[Experimental Example 1] Antioxidant activity test

The antioxidant activity of the skin whitening cosmetic composition according to the present invention was measured by measuring the free radical scavenging ratio.

The antioxidative effect of the compositions according to Examples 1 to 3 was evaluated by using DPPH (1,1-diphenyl-2-picrylhydrazyl) . [Experimental Example 2] Melanin production inhibition experiment

The degree of inhibition of melanin formation on the B16F1 melanocyte of the skin whitening cosmetic composition according to the present invention was examined. The B16F1 melanocyte used in this experiment is a mouse-derived cell line and is a cell that secretes a melanin pigment called melanin. During the artificial culture of these cells, samples were treated to compare the degree of reduction of melanin pigment.

To describe the specific experimental method, B16F1 melanocytes were dispensed into 6-well plates at a concentration of 2 per well, and after attaching the cells, the samples were treated at a concentration not causing toxicity and cultured for 72 hours. After incubation for 72 hours, the cells were detached with trypsin-EDTA, the number of cells was measured, and the cells were recovered by centrifugation.

Then, the cell pellet was washed once with PBS, and 1 ml of homogenization buffer (50 mM sodium phosphate, pH 6.8, 1% Triton X-100, 2 mM PMSF) was added and vortexed for 5 minutes to disrupt the cells. After centrifugation (3,000 rpm, 10 min), 1N NaOH (10% DMSO) was added to the cell filtrate to dissolve the extracted melanin, and the absorbance of melanin was measured at 490 nm using a microplate reader. Melanin formation inhibition rate (I) was calculated according to the following equation (2).

In addition, the inhibition rate of melanin formation was determined by the same method as above using kojic acid as a whitening ingredient. Experimental concentration of each sample according to the above example was set to 200 μg / ml, and the average value of the results of the experiment three times per each sample is shown in Table 2.

As shown in Table 2, the herbal medicine extract according to Example 1 of the present invention showed much higher inhibition of melanin formation (inhibition rate) on B16F1 melanocyte than kojic acid, which is a conventional whitening ingredient. [Experimental Example 3] Tyrosinase activity inhibition experiment

The skin whitening cosmetic composition according to the present invention was tested for inhibition of tyrosinase activity.

Each of the compositions according to Examples 1 to 3 of the present invention was diluted in a buffer solution to a concentration of 50 μg / ml, 0.5 ml of each sample thus prepared was diluted with a tyrosine solution (0.1 mg / ml solution prepared by dissolving tyrosine in 0.05 M sodium phosphate buffer (pH 6.8)), and the mixture was allowed to stand in a thermostat at 37 ° C. for 10 minutes. In order to test the whitening effect and moisturizing effect of the skin whitening cosmetic composition according to the present invention, the following Examples 4 to 6 and Comparative Examples were conducted.

The composition of Example 1 was mixed with 10 wt% of glycerin, 5 wt% of glycerin, 8 wt% of liquid paraffin, 2 wt% of polysorbate 60, 1 wt% of sorbitan sesquioleolate, 5 wt% of squalane, 2 wt% 0.5% by weight of amine, 1% by weight of propylene glycol, and 1.5% by weight of a carboxyl vinyl polymer are mixed, and the remaining amount is mixed with purified water to prepare an emulsion.

The composition of Example 2 was prepared by mixing 10 wt% of the composition, 5 wt% of glycerin, 8 wt% of liquid paraffin, 2 wt% of polysorbate 60, 1 wt% of sorbitan sesquioleolate, 5 wt% of squalane, 0.5% by weight of amine, 1% by weight of propylene glycol, and 1.5% by weight of a carboxyl vinyl polymer are mixed and the remaining amount is mixed with purified water to prepare an emulsion.

The composition of Example 3 was changed to 10 wt%, glycerin 5 wt%, liquid paraffin 8 wt%, polysorbate 60 2 wt%, sorbitan sesquirololate 1 wt%, squalane 5 wt%, wax 2 wt%, triethanol 0.5% by weight of amine, 1% by weight of propylene glycol, and 1.5% by weight of a carboxyl vinyl polymer are mixed, and the remaining amount is mixed with purified water to prepare an emulsion.

Comparative Example

Without mixing any of the compositions of Examples 1 to 3, only 5 wt% of glycerin, 8 wt% of liquid paraffin, 2 wt% of polysorbate 60, 1 wt% of sorbitan sesquioleolate, 5 wt% of squalane %, 2% by weight of beeswax, 0.5% by weight of triethanolamine, 1% by weight of propylene glycol and 1.5% by weight of a carboxyl vinyl polymer, and the remaining amount is mixed with purified water to prepare an emulsion.
