김치추출물과 과일산과의 배합물의 5-알파리닥타제에 대한 억제효과Inhibitory Effects of Kimchi Extracts and Fruit Acids on 5-alpha-dactactase

본 발명의 혼합 피부개선제조성물의 5-알파리닥타제 활성억제 실험은 메이슨(H.S. Mason) 및 나가스(T. Nagath)의 방법에 따라 시행하였다. 시험 피부개선제로서 과일산의 농도를 0.03%로 고정시키고 김치추출물의 농도를 0.02, 0.04, 0.10, 0.16 및 0.20%로 변화시킨 배합조성물을 사용하였다.상기 준비된 각각의 피부개선제 샘플에 5-알파리닥타제용액[제(5-알파리닥타제3400유니트/고체 ㎎) 5㎎을 1/15M인산염 완충용액 10㎖에 용해시킨 용액] 0.1㎖ 및 1/15M 인산염 완충용액(pH 6.8) 0.9㎖를 가하고, 이 혼합물을 37℃에서 10분 동안 예열한 다음, 0.03% Testosterone용액 1㎖를 가하여 37℃에서 10분 동안 반응시켰다. 이 때 대조군은 각 피부개선샘플 대신 1/15M 인산염 완충용액(pH 6.8) 1㎖를 가한다. 반응후, 흡수분광계(UVT06856, MOLECULAR DEVICE사의 MICROPLATE READER)를 사용하여 470nm에서 반응혼합물의 흡광도를 측정하였다. 각 피부개선제샘플의 5-알파리닥타제 활성저해율은 다음의 공식으로 구하였다.Inhibition of 5-alpha-dacactase activity of the mixed skin improvement composition of the present invention was carried out according to the method of Mason (H.S. 남자 20명을 실험대상자로 하여 오른쪽 얼굴에 2㎝×2㎝의 크기의 여드름이 있는 부위를 에탄올 70% 수용액으로 잘 세척하였다. 여드름 부위 한쪽줄 3곳에는 실시예 1, 4, 5의 조성물을, 다른 한줄에는 김치추출물과 과일산이 첨가되지 않은 것을 제외하고는 실시예 1, 4, 5 조성물과 동일한 각각의 기제를 1일 2회 4주간 도포하였다. 4주후에 육안으로 피부개선의 정도를 대조군(기제도포)과 비교하여 판정하였으며, 판정기준은 현저한 저해효과, 약간의 저해효과, 저해없음의 3 단계로 평가하였다. 결과는 하기 표 2에 나타내었다.Twenty males were used as test subjects, and the area of 2 cm × 2 cm acne on the right face was washed well with 70% ethanol solution. 표 2의 결과로부터 알 수 있듯이, 본 발명에 따른 실시예 1, 4, 5의 조성물의 경우 모두 최소한 16명 이상이 현저하거나 여드름 저해효과와 피부개선 효과가 있음을 확인하였다.As can be seen from the results of Table 2, in the case of the compositions of Examples 1, 4, and 5 according to the present invention, it was confirmed that at least 16 or more of them were remarkable or had acne inhibitory effects and skin improvement effects.
