sPLA2는 막 인지질을 가수분해시키는 아라키돈산 케스케이드에서 속도 제한 효소인 것으로 믿어진다. 따라서, sPLA2매개 지방산(예를 들면, 아라키돈산) 유리를 억제하는 화합물을 개발하는 것은 중요하다. 그러한 화합물은 패혈증 쇼크, 성인 호흡 곤란 증후군, 췌장염, 외상, 기관지 천식, 알러지성 비염 및 류마티스성 관절염 등과 같은 sPLA2의 과다 생산에 의해 유발되고(거나) 유지되는 질환의 일반적인 치료에서 중요할 것이다.
문헌[헤이덴 지. 비턴 (Hayden G. Beaton) 등, Journal of Medicinal Chemistry, 1994, 37권, No. 5]에는 비-인지질 sPLA2억제제의 다양한 신규한 (나프틸티오) 메틸 유사체가 기재되어 있다.
sPLA2에 의해 유발된 질병 치료용의 신규한 화합물 및 그 치료 방법을 개발하는 것이 요망된다.
본 발명은 패혈증 쇼크와 같은 질환에 있어서 sPLA2매개 지방산 유리를 억제하기 위해 유용한 신규한 나프틸 아세트아미드에 관한 것이다.
본 발명은 하기 화학식 I의 나프틸 아세트아미드 화합물로 공지되어 있는 화합물의 신규한 용도에 관한 것이다.
상기 식에서, R1, R2, R3및 X는 하기 정의하는 바와 같다. 이들 나프틸 아세트아미드 화합물은 사람의 sPLA2매개 지방산 유리를 억제하는데 효과적이다.
본 발명은 또한 사람 sPLA2의 억제제로서 강하고 선택적인 유효성을 갖는 신규한 종류의 나프틸 아세트아미드 화합물에 관한 것이다.
본 발명은 또한 나프틸 아세트아미드 화합물을 함유하는 제약 조성물에 관한 것이다.
본 발명은 또한, 본 발명의 나프틸 아세트아미드 화합물의 치료 유효량을 투여함에 의한 패혈증 쇼크, 성인 호흡 곤란 증후군, 췌장염, 외상, 기관지 천식, 알러지성 비염, 류마티스성 관절염 및 관련 질병의 예방 및 치료 방법에 관한 것이다.
정의:
본 발명의 나프틸 아세트아미드에서 사용되는 용어들을 다음과 같이 정의한다.
용어 "알킬"은 단독으로 또는 다른 치환체의 일부로서 달리 정의하지 않는 한 메틸, 에틸, n-프로필, 이소프로필, n-부틸, 3급 부틸, 이소부틸, 2급-부틸, n-펜틸 및 n-헥실과 같은 직쇄 또는 분지쇄 일가 탄화수소 라디칼을 의미한다.
용어 "알케닐"은 단독으로 또는 다른 용어와 함께 사용되어 진술된 수의 탄소 원자를 갖는 직쇄 또는 분지쇄 일가 탄화수소기를 의미하고, 비닐, 프로페닐, 크로토닐, 이소펜테닐 및 다양한 부테닐 이성체와 같은 기가 대표적이다.
용어 "알키닐"은 단독으로 또는 다른 용어와 함께 사용되어 진술된 수의 탄소 원자를 갖고 3중 결합을 갖는 직쇄 또는 분지쇄 탄화수소를 의미한다. 이 용어에는 아세틸렌, 프로핀, 다양한 부틴 이성체 등과 같은 기를 포함한다.
용어 "할로"는 플루오로, 클로로, 브로모 또는 요오도를 의미한다.

합성 방법
X가 산소인 본 발명의 화합물을 하기 반응식 I에 따라 제조할 수 있다.
상기 반응식의 제1 단계에서, 적절하게 치환된 1-브로모-4-메틸나프탈렌과 적절하게 치환된 페놀을 피리딘과 같은 비양성자성 극성 용매에 용해시킨다. 이 혼합물을 과량의 탄산칼륨과 과량의 구리-청동으로 처리하고, 질소 블랭킷 하에 환류시켜 화합물 (1)을 제조한다.
화합물 (1)을 촉매로서 2,2-아조비스이소부티로니트릴을 사용하여 사염화탄소와 같은 비극성 알킬 할로겐화물 용매 중에서 N-브로모숙신아미드와 같은 브롬화제와 함께 환류시킴으로써, 화합물 (1)을 화합물 (2)로 브롬화시킨다.
화합물 (2)를 시안화나트륨으로 처리하여 화합물 (3)을 제조한다. 이 반응은 약 60℃의 온도로 가열하면서, 디메틸 술폭시드(DMSO)와 같은 비양성자성 극성 용매 중에서 가장 잘 수행된다.
2단계에 걸쳐 시아노 화합물 (3)을 가수분해시켜 산 화합물 (4)를 제조한다. 공용매로서 디에틸렌 글리콜과 같은 극성 양성자성 용매를 사용하여, 시아노 화합물 (3)을 수산화칼륨과 같은 알칼리 금속 염기로 처리하고, 이 혼합물을 약 90 내지 95℃로 가열한다. 이어서, 생성된 화합물을 염산과 같은 강한 광산과 반응시킨다.
또 다른 2단계 과정으로 화합물 (4)를 원하는 나프틸 아세트아미드 화합물 (5)로 전환시킨다. 먼저, 산 화합물 (4)를 메틸렌 클로라이드와 같은 알킬 할로겐화물 용매에 용해시킨다. 산/알킬 할로겐화물 용액을 빙욕에서 냉각시킨 다음, 촉매로서 디메틸포름아미드(DMF)를 사용하여 옥살릴 클로라이드로 처리하여 산 염화물을 형성시킨다. 이 용액을 실온으로 가온시킨 다음, 실온에서 암모니아 기체로 처리하여 화합물 (5)를 제조한다.
원하는 화합물 (5)를 적합한 유기 용매, 바람직하게는 메틸렌 클로라이드/헥산 중에서 표준 재결정 방법을 이용하여 정제할 수 있다.
X가 메틸렌인 본 발명의 화합물을 하기 반응식 II에 따라 제조할 수 있다.
상기 화합물 (1a)는 그리나드(Grignard) 반응에 의해 제조한다. 적절하게 치환된 페닐 브롬화물을 마그네슘 및 에테르와 반응시켜 그리나드 시약 출발 물질을 제조한다. 이어서, 이 시약을 적절하게 치환된 나프틸 니트릴과 반응시키고, 생성된 화합물을 염산과 같은 수성산을 사용하여 가수분해시켜 벤조일 나프틸 (1a)를 형성한다.
화합물 (1a)를 몰과량의 붕화수소화나트륨과 같은 환원제로 처리하여 환원시킨다. 상기 반응을 트리플루오로아세트산과 같은 용매-촉매를 사용하여 빙욕 중에서 개시시킨 다음, 환원 반응이 진행함에 따라 실온으로 가온시킨다.
화합물 (2a)를 아세트산/인산 혼합물과 같은 극성 산성 용매 중에서 과량의 포름알데히드 및 진한 염산으로 처리하여 클로로메틸화시킨다. 이 반응은 약 90℃의 온도에서 가장 잘 수행된다.
염화물 화합물 (3a)를 시안화물을 사용하여 친핵 치환 반응시켜 니트릴 (4)을 제조한다. 화합물 (3a)를 디메틸포름아미드(DMF)와 같은 비양성자성 극성 용매 중에서 약간 몰과량의 시안화나트륨과 함께 약 5시간 동안 환류시킨 다음, 실온으로 냉각할 때까지 반응을 진행시킴으로써 이 반응을 수행한다.
이어서, 니트릴 (4a)로부터 3단계 공정으로 원하는 나프틸아미드 (5a)를 제조한다. DMSO와 같은 비양성자성 극성 용매 중에 용해시킨 니트릴 (4a)의 용액에, 탄산칼륨을 첨가하여 니트릴 용액을 약간 염기성으로 하였다. 이어서, 니트릴을 과산화수소 수용액으로 처리하여 가수분해시켰다. 과산화물 용액 중에 물을 첨가하여 나프틸 아세트아미드를 결정화시킬 수 있다.

당업계의 숙련인들은 치환된 페놀 및 페닐 브롬화물 출발 물질이 모두 상업적으로 입수가능하거나, 상업적으로 입수가능한 출발 물질로부터 공지 기술에 의해 쉽게 제조될 수 있음을 쉽게 이해할 것이다. 본 발명의 화합물을 제조하기 위해 사용된 모든 다른 반응물과 시약은 상업적으로 입수가능하다.

실시예 1
4-벤질-1-나프틸아세트아미드 (화합물 A)의 제조
A. 1-벤조일나프탈렌의 제조
디에틸 에테르 50 ㎖ 중 브로모벤젠 10.3 g(65 밀리몰)에 마그네슘 부스러기를 첨가하였다. 상기 브로모벤젠 용액 약 5 ㎖에 몇 개의 요오드 결정을 첨가하고 가볍게 가열함으로써 반응을 개시하였다. 이어서, 나머지 브로모벤젠을 완하한 환류를 유지하는 속도로 첨가하였다. 반응이 약해지면, 디에틸 에테르 50 ㎖ 중 1-나프토니트릴을 서서히 첨가하였다. 이 혼합물을 실온에서 밤새 교반하였다. 물을 첨가하고, 진한 염산을 사용하여 혼합물을 pH 1로 산성화시켰다. 메틸렌 클로라이드를 첨가하고, 혼합물을 환류 하에 밤새 가열하였다. 층들을 분리하고, 유기층을 염수로 세척하고, 황산마그네슘 상에서 건조시켰다. 메틸렌 클로라이드/헥산으로부터 하위표제 화합물을 결정화시킨 다음, 에틸 아세테이트:헥산(5:95)으로 용출시키는 실리카겔 상의 중압(medium pressure) 칼럼 크로마토그래피에 의해 더욱 정제하였다. 원하는 생성물을 백색 결정성 고체로서 수득하였다 (4.38 g, 29% 수율).

B. 1-벤질나프탈렌의 제조

C. 1-벤질-2-클로로메틸나프탈렌의 제조

실시예 2
4-페녹시나프트-1-일 아세트산 (화합물 B)의 제조
A. 1-메틸-4-페녹시나프탈렌의 제조
기계 교반기가 장착된 3가지 플라스크에 피리딘 600 ㎖(분자체 상에서 건조시킴), 4-브로모-1-메틸나프탈렌 100 g, 페놀 55.6 g, 탄산칼륨 83.7 g, 염화구리(I) 14.9g 및 구리 청동 2g을 첨가하였다. 질소 대기 하에, 혼합물을 격렬하게 교반하면서 72 시간 동안 환류시켰다. 반응 혼합물을 냉각시키고, 차가운 묽은 염산에 붓고, 생성된 혼합물을 에틸 아세테이트로 추출하였다. 유기층을 차가운 묽은 염산으로 2회 세척하고, 황산나트륨 상에서 건조시키고 여과하였다. 진공 하에 용매를 제거한 후, 생성물을 헥산으로 용출시키면서 실리카겔 상에서 플래시 크로마토그래피시켰다. 생성물을 분별 증류를 통하여 더욱 정제하여, 150℃ 및 0.075 토르에서 35 g (33%)의 원하는 1-메틸-4-페녹시나프탈렌 중간체를 수득하였다.

B. 1-브로모메틸-2-페녹시나프탈렌의 제조

실시예 3
4-페녹시나프트-1-일 아세트아미드 (화합물 C)의 제조
상기 실시예 2에서 제조한 나프틸 아세트산 0.250g를 메틸렌 클로라이드 20 ㎖에 용해시켰다. 용액을 빙욕 중에서 냉각시킨 후, 옥살릴 클로라이드 0.090 ㎖ 및 디메틸포름아미드 수적을 첨가하였다. 30분 후, 빙욕을 제거하고, 반응물을 30분에 걸쳐 실온이 되도록 하였다. 반응 혼합물을 용매를 진공 하에 제거하고, 생성물을 메틸렌 클로라이드 50 ㎖에 재용해시킨 다음, 진공 하에 증발시켜 나프틸아세틸 클로라이드를 오일로서 수득하였다. 이 생성물을 메틸렌 클로라이드 40 ㎖에 재용해시키고, 과량의 암모니아를 실온에서 15분에 걸쳐 버블링시켰다. 밤새 교반한 후, 반응 혼합물의 용매를 진공 하에 제거하였다. 반응 생성물을 에틸 아세테이트에 재용해시키고, 염수 및 묽은 탄산칼륨으로 세척하였다. 유기 용액을 황산나트륨 상에서 건조시키고, 여과하였다. 용매를 진공 하에 제거하고, 생성물을 최소의 메틸렌 클로라이드에 재용해시켰다. 헥산으로 흐려질 때까지 희석하고 냉각시킨 후, 130㎎(55%)의 4-페녹시나프트-1-일-아세트아미드를 수득하였다. 융점 = 119-122℃.

나프틸 아세트아미드 화합물의 치료 용도
본 명세서에 기재된 나프틸 아세트아미드 화합물은 아라키돈산이나 아라키돈산 케스케이드의 아라키돈산 아래의 다른 활성제(예를 들면, 5-리폭시게나제, 시클로옥시게나제 등)에 대한 길항제로서 작용함으로써가 아니라, 주로 사람의 sPLA2을 직접적으로 억제함으로써 유익한 치료 작용을 하는 것으로 믿어진다.
본 발명의 sPLA2매개 지방산 유리의 억제 방법은 sPLA2를 나프틸 아세트아미드 화합물, 그의 염 또는 프로드럭 유도체 치료 유효량과 접촉시키는 것을 포함한다.

본 발명의 제약 제제

본 발명의 화합물은 경구, 에어로졸, 직장, 경피, 피하, 정맥내, 근육내 및 경비 경로를 포함하는 다양한 경로로 투여할 수 있다.
본 발명의 제약 제제는 본 발명의 나프틸 아세트아미드 화합물 치료 유효량을 제약학적으로 허용되는 담체 또는 희석제와 합함으로써(예를 들면, 혼합함으로써) 제조된다. 본 발명의 제약 제제는 잘 공지되고 쉽게 입수가능한 성분을 사용하여 공지된 절차에 의해 제조된다.
본 발명의 조성물을 제조하는데 있어서, 일반적으로 활성 성분을 담체와 혼합하거나, 담체로 희석하거나, 또는 캡슐제, 사세제, 페이퍼제 또는 다른 용기의 형태일 수 있는 담체내에 밀폐할 것이다. 담체가 희석제 역할을 하는 경우, 담체는 비히클로서 작용하는 고체, 반고체 또는 액체 물질일 수 있고, 정제, 환제, 분말제, 로젠지제, 엘릭서제, 현탁액제, 유화액제, 용액제, 시럽제, 에어로졸제 (고체로서 또는 액체 매질 중), 또는 연고제(예를 들면, 10 중량% 이하의 활성 성분을 함유하는) 등의 형태일 수 있다. 본 발명의 화합물은 바람직하게는 투여하기 전에 제형화한다.
본 발명의 제약 제제에 당업계에 공지된 임의의 적합한 담체를 사용할 수 있다. 그러한 제제에서, 담체는 고체, 액체 또는 고체와 액체의 혼합물일 수 있다. 고체 형태의 제제는 분말제, 정제 및 캡슐제를 포함한다. 고체 담체는 또한 향미제, 윤활제, 가용화제, 현탁화제, 결합제, 정제 붕괴제 및 캡슐화 물질로서 작용할 수 있는 1종 이상의 성분일 수 있다.

분말제에서, 담체는 미분된 활성 성분과 혼합물로 존재하는 미분된 고체이다. 정제에서, 활성 성분은 필수적인 결합성을 갖는 담체와 적합한 비율로 혼합되어, 원하는 형태 및 크기로 압축된다. 분말제 및 정제는 바람직하게는 약 1 내지 약 99중량%의 본 발명의 신규한 화합물인 활성 성분을 함유한다. 적합한 고체 담체는 탄산마그네슘, 스테아르산마그네슘, 탈크, 당 락토스, 펙틴, 덱스트린, 전분, 젤라틴, 트라가칸트, 메틸 셀룰로스, 나트륨 카르복시메틸 셀룰로스, 저융점 왁스 및 코코아 버터이다.
멸균 액체 형태 제제는 현탁액제, 유화액제, 시럽제 및 엘리서제를 포함한다.
활성 성분은 멸균수, 멸균 유기 용매 또는 이 둘의 혼합물과 같은 제약학적으로 허용되는 담체 내에 용해되거나 현탁될 수 있다. 활성 성분은 종종 적합한 유기 용매(예를 들면, 수성 프로필렌 글리콜)에 용해될 수 있다. 다른 조성물은 미분된 활성 성분을 전분 또는 나트륨 카르복시메틸 셀룰로스 수용액 또는 적합한 오일 중에 분산시켜 제조할 수 있다.

활성 성분, 전분 및 셀룰로스를 45 메쉬 U.S. 시브를 통해 통과시키고, 충분히 혼합하였다. 폴리비닐피롤리돈을 함유하는 수용액을 생성된 분말과 혼합한 다음, 혼합물을 14 메쉬 U.S. 시브를 통하여 통과시켰다. 이렇게 하여 생성된 과립을 50℃에서 건조시키고, 18 메쉬 U.S. 시브를 통하여 통과시켰다. 이어서, 미리 60 메쉬 U.S. 시브를 통하여 통과시킨 나트륨 카르복시메틸 전분, 스테아르산마그네슘 및 탈크를 과립에 첨가하고, 혼합한 후 타정기에서 압축하여 각각 150 ㎎ 중량의 정제를 수득하였다.

각각 225 ㎎의 활성 성분을 함유하는 좌제를 하기와 같이 제조하였다:

활성 성분을 45 메쉬 U.S. 시브를 통하여 통과시키고, 나트륨 카르복시메틸 셀룰로스 및 시럽과 혼합하여 부드러운 페이스트를 형성하였다. 벤조산 용액, 방향제 및 착색제를 물 일부를 사용하여 희석하고, 교반하면서 첨가하였다. 이어서, 충분한 물을 첨가하여 필요한 부피를 채웠다.

정맥내 제제를 하기와 같이 제조할 수 있다:

분석 실험
크로모겐 분석
하기 크로모겐 분석 절차를 이용하여 재조합 사람 분비 포스포리파제 A2의 억제제를 확인하고 평가하였다. 본 명세서에 기재된 분석 방법은 96 웰 미세적정 평판을 사용하는 고용적 스크리닝을 위해 변경하였다. 이 분석 방법에 대한 일반적인 설명은 문헌["Analysis of Human Synovial Fluid Phospholipase A2on Short Chain Phosphatidylcholine-Mixed Micelles: Development of a Spectrophotometric Assay Suitable for a Microtiterplate Reader", 로라 제이. 레이놀즈 (Laura J. 100 ㎎/㎖의 농도로 클로로포름 중에 제공된 측정된 부피의 라세미 디헵타노일 티오 PC를 취하여 건조시키고, 10 밀리몰 트리톤 X-100TM비이온성 세제 수용액 중에 재용해시킨다. 반응 완충액을 상기 용액에 첨가한 다음, DTNB를 첨가하여 반응 혼합물을 수득하였다.
이렇게 수득한 반응 혼합물은 완충 수용액(pH 7.5) 중에 1mM 디헵타노일 티오-PC 기질, 0.29 밀리몰의 트리톤 X-100TM세제, 및 0.12 밀리몰의 DTMB를 함유한다.
분석 절차
1. 모든 웰에 반응 혼합물 0.2 ㎖을 첨가한다.
2. 적절한 웰에 시험 화합물 (또는 용매 블랭크) 10㎕을 첨가하고, 20초간 혼합한다.
3. 적절한 웰에 sPLA250ng(10 ㎕)을 첨가한다.
4. 평판을 40℃에서 30분 동안 인큐베이션한다.
5. 자동 평판 판독기를 사용하여 405 ㎚에서 웰의 흡광도를 판독한다.
본 발명의 모든 화합물을 3회 시험하였다. 전형적으로, 화합물을 5 ㎍/㎖의 최종 농도로 시험하였다. 화합물은 405 ㎚에서 측정될 때 비억제 대조 반응군에 비해 40% 이상의 억제를 나타낼 때 활성인 것으로 고려한다. 405 ㎚에서 색 전개가 없는 것이 억제의 증거가 된다. 처음에 활성인 것으로 밝혀진 화합물들을 재시험하여 그들의 활성을 확정하고, 충분히 활성인 경우 IC50값을 측정하였다. 전형적으로, 반응물 내의 최종 농도가 45 ㎍/㎖ 내지 0.35 ㎍/㎖ 범위에 있도록, 시험 화합물을 2배 계열 희석하여 IC50값(하기 표 I을 참조하시오)을 측정하였다. 더 강력한 억제제는 상당히 더 많은 희석을 필요로 하였다. 모든 경우에, 비억제 대조 반응군에 상대적인 억제제를 함유하는 효소 반응액에 의해 생성된 405 ㎚에서 측정한 억제(%)를 결정하였다. 각각의 샘플을 3회 적정하고, 결과의 값을 평균하여 플로팅하고 IC50값을 계산하였다. 10 내지 90% 억제율 범위의 억제 값에 대하여 로그 농도를 플로팅하여 IC50을 결정하였다.

Claims (10)
- 하기 화학식 I의 나프틸 아세트아미드 화합물 또는 제약학적으로 허용되는 그의 염, 용매화물 또는 프로드럭 유도체.<화학식 I>상기 식에서,R1및 R2는 각각 독립적으로 수소 또는 비간섭 치환체이며, 단, R1또는 R2중 적어도 하나는 반드시 수소이고;R3은 수소, -O(CH2)nY,,(여기에서, n은 2 내지 4의 수이고, Y는 -CO2H, -PO3H2또는 -SO3H이다)이며;X는 -O- 또는 -CH2-이다.
- 제 1 항에 있어서, R1및 R2가 각각 독립적으로 수소 또는 페닐이고, R3이 수소, -O(CH2)nY (여기에서, n은 2 내지 4이고, Y는 -CO2H, -PO3H2또는 -SO3H이다)인 화학식 I의 화합물.
