Abstract
The present invention relates to a preservative composition comprising the white rice extract as an active ingredient and a preparation method thereof.
Description
The present invention relates to a preservative composition comprising the white rice extract as an active ingredient and a method for preparing the same.
Food, industrial products, quasi-drugs, and medicines are the most used items in human life. For example, foods include instant foods, industrial products such as wipes and cosmetics, non-medicinal products such as toothpaste and pesticides, and medicines include anti-inflammatory analgesics.
These foods, industrial products, quasi-drugs and medicines are produced for a certain period of time from one day to several years before production and delivery to the actual consumer. After a certain period of time for the distribution of such foods, industrial products, quasi-drugs and pharmaceuticals, the product should be disposed of as bacteria or fungi develop and grow.
On the other hand, as described above, manufacturers of food, industrial products, quasi-drugs and pharmaceuticals add a preservative (preservative) in the product to increase the shelf life.
The preservatives used in the prior art have caused stability controversies in that they may be harmful to humans as synthetic preservatives.
It is an object of the present invention to provide a preservative composition comprising the white rice extract as an active ingredient and a preparation method thereof.
The object of the present invention is achieved by a preservative composition comprising an extract of Cynanchum atratum Bunge as an active ingredient.
The white rice extract may include beta amirin.
It may further comprise propanediol.
The propanediol may be 50 parts by weight to 400 parts by weight based on 100 parts by weight of the white rice extract.
Further comprising a natural component, the natural component may further comprise at least one of rosemary and moss.
An object of the present invention is a method for producing a preservative composition comprising a white rice extract, comprising: mixing the white rice with a solvent to obtain a mixed solution; Leaving the mixed liquor and filtering to obtain a filtrate.
The solvent may include at least one of chloroform, butanol, ethanol, methanol and propanediol.
The solvent may comprise 1,3 propanediol.
According to the present invention there is provided a preservative composition comprising the white rice extract as an active ingredient and a preparation method thereof.
Figure 1 shows the results of experiments on the antiseptic force against bacteria using a preservative composition prepared according to the present invention,
Figure 2 shows the results of experiments on the preservation of fungi (fungi) using the preservative composition prepared according to the present invention.
Hereinafter, the embodiments of the present invention will be described in more detail.
Prior to the description, the embodiments described below are intended to explain the present invention in more detail, and it is apparent that the scope of the present invention is not limited to these embodiments in accordance with the gist of the present invention.
White rice is a medicinal herb made from the roots of Cynanchum atratum Bunge or similar plants of the Panaxia family. In China, the root of the Cinnanchum versicolor Bunge is used. Roots and rhizomes are 15 ~ 50mm long, 5 ~ 12mm in diameter, with upper stems with round stem marks, and at the bottom end, several thin and long roots gather to form a horsetail. As used herein, 'white rice' includes Cynanchum atratum Bunge and Cynanchum versicolor Bunge.
Roots are slightly curved in a cylindrical shape, and the outer surface is yellowish brown to brown, and has a smooth or thin wrinkle pattern. The vagina is brittle and easy to bend. The plane is flat and the skin is yellowish white. The neck is relatively small and yellow. It has a slight smell and a bit bitter taste.
White rice makes urine go through the cold, cools the fever, cools the blood, detoxifies and treats skin diseases. The present invention uses white rice extract as a preservative composition in view that white rice contains antioxidant, anti-inflammatory and antibacterial ingredients.
Thus, the preservative composition according to the present invention comprises a white rice extract. The detailed method of obtaining white rice extract is mentioned later.
The white rice extract includes beta-amirin, and the beta-amirin may contain 0.002% to 1.0% by weight of white rice extract.
The preservative composition may further contain a solvent or natural ingredient, while containing the white rice extract as an active ingredient.
The solvent may be a solvent used in the extraction process or a solvent added to adjust the concentration. Natural ingredients can be ingredients with preservative performance or with other effects.
The solvent may comprise water (purified water) and / or propanediol, and propanediol may be 1,3 propanediol. Propanediol has an antiseptic effect on its own.
Natural ingredients can be rosemary and moss.
In addition to various natural preservatives such as protamine, chitosan, spices, special crops, enzymes such as lysozyme, iron-binding proteins such as lactoferin, polyphenols such as tannins and tea tannins, licorice extract, amino acids or derivatives thereof, and nisin, kojic It may further comprise a preservative component derived from microorganisms such as acid and polylysine.
In view of the composition ratio of the preservative composition, 100 parts by weight to 100 parts by weight of purified rice extract, 100 parts by weight to 10,000 parts by weight, 100 parts by weight to 5,000 parts by weight, 200 parts by weight to 5000 parts by weight, 300 parts by weight to 5000 parts by weight or 300 parts by weight Parts to 2000 parts by weight. Propanediol may be added to the preservative, and the amount may be 10 parts by weight to 1000 parts by weight, 50 parts by weight to 1000 parts by weight, 50 parts by weight to 400 parts by weight, or 100 parts by weight to 400 parts by weight.
The preservative composition according to the present invention is made of a natural component of the active ingredient is high in stability and excellent in preservation.
The preservative composition according to the present invention can be applied to various products such as food, medicine, quasi-drugs and cosmetics.
Hereinafter, a method for preparing a preservative composition according to the present invention will be described.
First, white rice is mixed with a solvent to obtain a mixed liquid.
The solvent may be chloroform, butanol, ethanol, methanol, propanediol, and the like, in particular 1, 3 propanediol.
The solvent may be used in an amount of 100 parts by weight to 10,000 parts by weight based on 100 parts by weight of white rice.
After mixing, the mixture is left at room temperature for a certain time (for example, about 24 hours) and then filtered to collect the filtrate. Filtration can be performed several times, for example three times.
The solvent in the filtrate is then concentrated under reduced pressure with a vacuum condenser to remove white rice extract. When a nonvolatile solvent such as 1,3 propanediol is used as the solvent, the solvent removal process may be omitted or reduced, and an insoluble portion of the white rice may be removed through a filtering process.
Purified water and / or natural ingredients are added to the obtained white rice extract (or a mixture of white rice extract and 1,3 propanediol and the like) to obtain a preservative composition. In other embodiments, additional processes such as purified water and / or natural ingredients may be omitted.
In addition, after obtaining a white rice extract with a solvent other than 1, 3 propanediol, 1, 3 propanediol may be added to obtain a white rice extract composition comprising white rice extract and 1,3 propanediol. In this case, after adding 1, 3 propanediol, it can be filtered to obtain a white rice extract composition.
The present invention will be described in more detail with reference to the following experimental examples.
White Rice Extract Production from White Rice and Solvent
30 g of white rice and 300 ml of solvent were mixed, and the mixture was left at room temperature for 24 hours and then filtered three times to obtain a filtrate. The filtrate was concentrated under reduced pressure with a vacuum concentrator to obtain a white rice extract.
Chloroform, butanol, ethanol, methanol and 1,3 propanediol were used as the solvent, respectively.
White Rice Extract Analysis
In order to analyze β-amyrin contained in the white rice extract, HPLC (High Performance Liquid Chromatography) analysis was performed under the conditions as shown in Table 1 below.
In order to analyze the content of beta-amirin contained in the white rice extract, a standard solution of beta-amirin was prepared.
The standard solution was dissolved in 10 mL of beta-amirin standard purchased from SIGMA ALDRICH in 1 mL of DMSO (dimethyl sulfoxide; dimethyl sulfoxide), and then diluted to a concentration of 1 ppm and 10 ppm using DMSO. .
The content of beta-amirin of the white rice extract obtained by mixing chloroform, butanol, ethanol, methanol and 1,3-propanediol with white rice is as follows.
As shown in Table 2, beta amirin was detected in all cases, especially when 1,3 propanediol was used, and a high content of beta amirin was detected.
Retention Test of White Rice Extract 1
White rice extract was obtained using three solvents, purified water, butanol and ethyl acetate. Purified water was added to the obtained white rice extract to prepare a preservative composition having a white rice extract of 3% by weight. The storage power test was done using the obtained preservative composition.
The retention test method was performed according to the CTFA Technical Guidelines "Preservation Testing of Cosmetics".
Inoculating microorganisms were 1) Bacteria: E. coli ATCC8739, P.aeruginosa ATCC9027, S.aureus ATCC25923, and 2) Fungi: Candida albicans ATCC10231, Aspergillus niger ATCC16404.
Bacteria or fungi were inoculated into the samples to constitute a bacterial medium and a fungal medium. Observations were made in units of 7 days from day 0 of inoculation.
As shown in Table 3, when purified water was used as the extraction solvent, the maintenance power was not exerted. This may be because the active ingredient of white rice was not extracted when purified water was used.
On the other hand, when butanol and ethyl acetate were used as extraction solvents, all of them showed a growth inhibitory effect against bacteria, and especially when ethyl acetate was used, they showed a growth inhibitory effect against fungi.
Of white rice extract Retention Test 2
The storage capacity (preservation) when 1,3-propanediol was used as a solvent was tested.
The sample was used by diluting the original preservative composition consisting of 30% by weight of white rice extract and 70% by weight of 1,3 propanediol with purified water.
The original preservative composition was obtained by adding 1,3 propanediol to the white rice extract obtained by using ethyl acetate and filtering.
Sample 1 consists of 10% by weight of the original preservative composition and 90% by weight of purified water, and Sample 2 consists of 20% by weight of the original preservative composition and 80% by weight of purified water.
The retention test results are shown in Table 4.
As shown in Table 4, it was confirmed that both Sample 1 and Sample 2 has a high growth inhibitory effect on bacteria. In addition, in the case of Sample 2 having a high content of white rice extract, it was confirmed that there was a high growth inhibitory effect against fungi. 1 and 2 show the growth inhibitory effect on bacteria and fungi, respectively, when sample 2 was used.
Thus, the natural composition containing white rice extract as an active ingredient can improve the food, industrial products, quasi-drugs or pharmaceutical preservation.
In particular, the cosmetic composition containing the white rice extract as an active ingredient can improve the shelf life of the cosmetic.
