Claims (47)
- 에피토프-택을 갖는(epitope-tagged) TATA-박스 결합 단백질(TBP)을 발현시킬 수 있는 능력을 갖는, 인간이 아닌 형질전이된(transgenic) 동물.
- 제 1 항에 있어서, TBP가 2개의 에피토프-택과의 융합 단백질로서 발현되는 인간이 아닌 형질전이된 동물.
- 제 1 항 또는 제 2 항에 있어서, 융합 단백질이 인간 TBP(hTBP)를 포함하는 인간이 아닌 형질전이된 동물.
- 제 1 항 내지 제 3 항중 어느 한 항에 있어서, 융합 단백질이 HA(헤마글루티닌)- 및 His-에피토프를 포함하는 인간이 아닌 형질전이된 동물.
- 제 4 항에 있어서, 융합 단백질이 서열 식별 번호 16의 서열을 갖는 인간이 아닌 형질전이된 동물.
- 제 1 항 내지 제 5 항중 어느 한 항에 있어서, 형질전이된 동물이 마우스인 인간이 아닌 형질전이된 동물.
- 인간이 아닌 동물의 배선(germline) 및/또는 체세포에 전이유전자(transgene)를 도입시킴을 포함하는 제 1 항 내지 제 6 항중 어느 한 항의 인간이 아닌 형질전이된 동물의 제조 방법.
- 제 7 항에 있어서, 형질전이된 DNA를 인간이 아닌 동물의 접합자에 미세주입함을 포함하는 인간이 아닌 형질전이된 동물의 제조 방법.
- 제 7 항에 있어서, 전이유전자를 함유하는 벡터로 포배를 형질감염시킴을 포함하는 인간이 아닌 형질전이된 동물의 제조 방법.
- 제 7 항에 있어서, 전이유전자를 배아간 세포에 도입시킴을 포함하는 인간이 아닌 형질전이된 동물의 제조 방법.
- 제 1 항 내지 제 6 항중 어느 한항의 인간이 아닌 형질전이된 동물을 제조하는데 사용될 수 있는 에피토프-택을 갖는 TBP를 코딩하는 전이유전자.
- 제 11 항에 있어서, 제 1 DNA 서열이 TBP를 코딩하고 제 2 DNA 서열이 하나이상의 에피토프-택을 코딩하는 전이유전자.
- 제 11 항 또는 제 12 항에 있어서, TBP를 코딩하는 DNA 서열이 cDNA인 전이유전자.
- 제 13 항에 있어서, DNA 서열이 인간 TBP(hTBP)의 cDNA인 전이유전자.
- 제 11 항 내지 제 14 항중 어느 한 항에 있어서, 제 2 DNA 서열이 2개의 에피토프-택을 코딩하는 전이유전자.
- 제 15 항에 있어서, 에피토프-택이 HA-에피토프 및 His-에피토프인 전이유전자.
- 제 15 항 또는 제 16 항에 있어서, HA-에피토프가 서열 식별 번호 1, 서열 식별 번호 2, 서열 식별 번호 3 또는 서열 식별 번호 4의 서열중 하나를 갖는 전이유전자.
- 제 11 항 내지 제 17 항중 어느 한 항에 있어서, 서열 식별 번호 13의 서열을 포함하는 전이유전자.
- 제 11 항 내지 제 18 항중 어느 한 항에 있어서, 유도성 또는 구성 프로모터의 DNA를 포함하는 전이유전자.
- 제 19 항에 있어서, 프로모터가 EF 유전자의 프로모터인 전이유전자.
- 제 19 항에 있어서, 프로모터가 MT 유전자의 프로모터인 전이유전자.
- 제 11 항 내지 제 21 항중 어느 한 항에 있어서, 서열 식별 번호 14 또는 서열 식별 번호 15의 서열을 갖는 전이유전자.
- 인간이 아닌 형질전이된 동물을 제조하기위한 제 11 항 내지 제 22 항중 어느 한 항의 전이유전자의 용도.
- 하나이상의 에피토프-택을 코딩하는 DNA 서열(들)을 TBP 단백질을 코딩하는 DNA 서열에 연결시킴을 포함하는 제 11 항 내지 제 22 항중 어느 한 항의 전이유전자의 제조 방법.
- 에피토프-택을 갖는 TBP를 제조하기위한 제 11 항 내지 제 22 항중 어느 한 항의 전이유전자의 용도.
- 에피토프-택을 갖는 TBP를 제조하기위한 제 1 항 내지 제 6 항중 어느 한 항의 형질전이된 동물의 용도.
- 제 1 항 내지 제 6 항중 어느 한 항의 형질전이된 동물에서 발현되는 에피토프-택을 갖는 TBP.
- 제 27 항에 있어서, 2개의 에피토프를 포함하는 에피토프-택을 갖는 TBP.
- 제 27 항 또는 제 28 항에 있어서, TBP가 hTBP인 에피토프-택을 갖는 TBP.
- 제 27 항 내지 제 29 항중 어느 한 항에 있어서, TBP가 HA-에피토프 및 His-에피토프에 연결된 에피토프-택을 갖는 TBP.

- 제 11 항 내지 제 22 항중 어느 한항의 전이유전자를 동물의 배선 및/또는 체세포에 도입시킴을 포함하는 제 27 항 내지 제 32 항중 어느 한 항의 에피토프-택을 갖는 TBP의 제조 방법.
- 제 33 항에 있어서, 구성 프로모터를 포함하는 전이유전자를 인간이 아닌 동물에게 도입시키고, 이때 에피토프-택을 갖는 TBP를 동물의 특정한 세포형 및/또는 조직으로부터, 선택적으로 동물의 특정한 발육 단계에서 발현시키는, 에피토프-택을 갖는 TBP의 제조 방법.
- 제 33 항에 있어서, 유도성 프로모터를 포함하는 전이유전자를 인간이 아닌 동물에 도입시키고, 프로모터 유도시 에피토프-택을 갖는 TBP를 발현시키는, 에피토프-택을 갖는 TBP의 제조 방법.

- 새롭고/새롭거나 특정한 TAF 및/또는 TAF-상호작용 인자를 확인하기위한 인간이 아닌 형질전이된 동물의 용도.
- 고 전사 복합체가 결합되는 에피토프-택을 갖는 TBP를 형질전이된 동물로부터 단리시킴을 포함하는 다양한 고 전사 복합체의 확인 및 특성화 방법.
- a) 제 11 항 내지 제 22 항중 어느 한항의 전이유전자를 인간이 아닌 동물에게 도입하는 단계,b) 에피토프-택을 갖는 TBP를 동물의 다양한 조직 및/또는 세포형으로부터, 선택적으로 동물의 상이한 발육 단계에서 단리하는 단계, 및c) 고 전사 복합체의 조성을 결정하는 단계를 포함하는,상이한 고 전사 복합체 조성의 특성화 방법.
- a) 제 11 항 내지 제 22 항중 어느 한항의 전이유전자를 인간이 아닌 동물에게 도입하는 단계,b) 에피토프-택을 갖는 TBP를 특정한 동물 조직 및/또는 동물의 특정한 세포형으로부터, 선택적으로는 특정한 발육 단계에서 단리하는 단계,c) 고 전사 복합체에서 에피토프-택을 갖는 TBP와 결합한 TAF 및/또는 TAF-상호작용 인자를 해리시키고 분리시키는 단계, 및d) 선택적으로 TAF 및/또는 TAF-상호작용 인자의 아미노산 서열을 결정하는 단계를 포함하는, 새롭고/새롭거나 특정한 TAF 및/또는 TAF-상호작용 인자의 확인 방법.
- TBP의 택인 하나이상의 에피토프를 이용하여 에피토프-택을 갖는 TBP를 친화성에 의해 정제할 때 에피토프-택을 갖는 TBP에 결합된 고 전사 복합체를 친화성에 의해 함께 정제하는, 제 1 항 내지 제 6 항중 어느 한항의 인간이 아닌 형질전이된 동물로부터 다양한 고 전사 복합체를 단리하는 방법.
- 제 43 항에 있어서,에피토프-택을 갖는 TBP를 그의 His 에피토프를 Ni2+-칼럼에 결합시킴으로써 정제하는 고 전사 복합체를 단리하는 방법.
- 제 43 항 또는 제 44 항에 있어서,에피토프-택을 갖는 TBP를, 그의 HA 에피토프를 항-HA 항체에 결합시킴으로써 정제하는, 고 전사 복합체를 단리하는 방법.
- 제 43 항 내지 제 45 항중 어느 한 항에 있어서, 서열 식별 번호 17의 서열을 갖고 에피토프-택을 갖는 TBP의 에피토프를 인식하는 항체가 있는 친화성 칼럼을 이용하여 고 전사 복합체를 단리시키는, 고 전사 복합체의 단리 방법.
- 서열 식별 번호 17의 서열을 갖는 에피토프-택을 갖는 TBP의 에피토프를 인식하는 항체.
