Claims (7)
- 식물세포의 이종 개체군으로부터의 단클론 식물세포주의 발생 방법으로서,
(a) 식물세포의 이종 개체군 준비;
(b) 상기 식물세포의 이종 개체군으로부터의 원형질체 준비 및 세포벽 재생의 개시;
(c) 액체 배지 내 배양보조세포 재료(feeder cell material)를 함유하는 세포 축적 기기의 제공;
(d) 상기 준비된 원형질체에 FACS를 적용하여 단일 원형질체 분리;
(e) 배양보조세포 재료를 함유하는 상기 세포 축적 기기의 액체 배지 내로 상기 단일 원형질체를 분류;
(f) 상기 배양보조세포 재료의 존재하에서 공배양에 의한, 마이크로콜로니(microcolony)가 형성될 때까지 분리된 단일 원형질체의 재생; 및
(g) 상기 배양보조세포 재료로부터 마이크로콜로니의 제거 및 단클론 식물세포주가 형성될 때까지 상기 마이크로콜로니의 배양을 포함하며,
상기 (b) 단계에서 준비된 원형질체는 형질전환된 것으로,
(i) 형광 마커 단백질 또는 폴리펩타이드를 생산할 수 있고/있거나,
(ii) 선택 시약의 존재하에 생존가능한 것인, 식물세포의 이종 개체군으로부터 단클론 식물세포주의 발생 방법.
- 청구항 1에 있어서, (g) 단계에서 획득한 단클론 식물세포주의 완전한 번식능력이 있는 식물로의 재생을 더 포함하는, 단클론 식물세포주의 발생 방법.
- 청구항 1 또는 2에 있어서, (b) 단계에서 준비된 원형질체가 형질전환되고 목적하는 생산물을 생산할 수 있는, 단클론 식물세포주의 발생 방법.
- 청구항 3에 있어서, 목적하는 생산물이 이종 단백질 또는 폴리펩타이드, 이차 대사물, 및 진단 또는 분석 목적을 위한 마커로 이루어진 그룹으로부터 선택되는, 단클론 식물세포주의 발생 방법.
