Abstract
Description
도 2는 본 발명에 따라 유도만능줄기세포로부터 각막윤부줄기세포로의 분화를 유도한 후, 현미경으로 세포의 형태를 관찰한 결과를 나타낸다. (A)는 유도만능줄기세포(저배율, x100), (B)는 유도만능줄기세포(고배율, x200), (C)는 BMP-4가 첨가되지 않은 배지에 유도만능줄기세포를 배양한 후의 분화된 각막윤부줄기세포(고배율, x200), (D)는 BMP-4가 첨가된 배지에서 유도만능줄기세포를 배양한 후의 분화된 각막윤부줄기세포(고배율, x200)를 나타낸다.
도 3은 본 발명에 따라 유도만능줄기세포로부터 각막윤부줄기세포로의 분화를 유도한 후, 각막상피세포 마커 K3와 각막윤부줄기세포 마커 ABCG2의 발현 변화를 확인한 결과를 나타낸다. (A)는 유도만능줄기세포, (B)는 BMP-4가 첨가되지 않은 배지에 배양한 후의 분화된 각막윤부줄기세포, (C)는 BMP-4가 첨가된 배지에서 배양한 후의 분화된 각막윤부줄기세포에서의 마커의 발현 변화를 확인한 면역조직화학염색 결과를 나타낸다. (D)는 BMP-4의 첨가 유무에 따른 마커의 발현 변화를 real time PCR로 확인한 결과를 나타내며, "PI"는 BMP-4가 첨가되지 않은 배지에 배양한 후의 분화된 각막윤부줄기세포, "BMP4"는 BMP-4가 첨가된 배지에서 배양한 후의 분화된 각막윤부줄기세포를 나타낸다.
도 4는 본 발명에 따라 유도만능줄기세포로부터 각막윤부줄기세포로의 분화를 유도한 후, 각막윤부줄기세포 마커인 ABCG2 및 p63의 발현 변화를 real time PCR로 확인한 결과를 나타낸다. "PI"는 BMP-4가 첨가되지 않은 배지에 배양한 후의 분화된 각막윤부줄기세포, "BMP4"는 BMP-4가 첨가된 배지에서 배양한 후의 분화된 각막윤부줄기세포를 나타낸다.
Claims (10)
- 유도만능줄기세포를 BMP-4(Bone morphogenetic protein-4)가 첨가된 혼합배지에서 배양하는 단계를 포함하며,
상기 혼합배지는, EGF(Epidermal Growth Factor), 인슐린(insulin), 하이드로코르티손(Hydrocortisone), 에탄올아민(Ethanolamine), 포스포에탄올아민(Phosphoethanolamine) 및 BPE(bovine pituitary extract)를 포함하는 배지와 FBS(Fetal bovine serum)가 보충된 IMDM(Iscove's modified Dulbecco's medium) 배지가 1:2 내지 2:1로 혼합된 것인, 유도만능줄기세포를 각막윤부줄기세포로 분화시키는 방법.
- 제1항에 있어서, 상기 배양 기간은 2일 내지 5일인, 방법.
- 제1항에 있어서, 상기 배양된 세포를, EGF, 인슐린, 하이드로코르티손, 에탄올아민, 포스포에탄올아민 및 BPE를 포함하는 배지와 FBS가 보충된 IMDM 배지가 1:2 내지 2:1로 혼합되어 있는 혼합배지에서 15일 내지 30일 동안 배양하는 단계를 추가로 포함하는, 방법.
- 제1항에 있어서, 상기 BMP-4는 혼합배지에 100 내지 200 ng/ml로 포함된 것인, 방법.
