레인 M: 100 bp DNA 마커
레인 1: DNA 앱타머로부터 PCR 기법을 통하여 증폭된 dsDNA 결과
레인 2, 3: PCR 기법으로 확보한 dsDNA를 열-냉각기법과 스트랩타비딘을 이용하여 제작한 ssDNA 결과
도 2는 본 발명의 일실시예에 따른 BoNT/C1 단백질과 특이적으로 결합하는 DNA 앱타머 제작을 위하여 실시한 각 SELEX 라운드에서 회수된 His 융합 BoNT/C1 단백질과 특이적으로 결합하는 DNA 앱타머 또는 BoNT/C1 단백질과 특이적으로 결합하는 DNA 앱타머 후보군들의 ssDNA 농도를 Nanodrop spectrophotometer를 이용하여 정량적으로 나타낸 결과이다.
도 3 및 도 4는 본 발명의 일실시예에 따른 BoNT/C1 단백질과 특이적으로 결합하는 DNA 앱타머 제작을 위한 SELEX 과정에서 선별된 15개의 BoNT/C1 단백질과 특이적으로 결합하는 DNA 앱타머 후보군 BoNT/C1-1 내지 BoNT/C1-15의 2차 구조를 나타낸 도면이다.
도 5는 카르복실기를 가지고 있는 CM5 센서 칩 표면에 BoNT/C1을 고정하는 단계를 나타낸 도면이다.
도 6은 BoNT/C1 결합 DNA 앱타머를 이용하여 ELISA 기법을 기반으로 BoNT/C1을 탐지하는 방법에 대한 모식도로서, 패널 [1]은 BoNT/C1 탐지를 위해 BoNT/C1 결합 DNA 앱타머를 이용한 ELISA 법의 모식도를 나타내며, 패널 [2]는 기존 항체를 이용하여 BoNT/C1을 탐지하는 ELISA 법의 모식도를 나타낸 것이다.
도 7은 BoNT/C1 단백질과 특이적으로 결합하는 앱타머를 이용한 조툴리즘 C형 표적인자인 BoNT/C1 검출 키트의 모식도를 나타낸 도면이다. 보툴리즘 C형으로 의심되는 가축의 시료를 BoNT/C1 단백질과 특이적으로 결합하는 앱타머가 고정되어 있는 진단 키트에 처리하였을 때, 샘플 내 BoNT/C1 단백질과 BoNT/C1 단백질 결합 앱타머의 결합에 의해 BoNT/C1을 진단하는 시스템을 모식도로 나타낸 것이다.
