본 발명에 따른 복합 추출물의 항산화 효과를 확인하기 위해 DPPH(1,1-diphenyl-2-picrylhydrazyl) 라디칼 소거능을 측정하였다. DPPH 항산화 측정을 위해, Blois의 방법(1958)을 변형하여 사용하였다. DPPH(1,1-diphenyl-2-picrylhydrazyl) 라디칼 소거능을 측정하기 위하여 본 발명의 천연물 추출물을 50 μg/ml이 되도록 순차 희석하여 사용하였다. 95% 에탄올에 용해시킨 0.2 mM DPPH 150 μl와 각 천연물 추출물 샘플을 50 μl를 96 웰 플레이트에 넣고 파이펫팅하여 혼합한 뒤 암실에서 30분간 반응시켰다. 남아있는 라디칼의 농도를 UV spectrophotometer를 사용하여 517 nm에서 측정하였다. 추출물의 DPPH에 대한 소거능은 남아있는 라디칼의 농도를 통해 소거된 라디칼을 계산하여 % 수치로 나타내었다.In order to confirm the antioxidant effect of the complex extract according to the present invention, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging ability was measured. 본 발명에 따른 복합 추출물에 대하여 NO(Nitric oxide) 생성억제 효과를 측정하였다. 구체적으로, raw264.7 세포를 24 well plate에 2×10^5 cells/ml의 농도로 분주하여 24 시간 배양한 후 추출물들과 Minoxidil을 100 μg/ml 또는 0.25 μM의 농도로 주입 후 1시간 뒤 1 μg/ml의 LPS로 자극 후 24시간 배양한 후 배양액을 수거하였다. 수거한 배양액 100 μL와 Griess reagent 100 μL를 96 well plate에 주입 후 15분간 반응시켰다. 반응이 끝난 후 540 nm 흡광도에서 측정하였다. 본 발명에 따른 복합 추출물에 대하여 웨스턴블롯 분석과 ELISA 분석을 통해 Steroid 5α-환원효소 2 억제효과를 측정하였다. 구체적으로, HFDPC (인간 모낭진피유두(모유두)세포, human follicle dermal papilla cell) 세포를 4×10^5 cells/ml의 농도로 분주하여 24시간 배양한 후 추출물들과 Minoxidil을 100 μg/ml의 또는 0.25 μM의 농도로 주입 후 1시간 뒤 Testosterone 100 μM농도로 자극 후 24시간 배양하였다. 그 후 ELISA kit 분석을 위하여 배양액을 수거하여 측정하였으며, 웨스턴블롯 분석을 위해 세포를 PBS로 2회 원심세척 후 RIPA 버퍼를 주입 후 4℃에서 15분간 12,000 rpm으로 원심분리하여 단백질을 추출하였다. 상기 추출한 각 단백질은 브래드포드 단백질 분석 시약(Bradford protein assay reagent)을 사용하여 595 nm에서 흡광도를 측정하여 정량하였다. 정량한 단백질을 SDS-PAGE를 통해 분리 후, 100 V로 1시간 동안 PVDF(polyvinylidene difluoride) 멤브레인(Bio-Rad)에 트랜스퍼하였다. 멤브레인은 5% BSA(bovine serum albumin)가 함유된 TBS-T(Tris buffered saline with 1% Tween 20)에 실온에서 블로킹 후 TBS-T로 10분간 3회 세척 후 1차 항체로 anti-5α-환원효소(5-AR) 및 anti-β-actin을 사용하여 4℃에서 16시간 반응시켰다. 그 후 TBS-T로 10분간 3회 세척 후 2차 항체(goat anti-rabbit IgG HRP, rabbit anti-mouse IgG HRP)를 5% BSA에서 실온에서 2시간 반응 후 TBS-T로 10분간 3회 세척하였다. 이후, 세척한 멤브레인은 detection system을 사용하여 이미지를 얻었다(도 5의 A).Steroid 5α-reductase 2 inhibitory effect was measured by Western blot analysis and ELISA analysis for the complex extract according to the present invention. The membrane was blocked in TBS-T (Tris buffered saline with 1% Tween 20) containing 5% BSA (bovine serum albumin) at room temperature, washed 3 times with TBS-T for 10 minutes, and then anti-5α-reduced with primary antibody Enzyme (5-AR) and anti-β-actin were used to react at 4° C. for 16 hours. Then, after washing 3 times for 10 minutes with TBS-T, the secondary antibody (goat anti-rabbit IgG HRP, rabbit anti-mouse IgG HRP) was reacted in 5% BSA at room temperature for 2 hours and washed 3 times for 10 minutes with TBS-T did.
