Description
본 분야에서 매우 상당한 정도의 진전이 이루어졌음에도 불구하고, 암 치료를 위해 현재 사용되고 있는 방법은 여전히 제한된 효율을 가진다. 방사선요법 및 화학요법은 확실히 암의 발생에 매우 바람직한 영향을 미친다. 그러나, 암의 치료에서 첨예한 문제점은 특정의 1차 종양의 무감각 및/또는 효과적 치료의 제 1 사이클 후에, 방사선- 및 화학요법에 내성을 띠는 종양 세포의 출현이다.
이러한 현상의 근원일 수 있는 분자 메카니즘을 밝히기 위한 다양한 연구가 시도되었다. 일반적으로, 연구는 화학요법제가 세포로 들어가는 방식 및 이들이 세포 표적과 반응하는 방식쪽으로 지향되어왔다[참조문헌: Chin et al., Adv. ancer Res. 60 (1993) 157-180; Chabner and Meyers in Cancer/Principles and practices of Oncology, De Vita et al., Eds., J. 본 출원인은 과대증식성 세포의 파괴에 특히 효과적인 치료의 신규한 방법을 개발하였다. 전술한 바와 같이, 본 발명에 따른 치료 방법은 본질적으로 2가지 유형의 치료제: 즉, 종양원성 세포 시그널 경로를 차단하는 유전자 및 화학요법제 및/또는 방사선요법제의 병용에 기초한다. 사실, 본 발명은 이러한 2가지 유형의 치료제의 병용과 관련된 특히 큰 효과적 상승 효과 입증의 산물이다.
본 발명은 과대증식성 병리 치료 분야에 관한 것이다. 특히 2가지 유형의 치료제의 병용에 기초한 과대증식성 병리 치료의 새로운 방법에 관한 것이다.
더욱 구체적으로 말해서, 본 발명은 종양원성 세포 시그널링 경로를 차단하는 유전자 및 화학요법제 및 또는 방사선요법제의 조합된 사용에 기초한 과대증식성 병리 치료의 신규한 방법에 관한 것이다. 본 발명에 따른 조합된 치료는 비교적 적은 용량으로 특히 과대증식 세포의 파괴에 유효한 효과가 있다. 따라서, 본 발명은 제한된 부작용으로 과대증식성 병리(암, 재협착증(restenosis) 등)의 치료의 특히 효과적인 신규한 방법을 제공한다.
따라서 본 발명의 제 1 목적은 과대증식성 병리의 치료에 동시에 또는 따로따로 사용하거나 시간 경과에 따라 확산시키기 위한, 적어도 부분적으로 종양원성 세포 시그널 경로를 억제하는 하나 이상의 핵산 및 항암치료제의 의약 배합물에 관한 것이다.
전술한 바와 같이, 본 발명은 본질적으로 특정 유전자의 산물과 항암 치료제 간의 상승 효과의 입증에 근거한다. 이러한 조합된 사용은 저용량의 약제로도 더 강력한 효과를 발휘한다. 따라서, 본 발명은 과대증식성 병리의 치료를 위한 특히 유리한 수단을 제공한다.

L. 본 발명의 제 1 바람직한 양태에 따라, 핵산은 적어도 부분적으로 종양원성 세포 시그널링 경로를 억제하는 폴리펩타이드 또는 단백질을 암호화하는 DNA또는 RNA이다. 더욱 특히, 폴리펩타이드 또는 단백질은 우세한 네가티브, ScFv 및 종양 억제제로부터 선택된다.
더욱 바람직하게는, 우세한 네가티브는 GAP 단백질, Gbr3-3 단백질 또는 Ets 단백질의 돌연변이체의 N-말단 영역의 구성 성분이다. ScFv의 경우, 이는 바람직하게는 돌연변이된 ras 단백질 또는 GAP 요소에 대한 ScFv이다. 종양 억제제 단백질은 유리하게는 p53, Rb, waf1, p21, DCC 또는 MTS이다.

안티센스 핵산의 용도
안티센스 핵산에 의한 표적 유전자의 발현의 조절은 증가된 발달을 통한 치료적 접근으로 구성된다. 이러한 접근은 다른 핵산의 상보 영역과 특이적으로 하이브리드화하고, 이에 따라 특정 유전자의 발현을 특이적으로 억제하는 핵산의 능력을 토대로 한다. 이러한 억제는 해독 수준 또는 전사 수준에서 발생할 수 있다.

리간드 RNA의 용도

우세한 네가티브
우세한 네가티브는 종양유전자 시그널링 경로의 폴리펩타이드 길항제이다. 이러한 길항작용은 폴리펩타이드가 종양발생 시그널링의 주요 요소와 접촉하여 위치하고 이 시그널링을 위해 세포내에 자연히 사용된 폴리펩타이드와 경쟁에 들어갈 때 발생한다. 사용된 폴리펩타이드 길항작용은 매우 자주 천연 폴리펩타이드의 모의품이지만 종양발생 시그널이 전파되도록 하는 영역이 결여되어 있다.
본 발명의 수행에 바람직한 우세한 네가티브 중에서, GAP 단백질의 NH2-말단 영역, Grb3-3 단백질 또는 ETS 단백질의 돌연변이된 형태를 암호화 하는 핵산이 언급될 수 있다.
특허 출원 WO94/03597에서 GAP-Ras 단백질의 NH2-말단 영역의 과발현이 변형된 세포의 종양형성성을 특이적으로 차단하고 이에 따라 돌연변이된 ras 유전자의 발현을 차단할 수 있다는 것을 나타냈다. 본 출원의 실시예 1은 GAP(170-702) 영역의 과발현이 사람 세포의 아포프토시스, 소위 폐의 비-소형 세포 암종(H460)을 유도하는 것을 나타낸다. 추가로, 실시예 2는 GAP(170-702) 작제물에 의해 유도된 아포프토시스 효과가 세포 생존능에 영향을 주지 않는 농도에서 사람 종양 세포의 배양 배지에 시스플라틴, 캠프토테신 또는 택소테르 같은 생성물의 추가에 의해 매우 증가됨을 보이고 있다.
또한, 본 출원의 실시예 1은 H460 세포내 grb3-3 유전자의 활성을 설명한다. Grb3-3의 서열 및 가정된 기능이 Science 1994에 기재되어 있다. 실시예 2는 또한 Grb3-3 유전자의 전달에 의해 유도된 아포프토시스 효과가 세포 생존능에 영향을 주지 않는 농도에서 사람 종양 세포의 배양 배지에 시스플라틴, 캠프토테신 또는 택소테르 같은 생성물을 첨가함으로써 현격히 증가된다는 것을 나타낸다.
이러한 예는 상이한 화학요법제가 유전자 전달에 의해 아포프토시스의 유도를 위한 전략과 효과적으로 조합할 수 있는 것을 분명하게 나타낸다.
ScFv
ScFv는 항체에 필적할만한 결합 성질을 지닌 세포내 활성 분자이다. 특히, ScFv는 항체의 경쇄 가변성 영역의 결합 부위에 상응하는 펩타이드로 구성된 분자로서 펩타이드 링커에 의해 항체의 중쇄 가변 영역의 결합 부위에 상응하는 펩타이드에 연결된다. 본 출원인에 의해 이러한 ScFv가 유전자 전달에 의해 생체내에서 생성될 수 있음이 나타났다(출원 WO94/29446 참조).
더욱 특히, 본 출원인은 상이한 세포 구획에서 ScFv를 발현시킴으로서 종양원성 단백질을 중화하는 것이 가능하다는 것을 나타낸다. 본 발명의 양태에 따라서, ras 단백질의 변형력을 중화시키는 ScFv의 세포내 생성을 허용하는 핵산이 화학요법제와 함께 사용된다. 이러한 조합은 상당한 상승 효과를 생성한다(실시예 2 참조).
종양 억제제
본 발명의 맥락에서 사용될 수 있는 종양 억제제 유전자중에서, p53, p21, Rb, rap1A, DDC, WAF 및 MTS 유전자는 더 특별하게 언급될 수 있다. 특히, p53, Rb 및 Waf 유전자가 사용된다.

Rb 유전자는 세포를 휴지 상태로 들어가게 함으로써 세포 분열을 억제하는 기능을 가진 약 927 아미노산의 핵 인단백질의 합성을 결정한다[참조문헌: Friend et al., Nature 323 (1986) 643]. Rb 유전자의 불활성화된 형태는 다양한 종양, 특히 망막아종 또는 골육종 같은 간엽암에 관련된다. 불활성화된 종양 세포내로 이러한 유전자의 재도입은 정상 상태로 되돌리고 종양형성성의 손실을 생성한다[참조문헌: Huang et al., Science 242 (1988) 1563]. 최근에 와서, 돌연변이된 형태가 아닌, 정상 Rb 단백질이 세포 증식을 위해 필수적인 유전자, c-fos proto-종양유전자의 발현을 억제하는 것으로 나타났다.

실시예 3은 탁솔 유도체와 p53 유전자 간의 조합의 효과적인 형태를 나타낸다. 탁솔은 배양액에서 다양한 종양 세포주에서 아포프토시스를 유도한다(Proceeding of American Association for cancer Research Vol. 35, march 1994, Bhalla et al., p306 Seiter et al., p314, Saunder et al., p317). p53은 다양한 세포 형태에서 아포프토시스를 유발한다. 현재 본 출원인은 탁솔 유도체와 p53의 조합체가 사람 종양 세포의 아포프토시스를 유도한다는 것을 보일 수 있게 되었다. 뚜렷하게, 특히 p53의 효과에 내성인 H460 세포의 클론이 탁소테르의 증가하는 용량의 존재하에서 배양된다. 실시예 3은 야생형 p53을 발현하지 않는 세포에 완전히 비효과적인 농도에서 탁소테르로 처리한 뒤 세포가 치사됨을 명백히 보여준다.
Waf 1[야생형 p53 활성화 단편 Cell 75, 817, 1993], 또는 p21[참조문헌: Nature 366,701, 1993]은 야생형 p53의 과발현에 의해 유도된다. Waf 1은 야생형 p53의 과발현에 이어서 G1 기 또는 아포프토시스에서 정지한 세포에서는 출현하지만, p53-독립 방식으로 G1 또는 아포프토시스에서 정지한 세포에서는 나타나지 않는다[참조문헌: Cancer Res, 54, 1169, 1994]. Waf 1이 야생형 p53과 같이 효과적으로 종양 세포의 성장을 감소시킨다. 항암 치료제
본 발명에 따른 조합된 치료에 사용될 수 있는 항암 치료제는 당해 기술 분야의 숙련인에게 공지된 모든 화학요법제 또는 방사선요법제로부터 선택될 수 있다. 특히, 가능한 약제는 시스플라틴, 탁소이드, 에토포사이드, TNF, 아드리아마이신, 캠프토테신, 유사분열 스핀들 포이즌 등이다. 이러한 다양한 약제는 시판되고 있다.

이러한 상이한 화합물은 예를 들면, 참고문헌으로 본 출원에서 인용된 출원 WO94/13654 및 WO92/09589에 기재된 방법에 따라 수득될 수 있다.
본 발명의 목적을 위해 탁솔, 도세탁솔 또는 파크리탁셀을 사용하는 것이 특히 유리하다.
핵산 투여용 벡터
핵산은 그 자체로 치료될 부위에 주사되거나, 파괴되거나 치료될 세포와 직접 배양시킬 수 있다. 사실, 네이키드 핵산이 특수 벡터없이 세포에 들어갈 수 있음이 보고되어 있다. 그러나, 본 발명의 맥락에서 (ⅰ) 세포 침투 효율, (ⅱ) 표적화 및 (ⅲ) 여분- 및 세포내 안정성이 향상되도록할 수 있는 투여 백터를 사용하는 것이 더 바람직하다.
상이한 유형의 벡터가 사용될 수 있다. 벡터는 바이러스성 또는 비-바이러스성일 수 있다.
바이러스 벡터
바이러스 벡터의 사용은 바이러스 형질감염의 천성에 기초한다. 따라서 아데노바이러스, 헤르페스바이러스, 레트로바이러스 및 더 최근에는 아데노-수반 바이러스를 사용하는 것이 가능하다. 이러한 벡터는 형질감염의 관점에서 볼 때 특히 효과적인 것으로 입증되었다.
더욱 특히 아데노바이러스의 경우, 다소 변화하는 상이한 혈청형, 다양한 구조 및 성질이 특징지워졌다. 이러한 혈청형 중, 본 발명의 맥락에서 사람 아데노바이러스 형 2 또는 5 (Ad 2 또는 Ad 5) 또는 동물 기원의 아데노바이러스를 사용하는 것이 바람직하다(출원 WO94/26914 참조). 본 발명의 맥락에서 사용될 수 있는 동물 기원의 아데노바이러스 중 개, 소, 쥐(예를 들면: Mavl, Beard et al., Virology 75 (1990) 81), 양, 돼지, 조류 또는 원숭이(예를 들면: SAV) 기원의 아데노바이러스가 언급될 수 있다. 바람직하게는, 동물 기원의 아데노바이러스는 개의 아데노바이러스이고, 더 바람직하게는 CAV2 아데노바이러스[예를 들면, 맨하탄 균주 또는 A26/61 (ATCC VR-800)]이다. 본 발명의 맥락에서 사람 또는 개 또는 혼합 기원의 아데노바이러스를 사용하는 것이 바람직하다.
바람직하게는, 본 발명의 결손 아데노바이러스는 ITR, 캡시드화를 허용하는 서열 및 해당 핵산으로 구성된다. 더 바람직하게는, 본 발명의 아데노바이러스의 게놈에서 적어도 E1 영역이 비-기능성이다. 본 바이러스 유전자는 당해 기술의 숙련인에게 공지된 여타의 기술, 특히 본 유전자에서 하나 이상의 염기의 제거, 치환, 부분 결실 또는 첨가에 의해 비-기능성이 될 수 있다. 이러한 변형은 시험관내(분리된 DNA 상) 또는 현장에서 예를 들면 유전 공학 기술 또는 돌연변이유발제에 의한 처리에 의해 수득될 수 있다. 다른 영역 또한 변형될 수 있다(E2, E3, E4, L1-L5 등).
본 발명에 따른 결손 재조합 아데노바이러스는 당해 기술의 숙련인에게 공지된 여타의 기술에 의해 제조될 수 있다[참조문헌: Lavrero et al., Gene 101 (1991) 195, EP 185 573; Graham, EMBO J. 3 (1984) 2917]. 특히, 결손 재조합 바이러스는 아데노바이러스, 및 특히 해당 DNA 서열을 운반하는 플라스미드 간의 상동 재조합에 의해 제조될 수 있다. 상동 재조합은 아데노바이러스 및 플라스미드를 적당한 세포주로 동시형질감염 시킨 후 발생한다. 사용된 세포주는 바람직하게는 (ⅰ) 전술한 요소에 의해 형질전환될 수 있고 (ⅱ) 결손 아데노바이러스의 게놈의 일부를 보완할 수 있는 서열을 바람직하게는 재조합의 위험을 피하기 위해 통합된 형태로 함유하여야 한다. 세포주의 예로서, 특히 자체의 게놈에 통합된 채로 Ad5 아데노바이러스의 게놈의 좌측 부분(12%)을 함유하는 사람 배의 신장 세포주 293[참조문헌: Graham et al., J. Gen. Virol. 36 (1977) 59]이 언급될 수 있다. 아데노바이러스로부터 유도된 벡터의 작제 전략은 또한 출원 제 WO 94/26914 및 FR 93 08596에 기재되어 있다.
이렇게 한 후, 증식시킨 아데노바이러스는 실시예에 나타낸 바와 같이, 분자 생물학의 표준 기술에 따라 회수되고 정제된다.
아데노-수반 바이러스(AAV)는 그 자체만으로는 안정하게 통합되고 세포의 게놈에서 부위-특이성 방법으로 비교적 작은-크기의 DNA 바이러스이다. 아데노-수반 바이러스는 세포 성장, 형태 또는 분화에 영향을 유도함없이 광범위한 세포를 감염시킬 수 있다. 또한, AAV는 사람의 병리에 연루된 것으로 보이지는 않는다. AAV 게놈은 클로닝되고, 서열분석되고 특성화되었다. AAV는 약 4700개의 염기를 포함하고, 각 말단에 바이러스를 위한 복제의 기원으로서 작용하는 약 145개의 염기로 이루어진 역 반복 영역(ITR)을 함유한다. 게놈의 나머지는 캡시드화 기능을 하는 2개의 필수 영역으로 나뉜다: 즉, 바이러스의 복제 및 바이러스 유전자의 발현에 관련된 rep 유전자를 함유하는 게놈의 좌측 부분; 바이러스의 캡시드 단백질을 암호화하는 cap 유전자를 함유하는 게놈의 우측 부분.
시험관내 및 생체내에서 유전자의 전달을 위해 AAV로부터 유도된 벡터의 사용이 문헌에 기재되어 있다[참조문헌: 특히 WO 91/18088; WO 93/09239; US 4,797,368, US 5,139,941, EP 488 528]. 이러한 출원은 rep 및/또는 cap 유전자가 결실되고 해당 유전자에 의해 치환된 AAV로부터 유도된 상이한 작제물 및 시험관내에서(배양액내 세포) 또는 생체내(신체내로 직접)에서 해당 유전자를 전달하기 위한 이들의 용도를 기술하고 있다. 본 발명에 따른, 결손 재조합 AAV는 AAV의 두 역 반복 영역(ITR)에 의해 플랭킹된 해당 핵산 서열을 함유한 플라스미드, 및 AAV의 캡시드화 유전자(rep 및 cap 유전자)를 운반하는 플라스미드를 사람 헬퍼 바이러스(예를 들면, 아데노바이러스)로 감염된 세포주 중으로 동시형질감염시킴으로써 제조될 수 있다. 생성된 재조합 AAV는 표준 기술에 의해 정제된다.
헤르페스바이러스 및 레트로바이러스에 관하여, 재조합 벡터의 작제 방법은 문헌에 충분히 기재되어 있다[참조: Breakfield et al., New Biologist 3 (1991) 203; EP 453242, EP 178220, Bernstein et al., Genet. Eng. 7 (1985) 235; McCormick, BioTechnology 3 (1985) 689 등]. 특히, 레트로바이러스는 분열 세포를 선택적으로 감염시켜 통합성 바이러스이다. 레트로바이러스는 암 적용을 위해 해당 벡터를 구성한다. 레트로바이러스 게놈은 본질적으로 2개의 ITR, 캡시드화 서열 및 3개의 암호화 영역(gag, pol 및 env)을 포함한다. 레트로바이러스로부터 유도된 재조합 벡터에 있어서, gag, pol 및 env 유전자는 일반적으로 전체 또는 부분적으로 결실되고, 해당 이종 핵산 서열에 의해 치환된다. 이러한 벡터는 예를 들면, MoMuLV(Moloney 쥐 백혈병 바이러스; MoMLV로도 불림), MSV(Moloney 쥐 육종 바이러스), HaSV(Harvey 육종 바이러스), SNV(비장 괴사 바이러스), RSV(Rous 육종 바이러스) 또는 Friend 바이러스 같은 레트로바이러스의 상이한 유형으로부터 생성될 수 있다.
해당 서열을 함유한 재조합 레트로바이러스를 작제하기 위해, 특히 LTR, 캡시드화 서열 및 해당 서열을 함유하는 플라스미드가 일반적으로 작제되고, 이어서 플라스미드에는 결핍되어 있는 레트로바이러스 기능을 트랜스로 제공할 수 있는 소위 캡시드화 세포주를 형질감염시키는데 사용한다. 일반적으로, 캡시드화 세포주는 gag, pol 및 env 유전자 발현할 수 있게 한다. 이러한 캡시드화 세포주는 특히 세포주 PA317( US4,861,719), 세포주 PsiCRIP( WO90/02806) 및 세포주 GP+envAm-12(WO89/07150)가 선행기술에 기재되어 있다. 또한, 재조합 레트로바이러스는 전사 활성 및 gag 유전자의 부분을 함유한 연장된 캡시드화 서열을 제거하기 위해 LTR에서 변형을 함유할 수 있다[참조문헌: Bender et al., J. Virol. 61 (1987) 1639]. 이어서, 생성된 재조합 레트로바이러스를 표준 기술에 의해 정제한다.
본 발명을 수행하기 위해, 결손 재조합 아데노바이러스 또는 레트로바이러스를 사용하는 것이 가장 유리하다. 사실, 이러한 벡터는 유전자의 종양 세포로의 전달을 위한 특히 유리한 성질을 지니고 있다.
비-바이러스성 벡터
본 발명에 따른 벡터는 또한 핵산의 진핵 세포로의 전달 및 이의 발현을 증진시킬 수 있는 비-바이러스성 제제일 수 있다. 화학적 또는 생화학적 벡터는 특히 편이성과 안정성 및 형질감염시킬 DNA의 크기와 관련한 이론적 제한이 없다는 이유로 천연 바이러스의 유리한 대안체이다.
이러한 합성 벡터는 2가지 주요 기능, 즉 형질감염시킬 핵산을 콤팩트하게 하는 기능 및 세포에 대한 결합을 증진 및 원형질 막과 경우에 따라 두 핵막을 통한 통과 촉진 기능을 보유한다. 핵산의 다중음이온성 성질을 보상하기 위해, 비-바이러스성 벡터 모두 다중양이온 전하를 가진다.
개발된 합성 벡터 중 폴리라이신의 양이온성 중합체, (LKLK)n, (LKKL)n, 폴리에틸렌이민과 DEAE-덱스트란 형, 또는 양이온성 지질 또는 리포펙턴트가 가장 유리하다. 이들은 DNA 압축 및 세포 막과의 관련을 증진시키는 성질을 가지고 있다. 후자의 화합물 중, 리포폴리아민(리포펙타민, 트랜스펙탐등) 및 다양한 양이온 또는 중성 지질(DOTMA, DOGS, DOPE 등)이 언급될 수 있다. 더 최근에는, 수용체-매개, 표적화된 형질감염의 개념이 개발되었고, 이러한 개념은 이식하고자 하는 세포 유형의 표면에 존재하는 막 수용체에 대한 양이온성 중합체 및 리간드 사이의 화학적 커플링의 결과로서 복합체의 막과의 결합을 지시하면서 양이온성 중합체에 의한 DNA 압축 원리를 잘 설명한다. 트랜스페린 또는 인슐린 수용체 또는 간세포의 아시알로글리코단백질 수용체의 표적화가 설명되었다.
투여 방법
본 발명에 따른 바람직한 투여 방법은 제일 먼저 핵산 이어서 치료제로 구성된다. 우선적 사용에서, 트랜스진의 투여는 분열 세포의 최대 수에서 최대 발현을 수득하기 위해 반복되고(예를 들면 5일 연속), 이어서 화학요법제가 투여된다. 유리하게는, 핵산(들)은 병변의 다수의 부위로 직접 종양내 주입에 의하여 병변과 접촉하여 투여되거나, 이러한 유형의 시술에 적당한 쿠숀에 의해 아테롬성 병변과 접촉하여 투여된다. 화학요법제는 임상 프로토콜에 따라 강제로 투여된다.
실시예 1
10%의 태 송아지 형청을 함유한 RPMI 1640 배지에서 배양한 H460 세포를 GAP[170-702] 영역 또는 Grb3-3 단백질을 암호화하는 cDNA와 양성 형질감염된 세포에 제네티신에 대한 내성을 부여하는 유전자(Neo)와 함께 형질감염시킨다. 플라스미드에 위치하고 발현이 바이러스 프로모터(pSV2-GAP[170-702], pSV2-Grb3-3 및 pSV2-Neo)의 조절하에 있는 이러한 cDNA를 형질감염제로서 리포펙타민을 사용하여 H460 세포로 도입한다. H460/NeoR세포(배양 배지에서 400 ㎍/㎖의 제네티신의 존재하에 내성)를 형질감염 15 내지 20일 후 선별하고 정량한다. 상이한 형질감염 조건하에서 NeoR콜로니 수의 정량을 대표하는 실험의 결과를 도 1에 요약하였다.(pSV2-Oli : 대상 임의의 cDNA를 보유하지 않고 따라서 제네티신에 의한 선별 효율 모니터링을 허용하는 대조군 플라스미드)
실시예 2
실시예 1에서 설명된 바와 같이 형질감염된 H460 세포는 제네티신에 의한 선별도 중 탁소테르, 시스플라틴 또는 캠프토테신의 상이한 농도에서 수일 동안처리된다. 화학요법제에 대해 내성인 H460/NeoR세포를 실시예 1에서 설명된 바와 같이 정량한다. pSV2-Neo(●), pSV2-Neo + pSV2-GAP[170-702](▲), 또는 pSV2-Neo + pSV2-Grb3-3(▼)으로 형질감염된 H460 세포의 탁소테르(A), 시스플라틴(B) 또는 캠프토테신(C)에 대한 감수성을 화학요법제의 처리없이 동일하게 형질감염된 세포에 대해 묘사하였다. 언급된 상이한 처리 후 콜로니 수 정량의 대표적인 실험 결과를 도 2에 요약하였다.
실시예 3
H460 세포를 CMV 프로모터의 제어하에서 플라스미드 pcDNA3에 위치한 야생형 p53 단백질(p53WT)를 암호화하는 cDNA로 형질감염시킨다. 플라스미드 pcDNA3은 또한 SV40 프로모터의 제어하에 놓인 Neo 유전자를 함유한다. pcDNA3 및 pcDNA3-p53WT로 형질감염된 세포를 실시예 1에서 기재된 바와 같이 선별하고 분리한다. 제네티신에 내성인 pcDNA3-p53WT로 형질감염된 세포에서 p53의 존재는 특이적 항체를 사용한 웨스턴 블로팅에 의해 증명된다. 도 3에는 pcDNA3 또는 pcDNA3-p53WT형질감염후 수득된 콜로니의 수(A)와 탁소테르로 처리하여 분리된 클론의 감수성(B)으로 묘사된 대표적 실시예의 결과가 요약되어 있다.
Claims (22)
- 과대증식성 병리의 치료를 위해 동시에, 별도로 사용하거나 시간 경과에 따라 확산시키기 위한 종양원성 세포 시그널링 경로를 적어도 부분적으로 억제하는 하나 이상의 핵산 및 항암치료제의 의약 배합물.
- 제 1 항에 있어서, 핵산이 종양원성 세포 시그널링 경로를 적어도 부분적으로 억제하는 생성물을 암호화하는 데옥시리보핵산(DNA) 또는 리보핵산(RNA) 임을 특징으로 하는 배합물.
- 제 2 항에 있어서, 핵산이 안티센스 RNA를 암호화하는 DNA임을 특징으로 하는 배합물.
- 제 2 항에 있어서, 핵산이 리간드 RNA를 암호화하는 DNA임을 특징으로 하는 배합물.
