Abstract
Description
The present invention relates to a cleansing composition for women containing natural ingredients.
The quality of the woman is the connection between the vulva and the uterus, which is the most humid and warm place in the female body. Women's vagina is a suitable environment for bacteria or fungi, which is the main cause of vaginitis, and the disease is likely to occur.
In normal conditions, many lactic acid bacteria live in the vagina, making the inside of the vagina mildly acidic to inhibit the growth of pathogens and fungi. However, the use of antibiotics or long-term use of birth control pills, tampons or loops, pregnancy or childbirth, When the balance inside the vagina breaks down due to hormonal balance changes, vaginitis may occur due to the proliferation of opportunistic pathogens that normally inhibit growth under normal conditions in the vagina.
Vaginitis has been reported as bacterial vaginosis, Candida vaginitis, Trichomonas vaginitis and Atrophic vaginitis.
The most common disease is bacterial vaginosis, caused by Gardnerella vaginalis . Candida albicans , common with bacterial vaginosis, is caused by the fungus Candida albicans .
To prevent female diseases, cleaners (vaginal cleansers) and vulva cleaners are often used. As a local cleaning agent commonly used to prevent vaginitis, a product obtained by sulfonating fatty acids or distilled lauryl alcohol and synthesizing emollients, thickeners, nutrients, foaming agents and organic disinfectants Are being sold.
However, the chemical quality detergent has a limitation in that the detergent effect is lowered and the germs are removed instead of removing the germs, and the odor and unpleasant odor removing effect are inhibited.
It is also known that polyene-based drugs such as nystatin, amphotericin B, azole-based drugs such as ketoconazole, miconazole, There are imidazole drugs and triazole drugs such as itraconazole and fluconazole but resistance to antibiotics may cause further difficulties in the subsequent treatment.
Although various drugs have been proposed as methods for treating vaginal cleansing and abnormal symptoms, it is still not enough to present a non-irritating detergent, a cleanser or a drug in consideration that the site of use is the most sensitive mucosal site for external substances . Thus, there is a need to develop new technologies to meet the ongoing need for cleaners using natural extracts without irritation and side effects.
Meanwhile, the cosmetics industry is developing a number of products using natural products to reduce skin irritation caused by various chemicals. Fermentation oil (Preparation Example 1)
Fermentation oil (Preparation Example 2)
Experimental Example 1: Skin safety test
The toxicity of the samples of the Examples and Comparative Examples to the cells was measured. Samples of the examples and comparative examples were suspended in purified water to give concentrations of 100, 50 and 25 μg / mL, respectively, and cell viability was measured by the following method.
Cytotoxicity was performed by modifying the method of Mosmann to measure cell viability using MTT {3- (4,5-dimethylthiazol-2-yl) -2-5-diphenyltetrazolium bromide} reagent. HDF (Human Dermal Fibroblast) cells were seeded in a 96-well plate at a concentration of 1 × 10 4 cells / well and stabilized at 37 ° C and 5% CO 2 for 24 hours.
Samples of the examples and comparative examples were treated for each concentration and cultured for 24 hours. After the medium was removed, MTT was treated at a concentration of 5 mg / mL and cultured for 2 hours. 100 μL of DMSO (dimethyl sulfoxide) was added, stirred for 10 minutes, and absorbance was measured at 540 nm.
As a result of the measurement, no cytotoxicity was observed at all concentrations of each sample. The results suggest that the composite composition is harmless to the human body and is excellent in safety.
Experimental Example 2: Evaluation of antibacterial activity
The antimicrobial activity of the female cleaner composition including the samples of the above Examples and Comparative Examples was confirmed by using the disk diffusion method.
Yeast strains caused by the target causes the fungus Candida albicans (Candida albicans ) and the common bacterium Gardnerella ( Gardnerella < RTI ID = 0.0 > vaginalis ) was used.
100 쨉 l of Candida albicans and Gardnerella vaginalis, which had been pre-cultured, were plated on a plate medium (Sabouraud Dextrose Agar. The compositions of the Examples and Comparative Examples were placed on a sterilized paper disk at a concentration of 5 mg / disc and placed on a medium.
Itraconazole, an antifungal agent for Candida albicans, and Ampicillin, an antibiotic for Gardnerella vaginalis were used as a positive control, and they were placed on a paper disk at a concentration of 100 μg / disk and then placed on a medium I left it.
After the plate culture was incubated at 28 ° C for 48 hours, the size (mm) of the growth inhibition zone (mm) was measured and the results are shown in Table 2 below.
Referring to Table 2, the compositions of Examples 1 to 8 showed strong growth inhibition effects against Candida albicans and Gardnerella vajinalis.
Particularly, the antifungal and antimicrobial effects were further enhanced when the composition was applied with Daemark Rosaceae fermentation oils (Examples 2 and 3) and black tea extract (Example 4).
In addition, the compositions (Examples 5 to 8) in which Lavender flowering, Bitter orange flowering, Pine flowering and Elder flowering were mixed showed remarkably excellent growth inhibition effect.
Experimental Example 3: Assessment of inhibition of microbial growth
Effects of Lactobacillus casei and Lactobacillus casei on the growth of Candida albicans and Gardnerella vaginalis , which are known as lactic acid bacteria that grow in female vagina, .
The presence of increased turbidity in the medium was confirmed and the IC 50 (half maximal inhibitory concentration) value required to inhibit the antimicrobial activity and the microorganism by 50% was measured using the Broth Microdilution Susceptibility Test Respectively.
Specifically, each composition was diluted with Sabouraud Dextrose medium to a cell number of 2 × 10 5 cells / ml, and the compositions of the examples and the comparative examples were treated at a concentration of 50 mg / ml in 96-wells. After incubation at 28 ° C for 24 hours, the absorbance was measured at a wavelength of 570 nm using a microplate reader (BIO-RAD) to determine IC 50 values. The results are shown in Table 3 below.
Referring to Table 3, the compositions of Examples 1 to 8 hardly affected the proliferation of lactobacillus casei, which is a lactic acid bacterium, but effectively inhibited the growth of Candida bacillus and Gardnerella vaginalis.
In particular, in Comparative Example 1 in which only damask rose oil was applied, the growth of Lactobacillus casei was partially affected, and the growth inhibitory effect on Candida albicans and Gardnerella vaginalis was somewhat insufficient.
The compositions of Examples 1 to 8 inhibited the growth of Candida bacillus and Gardnerella, but did not affect the growth of Lactobacillus casei. The results show that the microbial-specific antimicrobial effect of the compositions of Examples 1 to 8 Suggesting that it is very likely to be used as a female cleaner.
