Abstract
본 발명은 국내에서 유통되고 있는 벼 품종에 대하여 다형성을 나타내는 벼 품종을 식별하기 위한 마커, 벼 품종 식별 방법, 및 상기 마커를 포함하는 키트에 관한 것이다. 구체적으로, 본 발명은 국내에서 유통되고 있는 벼 300개 품종에 대해 높은 다형성을 갖는 초위성체 마커의 조합을 선별하고 이를 이용하여 국내에 육성된 벼 품종 식별을 식별할 수 있게 한다. 본 발명의 방법에서 사용된 벼 품종 식별용 분자 표지는 국내에서 육성된 벼 300개 품종을 식별할 수 있다. 따라서, 본 발명은 벼 품종의 진위성 규명, 보급종 및 원종 종자 생산시 혼종 여부의 검정, 품종 보호 출원시 대조 품종의 선정 및 침해 여부의 확인 등과 같은 여러 분야에 매우 유용하게 사용될 수 있다.The present invention relates to a marker for identifying rice varieties exhibiting polymorphism in rice varieties distributed in Korea, a rice cultivar identification method, and a kit including the marker. 국제 신품종 보호 동맹(UPOV)에서는 분자 마커의 활용에 대한 실험실 및 연구자 간의 신뢰성 높은 분석 결과를 도출하기 위하여, 유전자 분석 방법 및 DNA 마커의 선정, DNA 분리 방법 및 데이터베이스 구축 등에 대한 가이드라인을 제시하고 있다. 이 가이드라인에서는 분자 마커의 활용시 실험실 및 연구자 간에 반복 재현성이 높을 뿐만 아니라 공우성을 나타내며, 활용 분자 마커가 염색체상에 고르게 분포되어 있고 여러 가지 분석장비를 활용하더라도 동일한 결과를 나타내어야 한다는 전제 조건을 제시하고 있다. 이러한 요건에 가장 적합한 것이 분석 방법이 염색체상에 존재하는 반복염기서열의 차이를 활용하여 분석 재료간의 다형성을 확인하는 SSR(simple sequence repeat) 분석 방법이라고 할 수 있다. 실제로 유럽에서는 유럽품종보호사무소(CPVO) 주관 하에 감자, 장미, 토마토 등의 작물에 대하여 500품종에 대한 DNA 프로파일(profile) 데이터베이스를 구축하여 그 연구 결과를 보고하고 있다. 중국과 일본에서는 벼, 옥수수, 골풀, 딸기 등의 작물에 대하여 SSR 분석 기술을 활용한 품종별 DNA 프로파일을 데이터베이스화하여 품종보호 권리 침해와 자국 농산물의 보호 등과 같은 분야에 활용하고 있다. 한편, 우리나라의 경우 국립종자원에서 종자시장에서 중요도가 높거나 분쟁의 발생 가능성이 높은 무, 배추, 수박, 참외, 멜론, 오이, 벼 등과 같은 작물에 대하여 품종식별이 가능한 분자 마커를 활용하여 작물당 100~500품종에 대한 DNA 프로파일 데이터베이스를 구축하였거나 구축 중에 있으며, 실제로 품종별 DNA 프로파일 데이터베이스를 활용하여 품종보호 출원품종의 대조품종 선정 및 품종보호 재배심사시 구별성, 균일성 및 안정성을 확인하는데 이용하고 있으며, 최근에는 품종보호 등록품종의 특성 유지확인에도 유전자 분석 기술을 적극 활용하고 있다. 또한 품종식별 분자 마커는 농가와 종자 회사 및 종자회사 간에 발생하는 품종 진위성과 관련된 종자분쟁 발생시 이를 중재하기 위한 하나의 수단으로 적극 활용되고 있는 실정이다.The UPOV provides guidelines for the selection of genetic analysis methods, selection of DNA markers, DNA isolation methods, and database construction in order to obtain reliable analysis results between laboratories and researchers on the use of molecular markers .
