Abstract

Description
암은 동물 및 인간의 죽음을 유발하는 원인이다. 암의 정확한 원인은 밝혀지지 않았으나, 흡연 또는 발암 물질에의 노출과 같은 어떤 작용과 어떤 유형의 암 및 종양의 발생사이에 연관관계가 있는 것으로 다수의 연구자에 의해 밝혀졌다.

A. 정의

B. 항암성 조성물

C. 화학요법제

튜블린 상호작용제는 튜블린상에 있는 특정한 부위에, 포말 세포성 미소세관을 중합하는 단백질을 결합시킴으로써 작용한다. 미소세관은 임계적 세포 구조 단위체이다. 상호작용제가 단백질에 결합할 때, 세포는 미소세관을 형성할 수 있는 튜블린-상호작용제는 빈크리스틴 및 빈블라스틴, 이들 모두의 알칼로이드 및 팔크리탁셀을 포함한다.

D. 상승 작용제
상승 작용제 는 약학 조성물의 효능을 향상 또는 증가시키거나 면역계에 작용하는 임의의 물질일 수 있다. 이러한 상승 작용제는 화학요법제 및 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트와 병용되는 트리프롤리딘 및 이의 시스 이성체이다. 트리프롤리딘은 미국 특허 제 5,114,951(1992) 호에 기재되어 있다. 다른 상승 작용제는 프로코다졸, 1H-벤즈이미다졸-2-프로판산; [β-(2-벤즈이미다졸) 프로피온산; 2-(2-카르복시에틸)벤즈이미다졸; 프로파졸이다. 프로코다졸은 바이러스 및 박테리아 감염에 대해 비선택적 활성 면역보호제이며, 여기에서 청구된 조성물과 병용할 수 있다. 암, 종양, 백혈병 및 바이러스 감염의 치료에 있어서, N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트 만을 사용하거나 또는 화학요법제를 병용하는 것이 효과적이다.

E. 약제형

F. 약물 전달 제형

G. 치료 방법
치료 방법은 치료되는 특정한 형태의 바이러스 또는 종양의 치료에 유효한 적당한 임의의 방법일 수 있다. 치료는 경구, 직장, 국소, 비경구, 정맥 투여 또는 종양에 또는 그 부근에 주사등으로 수행할 수 있다. 유효량을 사용하는 방법은 또한 치료되는 종양에 따라 변한다. 적당한 담체, 첨가적인 암 억제용 화합물 또는 작용을 촉진하는 화합물 또는 희석제와 배합된 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트 화합물을 정맥내, 피하, 또는 근육내 투여에 의해 비경구적으로 치료하는 것이 화합물을 온혈 동물에 투여하는 바람직한 방법이 될 것이라고 믿어진다.
바이러스성 감염의 치료 방법은 경구, 직장, 국소, 비경구 또는 정맥내 투여에 의할 수 있다. 활성 시간 및 투여량은 처리되는 바이러스 및 원하는 혈액 수준에 달려있게될 것이다.
하기 실시예는 본 발명을 실시하기 위한 것이나 이에 한정되는 것은 아니다.
결장, 유방 및 폐 종양세포 테스트
하기의 세포 배양 테스트를 수행하여, 인간의 결장, 유방 및 폐 종양세포에 대한 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트 성분의 독성 테스트를 수행 하였다. 세포의 생존 능력은 MTT (3-[4,5-디메틸티아졸-2-일]-2,5-디페닐테트라졸륨 브로마이드) 의 환원 관찰을 통해 테스트하였다. MTT 효력 검정은 세포 생존능력을 측정하는 것으로 잘 알려져 있다.
결장 종양세포 (아메리칸 타입 컬쳐 콜렉션 (ATCC) 으로부터의 HT29) 및 유방 종양세포 (ATCC 로부터의 세포라인 유래 MX1) 을 10 % 소태아 혈청을 함유하는 이글스 최소 필수 배지 (Eagle's Minimal Essential Medium) 에서 배양하였다. 폐 종양세포 (ATCC 세포라인 유래 A549) 를 10 % 소태아 혈청을 함유하는 함스 에프 12 배지 (Ham's F12 Medium) 에서 배양하였다.
종양세포를 이주시켜 원하는 세포 농도로 배양 플라스크내에 접종하였다. 배양 배지를 제거하고, 세포 시트를 인산염 완충 식염수 (PBS) 로 2 회 세척하였다. 플라스크에 접종하기전에 세포를 트립신으로 처리하고 분쇄하였다. 달리 지시된 것이 없으면, 공기내 5±1% 이산화탄소의 습윤된 대기내에서 37±1℃ 로 인큐베이션시켰다. 배양액이 50 - 80 % 유착될때까지 배양액을 인큐베이션시켰다.
플라스크가 2 차 유착될때, 세포들을 2 차 배양하였다. 배지를 플라스크로부터 흡인하고, 세포 박판을 PBS 로 2 회 세척하였다. 다음에, 트립신 용액을 각각의 플라스크에 첨가하여, 세포 박판을 덮어씌었다. 상기 트립신 용액을 30-60 초후에 제거하고, 플라스크를 실온에서 2 내지 6 분간 인큐베이션시켰다. 세포의 90 %가 제거되었을때, 성장배지를 첨가하였다. 분쇄를 통해 세포를 제거하고, 무균 원심분리 튜브로 옮겼다. 현탁액내 세포의 농도를 결정하였으며, 적당하게 희석하여 5000 세포/ml 의 농도를 얻었다. 96-웰 바이오에세이 플레이트의 정해진 웰에서 상기 세포를 2 차 배양하였다 (200 마이크로리터 세포 현탁액/웰). PBS 를 잔존하는 모든 웰에 첨가하여 습도를 유지하였다. 그 다음, 테스트 물품 처리전에 상기 플레이트를 밤새 인큐베이션시켰다.
테스트 물품의 각 투여량은 배양액 4 중 웰을 각 희석액 100 마이크로리터로 처리하는 것을 통해 테스트하였다. 용매 대조군으로 정해진 그러한 웰에는 추가적으로 메탄올 대조구 100 마이크로리터를 공급하였으며 ; 음성 대조군 웰에는 추가적으로 처리 배지 100 마이크로리터를 공급하였다. PBS 를 테스트 물품이나 배지로 처리되지 않은 잔존 웰에 첨가하였다. 그 다음, 상기 플레이트를 약 5 일간 인큐베이션시켰다.
5 일간의 인큐베이션 말미에서, 각각의 투여량 군을 현미경으로 조사하여 독성을 평가하였다. 0.5 mg/ml MTT 희석액을 처리배지에서 제조하고, 상기 희석액을 0.45 마이크로미터 필터로 여과하여 비용해된 결정들을 제거하였다. 배지를 바이오에세이 플레이트의 웰로부터 제거하였다. 그후, 즉시, 상기 여과된 MTT 용액 2000 마이크로리터를 2 개의 비처리된 블랭크 테스트 웰을 제외한 모든 테스트웰에 첨가하였다. 상기 2 개의 블랭크 웰에는 처리 배지 200 마이크로리터를 공급하였다. 상기 플레이트를 약 3 시간 동안 인큐베이터로 다시 옮겼다. 인큐베이션후, MTT 함유 배지를 제거하였다. 과다한 배지를 각각의 웰에 첨가하고, 플레이트를 실온에서 약 2 시간 동안 교반시켰다.
분자 장치 (Menlo Park, CA) VMax 플레이트 판독기로, 각각의 웰에 대해 550 nm(OD550) 에서의 흡광도를 측정하였다.
용매 대조군 웰의 평균 OD550및 각각의 테스트 물품 희석액의 평균 OD550, 및 블랭크 웰 및 양성 대조군 각각의 평균 OD550을 계산하였다. 용매 대조군 웰 및 테스트 물품 웰 각각의 평균치로부터 블랭크 웰의 평균 OD550를 공제하여, 상응하는 평균 OD550를 제공하였다.

메탄올에 투여된 테스트 물품에 대해서, 별개의 반응들을 준비하여 메탄올 자료에 대해 보정하였다.
아드리아마이신을 양성 대조군으로 사용하였다. 모든 경우에 있어서, 이것은, 1 또는 2 번의 기록을 통해, 그 어느 테스트 물질 보다 더 독성이 있었다. 아드리아마이신은 현재 사용되는 것중에서 가장 강력한 제제중의 하나이며 중대한 부작용을 갖는 제제이다. 상당히 효과적인 다른 화학요법제의 피크 플라즈마 농도는 아드리아마이신 농도 보다 10 내지 50 배 더 높을 수 있다. EC-50 은 세포의 절반이 죽은 경우의 농도이다.

기타 작용
화학치료제 및 상승작용제와의 혼합 이외에, N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트는 살균체, 제조체 또는 다른 항바이러스제와 병용될 수 있다. 바람직한 제초제 및 살균제는 카르벤다짐, 플루코나졸, 베노밀, 글리포스페이트 및 프로피코다졸을 포함한다.

Claims (17)
- 하기 화학식 1 의 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트, 및 이들 화합물의 약학적으로 허용가능한 무기 또는 유기산 염을 안전하고 유효한 양 함유하는 바이러스 감염 및 암 또는 종양 치료용 약학 조성물:[화학식 1][식중, n 은 1 내지 3 이며; X 는 산소 및 황으로 이루어진 군에서 선택되며, R 은 수소, 저급 알킬 및 저급 알케닐, 시클로헥실, 페닐 및 탄소수 8 이하의 페닐알킬로 이루어진 군에서 선택된다.]
- 제 1 항에 있어서, 안전하고 유효한 양의 화학요법제를 더 함유하는 약학 조성물.
- 제 1 항 또는 2 항에 있어서, 약학적으로 허용 가능한 담체 및 안전하고 유효한 양의 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트를 함유하는 약학 조성물.
- 제 1 항, 2 항 또는 3 항 중 어느 한 항에 있어서, R 이 탄소수 1 내지 4 의 알킬이고, n 이 1 이며, X 가 O 이고 클로로가 페닐의 3 위치에 있는 약학 조성물.
- 제 2 항, 3 항 또는 4 항 중 어느 한 항에 있어서, 상기 화학요법제가 DNA-상호작용제, 대사길항제, 튜불린-상호작용제, 호르몬제, 아스파라기나아제 또는 히드록시우레아로 이루어진 군에서 선택된 약학 조성물.
- 제 5 항에 있어서, 상기 화학요법제가 아스파라기나아제, 히드록시우레아, 시스플라틴, 시클로포스파미드, 알트레타민, 블레오마이신, 댁티노마이신, 독소루비신, 에토포시드, 테니포시드, 탁솔, 플카미딘, 메토트렉세이트, 플루오로우라실, 플루오로데옥시유리딘, CB3717, 아자시티딘, 시타라빈, 플록스유리딘, 머캅토퓨린, 6-티오구아닌, 플루다라빈, 펜토스타틴, 시크트라빈 및 플루다라빈으로 이루어진 군에서 선택된 약학 조성물.
- 제 1 항, 2 항, 3 항, 4 항, 5 항 또는 6 항중 어느 한 항에 있어서, 상승작용제를 더 함유하는 약학 조성물.
- 약 2 mg/kg 체중 내지 약 400 mg/kg 의 제 1 항, 2 항, 3 항, 4 항, 5 항, 6 항 또는 7 항 중 어느 한항에 따른 약학 조성물을 투여하는 것을 특징으로 하는 온혈 동물의 암 치료 방법.
- 제 8 항에 있어서, 상기 N-클로로페닐카르바메이트을 경구, 장, 정맥, 비경구로 투여하거나 또는 종양에 또는 그 부근에 주사하여 투여하는 방법.
- 안전하고 유효한 담체 및 하기 화학식 1 의 N-클로로페닐카르바메이트 및 N-클로로페닐티오카르바메이트, 이들 화합물의 약학적으로 허용가능한 유기 또는 무기산 염을 함유하는 암, 종양 및 바이러스 감염 치료에 효과적인 단위 투여용 조성물:[화학식 1][식중, X 는 산소 및 황으로 이루어진 군에서 선택되며, n 은 1 내지 3 이고, R 은 수소, 저급 알킬 및 저급 알케닐, 시클로헥실, 탄소수 8 이하의 페닐알킬, 페닐로 이루어진 군에서 선택된다.]
- 제 10 항에 있어서, 상기 카르바메이트가 N-3-클로로페닐카르바메이트이며, 약 2 mg/kg 체중 내지 약 400 mg/kg 의 상기 N-3-클로로페닐카르바메이트가 투여되는 단위 투여용 조성물.
- 제 10 항 또는 11 항에 있어서, 상기 약학적으로 허용가능한 산 부가염이 히드로클로라이드, 포스페이트, 니트레이트, 아세테이트 및 살리실레이트로 이루어진 군에서 선택된 단위 투여용 조성물.
- 제 12 항에 있어서, 안전하고 유효한 양의 화학요법제를 더 함유하는 단위 투여용 조성물.
- 제 10 항, 11 항, 12 항 또는 13 항 중 어느 한 항에 있어서, 상기 화학요법제가 DNA-상호작용제, 대사길항제, 튜불린-상호작용제, 호르몬제, 아스파라기나아제 또는 히드록시우레아로 이루어진 군에서 선택된 단위 투여용 조성물.
- 제 14 항에 있어서, 상기 화학요법제가 아스파라기나아제, 히드록시우레아, 시스플라틴, 시클로포스파미드, 알트레타민, 블레오마이신, 댁티노마이신, 독소루비신, 에토포시드, 테니포시드, 탁솔, 플카미딘, 메토트렉세이트, 플루오로우라실, 플루오로데옥시유리딘, CB3717, 아자시티딘, 시타라빈, 플록스유리딘, 머캅토퓨린, 6-티오구아닌, 플루다라빈, 펜토스타틴, 시크트라빈 및 플루다라빈으로 이루어진 군에서 선택된 약학 조성물.
- 제 15 항 에 있어서, 약 2 mg/kg 체중 내지 약 400 mg/kg 의 상기 N-클로로페닐카르바메이트가 투여되는 단위 투여용 조성물.
- 제 11 항, 12 항, 13 항, 14 항 또는 15 항중 어느 한 항에 있어서, 안전하고 유효한 양의 상승작용제를 더 함유하는 단위 투여용 조성물.
