Abstract
본 발명은 애엽(artemisia asiatica) 추출물을 포함하는 항우울제 조성물에 대한 것으로, 종래에는 애엽의 95% 에탄올인 DA-9601(stillenTM) 가 항염증 기능을 가져서 위 점막 궤양에 유용하다는 것이 알려져 있었지만, 본 발명은 상기 DA-9601(stillenTM) 가 항우울제로서의 효능을 가진다는 것을 실험을 통하여 확인한 것이 특징이다.
Description
본 발명은 항우울제 조성물에 대한 것으로, 특히 애엽(artemisia asiatica) 추출물을 포함하는 것이며, 더욱 상세하게는 애엽의 95% 에탄올인 DA-9601(stillenTM) 가 항우울제로서의 효능을 가진다는 것을 실험을 통하여 확인한 것이다.
우울증은 전 세계적으로 상당한 장애 및 재정 비용을 발생시키는 심각한 일반적인 재발 정신장애이다(Arai et al., 2012; Lucca et al., 2009; Yi et al., 2013). 세계 보건 기구(WHO)는 우울증이 인간 장애-조절 수명(disability-adjusted life years)에서 손실의 두 번째 가장 큰 원인이 될 것이라고 예측한다(Murray and Lopez, 1997). 우울증은 또한 신체장애를 일으키고 높은 자살율을 가진다(Reeves et al., 2008).
현재 항우울제는 세로토닌성(serotonergic), 노르아드레날린성, 및 도파민성 시스템에 주로 영향을 미친다(Nestler et al., 2002; Leggio et al., 2013). 우울 장애의 치료에 사용되는 약물은 삼환계 항우울제(TCAs), 모노아민 산화효소 억제제 (MAOIs), 선택적 세로토닌 재흡수 억제제(SSRIs), 세로토닌 및 노르아드레날린 재흡수 억제제 (SNRIs), 및 노르아드레날린 및 도파민 재흡수 억제제(NDRI)이다. 지난 몇 년에 걸쳐, SSRIs 및 SNRIs는 일반 대중에 있어서 주로 이들의 향상된 내성 및 안전성 프로파일 때문에 우울 장애의 치료를 위한 선택 약물로서 점차로 TCAs를 대체해 왔다(Bauer et al., 2007; Stahl, 2008).
또한, 뇌 유래 신경 영양 인자(BDNF)는 기억 및 시냅스 가소성(synaptic plasticity)의 잠재적 조절자로서 작용하는 신경 영양 인자이다(Yan et al., 2010). 항우울제에 의하여 증가된 BDNF 농도는 뇌에서 신경발생(neurogenesis)을 유발함으로써, BDNF는 우울 장애에 있어서 유용한 효과를 가진다(Massa et al., 2010). 모노아민 신경전달물질, 5-하이드록시트립타민 (5-HT, 세로토닌)은 해마 조직에서 BDNF의 발현을 증가시킨다(Yi et al., 2013). 또한, 중추 신경계의 모노아민 신경전달물질의 결핍은 우울증의 병리학 및 생리학에서 중요한 역할을 한다(Delgado, 2000). 최근 연구는 중추신경계의 소교세포(microglia)의 활성화에 의해 유도되는 신경 염증성 반응이 BDNF 신호 전달체계(signal pathway)를 억제한다는 것을 나타낸다(Wager-Smith and Markou, 2011; Maes et al., 2011). 또한, 우울증에서 신경 염증성 시스템의 활성화는 인터루킨 (IL)-1β, IL-6, 및 종양 괴사 인자 (TNF)-α와 같은 염증성 매개물질의 방출에 의해 유도된다고 보고되었다(Wager-Smith and Markou, 2011).
그러나, 현재 치료에 사용되고 있는 항우울제는 거의 대부분 화합물의 합성에 의해 인공적으로 제조된 것이어서 성기능 장애와 체중 증가 등과 같은 부작용들을 나타내고 있는데, 천연물질이나 이것의 추출물을 이용하여 상기한 부작용을 극복할 수 있는 항우울제의 개발은 새로운 시장을 열 수 있을 것으로 전망된다.
한편, 애엽은 국화과(Compositae)에 속하는 다년생 초본식물로서, 독특한 향기와 맛을 갖고 있다. 어린잎은 식용으로서 쑥떡 등에 이용되어 왔고, 한방에서는 자궁을 따스하게 하여 자궁출혈, 임신 중의 출혈 등에 효과가 있다고 알려져 있으며, 강장보혈, 부인병, 설사치료제로 사용되어 왔다(동의보감).
애엽의 약리작용을 살펴보면, 애엽 추출물은 혈액응고시간을 연장시키며(약학회지 28(2): 69-77, 1984), 애엽으로부터 추출한 플라본(flavone)은 항암활성, 혈소판응집 억제작용(Zhongguo Zhongyao Zazhi, 17(6): 353, 1992), 항진균활성이 있으며, 항보체작용을 하여 숙주의 방어체계를 도와 알러지나 염증에 효과가 있고(Chem. Pharm. Bull. 33(5): 2028-2034, 1985), 혈압강하작용과 진정작용을 한다고 보고 된 바 있다.
그 외에도 애엽으로부터 유파틸린(eupatilin)이 분리 및 규명되었으며, 이에 대해 항종양제(WO 8803805), 건선치료제(WO 8803800), 알러지치료제(JP 84155314) 및 아구창(aphthous ulcer)치료제(CA 2065496 AA) 등의 용도가 특허로 출원된 바 있다. 한편, 애엽의 동속식물인 인진호(茵蔯蒿, Artemisia capillaris)의 대장염 치료효 과가 최근 보고 된 바 있으나(대한소화기학회지 28: 224, 1996) 인진호는 본 발명에서의 애엽과는 형태학적 및 약리학적으로 큰 차이를 보이며, 습열황달의 작용이 있어 예로부터 황달 치료제로 사용되어 온 생약이다(본초학 366, 1995, 대한약사회, 서울, 대한민국).
현재까지 알려진 Artemisia속의 쑥 및 그 추출물에 함유된 성분으로는 스코폴레틴 (scopletin), 세사민(sesamine), 유파폴린(eupafolin), 마트리카린 (matricarin), 베타시토스테롤(β-sitosterol), 알파아밀린(α-amyrin), 캐필린(capillin), 캐필라린(capillarin), 모노터페노이드류(monoterpenoids), 이소스코폴레틴(isoscopoletin) 아테필린 A, C(artepillin A, C), 아로마틱 화합물류(aromatic compounds) 등 무수히 많으나 개별 성분의 약리학적 활성에 대해서는 아직 제대로 밝혀져 있지 않은 실정이다.
이와 같이, 애엽 추출물에는 현재까지 밝혀진 성분 이외에 아직도 미지의 여러가지 성분이 함유되어 있으며 이들 중 일부 성분은 인체에 유효한 약리작용을 나타낸다. 그러나 현재까지는 애엽의 추출물이 위염 및 소화성궤양에 치료효과가 있다는 것만이 알려져 있을 뿐이다.
이에 따라, 천연물질인 애엽 및/또는 이것의 추출물이 가지는 다양한 약리 효과를 확인하여 아직 밝혀지지 않은 미지의 성분들에 의한 새로운 약학 조성물의 개발에 대한 필요성은 항시 존재하고 있는 실정이다.
본 발명은 상기한 문제점을 해결하기 위한 것으로, 종래에 항염증 기능과 위 점막 궤양에 유용한 애엽 추출물의 항우울제로서의 기능을 확인하는 것이 목적이다.
또한, 본 발명은 애엽 추출물을 포함하는 항우울제 조성물을 제공하기 위한 것이다.
상기한 목적을 달성하기 위한 본 발명에 따른 항 우울제 조성물은, 애엽(artemisia asiatica) 추출물을 유효성분으로 포함하는 것이다.
여기서, 상기 애엽 추출물은 DA-9601(stillenTM) 인 것이 바람직하다.
또한, 상기한 본 발명에 따른 조성물은 강제수영테스트(forced swimming test, FST)에서 부동시간(immobility time)을 감소시키는 것을 특징으로 할 수 있다.
또한, 세로토닌(serotonin)과 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF) 및 니슬 소체(nissl bodies) 중 하나 이상의 농도를 증가시키는 것이 가능하다.
또한, 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α) 중 하나 이상의 농도를 감소시키는 것이 바람직하다.
또한, 에스트로겐 수용체-β(estrogen receptor-β : ER-β)의 농도를 증가시키는 것일 수 있다.
이에 따라, 본 발명의 일 실시예는, 애엽(artemisia asiatica) 추출물을 유효성분으로 포함하고, 대상체의 강제수영테스트(forced swimming test, FST)에서의 부동시간(immobility time)을 감소시키며, 세로토닌(serotonin), 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF), 니슬 소체(nissl bodies) 및 에스트로겐 수용체-β(estrogen receptor-β : ER-β) 농도를 증가시키고, 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α)의 농도를 감소시키는 것을 특징으로 하는 항 우울제 조성물이다.
기타 실시예들의 구체적인 사항들은 상세한 설명 및 도면들에 포함되어 있다.
이러한 본 발명은 애엽(artemisia asiatica) 추출물을 유효성분으로 포함하는 것을 특징으로 하는 항우울제 조성물을 제공할 수 있다.
또한, 본 발명은 애엽 추출물을 포함하는 조성물에 의하여, 대상체의 강제수영테스트(forced swimming test, FST)에서의 부동시간(immobility time)을 감소시키며, 세로토닌(serotonin), 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF), 니슬 소체(Nissl bodies) 및 에스트로겐 수용체-β(estrogen receptor-β : ER-β) 농도를 증가시키고, 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α)의 농도를 감소시켜서, 우울증을 예방 및/또는 치료할 수 있는 효과가 있다.

본 발명은 다양한 변환을 가할 수 있고 여러 가지 실시 예를 가질 수 있는 바, 특정 실시 예들을 도면에 예시하고 상세한 설명에서 상세하게 설명하고자 한다. 그러나, 이는 본 발명을 특정한 실시 형태에 대해 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변환, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다. 본 발명을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 발명의 요지를 흐릴 수 있다고 판단되는 경우 그 상세한 설명을 생략한다.
본 출원에서 사용한 용어는 단지 특정한 실시예를 설명하기 위해 사용된 것으로, 본 발명을 한정하려는 의도가 아니다. 단수의 표현은 문맥상 명백하게 다르게 뜻하지 않는 한, 복수의 표현을 포함한다. 본 출원에서, "포함하다" 또는 "가지다" 등의 용어는 명세서상에 기재된 특징, 숫자, 단계, 동작, 구성요소, 부품 또는 이들을 조합한 것이 존재함을 지정하려는 것이지, 하나 또는 그 이상의 다른 특징들이나 숫자, 단계, 동작, 구성요소, 부품 또는 이들을 조합한 것들의 존재 또는 부가 가능성을 미리 배제하지 않는 것으로 이해되어야 한다.
제1, 제2 등의 용어는 다양한 구성요소들을 설명하는데 사용될 수 있지만, 상기 구성요소들은 상기 용어들에 의해 한정되어서는 안 된다. 상기 용어들은 하나의 구성요소를 다른 구성요소로부터 구별하는 목적으로만 사용된다.
본 발명은 애엽(artemisia asiatica) 추출물을 유효성분으로 포함하는 항우울제 조성물이다.
여기서, 상기 애엽 추출물은 애엽을 70~100%의 에탄올로 추출한 추출물인 것이 바람직하다. 그리고, 상기 애엽 추출물은 애엽을 70~100%의 에탄올로 10~36시간 동안 1∼2회 냉침추출하거나, 3∼8시간 동안 초음파추출하거나, 또는 4~10시간 동안 2∼3회 환류 추출한 후 건조하여 얻은 것이 가능하다. 또한, 상기 애엽 추출물은 애엽을 95%의 에탄올로 추출한 추출물일 수 있다. 또한, 상기 애엽 추출물은 DA-9601(stillenTM) 인 것이 바람직하다.
기존의 DA-9601(stillenTM)은 아르테미시아 아시아티카(Artemisia asiatica)의 추출물로서, 위점막 궤양을 가진 환자들을 치료하는 데 사용되어 왔다. 또한, DA-9601는 항 염증성 효과를 가지는 것으로 나타났다(Seo et al., 2002; Choi et al., 2006). 상기한 바와 같이, 신경 염증성 반응(neuroinflammatory reactions)은 우울증과 크게 관련되어 있을 수 있다. 이에 따라, 본 발명자들은 DA-9601이 우울증을 조절할지도 모른다고 가설을 세웠다. 그리고, 본 발명자들은 항우울제 치료의 효능을 예측하는 행동기능검사(behavioral test)인 강제수영테스트(FST) 등에 대한 DA-9601의 항우울제 같은 효과에 대해 연구하였다.
즉, 발명자들은 강제수영테스트(forced swimming test, FST)에 대한 DA-9601의 항우울 효과를 평가했다. 상기 FST에서 14 일간 DA-9601 투여군의 부동시간은 대조군에 비해 현저하게 감소하였다. 상기 DA-9601 군에서 세로토닌 (5-HT) 및 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF) 단백질의 농도는 해마 조직에서 증가되었다. 니슬 소체(Nissl bodies)는 DA-9601군의 해마 신경 세포질에서 또한 증가되었다. DA-9601은 해마 조직 및 혈청 대조군에 비해 mRNA 및 단백질 농도에서 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α)의 농도를 감소시켰다. 또한, 에스트로겐 수용체-β의 mRNA 발현은 해마 조직에서 DA-9601 투여에 의해 증가되었다. 이러한 결과는 본 발명에 따른 DA-9601이 항우울제 후보로서 간주될 수 있음을 시사한다.
이에 따라, 본 발명에 따른 조성물은 강제수영테스트(forced swimming test, FST)에서 부동시간(immobility time)을 감소시키는 것을 특징으로 할 수 있다. 또한, 세로토닌(serotonin)과 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF) 및 니슬 소체(nissl bodies) 중 하나 이상의 농도를 증가시키는 것이 가능하다. 또한, 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α) 중 하나 이상의 농도를 감소시키는 것이 바람직하다. 또한, 에스트로겐 수용체-β(estrogen receptor-β : ER-β)의 농도를 증가시키는 것일 수 있다.
구체적으로, 뇌의 세로토닌 시스템은 관련되고 중추 신경계의 기능성 5-HT 시스템의 결핍은 우울증의 병태 생리에 중요한 역할을 한다(Nestler et al., 2002; Delgado, 2000). 또한, SSRIs와 같은 항우울제 약물은 신경발생 및 BDNF 발현을 증가시키고(Hodes et al., 2010), 플루옥세틴은 5-HT의 농도를 증가시키고 우울증 증상을 완화시킨다(Hodes et al., 2010). 한편, 핀돌롤(pindolol)은 5-HT 신경에서 5-HT1A/1B 수용체를 활성화함으로써 우울증을 개선시키고(Artigas et al., 1994), 항우울제 같은 효과를 갖는 oleanolic 산의 만성 투여는 뇌의 5-HT 농도를 증가시킨다(Yi et al., 2013).
본 발명자들은 2 주 동안 DA-9601의 투여가 FST에서 부동시간을 현저하게 감소시켰고 해마 5-HT의 농도를 현저하게 증가시키던 것을 알아냈다. 따라서, 본 발명자들은 DA-9601의 항우울제와 같은 효과가 세로토닌 시스템과 부분적으로 관련되어 있음을 시사한다.
또한, BDNF는 신경 세포의 증식, 시냅스 활동, 기억에 중심 역할을 하고, BDNF 농도는 전두엽 피질 및 해마 우울증을 앓는 자살 희생자의 조직 영역에서 감소된다(Angelucci et al., 2005; Karege et al., 2005). 또한, Hodes 등은, 항우울제 약물의 효과가 세로토닌 수송체의 억제를 통해 BDNF 발현에 의해 매개되는 것으로 결론을 내렸다(2010). 최근 우울증 연구는 신경 세포 보호 효과 및 해마 치아 이랑에서 신경발생에 초점을 맞추고 있다(Hellweg et al., 2008; Boldrini et al., 2009). BDNF는 TrkB 수용체 신호 전달체계를 통해 ERK의 활성화를 증가시키고(Massa et al., 2010), ERK의 활성화는 향신경작용(neurotrophic action), 증식, 및 세포 생존과 관련이 있다(Creson et al., 2009). Sairanen 등은 BrdU-양성 세포의 수가 BDNF-/+ 마우스의 BrdU 주입 후 3 주 감소되었음을 알아내었고(2005), Xiong 등은 BDNF 발현이 예측할 수 없는 경도의 만성적 스트레스의 해마 조직에서 감소되었음을 보고했다(2011). 설사, 구토, 통증, 열과 같은 우울증의 증상은 염증인자에 의해 매개되며, 동물모델과 우울증 환자에게서 항염증제가 항우울 효과를 나타내었다(Wager-Smith and Markou, 2011). 또한, IL-6, 또는 IL-1 수용체 결핍 마우스가 우울 증상을 덜 나타내는 것으로 보고되었다(Chourbaji et al., 2006; Goshen et al., 2008). 다른 연구에서는, TNF-α 수용체 결핍 마우스에서 우울증 같은 행동이 완전히 사라졌다(Viana et al., 2010). 흥미롭게도, 최 등은, DA-9601이 염증 반응을 억제함을 보였으며(2006), 본 발명에서는 DA-9601이 해마조직의 IL-1β, IL-6, 및 TNF-α의 농도를 현저하게 감소시킴을 알 수 있었다. Wager-Smith and Markou (2011) 및 Maes 등(2011)은 신경 염증이 BDNF 신호 전달체계의 차단(blockade)에 의해 활성화되었음을 보고하였다. 또한, 미노싸이클린(minocyclin)은 환자와 동물 모델에서 항신경염증성 및 항우울제 효과를 가지는 것으로 보고되었다(Levine et al., 1999; Yrjanheikki et al., 1999). 에스트라디올은 BDNF 발현과 향상된 기억 및 우울증과 같은 행동을 증가시키는 것으로 보고 되었다(Brann et al., 2007). 중추 신경계 및 우울증에서, 에스트라디올은 뇌의 BDNF 발현과 관련되어 있다(Sohrabji and Lewis, 2006). 상기 언급된 연구 결과는 BDNF 발현이 항우울제 활성에 중요한 인자임을 나타낸다. 본 발명에서, DA-9601 투여에 의한 해마의 BDNF의 발현 증가, 니슬 소체 수의 증가 및 염증성 사이토카인 농도의 감소는 DA-9601이 BDNF 발현을 통해 항우울 효과를 나타냄을 의미한다.
결론적으로, 본 발명자들은 DA-9601이 강제수영모델에서 용량 의존적 방식으로 부동시간을 현저하게 감소함을 관찰하였다. 또한, DA-9601은 해마조직에서 5-HT 농도, BDNF 발현, ERK의 활성화, 및 ER-β 발현을 현저하게 증가시키고, 염증성 사이토카인의 농도를 현저하게 감소시킨다는 것을 확인하였다. 즉, 본 발명에 따른 DA-9601이 FST에서 항우울제와 같은 효과를 나타내며, 이러한 효과가 세로토닌 시스템과 해마 조직의 BDNF 신호 전달체계의 조절과 관련 있는 것을 보여준다. 이에 따라, 애엽 추출물은 항우울제로서 유용한 효과를 가질 수 있다.
상기한 바에 따르면, 본 발명의 일 실시예는, 애엽(artemisia asiatica) 추출물을 유효성분으로 포함하고, 대상체의 강제수영테스트(forced swimming test, FST)에서의 부동시간(immobility time)을 감소시키며, 세로토닌(serotonin), 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF), 니슬 소체(nissl bodies) 및 에스트로겐 수용체-β(estrogen receptor-β : ER-β) 농도를 증가시키고, 인터루킨(IL)-1β, IL-6 및 종양 괴사 인자-α(TNF-α)의 농도를 감소시키는 것을 특징으로 하는 항우울제 조성물이다.
본 발명의 애엽 추출물은 우울증의 치료 및/또는 예방에 탁월한 효과를 가지고 있고, 정제, 과립, 캡슐, 분말, 시럽, 연고, 좌제 및 피하, 근육, 정맥 또는 점적 주사제 등의 일반적인 약제학적 조성물의 형태로 투여될수 있으며, 1일 10∼1000mg을 1회 내지 수회에 나누어 복용할 수 있다.
본 발명에 따른 애엽 추출물을 유효성분으로 함유하는 약학적 조성물은 계면 활성제 및 공용매를 추가로 포함할 수 있다.
또한, 상기 조성물은 약효에 악영향을 미치지 아니하는 범위 내에서 약제학적으로 통상 사용되는 물질, 예를 들면 애엽 추출물의 용해도 및 위장관내 흡수를 증가시키고 경구 투여시에 물과 함께 분산 및 유화됨으로써 용출을 증가시키고 생체 이용율 향상에 널리 활용될 수 있는 지방산 또는 지방산 알코올과 같은 첨가제, 백당, 맥아이온 엿, 정백당, 젤라틴, 설탕 및 물엿과 같은 당류, 스테아린산 마그네슘, 탈크와 같은 활택제, 미세결정셀롤로우스, 인산일수소칼슘, 전분, 만니톨과 같은 부형제, 제제가 산화되는 것을 방지하는 항산화제, 착향제, 방부제, 방향제, 감미료, 색소, pH 조절제 및 점도 조절제를 추가로 포함할 수 있으며, 이들은 애엽 추출물에 대하여 통상적으로 사용되는 사용량으로 첨가하는 것이 바람직하다.
본 발명은 상기 애엽 추출물을 함유하는 약학적 조성물을 포함하는 경구용 제제를 제공한다.
상기 경구용 제제는 정제, 환제, 산제, 경질 캅셀제, 젤라틴 밴딩한 경질캅셀제 및 연질 캅셀제, 카라멜 또는 젤리 타입의 츄정 및 수용액을 함유하는 경구용 액제이다. 바람직하게는 상기 조성물을 포함하는 연질 캅셀제를 제공한다. 본 발명에 따른 상기 조성물은 애엽 추출물, 계면 활성제 및 공용매를 혼합하고, 80℃에서 가열 용해시킨 후 통상적인 방법을 이용하여 경구용 제제로 제형화 할 수 있다.
본 발명은 하기의 실시예에 의하여 보다 더 잘 이해 될 수 있으며, 하기의 실시예는 본 발명의 예시 목적을 위한 것이며, 첨부된 특허청구범위에 의하여 한정되는 보호범위를 제한하고자 하는 것은 아니다.

애엽
추출물을 포함하는 조성물
애엽(artemisia asiatica) 추출물을 포함하는 조성물은 다양한 방법으로 준비할 수 있다. 일례로, 애엽을 세절한 후 70~100%의 에탄올로 10~36시간 동안 1∼2회 냉침추출하거나, 3∼8시간 동안 초음파추출하거나, 또는 4~10시간 동안 2∼3회 환류 추출한 후 건조하여 얻을 수 있다.
본 실시예에서는 편의를 위하여, 애엽을 95%의 에탄올로 추출한 추출물을 유효성분으로 포함하는 DA-9601(stillenTM)을 시중에서 구입하여 이용하였다.
실험예
1 : 실험 동물의 준비
수컷 ICR 마우스(3 주령, 10~12 g)는 대한 실험 동물 센터(대전, 한국)로부터 구매하였고, 그 다음 경희대학교 한의과대학에서 관리하였다. 동물은 본 연구 내내, 07:00 h 동안 12:12 L/D 주기의 광 하에서 55±10%의 상대 습도에서 22±1℃의 온도로 유지된 층류(laminar air-flow)에서 케이지 당 5 내지 10 마리씩 보관했다. 음식과 물은 임의로 이용할 수 있게 하였다. 모든 조작은 09:00 내지 16:00 h 사이에 수행되하였으며, 동물은 한번 이상 사용하지 않았다. 모든 프로토콜은 경희대학교(KHUASP(SE)-10-032)의 기관 동물 관리 및 사용 위원회에 의해 승인되었다.
한편, 아래 본 실험예에서 통계 분석(Statistical Analysis)은 적어도 세 개의 독립적인 실험으로부터의 데이터를 종합하고 평균±S.D.(표준편차)로써 나타내었다. 통계적 유의성은 각각의 치료군 및 대조군 사이에 독립 t-검정 및 SPSS 통계 소프트웨어를 사용하여 Tukey 후 검정(post hoc test)과 ANOVA에 의해 비교되었다. 결과는 P < 0.05의 값으로 유의하였다.

본 발명자들은 강제수영테스트(FST)에서 부동시간(immobility time)에 대한 DA-9601의 효과를 연구함으로써, 항우울증 활성을 밝히기 위해 생체내(in vivo) 모델을 사용하였다. DA-9601(동아제약 연구소, 용인, 한국)은 증류수(D.W.)에 용해시켰다. 상기 DA-9601 (25, 50, 및 100 mg/kg) 또는 D.W.는 존대를 이용하여 2주 동안 하루 한 번씩 마우스 내에 경구 투여하였다. 플루옥세틴(10 mg/kg, 고전적 항우울제)은 양성 컨트롤(positive control)로서 사용하였다. 2주 동안의 경구 투여 후에, FST를 수행하였다. FST의 6 분 중에, 부동시간을 Porsolt 등에 의해 종래 언급된 바와 같은 방법으로 측정하였다(1977). 2 개의 플렉시유리 실린더(높이: 25 cm, 직경: 10 cm)로 이루어진 장치는 23~25℃에서 물로 채워진 마크롤론 케이지에 나란히 배치되었다. 2 마리의 마우스를 수직 플렉시 실린더 안쪽에 넣고 6 분 주기 동안 동시에 시험하였고, 상기 2 개의 실린더 사이에 놓여진 투명하지 않은 스크린은 마우스들이 서로 보는 것을 방지할 수 있었다. 부동의 총 지속시간은, 2 분 지연 후에, 4 분의 주기 중에 측정되었다. 각각의 마우스는 발버둥 치는 것을 멈추고 물에서 움직임 없이 떠있는 것을 유지할 때 부동이라고 간주하였으며, 이것은 물 위에 이들의 머리를 유지하기 위해 필요한 움직임만을 만든다.
이와 함께, 세로토닌 시스템은 우울증의 병태 생리 및 치료에 중심적인 역할을 한다(Nestler et al., 2002). 또한, 해마 조직의 5-HT의 농도의 증가는 항우울제 약물 치료에 따라 증가되었다(Yi et al., 2013). 이에 따라, 본 발명자들은 이것의 메카니즘을 조사하기 위해 FST 후에 마우스의 5-HT 농도를 조사하였다. 즉, 해마(Hippocampal) 5-HT 농도를 5-하이드록시트립타민 분석 키트(MyBiosource, San Diego, CA, USA)를 이용하여 제조업체의 사양에 의해 측정하였다.
그 결과, 마우스는 처음 실린더 내로 배치될 때, 격렬히 사방에서 수영하였고, 명백하게 출구를 찾았다. 2~3 분 후에는 이것들의 활동이 작아졌고, 마우스가 수동적으로 그냥 물 위에 머리와 반 수평으로 물에 떠있는 유지 기간이 증가되는 부동 기간으로 대체되었다. 부동시간의 첫 번째 측정 후, 상기 마우스는 상기 기록된 수영 시간을 기반으로 하는 대조군, 플루옥세틴군(기준 약물), 및 DA-9601 군으로 나누었다.
도 1은 본 발명의 일 실시예에 따른 DA-9601의 강제수영테스트(FST)에서의 부동시간(도 1의 A) 및 해마(hippocampus)에서의 5-HT 농도(도 1의 B)를 나타내는 그래프이고, 여기에 나타난 바와 같이, 14 일간 DA-9601 투여군(50 및 100 ㎎/kg)의 부동시간은 대조군에 비해 현저하게 감소하였다(도 1A, P < 0.05). 플루옥세틴군의 부동시간 또한 현저하게 감소했다(도 1A, P <0.05). 또한, 도 1B에 나타난 바와 같이, 5-HT의 농도는 대조군(P <0.05) 보다 DA-9601 투여군에서 현저하게 더 높았다.
즉, 도 1은 FST의 부동시간 및 해마 5-HT의 농도에 대한 DA-9601의 효과를 나타내고 있고, 도 1의 (A)에 나타난 바와 같이, DA-9601 또는 플루옥세틴(10 ㎎/㎏)을 14일 동안 투여하였다. DA-9601은 시험 전에 1 시간을 투여하였다. 도 1의 (B)에 나타난 바와 같이, 마지막 FST 후, 마우스에 케타민 (80 ㎎/kg) 및 자일라진 (4 ㎎/kg)을 복강 내 주사로 마취하였다. 해마 5-HT 농도는 분석 키트를 사용하여 측정하였다. 도 1에서 값은 평균±표준편차(S.D.)이다(* P < 0.01 대 D.W. 투여 대조군; # P < 0.05 대 정상 마우스 군).
이에 따르면, DA-9601을 14일 동안 경구 투여한 후 우울 동물모델인 강제수영시험(FST)을 실시하였을 때, Fluoxetine(양성대조군, 항우울제)과 DA-9601(50 mg/kg, 100 mg/kg) 그룹에서 유의적으로 부동시간이 감소함(P < 0.05)을 알 수 있었다. 또한, 뇌의 해마 조직에서 세로토닌(5-HT) 농도가 DA-9601 (100 mg/kg) 그룹에서 유의적으로 증가됨을 확인할 수 있었다.

3-1 : 면역화학조직검사법(

마우스는 PBS, 이어서 4% 빙냉(ice-cold) 파라포름알데하이드를 이용하여 관류시켰다. 뇌를 제거하고, 밤새 4% 파라포름알데하이드에서 후-고정(post-fixed)시키고, 3일 동안 PBS 중 30% 자당에 놓고, 자당으로부터 제거시키고 -70℃에서 냉동시켰다. 조직은 저온유지장치(cryostat) 상에서 절단하였다. PBS로 세척한 후에, 섹션은 10 분 동안 PBS에 1% H2O2에서 배양하였고, 3×10 분 동안 세척하였다. 섹션은 소혈청 알부민(bovine serum albumin)을 이용하여 블록킹 되었고 이후 1㎍/ml의 농도에서 항-BDNF 항체(Santa Cruz, CA, USA)를 이용하여 60 분 동안 배양시켰다. 이후 각 섹션은 실온에서 10 분 동안 3, 3'-diaminobenzidine 기질 용액을 이용하여 재반응 시켰다. 잔여 DAB는 세 PBS 세척을 이용하여 조각(slice)을 세척하였다. 모든 시편은 코드화되고, 광학 현미경 하에서 검사하며, 두 블라인드(blinded) 관찰자에 의해 무작위로 분석하였다.

해마 조직은 웨스턴 블럿 분석에 이용되었다. 샘플은 95℃에서 5 분 동안 가열하여 원심 분리 후에, 50㎍ 분액(aliquots)을 12% SDS-폴리아크릴아미드 겔 전기영동에 의하여 분석하였다. 상기 분석된 단백질을 25 V에서 25 mM 트리스, pH 8.5, 200 mM 글리세린, 및 20% 메탄올 중의 니트로셀룰로오즈 막에 밤새 electro-transferred 하였다. 블롯은 5% 탈지 분유를 함유하는 0.05% Tween 20을 함유하는 1×PBS를 이용하여 적어도 2 시간 동안 블로킹(blocked) 하였고, 이후 BDNF, 액틴, 세포외 신호 조절 단백질 키나아제(ERK), 및 인산화-ERK(phosphorylated-ERK, pERK) 항체(Santa Cruz, CA, USA)와 함께 1 시간 동안 실온에서 배양하였다. 퍼옥시다아제-결합 2차 항체, 및 단백질에 의해 개발된 블롯은 제조업체의 설명서에 따라서, 향상된 화학루미네선스 절차(GE Healthcare, Piscataway, NJ, USA)에 의하여 가시화되었다.

상기 FST 후에, 마우스에 케타민(ketamine)(80 mg/kg) 및 자일라진(xylazine) (4 mg/kg)을 복강내 주사로 주입하여 마취시켰고, 250 ml saline을 이용하여 관류시키었으며, 그 다음 30 분 동안 0.1 M PBS (pH 7.4) 중 4% 파라포름알데히드를 이용하여 관류시켰다. 마우스 뇌는 제거되었고 4℃에서 동일한 정착액 중에 24 h 동안 후-고정시켰고, 그리고 나서 4℃에서 3일 동안 30% 자당에 옮겼다. 상기 냉동된 섹션(15㎛ 두께)이 준비되었고 일반 뉴런 형태학의 평가를 위해 크레실 바이올렛(니슬)을 이용하여 염색하였다. 치아 이랑 영역(dentate gyrus region)에서 신경세포 밀도를 계산하였다. 상기 데이터는 mm 당 통합된 광학 밀도로 표현하였다. 섹션은 코드화되었고 두 개의 블라인드(blinded) 관찰자에 의하여 임의로 분석되었다.

즉, 도 2는 해마 조직의 BDNF 발현 및 ERK 활성화에 대한 DA-9601의 효과를 나타내는 것이다. 도 2의 (A)에서 해마 조직은 DAB(BDNF의 경우, 화살표)로 염색되었고, 치아 이랑(dentate gyrus, DG), 등지느러미(dorsal) 해마 조직의 CA1과 CA3 영역에서 BDNF 농도를 나타내며, BDNF 발현 (B) 및 ERK의 phosphorylations (C)는 웨스턴 블롯에 의해 분석된 결과이다(원래 배율 × 100, 200, 400, 스케일 바 = 50 ㎛).
강제수영시험 종료 후, 생쥐 뇌의 해마 조직에서 면역염색법으로 확인한 결과, DA-9601 (100 mg/kg) 그룹의 해마 조직에서 BDNF의 발현이 증가됨을 관찰할 수수 있었다. 또한, DA-9601 (100 mg/kg)은 BDNF에 의한 신경발생 (Neurogenesis) 기전에 중요한 ERK의 활성을 증가시키는 것을 확인할 수 있었다. BDNF는 중추신경 계의 발생에 중요한 역할을 하며, 특히 해마 치아 이랑(hippocampal dentate gyrus)에서 BDNF의 발현 결과는 신경손상과 신경발생의 억제를 초래한다(Xiong et al., 2011). 도 2A 및 도 B에 나타난 바와 같이, BDNF 농도는 대조군보다 DA-9601 투여군에서 더 높았고, DA-9601는 해마 ERK의 인산화를 증가시키는 것으로 확인되었다(도 2C).

이후, 본 발명자들은 해마 조직에서 신경 세포 손상에 대한 DA-9601의 효과를 조사하기 위하여 니슬(Nissl) 소체 염색을 수행하였다.
도 3은 본 발명의 일 실시예에 따른 DA-9601에 의한, 해마 조직에서의 니슬 소체(nissl bodies) 수에 대한 결과를 나타내는 사진 및 그래프이다. 즉, 도 3은 해마 조직에서 니슬 소체 수에 대한 DA-9601의 효과를 나타내는 것이다. 해마 조직은 니슬(Nissl) (화살표)로 염색하였다. 도 3의 (A)는 치아 이랑(dentate gyrus, DG)에서 니슬(Nissl) 염색 신경 세포, 등지느러미 해마 조직의 CA1과 CA3 영역을 나타내는 것이고, 도 3의 (B)는 영상 분석기를 이용하여 해마 DG의 니슬 소체 부량을 나타낸다. 여기서, IOD는 통합 광학 밀도이고, * P < 0.01 대 D.W. 투여 대조군(원래 배율 ×400, 스케일 바 = 50㎛)이다.
니슬 소체는 신경세포 손상 시 그 수가 감소되는데, 강제수영시험 종료 후 해마의 니슬 소체를 염색한 결과, DW 그룹에서 감소한 니슬 소체가 DA-9601 (100 mg/kg) 투여에 의해서 그 수가 유의적으로 증가하였다. 즉, 본 발명자들은 대조군(도 3, P < 0.05)과 비교하여 DA-9601 및 플루옥세틴이 치아 이랑(dentate gyrus) 영역에서 니슬 소체의 수가 현저하게 증가되었음을 알아냈다.

혈청 및 해마 조직의 사이토카인은 ELISAdp 의해 측정하였다. 상기 ELISA는 IL-1β, IL-6, 및 TNF-α용 capture Ab의 1㎍/well을 이용하여 96-well plates를 코팅함으로써 수행되었다(R&D Systems, Inc., Minneapolis, MN, USA). 분석에서 다음 단계 전에, 상기 코팅된 플레이트는 0.05% tween-20을 함유하는 phosphate-buffered saline (PBS)을 이용하여 2 번 세척하였다. 본 분석에서 사용된 모든 시약들 및 코팅된 웰(wells)은 실온에서 2 시간 동안 배양하였다. 표준 곡선은 제조업체에 의해 제공된 것으로서, 사이토카인의 공지된 농도로부터 발생되었다.

Total RNA는 easy-BLUETM RNA 추출 키트(iNtRON Biotech, Korea)를 이용하여 상기 제조업체의 사양에 의해 조직으로부터 분리되었다. 총 용출에서 상기 total RNA의 농도를 분광 광도계에 의하여 측정하였다. Total RNA (2.5 mg)는 65℃에서 10 분 동안 가열하고 이후 얼음 상에서 냉동시켰다. 각 샘플은 cDNA 합성 키트(Amersham Pharmacia Biotech, Piscataway, NJ, USA)를 이용하여 37℃에서 90 분 동안 cDNA에 역전사 되었다. PCR은 하기 프라이머를 이용하여 마우스 IL-1β(5'-AGG CCA CAG GTA TTT TGT CG-3'; 5'-GCC CAT CCT CTG TGA CTC AT-3'), 마우스 IL-6 (5'-CGG GAT CCA TGT TCC CTA CTT CAC AA-3'; 5'-CCC AAG CTT CTA CGT TTG CCG AGT AGA-3'), 마우스 TNF-α(5'-ATG AGA ACA GAA AGC ATG ATC-3'; 5'-TAC AGG CTT G TC ACT CGA ATT-3'), 마우스 에스트로겐 수용체 (ER-β (5'- ACA GTC CTG CTG TGA TGA AC-3'; 5'- ACT AGT AAC AGG GCT GGC AC-3')에 대해 수행되었고, 및 마우스 GAPDH (5'-TTC ACC ACC ATG GAG AAG GC-3' ; 5'-GGC ATG GAC TGT GGT CAT GA-3')은 RNA의 동등한 양이 상이한 실험 조건으로부터 역전사 및 PCR 증폭을 위해 사용되었는지를 확인하기 위하여 사용되었다. 어닐링 온도는 각각 IL-1β 대해 50℃, IL-6에 대해 62℃, TNF-α및 GAPDH 에 대해 60℃, 및 ER-β 대해 55℃였다. 생성물은 1.5% 아가로스 겔 상에서 전기영동하고 브롬산 에티디움을 이용하여 염색함으로써 가시화하였다.

중추 신경계의 염증성 사이토카인은 BDNF 신호 전달체계를 봉쇄(blockading)함으로써 우울증을 유도하는 것으로 알려져 있다(Wager-Smith and Markou, 2011; Maes et al., 2011).
도 4는 본 발명의 일 실시예에 따른 DA-9601에 의한, 해마 조직에서의 염증성 사이토카인 단백질과 mRNA 농도에 대한 결과를 나타내는 그래프 및 사진이고, 도 5는 본 발명의 일 실시예에 따른 DA-9601에 의한, 혈청 염증성 사이토카인의 농도에 대한 결과를 나타내는 그래프이다.
구체적으로, 도 4는 해마 조직의 염증성 사이토카인의 단백질과 mRNA 농도에 대한 DA-9601의 효과를 나타낸다. 마지막 FST 후, 마우스에 케타민 (80 ㎎/㎏)과 자일라진(4 ㎎/㎏)을 복강 내 주사하여 마취하였다. 도 4의 (A-C)는 해마 조직에서의 IL-1β, IL-6 및 TNF-α단백질 수준을 ELISA로 측정한 결과이고, 도 4의 (D)는 IL-1β, IL-6 및 TNF-α mRNA 농도를 RT-PCR에 의해 측정한 결과이다. 도 4에서, 값(value)은 평균±표준편차(S.D.)이다(* P < 0.01 대 D.W. 투여 대조군; 독감, 플루옥세틴(10 ㎎/㎏/일, p.o.) 투여군). 도 4에 나타난 바와 같이, 강제수영시험 종료 후 생쥐 뇌의 해마 조직에서 염증성 사이토카인 농도를 정량한 결과, DA-9601 (100 mg/kg) 경구 투여 그룹에서 IL-1β, IL-6 농도가 유의적으로 감소하였고(P < 0.05). 또한 DA-9601(25, 50, 100 mg/kg)는 TNF-α 농도도 유의적으로 감소시키었으며(P < 0.05), DA-9601은 염증성 사이토카인 mRNA 발현도 감소시키었다.
도 5는 혈청 염증성 사이토카인 농도에 대한 DA-9601의 효과를 나타낸다. 마지막 FST 후, 마우스에 케타민(80 ㎎/㎏)과 자일라진(4 ㎎/㎏)을 복강 내 주사로 마취하였다. 이후 혈액은 강제 수영을 실시하는 마우스의 심장에서 주사기 내로 추출하였다. IL-1β IL-6 및 TNF-α 농도는 ELISA 방법을 사용하여 측정하였다. 도 5에서, 값(value)은 평균±표준편차(S.D.)이다(* P < 0.01 대 D.W. 투여 대조군; 독감, 플루옥세틴(10 ㎎/㎏/일, p.o.) 투여군). 도 5에 나타난 바와 같이, 우울증 환자에서 염증성 사이토카인 농도가 증가하였다. 즉. 강제수영시험 종료 후 생쥐의 혈청에서 염증성 사이토카인 농도를 정량한 결과, DA-9601(50 mg/kg, 100 mg/kg) 그룹에서 IL-6와 TNF-α 농도가 유의적으로 감소하였고(P < 0.05). IL-1β 농도는 감소하는 경향을 보였으나 유의적인 차이는 없었다.
상기 도 4 및 도 5에 나타난 바와 같이, 염증성 사이토카인, 즉, IL-1β, IL-6 및 TNF-α의 단백질 및 mRNA 농도는 해마 조직 및 혈청에서 DA-9601 투여에 의해 감소되었다.

도 6은 본 발명의 일 실시예에 따른 DA-9601에 의한, 해마 에스트로겐 수용체-β(estrogen receptor-β : ER-β) 단백질의 농도에 대한 결과를 나타내는 사진 및 그래프이다.
즉, 도 6은 해마 ER-β mRNA에 대한 DA-9601의 효과를 나타낸다. 마지막 FST 후, 마우스에 케타민(80 ㎎/㎏)과 자일라진(4 ㎎/㎏)을 복강 내 주사하여 마취하였다. 도 6의 (A)는 해마 ER-β mRNA를 RT-PCR에 의해 측정한 결과이고, (B)는 ER-β 농도를 농도계에 의해 정량화한 것이다. 도 6에서, 값(value)은 평균±표준편차(S.D.)이다(* P < 0.01 대 D.W. 투여 대조군; 독감, 플루옥세틴(10 ㎎/㎏/일, p.o.) 투여군).
도 6의 Estrogen receptor beta(ER beta)는 강제수영시험에 있어서 항우울 효과를 가지는 선택적인 에스트로젠 수용체이다. 에스트라디올(estradiol)은 여성의 우울증 증상을 개선하고 설치류의 우울증 행동을 감소시킨다(Walf et al., 2004). 또한, Walf 등은 에스트라디올의 항우울제의 효과가 FST에서 ER-β 활성화에 의해 매개되는 것으로 알려져 있다(2004).
본 발명자들은 DA-9601의 항우울제와 같은 효과가 ER-β 활성화과 관련이 있는지 여부를 확인하기 위하여, 해마 조직의 ER-β mRNA 발현을 조사하였다. 강제수영시험 종료 후 생쥐 뇌의 해마 조직에서 ERbeta mRNA 발현을 RT-PCR로 측정한 결과, DA-9601 투여가 ERbeta mRNA 발현을 증가시킴을 확인할 수 있었다. 즉, 도 6에 도시된 바와 같이, DA-9601 및 플루옥세틴은 ER-β mRNA의 발현(P <0.05)을 현저하게 증가시켰다.
상기에서는 본 발명을 특정의 바람직한 실시예에 관련하여 도시하고 설명하였지만, 이하의 특허청구범위에 의해 마련되는 본 발명의 기술적 특징이나 분야를 이탈하지 않는 한도 내에서 본 발명이 다양하게 개조 및 변화될 수 있다는 것은 당업계에서 통상의 지식을 가진 자에게 명백한 것이다.
Claims (10)
- 아르테미시아 아시아티카(Artemisia asiatica)를 에탄올로 추출한 추출물을 유효성분으로 포함하고,
세로토닌(serotonin)과 뇌 유래 신경 영양 인자(brain-derived neurotrophic factor, BDNF) 및 니슬 소체(nissl bodies) 중 하나 이상의 농도를 증가시키는 것을 특징으로 하는 항우울제 조성물.
- 제1항에 있어서,
상기 추출물은 아르테미시아 아시아티카(Artemisia asiatica)를 70~100%의 에탄올로 추출한 추출물인 것을 특징으로 하는 항우울제 조성물.
- 제2항에 있어서,
상기 추출물은 아르테미시아 아시아티카(Artemisia asiatica)를 70~100%의 에탄올로 10~36시간 동안 1∼2회 냉침추출하거나, 3∼8시간 동안 초음파추출하거나, 또는 4~10시간 동안 2∼3회 환류 추출한 후 건조하여 얻은 것임을 특징으로 하는 항우울제 조성물.
- 제1항에 있어서,
상기 추출물은 아르테미시아 아시아티카(Artemisia asiatica)를 95%의 에탄올로 추출한 추출물인 것을 특징으로 하는 항우울제 조성물.

강제수영테스트(forced swimming test, FST)에서 부동시간(immobility time)을 감소시키는 것을 특징으로 하는 항우울제 조성물.
