피부보습관련 유전자 히알루론산 합성효소 HAS-1, HAS-2, HAS-3와 FLG는 RT-PCR 방법을 이용하여 확인하였다. HaCaT 세포를 3X105 cells/mL의 밀도로 조정하고 6 well plate (SPL Life Science)에 2 L씩 분주하였다. 배양기에서 24시간 배양 후, 무형청 배지로 교체하여 24시간 추가 배양하였다. 무혈청 배지에 희석한 농도별 톳 추출물을 처리하고 24시간 동안 재배양하였다. 상등액 제거 후, Trizol reagent (Invitrogen)를 이용하여 Total RNA 추출을 진행하고, RNA정량은 Nano drop (Thermo)을 이용하여 측정하였다. 바이오니아 AccuPower® RT PreMix를 이용하여 cDNA를 합성하였고, specific primer 들을 이용한 PCR방법을 통하여 cDNA를 증폭하였다. PCR 증폭을 하기 위하여 바이오니아 AccuPower® PCR PreMix를 이용하였으며, 반응조건은 95℃에서 5분간 초기 denaturation 시킨 후 30 cycle을 증폭시켰다. 각각의 cycle은 95℃에서 30초 동안 denaturation, 55℃-60℃에서 30초 동안 annealing, 72℃에서 30초 동안 extension하였다. PCR product는 1.5 % agarose gel에서 전기영동 하였고, ethidium bromide (EtBr)로 염색을 한 후 Gel Doc (Bio-Rad, USA)을 사용하여 관찰하였다. GAPDH를 internal control로 사용하여 FLG, HAS-1, HAS-2, HAS-3 유전자 발현을 상대적으로 평가하였다. The PCR product was electrophoresed on a 1.5% agarose gel, stained with ethidium bromide (EtBr), and observed using Gel Doc (Bio-Rad, USA). Using GAPDH as an internal control, the relative expression of FLG, HAS-1, HAS-2, and HAS-3 genes was evaluated. The primers used in PCR are shown in Table 1 below.

상기 실시예 2에서 제조된 화장료 조성물의 보습 효과를 확인하기 위하여 다음과 같은 실험을 수행하였다. 설문 조사를 통하여 피부가 건조하다고 느끼는 건강한 성인남녀 100명을 무작위로 선정하여 실시예 2의 화장료 조성물을 각각 1일 2회씩 8주간 안면에 도포한 후 물로 씻어 냈다. 보습 효과는 실사용 시험 시작 후부터 매 1주간, 그리고 종료 후의 개선 효과를 수분 함유량 측정기기인 corneometer CM 820(Corage +Khazaka, Germany)를 이용하여 평가하였다. 보습 효과 측정 결과를 하기 표 3에 나타내었다.The following experiment was performed to confirm the moisturizing effect of the cosmetic composition prepared in Example 2.
