Abstract

Transgenes que comprende ADN que codifica una proteína de union a la TATA -box, epitopicamente marcada (TBP), la produccion de animales transgénicos queexpresan TBP epitopicamente marcada y el uso de la misma para purificacion de afinidad de factores de transcripcion y complejos de transcripcion (nuevos)de una variedad de tejidos y tipos de células eucarioticas. Animales transgénicos que expresan una proteína de union a la caja TATA (TBP) ligadaa ninguna, una o más marcas de epítope como por ejemplo la epítope HA, o epítope His así como la preparacion y uso de los animales transgénicos. Lasespecies de la cuales se obtienen el TBP podrían ser de diferentes orígenes como por ejemplo: el humano. El transgen es expresado o sobreexpresado en unamanera constitutiva o inducida por uso de promotores apropiados como los promotores del gen-EF-, o -MT-. Los complejos TBP marcados epitopicamente enlos animales transgénicos son usados para la identificacion de TAF y/o factores de interaccion TAF por medio de diferentes métodos como lacromatografía de afinidad o uso de anticuerpos.
