PCR 증폭을 하기 위하여 바이오니아 AccuPower® PCR PreMix를 이용하였으며, 반응조건은 95℃에서 5분간 초기 denaturation 시킨 후 30 cycle을 증폭시켰다. 각각의 cycle은 95℃에서 30초 동안 denaturation, 55℃-60℃에서 30초 동안 annealing, 72℃에서 30초 동안 extension하였다. PCR product는 1.5 % agarose gel에서 전기영동 하였고, ethidium bromide (EtBr)로 염색을 한 후 Gel Doc (Bio-Rad, USA)을 사용하여 관찰하였다. GAPDH를 internal control로 사용하여 FLG, HAS-1, HAS-2, HAS-3 유전자 발현을 상대적으로 평가하였다. 상기 실시예 2에서 제조된 화장료 조성물의 피부 주름 개선 효과를 확인하기 위하여 다음과 같은 실험을 수행하였다. 구체적으로, 연령이 40-50대인 피시험자 25명 각각은 아침과 저녁으로 매일 2회에 걸쳐 약 3-4 g의 화장료 조성물을 얼굴에 고루 펴서 바르는 식으로 40일간 시험을 실시하였다. 40일의 시험 종료 후에 주름 개선의 정도를 관찰자의 육안을 이용하여 평가하였다. 주름 개선의 정도는 아래와 같이 1-4번 항목으로 평가하였고, 그 결과를 하기 표 2에 나타내었다.The following experiment was performed to confirm the skin wrinkle improvement effect of the cosmetic composition prepared in Example 2.
