 DNA 가 회수되었다.
(실시예 6 ; SCC-A 유전자 상류 서열의 서열화)
상기 B612, C618 의 양 클론중, 서열번호 2 에 나타내는 서열에서 상류서열을 서열화하였다.
그 결과, C618 에서 서열번호 5, B612 에서 서열번호 6 에 나타내는 DNA 서열이 얻어졌다. 상동성 검색 결과, 서열번호 5 의 3' 말단영역은 사람 편평상피세포암 항원 1 (Human squamous cell carcinoma antigen 1: SCC-A1) 유전자의 엑손 1 및 2 의 서열과, 서열번호 6 의 3' 말단영역은 사람 편평상피세포암 항원 1 (SCC-A2) 유전자의 엑손 1 및 2 의 서열과 상동성이 높았다. 이러한 점에서 서열번호 5 는 SCC-A1 유전자 게놈 상류영역 및 SCC-A1 유전자의 일부를 포함하는 서열, 서열번호 6 은 SCC-A2 유전자 게놈 상류영역 및 SCC-A2 유전자의 일부를 포함하는 서열인 것이 판명되었다.
(실시예 7 ; Cap 사이트의 결정)
편평상피암세포인 SKGⅢa 세포 (전 일본 암연구자원 뱅크사, 현 휴먼사이언스 연구자원 뱅크사) 에서 ISOGEN (니뽕진사) 을 사용하여 total RNA 를 조제하였다. 역전사반응은 SMART PCR cDNA SYNTHESIS KIT (Clontech사) 및, SuperScript Ⅱ RNaseH 역전사효소 (GIBCO사) 를 사용하였다. 이로써 5' 말단에 서열번호가 7 인 서열을 부가한 cDNA 를 제조하였다. 이 역전사반응물을 주형으로 하고, 서열번호 8 (SMART PCR cDNA SYNTHESIS KIT 첨부), 서열번호 9 (스워디 테크놀러지사) 의 PCR 프라이머를 사용하여 PCR 을 실시하였다.
반응후의 혼액을 0.8 % 아가로오스 겔 전기영동에 가하여 특이적으로 증폭된 PCR 산물을 겔편마다 회수하고, QIAquick Gel extraction kit (QIAGEN사) 를 사용하여 겔에서 추출하였다.
0.1 ㎍ 의 Bluscript Ⅱ T-벡터 (도요보사) 와, 정제한 PCR 산물을 혼합하고, T4 DNA Ligase (GiBCO사) 를 사용하여 4 ℃ 에서 하룻밤 라이게이션반응을 실시하였다.
컴피텐트 셀 E. coli XLI-blue (다카라슈조사) 200 ㎕ 를 얼음중에서 녹이고, 상기 5 ㎕ 의 라이게이션 반응액을 첨가하여 30 분간 방치하였다.
42 ℃ 에서 60 초간 인큐베이트한 후, 재차 얼음중에 5 분간 두어 SOC 배지 (GIBCO사) 를 800 ㎕ 첨가하고, 다시 37 ℃ 에서 1 시간 인큐베이트하였다. 50 ㎍/㎖ 의 암피실린 (와코쥰야쿠사) 을 포함하는 LB 한천배지에 100 ㎕ 를 파종하여 37 ℃ 에서 하룻밤 인큐베이트하였다. 단독 콜로니를 5 클론 선택하여 50 ㎍/㎖ 의 암피실린을 포함하는 LB 액체배지에 첨가하고, 다시 37 ℃ 에서 하룻밤 배양하였다.
키아겐 플라스미드 MINI kit (QIAGEN사) 를 사용하여 픽업한 5 클론의 플라스미드정제를 실시하였다.
정제한 5 클론중의 상류서열을 DNA 시퀀서 타입 310 (ABI사) 을 사용하여 확인하였다.
그 결과, SCC-A1 유전자의 Cap 사이트는 서열번호 5 의 4296 번째의 염기인 것이 판명되었다.
(실시예 8 ; 발현 플라스미드의 구축 I)
실시예 5 에서 얻어진 파지 DNA (C618) 를 주형으로 하고, 표 1 에 나타내는 조합의 PCR 프라이머를 사용하여 서열번호 16 ∼ 20 에 나타나는 5 종이 상이한 길이의 SCC-A1 유전자 상류영역 DNA 단편을 PCR 에 의해 조제하였다. 반응후의 혼액을 0.8 % 아가로오스 겔 전기영동에 가하여 특이적으로 증폭된 PCR 산물을 회수하고, QIAquick Gel extraction kit 를 사용하여 겔에서 추출하였다. 결과를 표 1 에 함께 나타낸다.

0.1 ㎍ 의 베이식 벡터 (니뽕진사) 를 Mlu I (다카라슈조사) 및 Bg1 Ⅱ (다카라슈조사) 제한효소처리하였다. 상기 벡터와 각각의 PCR 산물을 혼합하여 DNA Ligation kit (다카라슈조사) 를 사용하여 16 ℃ 에서 하룻밤 라이게이션 반응을 실시하였다.
이하, 실시예 7 에 나타낸 수법을 사용하여 트랜스포메이션을 실시하고, 키아겐 플라스미드 MINI kit 를 사용하여 플라스미드를 정제하였다.
서열번호 16 ∼ 20 의 서열을 삽입한 플라스미드의 유전자 서열을, DNA 시퀀서 타입 310 (ABI사) 을 사용하여 확인하였다. 이상과 같은 조작에 의해, 도 1 에 나타내는 서열번호 16 ∼ 20 이 상이한 길이의 SCC-A1 유전자 상류영역을 갖는 발현 벡터를 조제하였다.
(실시예 9 ; 유전자 발현성 시험 I)
정상 편평상피세포인 정상 사람피부각화세포 (다이니뽕세이야쿠사), 편평상피암세포인 SKGⅢa 세포 및 비편평상피암세포인 난소암세포 (SKOV3 세포 (ATCC사))를 12 웰 플레이트에 1.0 ×106세포/웰이 되도록 세포를 파종하여 하룻밤 배양하였다.
서열번호 16 ∼ 20 의 서열을 갖는 5 종의 플라스미드 및 양성대조로서 SV40 프로모터를 갖는 컨트롤 벡터 (니뽕진사), 음성대조로서 프로모터서열을 갖지 않는 베이식 벡터를 각각 실험에 사용하였다. 각각의 플라스미드 1 ㎍ 에 6 ㎍ 의 DOTAP 리포좀 형질감염 시약 (liposomal transfection reagent: Boheringer-Mannheim사) 을 첨가하고, 실온에서 15 분간 방치하여 형질감염 혼액을 조제하였다. 이 혼액을 피부각화세포에 첨가하는 군은 정상 사람피부각화세포용 무혈청배지 (다이니뽕세이야쿠사) 1 ㎖ 에, SKGⅢa 세포 및 SKOV3 세포에 첨가하는 군은 10 % FCS (GIBCO사) 를 포함하는 RPMI 1640 배지 (GIBCO사) 1 ㎖ 에 첨가하고, 이것을 각 세포를 파종한 12 웰 플레이트에 첨가하여 37 ℃, 5 % CO2분위기하, 인큐베이트하였다. 6 시간후, 각각의 증식배지를 교환하여 37 ℃, 5 % CO2분위기하, 다시 48 시간 인큐베이트하였다.
48 시간후, PBS (-) 액에서 5 배로 희석한 PicaGene Cell Culture Lysis Regent Luc (니뽕진사) 를 20 ㎕/웰 첨가하여 세포를 용해하고, PicaGene Luminescence Kit (니뽕진사) 를 사용하여 루시페라아제 활성을 측정하였다. 루미노미터는 Lumicounter 700 (마이크로텍사) 을 사용하였다.
결과를 표 2 에 나타낸다. 서열번호 16 ∼ 20 의 서열을 갖는 5 종의 플라스미드는 피부각화세포 및 SKGⅢa 세포에서 루시페라아제 활성의 상승이 발견되었다. 한편, SKOV3 세포에서는 거의 루시페라아제 활성이 검출되지 않고, 프로모터를 갖지 않는 베이식 벡터와 동일한 정도였다. SV40 프로모터를 갖는 컨트롤 벡터는 어느 세포에서도 동일한 정도의 높은 루시페라아제 활성이 검출되며 세포종특이성을 보이지 않았다. 이러한 점에서 서열번호가 16 ∼ 20 인 서열을 갖는 5 종의 플라스미드는 편평상피세포 특이적인 유전자 발현 활성을 갖는 것이 판명되었다. 특히, 이들 서열번호 16 ∼ 20 의 서열을 갖는 5 종의 플라스미드 중, 서열번호 16 ∼ 18 의 서열을 갖는 플라스미드가 강한 유전자 발현 활성을 갖는 것도 판명되었다. 또한 서열번호가 17 인 서열을 갖는 플라스미드가 가장 강한 유전자 발현성을 나타냄을 알 수 있었다.
(실시예 10 ; 발현 플라스미드의 구축 Ⅱ)
실시예 9 에서 가장 유전자 발현 활성이 높았던 서열번호 17 의 특이성을 더 검토할 목적으로, 서열번호 24 ∼ 26 의 서열을 갖는 DNA 단편을 SCC-A1 유전자 상류영역 (서열번호 5) 으로부터 각각 조제하여 발현 플라스미드를 구축하였다. 구체적으로는 발현 플라스미드를 실시예 8 과 동일한 수법에 따라 표 2 에 나타내는 조합의 PCR 프라이머를 사용함으로써 구축하였다. 또 마찬가지로 SCC-A2 유전자 상류영역 (서열번호 6) 을 주형으로 하고, 서열번호 16 ∼ 18 및 서열번호 24 ∼ 26 의 서열을 갖는 DNA 단편과 동일한 길이의 DNA 단편 (서열번호 27 ∼ 32) 을 갖는 발현 플라스미드를 구축하였다. 결과를 각각 표 2 및 표 3 에 나타낸다.

실시예 9 에서 조제한 SCC-A1 유전자 상류영역 유래의 DNA 단편 (서열번호 16 ∼ 18) 을 갖는 발현 플라스미드, 실시예 10 에서 조제한 SCC-A1 유전자 상류영역 유래의 DNA 단편 (서열번호 24 ∼ 26) 및 SCC-A2 유전자 상류영역 유래의 DNA 단편 (서열번호 27 ∼ 32) 을 갖는 발현 플라스미드의 유전자 발현 활성을 측정하였다.
그 결과를 도 3 ∼ 도 5 에 나타낸다. 도 3 은 각 발현 플라스미드의 피부각화세포에서의 유전자 발현 활성을, 도 4 는 각 발현 플라스미드의 SKGⅢa 세포에서의 유전자 발현 화성을, 도 5 는 각 발현 플라스미드의 난소암세포에서의 유전자 발현 활성을 각각 나타낸다.
서열번호 16 ∼ 18 및 서열번호 24 ∼ 26 의 서열을 갖는, SCC-A1 유전자 상류영역 유래의 DNA 단편을 갖는 6 종의 발현 플라스미드 및 서열번호 27 ∼ 32 의 서열을 갖는 SCC-A2 유전자 상류영역 유래의 DNA 단편을 갖는 6 종의 발현 플라스미드는, 모두 SKOV3 세포에서는 거의 루시페라아제 활성을 나타내지 않고, 프로모터 활성을 갖지 않는 베이식 벡터와 동일한 정도였다 (도 5). 또 상기 모든 발현 플라스미드에 대하여 피부각화세포 및 SKGⅢa 세포에서는 베이식 벡터와 비교하여 루시페라아제 활성의 상승이 보였다 (도 3 및 도 4). 또한, 서열번호 17 의 발현 플라스미드는 SKGⅢa 세포에서 세포 특이성을 보이지 않았던 SV40 프로모터를 갖는 컨트롤 벡터와 동일한 정도의 높은 유전자 발현 활성을 나타내었다.
이상, 사용한 SCC-A1 및 SCC-A2 유전자 상류영역에서 유래하는 염기서열을 포함하는 12 종의 발현 플라스미드는 모두 세포종특이적, 특히 편평상피세포 특이적인 유전자 발현 활성을 갖는 것이 판명되었다. 또 양자의 발현 플라스미드군을 비교하면, 전자 (즉 SCC-A1 유전자 상류영역 유래의 염기서열을 포함함) 발현 플라스미드군의 유전자 발현 활성이 높았다. 또한 서열번호 17 의 서열을 갖는 SCC-A1 유전자 상류영역 유래의 DNA 단편을 삽입한 플라스미드는 그 이외의 SCC-A1 유전자 상류영역 유래의 DNA 단편을 갖는 다른 발현 플라스미드 및 SCC-A2 유전자 상류영역 유래의 DNA 단편을 갖는 모든 발현 플라스미드와 비교하여 가장 강한 유전자 발현 활성을 나타냄을 알 수 있었다.
본 발명의 치료용 유전자 발현 염기서열은 SCC-A 유전자의 상류영역의 염기서열이며, 편평상피세포에 특정 유전자를 발현시키는 프로모터 작용을 갖는다.
본 발명에 따르면 이와 같은 염기서열의 하류영역에 치료용 유전자를 도입함으로써 편평상피세포에 특이적으로 치료용 유전자를 발현시킬 수 있다. 또한, 본발명에 따르면 상기 염기서열의 하류영역에 치료용 유전자를 도입한 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제를 얻을 수 있다.
Claims (3)
- 편평상피세포에 치료용 유전자를 특이적으로 발현시키는, 하기 (a) ∼ (d) 중 어느 하나의 치료용 유전자 발현 염기서열.(a) 서열번호 1 로 표시되는 염기서열(b) 서열번호 18 로 표시되는 염기서열(c) 서열번호 17 로 표시되는 염기서열(d) 서열번호 16 로 표시되는 염기서열.
- 하기 (a) ∼ (d) 중 어느 하나의 염기서열에서 1개 또는 수개의 염기가 결실, 치환 또는 부가된 염기서열로서, 편평상피세포에 치료용 유전자를 특이적으로 발현시키는 치료용 유전자 발현 염기서열.(a) 서열번호 1 로 표시되는 염기서열(b) 서열번호 18 로 표시되는 염기서열(c) 서열번호 17 로 표시되는 염기서열(d) 서열번호 16 로 표시되는 염기서열.
- 제 1 항 또는 제 2 항에 기재된 치료용 유전자 발현 염기서열의 하류영역에 치료용 유전자를 갖고, 치료용 유전자를 편평상피세포에 특이적으로 발현시키는 유전자 치료용 약제.
