Abstract
본 발명은 메틸영양세균과 메틸영양세균의 1,2-프로판다이올 생산을 촉진하는 성분을 함유한 배지를 포함하는 1,2-프로판다이올 생산 촉진용 조성물 및 이를 이용하여 1,2-프로판다이올을 대량으로 생산하는 방법에 관한 것으로, 본 발명의 배지 및 생산 방법을 이용하면 1,2-프로판다이올을 신속하고 효율적으로 대량 생산할 수 있다.
Description
본 발명은 메틸영양세균; 메틸영양세균의 1,2-프로판다이올 생산을 촉진하는 성분을 함유한 2종의 배지;를 포함하는 1,2-프로판다이올 생산 촉진용 조성물 및 이를 이용한 1,2-프로판다이올을 효율적으로 대량 생산하는 생산 방법에 관한 것이다.
메탄올은 천연가스, 석탄 등의 원료로 생산되며 미국, 덴마크 등을 중심으로 메탄올 수율 향상을 위한 촉매 성능 개선, 신규 촉매 개발이 진행 중이며, 이 외에도 생촉매, 인공 광합성 관련 연구가 활발히 이루어지고 있다. 이란을 포함한 세계 메탄올 생산력 증가는 1,033만톤으로 공급 과잉에 들어갈 것으로 전망되며 중장기적으로도 셰일 가스를 활용한 북미의 메탄올 생산 능력이 확대될 전망이다.
메탄올은 생물 공학적 적용 (biotechnological applications)을 위한 탄소원으로 이용 시, 앞서 언급한 바와 같이 풍부함 (abundance)과 저렴한 가격 (low price)의 이점을 지닌다. 생물 공학적 공정 (Biotechnological processes)은 total cost에서 원료가 차지하는 비중이 높기 때문에 저렴한 원료를 사용하기 위하여 노력하며, 버개스 (bagasse), 사탕수수 당밀 (sugar cane molasses), 옥수수 전분 가수 분해물 (corn starch hydrolysates) 등이 주로 이용된다. 하지만 이와 같은 곡물 유래의 원료는 식량 문제를 피하기 어렵다. 반면에 메탄올은 앞서 언급한 원료들과 달리 식량 문제에서 자유로울 뿐만 아니라 배양 시 오염의 위험성을 줄일 수 있으며 글루코스 (glucose) 대비 높은 환원력 (reducing power)을 가지고 있어 생산물 수율 (yield)에 이점을 가진다.
메탄올은 2020년 6월 Methanex의 Asia Pacific 지역 기준 USD 215/ton이다. 반면에 1,2-프로판다이올 (1,2-propanediol, 1,2-PDO)은 주요 공급자 중 하나인 Dow의 Industrial grade 기준 USD 1,650/ton 이며 작년과 올해 가격인상 공지를 감안하면 꾸준한 가격 인상을 예측할 수 있다. 이처럼 메탄올은 1,2-PDO로 전환 시 약 7.7배 높은 가격을 가지므로, 고부가화 가능하다.
1,2-PDO가 사용되는 대표적인 예로 불포화 폴리에스터 수지 (Unsaturated poylester resin), 부동액 (Antifreeze), 세제 (Detergent) 등을 들 수 있으며 이외에도 제약, 식품, 화장품, Functional fluid 등의 광범위한 분야에서 활용되는 물질이다. 2019년 기준 1,2-PDO 시장규모는 4.3 billion dollar 규모로 추정되고, 2025년까지 아시아태평양 지역과 중동, 아프리카의 산업성장과 더불어 그 수요량이 증가하여 2025년까지 5.8 billion dollar 규모로 성장할 것으로 기대된다.
1,2-PDO는 전통적으로 프로필렌 옥사이드 (propylene oxide)를 산화시켜 생산하며 이러한 석유화학 기반의 생산공정은 고온고압일 뿐만 아니라 유독한 중간체와 부산물이 발생한다는 단점을 지닌다. 2000년대 들어 바이오 기반의 1,2-PDO 생산공정이 ADM 등에서 상용화 되고 있으나 원료로 콩이나 옥수수 같은 재생 가능한 공급원 (renewable source)이 이용되었을 뿐 공정 측면에서는 여전히 고온 고압에서 금속 촉매를 이용하고 있다.
미생물 기반의 1,2-PDO 생산 촉매는 상온 상압이라는 공정 측면의 이점을 지니며 다양한 탄소원을 이용 가능하다. 기존의 미생물을 이용한 1,2-PDO 연구는 크게 람노스 (Rhamnose)와 푸코스 (Fucose) 전환과 DHAP 전환으로 나뉜다. 람노스와 푸코스는 고가이기 때문에 산업적 활용이 어렵다. DHAP 전환은 그 기질로 글루코스 (Glucose), 글리세롤 (Glycerol), 자일로스 (Xylose) 등이 이용되었다는 보고는 있으나, 메탄올 (methanol)을 이용한 논문은 거의 전무하다.
이에 본 발명자들은 외래 유전자를 도입하여 1,2-프로판다이올 생합성 경로를 구축하거나, 내재된 유전자를 결실시킨 메틸로박테리움 엑스토르켄스 균주와 2종의 배지를 포함하는 1,2-프로판다이올 생산 촉진용 조성물을 이용한 결과, 1,2-프로판다이올의 생산이 촉진됨을 확인하였다.
이에, 본 발명의 목적은 메틸영양세균, 제1배지 및 제2배지를 포함하는 1,2-프로판다이올 생산 촉진용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 제1배양 단계 및 제2배양 단계를 포함하는 1,2-프로판다이올 생산 촉진 방법을 제공하는 것이다.
본 발명은 메틸영양세균; 메틸영양세균의 1,2-프로판다이올 생산을 촉진하는 성분을 함유한 2종의 배지;를 포함하는 1,2-프로판다이올 생산 촉진용 조성물 및 이를 이용한 1,2-프로판다이올을 효율적으로 대량 생산하는 생산 방법에 관한 것이다.
이하 본 발명을 더욱 자세히 설명하고자 한다.
본 발명의 일 예는 메틸영양세균; 제1배지; 및 제2배지;를 포함하는 1,2-프로판다이올 생산 촉진용 조성물에 관한 것이다.
본 발명에 있어서 용어 “메틸영양세균 (methylotroph)”은 탄소원을 이용하여 물질대사하는 세균을 의미하는 것일 수 있다.
본 발명에 있어서 메틸영양세균은 메틸글리옥살 합성효소 (methylglyoxal synthase, mgsA)를 코딩하는 핵산 서열, 메틸글리옥살 환원효소 (methylglyoxal reductase, yqhD)를 코딩하는 핵산 서열, 글리세롤 탈수소효소 (glycerol dehydrogenase, gldA)를 코딩하는 핵산 서열, 포몰레이즈 (formolase, fls)를 코딩하는 핵산 서열 및 디하이드록시아세톤 인산화효소 (dihydroxyacetone kinase, dak)를 코딩하는 핵산 서열로 이루어진 군으로부터 선택되는 1종 이상의 핵산 서열을 포함하는 재조합 벡터로 형질전환되고, 락토일글루타티온 분해효소(Lactoylglutathione lyase, gloA)를 코딩하는 핵산 서열을 포함하지 않는 것일 수 있다.
본 발명에 있어서 용어 “메틸글리옥살 합성효소 (methylglyoxal synthase)”는 디하이드록시아세톤 포스페이트 (dihydroxyacetone phosphate, DHAP)를 메틸글리옥살 (methylglyoxal, MG)로 전환하는 효소를 의미하는 것일 수 있고, 서열번호 2로 표시되는 핵산 서열로 이루어진 mgsA 유전자에 의해 코딩되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 용어 “메틸글리옥살 환원효소 (methylglyoxal reductase)”는 메틸글리옥살을 하이드록시아세톤으로 전환하는 효소인 메틸글리옥살 환원효소 (methylglyoxal reductase)를 의미하는 것일 수 있고, 서열번호 3으로 표시되는 핵산 서열로 이루어진 yqhD 유전자에 의해 코딩되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 용어 “글리세롤 탈수소효소 (glycerol dehydrogenase)”는 하이드록시아세톤을 1,2-PDO로 전환하는 효소를 의미하는 것일 수 있고, 서열번호 4로 표시되는 핵산 서열로 이루어진 gldA 유전자에 의해 코딩되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 용어 “포몰레이즈 (formolase)”는 포름알데히드를 디하이드록시아세톤 (dihydroxyacetone, DHA)으로 전환시키는 효소를 의미하는 것일 수 있고, 서열번호 11로 표시되는 핵산 서열로 이루어진 fls 유전자에 의해 코딩되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 용어 “디하이드록시아세톤 인산화효소 (dihydroxyacetone kinase)”는 디하이드록시아세톤(dihydroxyacetone, DHA)을 디하이드록시아세톤 포스페이트 (dihydroxyacetone phosphate)로 전환시키는 효소를 의미하는 것일 수 있고, 서열번호 12으로 표시되는 핵산 서열로 이루어진 사카로미세스 세레비시에(Saccharomyces cerevisiae) 유래의 dak1 유전자에 의해 코딩되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 용어 “벡터 (vector)”는 숙주 세포에서 목적 유전자를 발현시키기 위한 수단을 의미한다. 예를 들어, 플라스미드 벡터, 코즈미드 벡터 및 박테리오파아지 벡터, 아데노바이러스 벡터, 레트로바이러스 벡터 또는 아데노연관 바이러스 벡터와 같은 바이러스 벡터를 포함하는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 재조합 벡터로 사용될 수 있는 벡터는 당업계에서 종종 사용되는 플라스미드 (예를 들면, pSC101, pGV1106, pACYC177, ColE1, pKT230, pME290, pBR322, pUC8/9, pUC6, pBD9, pHC79, pIJ61, pLAFR1, pHV14, pGEX 시리즈, pET 시리즈 또는 pUC19 등), 파지 (예를 들면, λgt4λB, λ-Charon, λ△z1 또는 M13 등) 또는 바이러스 (예를 들면, SV40 등)를 조작하여 제작되는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 재조합 벡터는 전형적으로 클로닝을 위한 벡터 또는 발현을 위한 벡터로서 구축되는 것일 수 있다. 상기 발현용 벡터는 당업계에서 식물, 동물 또는 미생물에서 외래의 단백질을 발현하는 데 사용되는 통상의 것을 사용할 수 있고, 당업계에 공지된 다양한 방법을 통해 구축되는 것일 수 있다.
본 발명에 있어서 재조합 벡터는 원핵 세포 또는 진핵 세포를 숙주로 하여 구축되는 것일 수 있다. 예를 들어, 사용되는 벡터가 발현 벡터이고, 원핵 세포를 숙주로 하는 경우에는, 전사를 진행시킬 수 있는 강력한 프로모터 (예를 들어, pLλ프로모터, CMV 프로모터, trp 프로모터, lac 프로모터, tac 프로모터, T7 프로모터 등), 해독의 개시를 위한 라이보좀 결합 자리 및 전사/해독 종결 서열을 포함하는 것이 일반적이다. 진핵 세포를 숙주로 하는 경우에는, 벡터에 포함되는 진핵 세포에서 작동하는 복제원점은 f1 복제원점, SV40 복제원점, pMB1 복제원점, 아데노 복제원점, AAV 복제원점 및 BBV 복제원점 등을 포함하나, 이에 한정되는 것은 아니다. 또한, 포유동물 세포의 게놈으로부터 유래된 프로모터 (예를 들어, 메탈로티오닌 프로모터) 또는 포유동물 바이러스로부터 유래된 프로모터 (예를 들어, 아데노바이러스 후기 프로모터, 백시니아 바이러스 7.5K 프로모터, SV40 프로모터, 사이토메갈로바이러스 프로모터 및 HSV의 tk 프로모터)가 이용될 수 있으며, 전사 종결 서열로서 폴리아데닐화 서열을 일반적으로 갖는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 일 구체예에서, 형질전환체는 재조합 벡터를 적절한 숙주세포에 삽입됨으로써 만드는 것일 수 있다.
본 발명에 있어서 숙주세포는 대장균, 효모, 동물세포, 식물세포 또는 곤충세포 등일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 숙주세포는 원핵세포로서 예를 들어, E. coli JM109, E. coli BL21, E. coli RR1, E. coli LE392, E. coli B, E. coli X 1776, E. coli W3110, 바실러스 서브틸리스, 바실러스 츄린겐시스와 같은 바실러스 속 균주, 그리고 살모넬라 티피무리움, 세라티아 마르세슨스 및 다양한 슈도모나스 종과 같은 장내균과 균주 등일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 숙주세포는 진핵 세포로서 효모(Saccharomyce cerevisiae), 곤충 세포, 식물 세포 및 동물 세포 등일 수 있으며, 예를 들어, Sp2/0, CHO(Chinese hamster ovary) K1, CHO DG44, PER.C6, W138, BHK, COS7, 293, HepG2, Huh7, 3T3, RIN, MDCK 세포주 등일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 재조합 벡터는 숙주 세포 내로의 운반 (도입)되기 위하여, 당업계에 널리 알려진 운반 방법을 사용하는 것일 수 있다.
본 발명에 있어서 운반 (도입)은 예를 들어, 숙주 세포가 원핵 세포인 경우, CaCl2 방법 또는 전기 천공 방법 등을 사용하는 것일 수 있고, 숙주 세포가 진핵 세포인 경우에는, 미세 주입법, 칼슘 포스페이트 침전법, 전기 천공법, 리포좀매개 형질감염법 및 유전자 밤바드먼트 등을 사용하는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 재조합 벡터가 숙주 세포 내로 운반되어 형질 전환된 숙주 세포를 선별하는 방법은 선택 표지에 의해 발현되는 표현형을 이용하여, 당업계에 널리 알려진 방법에 따라 용이하게 실시할 수 있으며, 예를 들어, 선택 표지가 특정 항생제 내성 유전자인 경우에는, 상기 특정 항생제가 함유된 배지에서 형질전환체를 배양함으로써 형질전환체를 용이하게 선별하는 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 메틸영양세균은 락토일글루타티온 분해효소 (Lactoylglutathione lyase)를 코딩하는 gloA 유전자를 포함하지 않는 것일 수 있다.
본 발명에 있어서 유전자 “gloA”는 MG를 락토일글루타티온으로 전환하는 락토일글루타티온 분해효소 (Lactoylglutathione lyase)를 코딩하는 핵산 서열일 수 있고, 서열번호 17 및 18로 표시되는 핵산 서열로 이루어진 것일 수 있다.

본 발명에 있어서 1,2-프로판다이올 생산 촉진용 조성물은 메틸영양세균의 1,2-PDO Max titer에 도달하는 시간을 감소시키거나, 또는 시간을 감소시키면서 1,2-PDO Max titer 값을 향상시킴으로써, 1,2-프로판다이올의 생산을 촉진할 수 있다.
본 발명의 다른 일 예는 다음의 단계를 포함하는 1,2-프로판다이올 생산 촉진 방법에 관한 것이다:
탄소원 (carbon source)을 포함하는 제1배지에서, 메틸영양세균을 배양하는 제1배양 단계; 및
탄소원 (carbon source) 및 질소원 (nitrogen source)을 포함하는 제2배지에서, 상기 제1배양 단계에서 배양한 메틸영양세균을 배양하는 제2배양 단계.
본 발명에 있어서 제1배양 단계는 메틸영양세균의 대량 배양을 위하여, 또는 일정한 배양액 조건 하에서 일정한 메틸영양세균을 얻기 위하여 상기 메틸영양세균을 미리 배양하는 것 (전배양)을 의미할 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 제1배양 단계는 제1배지를 이용하는 것일 수 있다.
본 발명에 있어서 제1배양 단계는 5 내지 40 ℃, 5 내지 35 ℃, 10 내지 40 ℃, 10 내지 35 ℃, 15 내지 40 ℃, 15 내지 35 ℃, 20 내지 40 ℃, 20 내지 35 ℃, 25 내지 40 ℃ 또는 25 내지 35 ℃의 온도 조건 하에서 수행되는 것일 수 있으며, 예를 들어, 25 내지 35 ℃의 온도 조건 하에서 수행되는 것일 수 있으나, 이에 한정되는 것은 아니고, 메틸영양세균의 최적 성장 온도 (optimal growth temperature)를 고려하여 적절한 온도 조건 하에서 수행되는 것일 수 있다.
본 발명에 있어서 제1배양 단계는 세균 성장 곡선 (bacterial growth curve)을 기준으로, 메틸영양세균이 중기-지수상 (Mid-exponential phase)과 초기-정지상 (Early-Stationary phase)에 도달할 때까지 메틸영양세균을 배양하는 것일 수 있다.
본 발명에 있어서 제2배양 단계는 메틸영양세균이 1,2-프로판다이올을 생산 하기 위하여 메틸영양세균을 배양하는 것 (본배양)을 의미할 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 제2배양 단계는 제2배지를 이용하는 것일 수 있다.
본 발명에 있어서 제2배양 단계는 5 내지 40 ℃, 5 내지 35 ℃, 10 내지 40 ℃, 10 내지 35 ℃, 15 내지 40 ℃, 15 내지 35 ℃, 20 내지 40 ℃, 20 내지 35 ℃, 25 내지 40 ℃ 또는 25 내지 35 ℃의 온도 조건 하에서 수행되는 것일 수 있으며, 예를 들어, 25 내지 35 ℃의 온도 조건 하에서 수행되는 것일 수 있으나, 이에 한정되는 것은 아니고, 메틸영양세균의 최적 성장 온도 (optimal growth temperature)를 고려하여 적절한 온도 조건 하에서 수행되는 것일 수 있다.
본 발명에 있어서 제2배양 단계는 회분식 배양 (batch culture)으로서, 제2배지에 포함되는 탄소원이 고갈되지 않도록, 탄소원이 고갈된 경우, 탄소원을 제2배지와 혼합하는 탄소원 혼합 단계를 포함할 수 있다.
본 발명의 1,2-프로판다이올 생산 촉진 방법은 메틸영양세균이 일정한 속도로 1,2-PDO를 생산하지 않고, 세균 성장 곡선 (bacterial growth curve)을 기준으로, 중기-지수상 (Mid-exponential phase) 이후부터 빠르게 1,2-PDO를 생산한 후, 메탄올이 고갈되는 부근에서 최대 1,2-PDO titer를 나타내는 것을 고려하여, 제2배양 단계가 탄소원 혼합 단계를 포함하는 것일 수 있다.
본 발명에 있어서 탄소원 혼합 단계는 제2배양 단계 전반에 걸쳐 제2배지의 탄소원이 고갈되지 않게 함으로써 1,2-PDO titer가 낮아지지 않도록 유지시킬 수 있다.
본 발명에 있어서 1,2-프로판다이올 생산 촉진 방법은 제2배양 단계 이후, 제2배지와 pH 조절제를 혼합하는 pH 조절 단계를 더 포함할 수 있다.
본 발명에 있어서 pH 조절제는 포름산 (HCOOH), 젖산 (CH3CHOHCOOH), 아세트산 (CH3COOH), 프로피온산 (CH3CH2COOH), 옥살산 (H2C2O4) 및 글리콜산 (CH2OHCOOH)으로 이루어진 군으로부터 선택되는 1종 이상일 수 있으며, 예를 들어, 포름산일 수 있으나, 이에 한정되는 것은 아니다.
또한, 본 발명에 있어서 pH 조절제는 염산 (HCl), 질산 (HNO3), 황산 (H2SO4) 및 인산 (H3PO4)으로 이루어진 군으로부터 선택되는 1종 이상일 수 있으며, 예를 들어, 염산일 수 있으나, 이에 한정되는 것은 아니다.
또한, 본 발명에 있어서 pH 조절제는 수산화나트륨 (NaOH), 수산화칼륨 (KOH), 수산화칼슘 [Ca(OH)2] 및 수산화바륨 [Ba(OH)2]으로 이루어진 군으로부터 선택되는 1종 이상일 수 있으며, 예를 들어, 수산화나트륨일 수 있으나, 이에 한정되는 것은 아니다.
pH 조절 단계는 제2배지와 pH 조절제를 혼합함으로써, 제2배양 단계에서 메틸영양세균이 생산한 1,2-PDO의 분해를 억제할 수 있다.
본 발명은 메틸영양세균; 메틸영양세균의 1,2-프로판다이올 (1,2-PDO)의 생산을 촉진하는 성분을 함유한 배지를 포함하는 1,2-프로판다이올 생산 촉진용 조성물 및 이를 이용한 1,2-프로판다이올 생산 촉진 방법에 관한 것으로, 본 발명의 조성물 및 생산 방법을 이용하면 1,2-프로판다이올을 신속하고 효율적으로 대량 생산할 수 있다.
도 1은 본 발명의 일 구현예에 따라 메틸글리옥살 합성효소(methylglyoxal synthase, mgsA) 및 글리세롤 탈수소효소(glycerol dehydrogenase, gldA)를 활성화시키고, 메틸글리옥살 환원효소(methylglyoxal reductase) 또는 락트알데히드 환원효소(lactaldehyde reductase, fucO)를 활성화시킴으로써 구축한 1,2-프로필렌글라이콜(propanediol, PDO) 생합성 경로의 모식도이다.

재조합 호스트(Host)로는 메탄올을 탄소원으로 이용 가능하며 대사작용이 알려져 있고 유전자 조작 툴이 개발되어 있는 메틸로박테리움 엑스토르켄스 (Methylobacterium extorquens) AM1 를 사용하였다. M. extorquens AM1 wild type (야생형)은 1,2-PDO를 생산하지 못하므로, 외래 유전자를 도입하여 1,2-PDO 생합성 경로를 도 1과 같이 구축하였다. 파란색으로 표시한 화살표는 외래 유전자를 도입한 경로를 나타낸 것이다.
1,2-PDO 생산 경로는 하이드록시아세톤 (hydroxyacetone)을 거치는 경로와 L-락트알데히드 (lactaldehyde)를 거치는 경로가 있다. 하이드록시아세톤을 거치는 경로 또는 L- 락트알데히드를 거치는 경로를 지닌 재조합 균주를 제작하였다.
1-1. 하이드록시아세톤 경로를 지닌 재조합 균주의 제조
하이드록시아세톤을 거치는 경로는 바실러스 서브틸리스 (Bacillus subtilis) 유래의 디하이드록시아세톤 포스페이트 (dihydroxyacetone phosphate, DHAP)를 메틸글리옥살 (methylglyoxal, MG)로 전환하는 효소인 메틸글리옥살 합성효소(synthase)를 코딩하는 mgsA 유전자, 대장균 (Escherichia coli) 유래의 MG를 하이드록시아세톤으로 전환하는 효소인 메틸글리옥살 환원효소 (methylglyoxal reductase)를 코딩하는 yqhD 유전자, 대장균 유래의 하이드록시아세톤을 1,2-PDO로 전환하는 효소인 글리세롤 탈수소효소 (glycerol dehydrogenase)를 코딩하는 gldA 유전자를 pMEV 벡터에 도입하여, pMEV::mgsA::yqhD::gldA 플라스미드 벡터를 구축하였다.
pMEV 벡터의 서열정보는 서열번호 1과 같다. mgsA 유전자의 서열정보는 서열번호 2, yghD 유전자의 서열정보는 서열번호 3, gldA 유전자의 서열정보는 서열번호 4와 같으며 하기 표 1에 나타내었다.

균주 제작은 인퓨젼 (Infusion) 효소를 이용하여 클로닝 (Multi-insert Cloning)하였으며 제한효소로는 EcoR1과 Sac1을 이용하였다. M. extorquens AM1::pMEV::mgsA::yqhD::gldA 균주 (MPG001)의 클로닝에 사용한 프라이머는 서열번호 5 내지 10으로, 하기 표 2와 같다. 밑줄 친 서열은 리보솜 결합부위 (ribosome binding site, RBS)이다.

1-2. DHAP 흐름을 증가시킨 재조합 균주의 제조
DHAP는 M. extorquens AM1의 대사산물 (metabolite)이자 앞서 삽입한 1,2-PDO 생합성 경로의 전구체로 메틸글리옥살 (Methylglyoxal, MG) 생합성에 의해 메틸글리옥살로 전환된다. M. extorquens AM1이 탄소원인 메탄올을 DHAP로 전환시키기 위해서는 복잡한 단계를 거쳐야 한다.
이에 보다 직접적인 DHAP 합성 경로를 도입하여 DHAP 흐름 (flux)을 증진시켰다. M. extorquens AM1은 풍부한 포름알데히드 (formaldehyde) 흐름을 지니므로 포름알데히드를 디하이드록시아세톤 (dihydroxyacetone, DHA)으로 전환시키는 효소 포몰레이즈 (Formolase) 및 DHA를 디하이드록시아세톤 포스페이트 (dihydroxyacetone phosphate)로 전환시키는 효소 디하이드록시아세톤 인산화효소 (dihydroxyacetone kinase)를 과발현시켰다.
포몰레이즈는 새로운 탄소 동화 경로 (carbon assimilation pathway)를 가능케하는 관점에서는 논문이 보고 되었지만 이처럼 특정 물질을 생산하는 목적으로 이용된 케이스는 보고된 바가 없으며 E. coli 이외의 균주에서 포몰레이즈를 작동시킨 전례는 없는 것으로 추정된다.
포몰레이즈를 코딩하는 유전자 fls는 AM1에 맞춰 코돈 최적화 (codon optimization)시켰다. 디하이드록시아세톤 인산화효소를 코딩하는 유전자는 다양하며 이 연구에서는 사카로미세스 세레비시에 (Saccharomyces cerevisiae) 유래의 dak1 유전자를 사용하였다. 기존에 구축한 플라스미드 pMEV::mgsA::yqhD::gldA에 fls와 dak1을 추가로 도입하여 새로운 플라스미드 pMEV::mgsA::yqhD::gldA::fls::dak1를 구축하였고 이 플라스미드를 지닌 M. extorquens AM1::pMEV::mgsA::yqhD::gldA::fls::dak1 균주 (MPG011)를 준비하였다.
코돈 최적화된 fls 유전자의 서열정보는 서열번호 11, dak1 유전자의 서열정보는 서열번호 12와 같으며 하기 표 3에 나타내었다.

1-3. MG로의 경쟁 경로를 결실시킨 재조합 균주의 제조
M. extorquens AM1은 MG에서 락토일글루타티온 (Lactoylglutathione)을 거쳐 락테이트 (lactate)로 흐르는 경로를 보유하고 있다. 따라서, M. extorquens AM1에서 생성된 MG는 과발현시킨 1,2-PDO 생산 경로의 하이드록시아세톤과 락테이트로 흐름이 나누어지게 된다. MG를 락토일글루타티온으로 전환시키는 효소 락토일글루타티온 분해효소 (Lactoylglutathione lyase)를 코딩하는 유전자 gloA를 결실시킴으로써 락테이트로 흐름 손실이 일어나지 않는 M. extorquens AM1ΔgloA::pMEV::mgsA::yqhD::gldA 균주 (MPG101) 및 M. extorquens AM1ΔgloA::pMEV::mgsA::yqhD::gldA::fls::dak1 균주 (MPG111)를 준비하였다.
M. extorquens AM1의 Mex_1p0359 및 Mex_1p4683 2개의 유전자가 gloA에 해당하므로 pk19mobsacB 벡터를 이용하여 Mex_1p0359 (서열번호 17) 및 Mex_1p4683 (서열번호 18)을 결실시켰고, 서열정보는 하기 표 5에 나타내었다. pk19mobsacB 벡터의 서열은 서열번호 19의 서열과 같다.

실험예 1. NMS (Nitrate Mineral Salts) 배지 환경에서 재조합 균주의 1,2-PDO 생산량 확인
3 ml의 NMS 배지 (제2배지)를 포함하는 14 ml 둥근바닥 테스트 튜브(14ml round bottom test tube)에서 30 ℃이 200 rpm 조건으로 제조예 1에서 제조한 MPG111, MPG001, MPG011 및 MPG101 균주를 배양하였다. 이후 균주를 100 ml의 NMS 배지 (제2배지)를 포함하는 500 ml 배플 플라스크에 접종하여 30 ℃ 및 200 rpm의 조건으로 균주를 다시 배양하였다 (전배양).
전배양을 마친 MPG111, MPG001, MPG011 및 MPG101 균주를 수확하여 100 ml의 NMS 배지 (제2배지)를 포함하는 500 ml 배플 플라스크에 재현탁하고 30 ℃ 및 200 rpm의 조건으로 균주를 배양하였다 (본배양).
상기 전배양 및 본배양을 3회 반복하여, 1,2-PDO를 생산하였다. 3회 반복한 실험값의 평균은 표 11의 average에 나타내었고, 3회 반복한 실험값 중 최대값을 표 11의 Max에, 전체 실험데이터를 도 3a 내지 3d 및 표 11에 나타내었다.

1-2. Minimal 배지 환경에서 재조합 균주의 1,2-PDO 생산량 확인
50 ml의 Minimal 배지 (제1배지)를 포함하는 250 ml 배플 플라스크에서 30 ℃ 및 200 rpm의 조건으로 제조예 1에서 제조한 MPG111 균주를 배양하였다 (전배양).
전배양을 마친 MPG111 균주를 수확하여 50 ml의 Minimal 배지 (제1배지)를 포함하는 250 ml 배플 플라스크에 재현탁하고 30 ℃ 및 200 rpm의 조건으로 배양하여 (본배양) 1,2-PDO를 생산하였다.
실험예 1-1에서 생산한 1,2-PDO titer 대비 실험예 1-2에서 생산한 1,2-PDO titer를 도 4에 나타내었다.
도 4로부터 확인할 수 있듯이, 전배양 및 본배양시 모두 제1배지에서 배양된 MPG111 균주는 1,2-PDO를 생산하지 않았다. 즉, 1,2-PDO의 생산을 위해서는 Minimal 배지에서 배양하는 과정만으로는 1,2-PDO를 생산할 수 없고, 반드시 NMS 배지에서 배양하는 과정이 포함되어야 1,2-PDO를 생산할 수 있는 것으로 판단하였다.
실험예 2. Minimal 배지 및 NMS 배지를 이용한 재조합 균주의 1,2-프로판다이올 생산량 확인
전배양 및 본배양 단계 모두 동일한 배지를 사용하는 종래 방법을 변경하여, 전배양 (제1배양 단계)은 세포 성장 속도가 빠르고 안정적인 Minimal 배지 (제1배지)를, 본배양 (제2배양 단계)은 1,2-프로판다이올의 역가 (titer)가 우수한 NMS 배지 (제2배지)를 이용하였다. 구체적인 실험 조건은 다음과 같다.
3 ml의 Minimal 배지 (제2배지)를 포함하는 14 ml 둥근바닥 테스트 튜브에서 30 ℃이 230 rpm 조건으로 제조예 1에서 제조한 MPG111, MPG001, MPG011 및 MPG101 균주를 배양하였다. 이후 균주를 50 ml의 NMS 배지 (제2배지)를 포함하는 250 ml 배플 플라스크에 접종하여 30 ℃ 및 230 rpm의 조건으로 균주를 다시 배양하였다 (전배양).
전배양이 중기-지수상 (Mid-exponential phase)과 초기-정지상 (Early-Stationary phase) 초입에 도달하면, 본배양에 필요한 만큼 MPG111, MPG001, MPG011 및 MPG101 균주를 수확하여 50 ml의 NMS 배지 (제2배지)를 포함하는 250 ml 배플 플라스크에 재현탁하고 30 ℃ 및 230 rpm의 조건으로 균주를 배양하였다 (본배양).
균주의 수확은 본배양시 원하는 초기 OD 값을 기초로, 필요한 Minimal 전배양액을 채취하고 원심분리후 상등액을 제거하고 NMS 배지로 재현탁 (resuspension)함으로써 수행될 수 있다.
상기 전배양 및 본배양을 3회 반복하여 1,2-PDO를 생산한 후, 그 결과를 도 5a 내지 5d 및 표 12에 나타내었다.
MPG111 균주의 1,2-프로판다이올 생산 결과는 5a에, MPG001 균주의 1,2-프로판다이올 생산 결과는 도 5b에, MPG011 균주의 1,2-프로판다이올 생산 결과는 도 5c에, MPG101 균주의 1,2-프로판다이올 생산 결과는 도 5d에 나타내었다.

전배양 및 본배양시 동일하게 NMS 배지를 사용한 표 11의 실험데이터와 전배양시 Minimal 배지 및 본배양시 NMS 배지를 사용한 표 13의 실험데이터를 비교하면 확인할 수 있듯이, MPG111 균주의 1,2-PDO average titer는 58.8 mg/L에서 137.610 mg/L로 약 +134.0% 증가하였고, Max titer는 61.6 mg/L에서 159.101 mg/L로 +158.3% 증가하였다.
MPG001 균주의 1,2-PDO average titer는 8.6 mg/L에서 38.731 mg/L로 약 +360.4% 증가하였고, Max titer는 10.1 mg/L에서 47.352 mg/L로 +368.8% 증가하였다.
MPG011 균주의 1,2-PDO average titer는 9.8 mg/L에서 28.276 mg/L로 약 +188.5% 증가하였고, Max titer는 10.8 mg/L에서 34.310 mg/L로 +217.7% 증가하였다.
MPG101 균주의 1,2-PDO average titer는 43.8 mg/L에서 208.650 mg/L로 약 +376.4% 증가하였고, Max titer는 54.1 mg/L에서 219.105 mg/L로 +305.0% 증가하였다.
또한, 전배양 및 본배양시 사용하는 배지를 변경함으로써, 배지를 변경하지 않는 것에 비하여 Max titer까지 도달하는 시간에 대하여, MPG111 균주는 32 시간에서 18 시간으로 -43.75%, MPG001 균주는 20 시간에서 18 시간으로 -10%, MPG011 균주 24 시간에서 18 시간으로 -25%, MPG101 균주는 69 시간에서 20 시간으로 -71.01% 단축되었다.
실험예 3. MPG101 균주의 배양 조건 구체화
3-1. 전배양 온도 조건 변화
1,2-PDO 생산량이 가장 높은 MPG101 균주로 1,2-PDO 배양 조건을 구체화하였다. 전술한 실험예 2의 조건과 동일하되, 전배양은 20 ℃, 25 ℃, 30 ℃ 또는 35 ℃의 조건으로 수행하였고, 본배양은 30 ℃의 조건으로 수행하여, 1,2-PDO를 생산한 결과를 도 6a 내지 6d에 나타내었다. 그리고, 평균 생산량 최대값 (Average ProductionMAX) 및 평균 생산효율 최대값 (Average ProductivityMAX)을 계산하여, 표 14에 나타내었다.

도 6a 내지 6e 및 표 14에서 확인할 수 있듯이, 1,2-PDO 평균 생산량 및 평균 생산효율 최대값은 전배양 온도가 30 ℃인 경우 가장 높았고, 35 ℃인 경우 가장 낮았다.
3-2. 본배양 온도 조건 변화
실험예 3-1과 달리, 전배양은 30 ℃의 조건으로 수행하였고, 본배양은 20 ℃, 25 ℃, 30 ℃ 또는 35 ℃의 조건으로 수행하여, 1,2-PDO를 생산한 결과를 도 7a 내지 7d에 나타내었다. 그리고, 평균 생산량 최대값 (Average ProductionMAX) 및 평균 생산효율 최대값 (Average ProductivityMAX)을 계산하여, 표 15에 나타내었다.

표 15의 30 ℃ Average ProductionMAX를 기준으로, 20 ℃, 25 ℃ 및 35 ℃의 Average ProductionMAX 상대값을 도 7e에 나타내었고, 30 ℃ Average ProductivityMAX를 기준으로, 20 ℃, 25 ℃ 및 35 ℃의 Average ProductivityMAX 상대값을 도 7f에 나타내었다.
도 7a 내지 7f 및 표 15에서 확인할 수 있듯이, 1,2-PDO 평균 생산량은 본배양 온도가 20 ℃인 경우 가장 높은 반면, 균주의 생장속도가 느려 생산효율이 낮았다. 그러나, 본배양 온도가 30 ℃인 경우에는 균주의 생장속도가 빨라 평균 생산효율 최대값이 가장 높았다.
3-3. 완충용액 (buffer) 또는 질소원 변화
본배양시 제2배지의 완충용액 또는 질소원의 조성을 변화하였다.
먼저, 기존 실험예 2의 제2배지 (KH2PO4 및 Na2HPO4ㆍ7H2O를 포함하는 NMS buffer 이용)로 1,2-PDO 생산한 것을 Control (실시예 4)로 나타내었고, Control에서 완충용량 (capacity)을 증가시키기 위해 상기 NMS buffer 대신 Minimal buffer (K2HPO4 및 NaH2PO4ㆍ2H2O 포함)를 이용한 제2배지로 1,2-PDO를 생산하여 실시예 3으로 나타내었다. 완충용액 조성 변화시 제2배지에 포함된 K2HPO4 및 NaH2PO4ㆍ2H2O의 농도는 제1배지와 동일하게 설정하였다.
다음으로, 실시예 3에서 질소원으로서 질산칼륨 (Potassium nitrate; KNO3)을 이용하는 대신, 요소 (Urea) 또는 염화암모늄 (Ammonium chloride; NH4Cl)으로 1,2-PDO를 생산하여 실시예 1, 2에 나타내었고, 효모 추출물 (Yeast extract) 또는 펩톤 (Peptone)으로 1,2-PDO를 생산하여 비교예 1, 2에 나타내었다. 분자식이 알려진 물질인 경우, 질소원의 양은 이온화를 고려하여 몰수를 동일하게 설정하였고, 효모 추출물 (Yeast extract) 및 펩톤 (Peptone)은 1 g/L로 설정하였다.

도 8a 내지 8f 및 표 17에서 확인할 수 있듯이, 1,2-PDO 평균 생산량 측면에서, Urea (실시예 1)는 Control (실시예 4)에 비해 +52% 증가하여, 가장 높은 평균 생산량을 나타내었다. 또한, Control에 비해 염화암모늄 (실시예 2)은 +40%, 질산칼륨 (실시예 3)은 +15% 증가하였다. 본배양시 제2배지가 효모 추출물 및 펩톤을 포함하는 경우에는 메탄올이 일부만 소모되어, 1,2-PDO가 거의 생산되지 않았다.
그리고, 1,2-PDO 평균 생산효율 측면에서, Urea (실시예 1)는 Control (실시예 4)에 비해 +37% 증가하여, 가장 높은 생산효율을 나타내었다. 또한, Control에 비해 염화암모늄 (실시예 2)은 +20%, 질산칼륨 (실시예 3)은 +6% 증가하였다.
3-4. 본배양시 메탄올 농도 조건 변화
전술한 실험예 2의 조건과 동일하되, 본배양시 제2배지의 메탄올 농도가 60 mM, 125 mM, 185 mM 또는 250 mM인 조건에서 1,2-PDO를 생산하여, 그 결과를 도 9a 내지 9d에 나타내었다. 그리고, 평균 생산량 최대값 (Average ProductionMAX) 및 평균 생산효율 최대값 (Average ProductivityMAX)을 계산하여, 도 9e 및 표 18에 나타내었다.

도 9a 내지 9e, 표 18에서 확인할 수 있듯이, 1,2-PDO 평균 생산량은 본배양 배지가 250 mM의 메탄올을 포함하는 경우 가장 높았고, 평균 생산효율은 125 mM의 메탄올을 포함하는 경우에 가장 높았다.
메탄올 농도가 60 mM에서 250 mM로 높아질수록 1,2-PDO의 생산량은 증가하였으나, 생산효율은 125 mM에서 최대치를 나타내었다. 도 9e 및 표 18에 나타낸 1,2-PDO의 생산량 및 생산효율의 상관관계를 고려하여, 균주가 생장할 수 있는 범위 내에서 메탄올 농도를 조절하는 것이 필요하다.
실험예 4. 1,2-프로판다이올 생산 종결
전술한 실험예 2 및 실험예 3에서 확인할 수 있듯이, 1,2-PDO는 최대 역가를 달성한 후 분해된다. 1,2-PDO의 분해를 방지하기 위해, 먼저 실험예 2의 실험조건과 동일하게 1,2-PDO를 생산하였다. 최대 역가를 달성한 이후 50 mM 포름산 (Formic acid)을 첨가하였다. 1,2-PDO는 분해가 억제됨을 확인하여, 그 결과를 도 10a 및 10b에 나타내었다.
다음으로, 25 mM의 포름산, 염산 (Hydrochloric acid; HCl) 및 수산화나트륨 (Sodium hydroxide; NaOH)을 첨가하여, 마찬가지로 1,2-PDO 분해가 억제됨을 확인하여, 그 결과를 도 10c 내지 10e에 나타내었다. (Add로 나타낸 시점에 산과 염기를 첨가하였다.)
도 10a 내지 10e에서 확인할 수 있듯이, 1,2-PDO의 분해 종결은 pH 변화를 이용하는 것으로, 반드시 포름산과 같은 유기산에 한정되는 것이 아니라, 무기산이나 염기에 의해서도 가능하다.
따라서, 본 발명의 1,2-PDO 생산 촉진용 조성물 및 이를 이용한 1,2-PDO 대량 생산 방법을 이용하면 신속하고 효율적으로 1,2-프로판다이올을 대량 생산할 수 있다.

- 메틸영양세균;
탄소원 (carbon source)을 포함하는 제1배지; 및
탄소원 (carbon source) 및 질소원 (nitrogen source)을 포함하는 제2배지;
를 포함하고,
상기 메틸영양세균은 메틸로박테리움 엑스토르켄스 (Methylobacterium extorquens)인 것이고,
상기 탄소원은 메탄올 (methanol)인 것이고,
상기 메틸로박테리움 엑스토르켄스는 메틸글리옥살 합성효소 (methylglyoxal synthase, mgsA)를 코딩하는 핵산 서열, 메틸글리옥살 환원효소 (methylglyoxal reductase, yqhD)를 코딩하는 핵산 서열 및 글리세롤 탈수소효소 (glycerol dehydrogenase, gldA)를 코딩하는 핵산 서열을 포함하는 재조합 벡터로 형질전환된 것이고,
상기 형질전환된 메틸로박테리움 엑스토르켄스는 락토일글루타티온 분해효소(Lactoylglutathione lyase, gloA)를 코딩하는 핵산 서열을 포함하지 않는 것인, 1,2-프로판다이올 생산 촉진용 조성물. - 삭제
- 제1항에 있어서, 상기 형질전환된 메틸영양세균은 포몰레이즈 (formolase, fls)를 코딩하는 핵산 서열 및 디하이드록시아세톤 인산화효소 (dihydroxyacetone kinase, dak)를 코딩하는 핵산 서열을 포함하는 재조합 벡터로 추가로 형질전환된 것인, 1,2-프로판다이올 생산 촉진용 조성물.

- 다음의 단계를 포함하는 1,2-프로판다이올 생산 촉진 방법:
탄소원 (carbon source)을 포함하는 제1배지에서, 메틸영양세균을 배양하는 제1배양 단계; 및
탄소원 (carbon source) 및 질소원 (nitrogen source)을 포함하는 제2배지에서, 상기 제1배양 단계에서 배양한 메틸영양세균을 배양하는 제2배양 단계;를 포함하고,
상기 메틸영양세균은 메틸로박테리움 엑스토르켄스 (Methylobacterium extorquens)인 것이고,
상기 탄소원은 메탄올 (methanol)인 것이고,

- 제7항에 있어서, 상기 제2배양 단계는 회분식 배양 (batch culture)으로서,
제2배지에 포함되는 탄소원이 고갈되지 않도록 탄소원을 제2배지와 혼합하는 탄소원 혼합 단계를 포함하는 것인, 1,2-프로판다이올 생산 촉진 방법. - 제7항에 있어서, 상기 방법은 제2배양 단계 이후,
제2배지와 pH 조절제를 혼합하는 pH 조절 단계를 더 포함하는 것인, 1,2-프로판다이올 생산 촉진 방법.
