Abstract
본 발명은 자연 살해세포의 배양 시 활성산소 억제제 및/또는 p53 억제제를 처리하는 단계를 포함하는 배양방법 및 이를 포함하는 조성물에 관한 것으로, 본 발명에 의하는 경우 활성산소와 p53 단백질의 작용을 억제함으로서 암세포 살상 기능을 유지하면서도 자연 살해세포의 증식 효율을 높일 수 있다.
Description
본 발명은 자연 살해세포를 증식하는 방법 및 배양용 조성물에 관한 것이다.
인체의 면역계는 복잡한 기전에 의해서 조절이 되며 면역계에 이상이 생길 경우 면역시스템에 불균형이 초래되어 암과 같은 각종 난치성 질환이 발생할 수 있다. 따라서 면역계에서 발생하는 불균형을 해소하여 정상상태로 회복, 강화시켜 면역관련 질환을 치료하는 방법인 면역세포치료제의 개발이 각광을 받고 있다.
우리 몸의 면역계는 선천성 면역계와 후천성 면역계로 나뉘며 선천면역계는 우리 몸에 외부 항원이 들어오면 제일 먼저 공격을 하는 세포들로 구성되어 있으며 대표적인 세포로는 자연 살해 세포가 있고 다양한 종류의 암세포의 살상이 가능하고 항원유무에 관계없이 암세포를 인식할 수 있다는 장점이 있기 때문에 세포치료제로 주목을 받고 있다.
면역세포를 세포치료제로 사용하기 위해 필요한 것은 다량의 세포 수 확보, 세포의 높은 항암능력이 무엇보다 중요하다. 또한, 이렇게 확보된 세포들이 환자의 체내 주입 시 효과적으로 체내에서 생존을 해야 한다.
하지만 체외에서 배양된 면역 세포는 사실 그리 오래 활성을 유지하지 못한다. 특히나 과도하게 확장되다 보면 필연적으로 세포사 또는 세포 노화에 빠지기 쉬워서 배양된 면역 세포가 환자의 체내에서 오랫동안 안정성을 유지해야만 세포치료제로써 제대로 효력을 발휘할 수 있다.
일반적으로 세포사 및 세포노화는 다양한 세포성 스트레스에 의해 야기된다.
활성산소는 산소의 정상적인 대사작용에 의해 자연스럽게 생기며 세포신호와 항상성에 중요한 역할을 한다. 하지만 환경적 스트레스로 인해 빠르게 증가하는 활성산소는 세포에게 산화적 스트레스를 주고 세포사 또는 노화와 같은 문제를 야기한다.
활성산소는 주로 미토콘드리아 또는 NADPH 산화효소(NADPH oxidase)에 의해 발생되며 낮은 수준의 활성산소는 세포 내 신호전달 물질로 작용하지만 과량으로 존재하는 활성산소는 DNA, 단백질, 지질과 같은 거대분자에 손상을 줄 수 있으며 최종적으로 세포 노화나 세포사를 유도할 수 있다.
p53 유전자는 인체의 모든 세포에 들어 있는 유전자로 알려져 있고 암세포에서의 역할이 많이 보고되고 있으나 면역세포 관련하여 연구가 미비하다. p53은 종양 억제 유전자 중 하나로 p53의 변이가 생겨서 DNA repair, apoptosis, autophagy, metabolism에 문제가 발생되어 암이 발생되는 것으로 보고되고 있다.
현재 암환자의 50% 이상이 p53유전자가 기능을 상실해 생기는 것으로 알려져 있으며 p53 유전자를 사람의 방광암 세포에 주입한 결과 세포에 노화가 일어나서 암세포의 증식이 완전히 억제가 된다는 보고가 있다.
본 발명의 목적은 활성산소 억제제와 p53 억제제를 처리하여 인간유래 자연 살해세포의 확장능을 현재보다 효율적으로 증대 및 활성을 유지시키는 방법을 제공하는 데 있다..
본 발명의 다른 목적은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는 자연 살해세포(NK cell) 배양 또는 보관용 조성물을 제공하는데 있다.
본 발명의 또 다른 목적은 상기 방법으로 제조된 자연 살해세포를 포함하는 암의 치료용 조성물을 제공하는 데 있다.
상기 목적을 달성하기 위하여, 본 발명자는 자연 살해 세포를 확장 배양하는 과정 중 발생하는 세포사를 극복하여 더 많은 수의 자연살해 세포를 획득하고자 연구하였으며, 다수의 활성산소 및 p53과 관련된 다수의 저해제들을 이용하여 체외 자연 살해세포 자연 살해세포의 암세포 살상능은 유지되며 자연 살해세포의 세포사를 극복하여 배양효율을 높일 수 있는 물질을 발굴하고, 나아가 상기 저해제들을 테스트하여 본 발명을 완성하였다.
본 발명은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 처리하는 단계;를 포함하는 자연 살해세포(NK cell)의 증식 방법을 제공한다.
또한, 본 발명은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 유효성분으로 자연 살해세포(NK cell) 배양용 또는 보관용 조성물을 제공한다.
또한, 본 발명은 상기 방법에 따라 얻어진 자연 살해세포를 유효성분으로 포함하는 암의 예방 또는 치료용 약학 조성물을 제공한다.
본 발명의 자연 살해세포 대량증식 방법에 따라 활성산소 억제제 및/또는 p53 억제제를 처리하여 자연 살해세포를 배양하면 자연 살해세포의 기능에는 저하가 없으면서 기존 배양 방법에 비해 더 많은 세포수의 자연 살해세포를 얻을 수 있어 세포치료제의 생산을 위한 자연 살해세포를 효율적으로 확보 할 수 있다.

본 발명은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 처리하는 단계;를 포함하는 자연 살해세포(NK cell)의 증식 방법 및 상기 방법에 의하여 얻어진 자연 살해세포를 포함하는 암의 예방 또는 치료용 조성물, 그리고 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는 자연 살해세포의 배양 또는 보관용 조성물을 제공한다.
이하, 본 발명의 구성을 구체적으로 설명한다.
본 명세서에 있어서, 특별한 언급이 없는 한, 용어 "치료"는 질환과 관련된 임상적 상황을 억제하거나 완화하거나 이롭게 변경하는 모든 행위를 의미한다. 또한, 치료는 치료를 받지 않은 경우 예상되는 생존율과 비교하여 증가된 생존을 의미할 수 있다. 치료는 치료적 수단 이외에 예방적 수단을 동시에 포함한다.
본 명세서에 있어서, 특별한 언급이 없는 한, 용어 "개체"는 척추동물, 바람직하게는 포유동물, 예를 들어, 개, 고양이, 생쥐, 인간 등일 수 있다.
본 발명은 자연 살해세포의 증식 방법을 제공한다.
본 발명의 증식 방법은 자연 살해세포의 수득량을 늘릴 수 있는 자연 살해세포의 배양방법 또는 자연 살해세포를 대량으로 증식하는 방법 일 수 있다. 또한, 상기 증식 방법은 자연 살해세포의 노화 또는 세포사를 방지하여 수득량을 증가시킬 수 있으므로, 본 발명의 증식방법은 자연 살해세포의 노화 또는 세포사 억제를 위한 처리방법이거나, 증식 또는 배양된 자연 살해세포의 활성을 유지시키기 위한 보관방법 일 수 있다.
상기 증식 방법은 바람직하게 체외에서 증식하는 방법을 의미한다.
상기 자연 살해세포는 인간유래 일 수 있고, 바람직하게는 인간 말초혈액단핵구 유래 자연 살해세포 일 수 있다. 상기 말초혈액단핵구(Peripheral Blood Mononuclear Cell, PBMC)는 항암면역치료를 위하여 통상적으로 사용되는 말초혈액(Peripheral Blood)으로부터 분리된 단핵구(Mononuclear Cell)를 의미한다. 말초혈액단핵구는 채혈한 사람의 혈액을 피콜-하이팩 밀도구배 분리법(Ficoll-Hypaque density gradient method)과 같은 공지의 방법을 사용하여 수득할 수 있다.
본 발명에서 사용되는 말초혈액단핵구는 정상인, 암의 위험성을 갖는 환자 또는 암 환자로부터 얻을 수 있고, 반드시 자가 유래일 필요는 없으며, 동종 유래의 말초혈액 단핵구라면 본 발명에 따른 항암면역치료를 위한 자연 살해세포의 유도 및 증식을 위해 사용할 수 있다. 상기 말초혈액단핵구의 공여자(dornor)에 따라서, 증식되는 세포의 절대 수에 차이가 있을 수 있으나, 동일 공여자에서 자연 살해세포에 대하여 배양방법에 따른 증식 효율은 본 발명의 억제제를 처리하는 경우가 가장 우수함을 확인하였다..
상기 증식 방법은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상의 억제제를 처리하는 단계를 포함할 수 있다.
본 발명의 일 실시예에서, 상기 처리는 말초혈액단핵구로부터 유도 및 증식된 자연 살해세포를 1 내지 3주 동안 활성산소 억제제 및 p53 억제제 중 하나 이상의 존재하에서 배양하는 것 일 수 있다. 더욱 구체적으로, 상기 억제제 중 하나 이상을 3일 간격으로 세포에 처리하여 배양하는 것일 수 있다.
따라서, 상기 증식 방법은 말초혈액단핵구로부터 자연 살해세포를 유도 및 증식하는 단계; 및 상기 유도 및 증식된 자연 살해세포를 활성산소 억제제 및 p53 억제제 중 하나 이상의 존재하에서 1 내지 3주 동안 인큐베이션하는 단계;를 포함할 수 있다.
상기 활성산소 억제제는 세포 내에서 활성산소(oxygen free radical, ROS)의 생성을 저해하는 물질을 의미하는 것으로, 상기 활성산소란 호흡과정에서 몸속으로 들어간 산소가 산화과정에 이용되면서 여러 대사과정에서 생성될 수 있는 불안정한 상태의 산소로, 몸속에서 산화작용을 일으키고, 세포 구조의 손상을 초래할 수 있다.
상기 활성산소 억제제는 활성산소의 발생을 억제할 수 있는 것이라면 그 종류에 한정되지 않고, 구체적으로, 아포시닌(apocynin, 1-(4-hydroxy-3-methoxyphenyl)-ethanone), 트롤록스(trolox, 6-hydroxy-2,5,7,8-tetramethylchroman-2-Carboxylic Acid), 피롤리딘 디티오카바메이트(PDTC, pyrrolidine dithiocarbamate), 글루타치온(GSH, glutathione), 카탈라아제(catalase), 망간-초과산화물불균등화 효소(MnSOD, manganese superoxide dismutase), 비타민 E(Vitamin E) 및 퀘르세틴(Quercetin)으로 이루어진 군에서 선택된 하나 이상일 수 있으며, 바람직하게는 아포시닌, 트롤록스 및 이들의 조합 중 어느 하나 일 수 있다. 상기 억제제를 처리하여 자연 살해세포를 배양하는 경우 세포사 및 노화를 막아 세포의 생존률을 높여주며, 이로 인하여 배양에 의한 증식 효율을 높일 수 있다.
상기 아포시닌(apocynin)(4-hydroxy-3-methoxyacetophenone)은 포시눔 칸나비눔(pocynum cannabinum)에서 처음 분리한 물질로 활성산소 형성 관련 효소인 NADPH oxidases 억제제로 사용되는 화합물이다. 따라서 세포에 아포시닌(apocynin)을 처리 시 NADPH oxidase를 억제함으로 최종적으로 활성산소의 생산을 억제하는 것으로 알려져있다.
트롤록스(trolox)(6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid)는 Vitamin E의 아날로그(analog)로 수용성이며 처리 시 산화 스트레스 또는 손상을 감소시키는 역할을 할 수 있다.
상기 p53 억제제는 p53 유전자 또는 단밸질의 발현을 억제 또는 저해할 수 있다면 그 종류에 한정되지 않고, 일 예로 피피트린-α(pifithrin-α, 2-(2-Imino-4,5,6,7-tetrahydrobenzothiazol-3-yl)-1-p-tolylethanone hydrobromide) 및 피피트린-μ(Pifithrin-μ)로 이루어진 군에서 선택된 하나 이상일 수 있고, 바람직하게는 피피트린-α일 수 있다. 상기 p53 억제제를 처리하여 자연 살해세포를 배양하는 경우 세포사 및 노화를 막아 세포의 생존률을 높여주며, 이로 인하여 배양에 의한 증식 효율을 높일 수 있다.
피피트린-α(pifithrin-α)는 p53 저해제(또는 억제제, inhibitor)로 일반적으로 p53 매개 세포사, p53 의존적 유전자 (cyclin G, p21/waf1) 전사를 조절하여 세포의 생존을 증가시키는 것으로 알려져있다.
또한, 상기 억제제는 활성산소 억제제와 p53 억제제를 모두 포함할 수 있다. 일 예로, 트롤록스와 피피트린-α를 50 내지 150 : 1의 몰비, 바람직하게는 80 내지 120 : 1의 몰비로 포함하여 처리될 수 있다. 상기와 같이 활성산소 억제제와 p53 억제제를 함께 처리하는 경우, 자연 살해세포의 배양효율을 더욱 높일 수 있다.
본 발명의 일 실시예에서, 상기 억제제의 자연 살해세포 배양에 미치는 영향을 알아보기 위하여, 상기 자연 살해세포에 활성산소 억제제 또는 p53 억제제를 각각 단독처리 또는 병용처리하여 배양한 군과 아무런 억제제도 처리하지 않은 군(대조군)과 배양일 수에 따른 세포의 수득량을 확인하였고, 억제제 처리군 모두에서 대조군보다 많은 세포가 얻어졌음을 실험적으로 확인 하였으며(도 3 내지 도 6), 상기 배양방법에 의하여 얻어진 자연 살해세포의 경우 살상능 등이 종래 자연 살해세포 수준과 거의 동등 또는 우수하면서도, 활성 산소 생산량이 낮아 노화 또는 세포사에 더욱 강한 것을 확인 하였다(도 7 내지 도 9).
상기 억제제는 세포수 2.5x106 cells를 기준으로 상기 억제제를 1nM 내지 500 uM로 처리하는 것 일 수 있다. 상기와 같이 억제제를 처리하여 자연 살해세포를 배양하면 세포의 기능에는 변화가 없으면서 기존 배양 방법에 비해 더 많은 세포수를 얻을 수 있어 세포치료제의 생산 필요한 자연 살해세포를 더욱 효율적으로 확보 할 수 있다.
구체적으로, 세포수 2.5x106 cells를 기준으로, 아포시닌의 경우 100 uM 내지 800 uM, 200 uM 내지 600 uM, 또는 300 uM 내지 500 uM을 처리할 수 있고, 트롤록스는 10 내지 80 uM, 25 uM 내지 65 uM, 또는 35 uM 내지 55 uM을 처리할 수 있고, 피피트린-α의 경우 0.05 uM 내자 20 uM, 0.5 uM 내지 5 uM, 또는 0.7 uM 내지 3 uM을 처리할 수 있다. 또한, 트롤록스와 피피트린-α를 병용처리하는 경우, 트롤록스 25 uM 내지 65 uM, 또는 35 uM 내지 55 uM 와 피피트린-α를 100 nM 내지 800 nM, 또는 300 nM 내지 600 nM을 병용처리 할 수 있다.
상기 억제제의 처리는 1일 내지 10일 간격으로 하루에 한 번 이상 처리할 수 있고, 바람직하게는 1일 내지 5일 간격으로 하루에 한 번 이상 처리할 수 있으며, 또는 바람직하게는 2일 내지 4일 간격으로 하루에 한 번 이상 처리하면서 수행될 수 있다. 상기의 주기로 억제제를 처리하는 경우, 억제제의 독성이 자연 살해세포에 미치지 않아, 증식에 있어서 배양 후 수득률을 증가시킬 수 있다.
상기 처리는 자연 살해세포 또는 상기 세포의 배양배지 또는 배양조성물에 처리하는 것을 모두 포함한다.
상기 증식 방법은 상기 자연 살해세포를 배양하는 단계 전에, 말초혈액단핵구(PBMC)로부터 자연 살해세포를 유도 및 증식하는 단계를 더 포함할 수 있다. 상기 단계는 1일 내지 20일 동안, 바람직하게는 8일 내지 12일 동안 수행될 수 있고, 체외에서 배양하는 것 일 수 있다.
또한, 유도 및 증식단계는 말초혈액단핵구를 영양세포와 공배양 하는 것 일 수 있고, 바람직하게는 방사선 조사된 영양세포와 공배양 하는 것 일 수 있으며, 더욱 바람직하게 사이토카인의 존재하에서 공배양하는 것 일 수 있다. 상기 영양세포는 Jurkat 세포주, EBV-LCL 및 이들의 조합으로 이루어진 군에서 선택된 어느 하나 일 수 있다. 상기 영양세포와 공배양하는 경우 자연 살해세포로의 유도 및 증식 효율을 더욱 높일 수 있다.
상기 “Jurkat 세포” 또는 “Jurkat 세포주”는 혈액암(immortalized acute T cell leukemia) 세포주로 University of California at San Francisco의 Dr. Arthur Weiss가 개발한 세포주이다. 다양한 케모카인 수용체가 발현하고 있으며 IL-2를 생산할 수 있는 세포로서, 이는 세포 표면에 자연 살해세포 활성억제 인자인 MHC class I을 높이 발현하기 때문에 항암면역치료를 위한 영양세포(feeder cells)의 후보로 가능성이 전혀 대두되지 못했던 세포주였으나, 본 발명자들이 말초혈액 단핵구로부터 자연 살해세포로의 분화 및 증식을 위해 다수의 혈액암 세포주를 스크리닝하여 찾아낸 결과 Jurkat 세포를 영양세포로서 사용할 수 있음은 밝힌 바 있다 (국내 특허출원 제10-2010-0078777호 참조). 본 발명에 사용되는 Jurkat 세포는 ATCC로부터 입수할 수 있다(ATCC TIB-152).
본 발명에 있어서, 배지는 자연 살해세포의 배양 또는 말초혈액단핵구로부터 자연 살해세포로의 유도 및 증식에 사용하고 있는 통상의 배지를 제한 없이 사용할 수 있고, 일 예로 RPMI, DMEM, xvivo10, x-vivo20, cellgro SCGM 및 RPM1640 배지를 사용할 수 있으나 이에 한정되는 것은 아니다.
본 발명에 있어서, 그 외 배양조건은 통상의 자연 살해세포 배양 조건에 따른다.
또한, 본 발명은 활성산소 억제제 및 p53 억제제로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는 자연 살해세포(NK cell) 배양용 조성물을 제공한다.
상기 배양용 조성물을 이용하여 자연 살해세포를 배양하는 경우, 세포의 살상능을 향상시키면서, 세포의 사멸 및 노화를 예방하여 더욱 많은 자연 살해세포를 증식 및 수득할 수 있는 효과가 있음을 본 발명의 일 실시예에서 실험적으로 확인하였다.
상기 배양용 조성물의 사용법에 제한은 없고, 일 예로 자연 살해세포에 직접 처리하여 사용할 수 있고, 자연 살해세포를 배양하는 배양배지 또는 배양 조성물에 혼합하여 사용할 수 있다.
상기 배양을 위한 배지 및 그 외 배양 조건은 본 명세서에서 별도로 정의 또는 제한하지 않는 한 상기와 같이 통상적으로 사용되는 배지 및 조건에 따라 이용할 수 있다.
또한, 본 발명은 본 발명의 증식 방법으로 얻어진 자연 살해세포를 유효성분으로 포함하는 암의 예방 또는 치료용 조성물, 암의 예방 또는 치료를 위한 약학 조성물의 제조를 위한 상기 자연 살해세포의 용도, 또한, 본 발명의 증식 방법으로 얻어진 자연 살해세포 또는 이를 포함하는 조성물의 유효량을 대상체에 투여하는 것을 포함하는 암의 예방 및 치료 방법을 제공한다.
본 발명의 일 실시예에서, 본 발명의 증식방법에 의하여 얻어진 자연 살해세포의 경우 세포의 살상능 등에 있어서 기능저하가 없고, 활성산소의 생성도 낮아 노화에 따른 세포의 사멸위험이 적음을 실험적으로 확인하였는바, 본 발명에 따른 자연 살해세포는 암의 예방 및 치료를 위한 용도로 유용하게 사용될 수 있다. 특히 세포치료제의 효과 및 안정성의 향상에 더욱 효과적 일 수 있다.
상기 암은 암으로 분류되는 것이라는 모두 포함하고, 고형암, 혈액암 등 그 종류에 특별히 제한되지 않는다.
상기 대상체는 암의 예방 및/또는 치료가 필요한 하는 인간 일 수 있고, 암환자뿐 아니라 암의 발명 위험을 갖는 환자 또는 정상인도 모두 포함한다.
상기 약학 조성물은 인 비트로, 인 비보 또는 엑스 비보에서 진단적 또는 치료적 용도에 적합한 조성물을 이루는 활성성분 및 활성 또는 무활성 약학적으로 허용 가능한 담체를 포함할 수 있다.
상기 약학 조성물은 약학적으로 허용 가능한 담체를 추가로 포함하여 제제화될 수 있다.
상기 "약학적으로 허용 가능한 담체"란 생물체를 상당히 자극하지 않고 투여 성분의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 본 발명에서의 약학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 또는 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액 및 정균제 등 다른 통상의 첨가제를 첨가하여, 조직 또는 장기에 주입하기에 적합한 주사제의 형태로 제형화할 수 있다. 또한, 등장성 멸균 용액, 또는 경우에 따라 멸균수나 생리식염수를 첨가하여 주사 가능한 용액이 될 수 있는 건조 제제(특히 동결 건조제제)로 제형화할 수도 있고, 필요에 따라 적절한 성분을 포함하는 수용액에 적절한 농도로 상기 자연 살해세포가 현탁되어 있는 형태로 제제화 될 수 있다.
또한, 바람직하게 본 발명의 조성물은 충진제, 부형제, 붕해제, 결합제 및 활택제 등을 추가로 포함할 수 있다. 또한, 본 발명의 조성물은 포유동물에 투여된 후 활성성분의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 당업계에 공지된 방법을 사용하여 제형화 될 수 있다.
본 발명에서 용어, "투여"는 어떠한 적절한 방법으로 환자에게 본 발명의 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다. 복강내 투여, 정맥내 투여, 동맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 직장내 투여 될 수 있으나, 이에 제한되지는 않는다.
상기 유효량은 목적하는 치료되어야 할 특정 질환의 발병 또는 진행을 지연하거나 전적으로 중지, 또는 치료 효과를 이루는데 필요한 양을 의미한다. 본 발명에서 조성물은 약학적 유효량으로 투여될 수 있다. 적합한 총 1일 사용량은 올바른 의학적 판단범위 내에서 결정될 수 있다는 것은 당업자에게 자명한 일이다. 본 발명의 목적상, 특정 환자에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적인 질환의 종류, 중증도, 조성물에 함유된 다른 성분의 종류 및 함량, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다. 일 예로 성인의 경우 1일 1회 내지 수회 투여 시, 본 발명의 말초혈액단핵구 유래 자연 살해세포는 1X106 cells/kg 내지 1X1011 cells/kg, 예컨대 1X106 cells/kg 내지 1X108 cells/kg의 용량으로 투여할 수 있으나, 이에 제한되는 것은 아니다.
이하, 본 발명을 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.

실시예
]
준비예 1: 자연 살해세포의 제조 및 배양
사람의 혈액을 채혈한 후, Ficoll(Ficoll-paqueTM PLUS, GE healthcare)을 이용하여 2500 rpm에서 30분간 원심분리 한 후 연막층(buffy coat)에서 말초혈액 단핵세포를 분리하였다.
그 후 100 Gy로 방사선 조사한 Jurkat 세포주와 EBV-LCL 세포주를 사용하여 IL-2 500 U/ml 존재 하에 PBMC: KL-1: EBV-LCL을 1:0.5:0.5의 비율로, RPMI1640 배지에 10% FBS와 1% 페니실린/스트렙토마이신(penicillin/streptomycin)을 넣은 hRPMI 배지에 공배양하여 2-3일 마다 한번씩 IL-2가 500 U/ml로 첨가된 hRPMI 배지로 교환을 하면서 배양을 하였다.
시험예
1: 활성산소 및/또는 p53 억제제가 자연
살해세포의
배양에 미치는 영향 확인
준비예 1과 같이 자연 살해세포를 배양함에 따라 자연살해세포에 발현하는 단백질에 어떠한 변화가 있는지 확인하고자 하였다. 세포주기 관련 마커인 pRb, 세포노화 마커인 P53, P21, ER stress 마커인 Bip, CHOP, DNA damage 마커인 γH2AX, 세포사 마커인 PARP, cl-caspase-7을 확인하였다.
그 결과를 도 2에 나타내었고, 대부분의 마커들이 13 내지 16일을세포 기점으로 피크를 이루는 것을 확인하였다.
활성산소 억제제가 자연 살해세포에 미치는 영향을 보기 위하여 준비예 1의 조건으로 10일간 자연 살해세포를 배양한 후, 활성산소 억제제(아포시닌(apocynin, Calbiochem) 400uM, 트롤록스(trolox, Santa cruz) 50uM) 또는 p53 억제제인 피피트린-α(pifithrin-α, Santa cruz) 1uM를 각각 처리하거나, 또는 50 uM 트롤록스와 500 nM 피피트린-α를 병용 처리하였다. 3일 간격으로 처리하며 총 2주간 배양한 후 배양된 세포수를 확인하였다. 이 때, 배지는 3일에 한 번씩 IL-2가 500 U/ml로 첨가된 hRPMI 배지로 교환하였고, 세포수는 헤마토사이토미터를 이용하여 계수하였다. 억제제의 처리 유무에 따른 배양된 세포수 변화를 비교하여 그 결과는 도 3 내지 도 6에 나타내었다.
도 3 및 도 4에 나타낸 바와 같이, 활성산소 억제제인 아포시닌(apocynin) 400 uM로 처리한 그룹과 트롤록스(trolox) 처리 그룹 각각 모두 대조군에 비하여 배양된 세포수가 통계적으로 유의하게 증가되는 것을 알 수 있었고, 도 5에 나타낸 바와 같이, p53 억제제인 피피트린 처리 시 대조군에 비하여 배양된 세포수가 통계적으로 유의하게 증가됨을 확인 하였다.
또한, 도 6에 나타낸 바와 같이, 50 uM 트롤록스와 500 nM 피피트린-α을 병용 처리한 그룹에서 대조군에 비하여 세포수가 통계적으로 유의하게 증가되는 것을 알 수 있었다.
시험예 2: 자연 살해세포의 암세포 살상능 및 기능 변화 측정
물질 처리 후 자연 살해세포의 암세포 살사능을 확인하기 위한 실험을 실시하였다.
타겟(Target) 세포로 K562, A375 세포를 준비한 후 크롬(Chromium)으로 1시간 동안 표지(labeling)한 후 자연 살해세포와 적절한 비율로 공동 배양하고, 4시간 후 상층액을 취해서 감마 카운터(gamma counter)로 동위원소 값을 확인하였다. 아무것도 처리하지 않는 대조군과 아포시닌(apocynin), 트롤록스(trolox), 피피트린-α(pifithrin-α) 및 트롤록스+피피트린-α를 각각 처리한 군으로 나누어 실험을 실시하였고, 그 결과를 도 7에 나타내었다.
도 7에 나타낸 바와 같이, 본 발명에 따른 활성산소 억제제 및/또는 p53 억제제로 처리하여 배양한 경우에도, 아무것도 처리하지 않은 대조군과 동일한 수준으로 암세포 살상능이 유지되는 것을 알 수 있다.
또한, Target 세포로 K562, A375 세포를 준비한 후 자연 살해 세포와 target 세포를 1:1 비율로 섞어 1시간 동안 37℃에서 배양하고 FITC labeled anti CD107a mAb를 2.5ul 넣고 Golgi stop(BD pharmingen)을 넣은 후 5시간 동안 37℃에서 배양한다. 배양이 끝난 후 Percp-표지된 CD3 mAb, APC-표지된 CD56mAb를 넣고 4℃에서 20분간 반응하였다. Intracellular FACS를 위해 세포를 FACS buffer로 세척하고 고정시킨 후 BD cytoperm/cytofix kit(BD Pharmingen, San Diego, CA)로 투과시켜 PE-표지된 IFN-g mAb로 염색한 후 유세포 분석기를 이용하여 분석하였다. 아무것도 처리하지 않는 미처리군과 아포시닌(apocynin), 트롤록스(trolox), 피피트린-α(pifithrin-α) 및 트롤록스+피피트린-α를 각각 처리한 군으로 나누어 실험을 실시하였고, 그 결과를 도 8에 나타내었다.
도 8에 나타낸 바와 같이, 활성산소 억제제 및/또는 p53 억제제를 처리하여 배양한 경우에도, 자연 살해세포의 CD107a(살상능 척도)의 발현 및 IFN-g(사이토카인)의 분비에 있어서도 저하됨 없이 자연 살해세포의 살상능 등이 유지되는 것을 실험적으로 확인 하였다.
실험예 3: ROS 형성 측정
체외 확장시킨 자연 살해세포의 활성 산소 형성을 측정하기 위하여, 각각의 억제제를 단독 또는 병용 처리하여 배양한 자연 살해세포를 수거한 후, 1x106 개의 세포를 24 well plate에서 세포를 넣고, DCF-DA 100uM을 5% CO2 배양기에 30분 동안 배양한 후 세포를 수거하여 PBS 1ml로 1회 세척한 다음 유세포 분석기로 분석하였다. 아무것도 처리하지 않는 대조군과 아포시닌(apocynin), 트롤록스(trolox), 피피트린-α(pifithrin-α) 및 트롤록스+피피트린-α를 각각 처리한 군으로 나누어 실험을 실시하였고, 그 결과를 도 9에 나타내었다.
도 9에 나타낸 바와 같이, 트롤록스(trolox) 처리군에서 대조군에 비해 월등히 ROS 형성이 감소하는 것을 알 수 있었고, 트롤록스(trolox) < 피피트린-α(pifithrin-α) < 트롤록스+피피트린-α < 아포시닌(apocynin) < 대조군(미처리) 순서로 ROS 형성이 감소됨을 확인하였다.
따라서 본 발명의 억제제를 처리하는 경우, 기존의 배양방법 더 많은 수의 세포를 수확할 수 있고, 더욱이 본 발명에 따라 배양된 세포는 활성산소 형성은 감소되면서 살상능은 유지되어 기능적으로 더 우수하였으므로, 암의 예방 또는 치료를 위한 세포치료제 등에 효과적으로 사용할 수 있음을 실험적으로 확인하였다.
