Abstract
Description
도 2는 (A) 대나무 성체에서 측아 부위를 포함한 가지를 Y자형으로 마디 절편을 채취하여 멸균한 후 신초 유도배지에 치상한 모습; (B) 치상 후 2주 후에 측아에서 신초가 발아하여 자라난 모습이다.
도 3은 (A) 발아하여 2마디 이상 생장한 신초를 신초 증식배지에 옮긴 후 모습; (B) 신초가 생장하는 모습이다.
도 4는 신초를 매 3주마다 계대 배양하면서 (A) 계대배양 초기와, (B) 2차 계대배양 후 다중 신초 다발(다발성 신초)이 발생하는 모습을 나타내었다.
도 5 (A) 및 (B)는 다중 신초 다발(다발성 신초)을 2~3개의 신초 다발로 나누어 다시 계대배양한 모습이다.
도 6은 여러 개로 분화한 신초에 식물호르몬(NAA 0.5, 1, 2, 4, 6㎎/L)을 처리하면서 뿌리의 발근상태를 관찰한 모습이다: (A) 발근 초기모습, (B) 3주 후 유식물체로 발달한 모습.

- (a) 대나무의 가지에서 측아를 포함하는 마디 절편을 채취하여 멸균시키는 단계;
(b) 4~6 ㎎/L BAP, 2~4% 수크로오스 및 3~5 g/L 젤라이트가 첨가되고, pH 5.6~6.0으로 조절된 MS 배지에서 상기 멸균한 마디 절편을 배양하여 신초를 발생시키는 단계;
(c) 2~4 ㎎/L BAP, 2~4% 수크로오스 및 3~5 g/L 젤라이트가 첨가되고, pH 5.6~6.0으로 조절된 MS 배지에서 상기 발생한 신초를 계대배양하여 다발성 신초를 유도하는 단계;
(d) 0.5~1 mg/L BAP, 1~4 mg/L NAA, 1~2% 수크로오스 및 3~4 g/L 젤라이트가 첨가되고, pH 5.6~6.0으로 조절된 1/2 MS 배지에서 상기 유도된 다발성 신초를 배양하여 발근을 유도하는 단계; 및
(e) 0.5~1 mg/L BAP, 0.5~1 mg/L NAA, 1~2% 수크로오스 및 3~5 g/L 젤라이트가 첨가되고, pH 5.6~6.0으로 조절된 1/2 MS 배지에서 상기 발근이 유도된 다발성 신초를 배양하여 뿌리와 줄기를 성장시켜 유식물체를 얻는 단계;를 포함하는 기내배양에 의한 대나무 대량증식 방법.
- 4~6 ㎎/L BAP, 2~4% 수크로오스 및 3~5 g/L 젤라이트가 첨가된 MS 배지를 유효성분으로 함유하며, pH가 5.6~6.0으로 조절된, 대나무의 신초 유도용 배지 조성물.
- 2~4 ㎎/L BAP, 2~4% 수크로오스 및 3~5 g/L 젤라이트가 첨가된 MS 배지를 유효성분으로 함유하며, pH가 5.6~6.0으로 조절된, 대나무의 신초 증식용 배지 조성물.
- 0.5~1 mg/L BAP, 1~4 mg/L NAA, 1~2% 수크로오스 및 3~4 g/L 젤라이트가 첨가된 1/2 MS 배지를 유효성분으로 함유하며, pH 5.6~6.0으로 조절된, 대나무의 발근 유도용 배지 조성물.
