Abstract

Description

따라서, 본 발명의 일면은 대상 내의 구강 박테리아 또는 진균류에 의해 초래되는 질환 및 장애를 치료 및 예방하기 위한, 구강으로 갈륨 화합물을 적용하는 것을 포함하는 방법을 제공한다. 특정 구현예에서, 질환 및 장애는 충치, 치은염, 구취 및 아구창(oral thrush) 등을 포함한다. 또한, 본 발명은 대상 내의 표면적 감염을 치료 및 예방하기 위한, 감염된 부위에 갈륨 화합물을 국소적으로 적용하는 것을 포함하는 방법을 제공한다. 특정 구현예에서, 상기 표면적 감염은 피부 감염, 질 효모 감염 및 눈 감염 등을 포함한다.

5.1 철 수송 및 갈륨
대부분 미생물은, 일부 예외(예를 들어, Lactobacillus spp . 5.2 갈륨 화합물
본 발명의 사용에 적합한 갈륨 화합물은 약학적으로 수용가능하고 포유류 사용, 특히 사람 사용에 안전한 모든 갈륨-함유 화합물을 포함한다. 갈륨 화합물은 사람에 있어 진단용으로 그리고 치료용으로 사용되어 왔고, 사람 사용에 매우 안전한 것으로 알려져 있다(Foster 등, 1986, 상기; Todd 등, 1991, Drugs 42:261-273; Johnkoff 등, 1993, Br J Cancer 67:693-700).
본 발명의 사용에 적합한 약학적으로 수용가능한 갈륨 화합물로는, 한정하는 것은 아니지만, 갈륨 나이트레이트, 갈륨 말톨레이트(maltolate), 갈륨 시트레이트, 갈륨 포스페이트, 갈륨 클로라이드, 갈륨 플루오라이드, 갈륨 카보네이트, 갈륨 포르메이트, 갈륨 아세테이트, 갈륨 설페이트, 갈륨 타르트레이트(tartrate), 갈륨 옥살레이트, 갈륨 옥사이드 및 다양한 적용에 있어 효과적인 수준의 갈륨 원소를 안전하게 제공할 수 있는 다른 갈륨 화합물을 포함한다. 또한, 갈륨 피론(pyrone), 갈륨 피리돈(pyridone) 및 갈륨 옥심과 같은 갈륨 착물뿐만 아니라, 트랜스페린 및 락토페린과 같은 단백질과 결합한 갈륨, 또는 히드록사메이트, 폴리카르복실레이트, 페톨레이트-카테콜레이트 및 데스페리옥사민과 같은 철포획체 및 시스테인, α-케토산, 히드록시 산 및 피리독살 이소니코틸닐 히드라존(pyridoxal isonicotinyl hydrazone) 클래스(Richardson 등, 1997, Antimicrobial Agents and Chemotherapy 41(9):2061-2063)와 같은 기타 철-킬레이터 등과 결합한 갈륨도 본 발명의 사용에 적합하다.
갈륨 화합물의 치료적 유효량(즉, 투약량)은 치료될 질환 또는 장애의 특질 및 심각성, 감염된 영역의 위치, 대상의 연령 및 면역학적 배경, 사용되는 갈륨 화합물의 유형뿐만 아니라, 기술분야의 당업자에게 명백한 기타 요인에 기초하여 변할 수 있다. 전형적으로, 갈륨 화합물의 치료적 유효량은, 적용 부위에 적어도 약 1μM, 적어도 약 50μM, 적어도 약 100μM, 적어도 약 500μM, 적어도 약 1 mM, 적어도 약 10 mM, 적어도 약 50 mM, 적어도 약 100 mM, 적어도 약 200 mM 및 약 500 mM까지 갈륨 이온 농도를 제공하는 양일 수 있다. 갈륨의 낮은 독성 때문에, 상기 양은 500 mM 이상까지, 그러나 어떤한 독성이라도 초래하는 양 미만으로, 자유롭게 증가될 수 있다. 참고적으로, 건강한 성인은 적어도 7일 동안 적어도 약 200㎎/㎡/일의 갈륨 나이트레이트를 혈관내 주입을 견딜 수 있다고 보고되어있다(Schlesinger 등, 상기 참조).

5.3 갈륨 화합물의 제형화
본 발명에 적합한 갈륨 화합물은 그 적용에 따라 다양한 형태로 제형화 될 수 있다. 따라서, 본 발명은 철-의존성 병원성 미생물의 성장을 제어하기 위한 갈륨 화합물을 포함하는 제형을 제공한다. 특정 구현예에서, 갈륨 화합물은 구강에 적용하기 위한 치약, 구강세정제, 젤 또는 가글(gargle)로 제형화된다. 또 다른 특정 구현예에서, 갈륨 화합물은 국소적 또는 국부적 적용을 위해 연고, 로션, 크림, 젤, 용액, 에어로졸 및 좌약 등으로 제형화된다. 전형적으로 국소적 또는 국부적 적용을 위해 사용되는 임의의 기타 제형이 본 발명에 적용 가능하다.
또 다른 바람직한 구현예에서, 본 발명의 제형은 갈륨 화합물에 추가하여 프로바이오틱 미생물을 더 포함할 수 있다. 상기 제형이 감염된 영역에 적용될 때, 갈륨 화합물이 병원성 생물의 성장을 감소 또는 금지하거나 병원성 생물을 사멸시키는 동안 프로바이오틱은 미소식물을 인계받을 수 있다.
본 발명의 갈륨 화합물 제형은 추가로 기타 활성 또는 비활성 성분을 포함할 수 있다. 치약의 경우, 상기 기타 성분으로는, 한정하는 것은 아니지만, 질산칼륨(치아 민감성 감소를 위해), 불소화물, 수화된 실리카와 같은 연마재, 라우릴황산나트륨 및 라우릴황산암모늄과 같은 계면활성제, 테트라소듐 피로포스페이트(tetrasodium pyrophosphate, TSPP)와 같은 격리제(sequestering agent), 아크릴성 PVM/MA 코폴리머, 폴리에틸렌글리콜(PEG) 및 폴리프로필렌글리콜(PEG)과 같은 폴리머, 과산화 탄산나트륨과 같은 치아 미백제, 감미료, 중탄산나트륨뿐만 아니라, 기타 항세균성 제형 및 충진재를 포함한다. 크림 및 로션의 경우, 상기 기타 성분으로는, 한정하는 것은 아니지만, 완화제, 가습제, 비타민, 항산화제 및 계면활성제 등을 포함한다.
5.4 충치
지난 20년간 수돗물의 불소화 뿐만 아니라, 불화물-함유 제품의 사용은 미국에서 충치가 퍼지는 것을 감소시켜 왔다(이 주제의 검토를 위해 Featherstone, 2000, JADA 131:887-898). 그러나, 최근 보고에서 치아가 있는 미국의 18세 이상의 모든 성인의 약 94%가 치료 또는 비치료된 치관측 충치(coronal caries)의 흔적을 갖는다고 보고되었으며(Winn 등, 1996, J Dent Res 75:642-51), 따라서 치아가 썩는 것은 미국 내 아동뿐만 아니라 성인에게 주요 문제로 남아 있다.
충치는 저하된 침의 기능 및 발효될 수 있는 탄수화물의 빈번한 사용과 같은 기타 병원성 요인과 결합하여 박테리아적으로 발생하는 전염성 질환이다(Featherstone, 2000, 상기). 상주하는 구강 박테리아 중에서, 고수준의 스트렙토코커스 뮤탄스(Streptococcus mutans ) 및 락토바실리(lactobacilli)는 충치 형성에서 주된 병인을 구성한다. 그러나, 항세균적 접근은 충치의 진전을 예방하거나 정지하기 위한 미국에서 채택된 정기적인 구강 위생 방책의 초점이 아니었다. 예외로서, 고수준의 박테리아가 있는 환자를 위해 클로르헥시딘 글루코네이트(chlorhexidine gluconate) 린스 또는 젤의 처방이 있지만, 임상의가 구강에서 우식성 박테리아의 수준을 결정하는 것은 어렵다.
또한, 스트렙토코커스 뮤탄스(S. mutans)는 성장 및 생존을 위해 철 섭취가 요구된다고 알려져 있고, 다수의 철-응답성 단백질을 발현한다(Spatafora 등, 2001, Microbiology 147:1599-1610). 철-응답성 단백질 중의 하나인 fimA의 널 돌연변이(null mutation)는 S. mutans을 죽이지 않으며, 이는 철 섭취 및 수송에 대한 다른 메카니즘이 이 병원성 생물에서 작동하고 있다는 것을 나타낸다(Spatafora 등, 2001, 상기). 따라서, 본 발명은 대상의 치아 표면 및 구강에 예방적 유효량의 갈륨 화합물을 적용하는 것을 포함하는, 충치를 예방하기 위한 방법을 제공한다. 바람직한 일 구현예에서, 갈륨 화합물은 갈륨 화합물을 포함하는 치약으로 제형화되고, 브러싱(brushing)에 의해 치아 표면 및 구강에 적용된다. 또 다른 바람직한 구현예에서, 갈륨 화합물은 갈륨 화합물을 포함하는 구강세정제로 제형화되고, 구강을 헹굼(rinsing)으로써 치아 표면에 적용된다. 또 다른 바람직한 구현예에서, 갈륨 화합물은 치아 표면에 적용되기 위한 연고 또는 젤로 제형화될 수 있다.
5.5 질 감염
질 감염(또는 질염)은 여성에게 있어 가장 통상적인 의학적 관심사 중 하나이다. 질 감염 중에, 가장 통상적인 것은 질 내에 보통 적은 수가 상주하는 칸디다 알비칸스(C. albicans)의 과잉성장에 의해 초래되는 효모 감염이다. sit1(siderophore iron transport 1) 유전자 생성물에 의한 철포획체 흡수는 C. albicans가 상피층을 침입하여 관통하는 것이 요구되며, 이는 철 흡수가 생물의 병원성 요인에 직접 관련되어 있다는 것을 나타낸다고 보고되어 왔다(Heymann 등, 2002, Infection and Immunity 70(9):5246-5255). 따라서, 철을 갈륨 화합물로 대신하는 것은 질 내에서 C. albicans의 성장을 효과적으로 제어할 수 있다. 결과적으로, 특정 구현예에서, 본 발명은 갈륨 화합물을 대상의 질에 적용하는 것을 포함하는, 질 효모 감염를 치료하기 위한 방법을 제공한다. 상기 갈륨 화합물은 크림, 연고, 젤, 주수(douche) 또는 좌약으로 제형화될 수 있다.
또한, 본 발명은, 한정하는 것은 아니지만, 가드네렐라 바지날리스( Gardnerella vaginalis ), 박테로이데스 속의 종(Bacteroides spp .), 모빌룬커스 속의 종(Mobiluncus spp .) 및 미코플라스마 호미니스(Mycoplasma Hominis)와 같은 병원성 박테리아에 의해 초래되는 박테리아성 질염 및 트리코모나스 바지날리스(Trichomonas vaginalis)에 의해 초래되는 기생충성 질염을 포함하는 기타 유형의 질 감염을 질에 다양한 제형 중의 갈륨 화합물을 적용함으로써 치료하는 방법을 제공한다.
대부분의 질 감염은 질의 일반 생물상(flora)의 개체수에 있어 변화의 결과이기 때문에, 그 영역에 락토바실러스 속의 종(Lactobacillus spp .)과 같은 비-병원성 일반 상주물이 거주하게 하는 것은 병원성 생물의 과성장을 방지할 수도 있다. 따라서, 바람직한 일 구현예에서, 본 발명은 락토바실러스 아시도필러스(Lactobacillus acidophilus), 락토바실러스 플란타룸(Lactobacillus plantarum), 락토바실러스 레이크만니(Lactobacillus leichmannii ), 락토바실러스 존스니(Lactobacillus johnsonni ), 락토바실러스 델브루엑키(Lactobacillus delbrueckii)와 같은 프로바이오틱과 조합하여 갈륨 화합물을 적용하는 것을 포함하는, 질 감염을 치료하는 방법을 제공한다.
5.6 무생물 또는 생물 물체의 소독
갈륨 화합물의 항미생물적 활성은 무생물 또는 생물 물체의 표면을 미생물적 오염으로부터 소독 또는 보호하기 위해서 사용될 수도 있다. 따라서, 본 발명은 유효량의 갈륨 화합물을 물체의 표면에 적용하는 것을 포함하는, 무생물 또는 생물 물체의 표면 상에 미생물의 성장을 제어하는 방법을 제공한다.
예를 들어, 칫솔, 의치 및 치아 교정기는 자주 그 위로 이송되는 구강 미생물의 서식처이고, 위생 관점에서 상기 제품의 표면 상에 상기 생물의 성장을 방지하는 것이 바람직하다. 따라서, 특정 구현예에서, 본 발명은 물체의 표면에 유효량의 갈륨 화합물을 적용함으로써 칫솔, 의치, 치아 교정기 및 기타 치아관련 기구와 같은 물체를 소독하는 방법을 제공한다. 상기 갈륨 화합물은 침지, 브러싱, 분사 및 코팅에 의해 상기 물체의 표면에 적용될 수 있다. 상기 코팅은 미생물이 상기 물체의 표면에서 성장하지 못하는 속도로 서서히 갈륨 화합물이 확산되도록 하는 어떤 폴리머에 기초할 수 있다. 폴리머 코팅의 예로는, 한정하는 것은 아니지만, 히드록시아파타이트(hydroxyapatite), 폴리메타크릴레이트, 트리칼슘포스테이트, 폴리안하이드라이드, 하이드로카본 엘라스토머, 메틸- 및 에틸 셀룰로스, 히드록시프로필 셀룰로스, 셀룰로스 아세테이트 및 프탈레이트 등을 포함한다. 다른 방식으로, 갈륨 화합물은 예를 들어, 폴리머(예를 들어 열가소성, 열경화성 수지 등) 및 금속 등의, 그 자체가 물체(예를 들어, 칫솔의 털, 카테터(catheter), 의학적 장치 및 주사 바늘 등)를 형성하는 재료에 함입될 수 있다.

또 다른 특정 구현예에서, 본 발명은 곡물, 야채, 및 어류, 알, 낙농 제품 및 육류 등의 동물성 제품을 포함하는 음료 및 식품 내의 미생물의 성장을 갈륨 화합물을 세척, 담금 및/또는 분사에 의해 직접 상기 제품에 적용하여 제어하는 방법을 제공한다. 상기와 같은 식품의 처리는 소비자를 식중독으로부터 효과적으로 보호할 수 있다.
본 발명은 또한, 갈륨 화합물이 표면 코팅된 의료용 인공삽입물, 붕대(bandage) 및 상처 치료재료(wound dressing)을 제공한다.
또한, 본 발명은 가정, 병원 및 학교 등에서 다양한 주위의 표면에 갈륨 화합물을 적용함으로써 미생물 성장을 제어하는 방법을 제공한다. 상기 갈륨 화합물은 본 명세서를 통해 논의된 바와 같이(발명의 상세한 설명 및 상기 5.4 및 5.5 그리고 하기 5.7 및 5.8 참조), 일선의 항생제에 내성을 갖게 된다고 알려진 미생물을 포함한 모든 갈륨-감수성 병원성 미생물에 대하여 사용될 수 있다. 상기 미생물로는 대장균(Escherichia coli) O157(위장염, 출혈성 대장염 또는 비뇨기 및 생식기 관 감염의 원인 생물), 반코마이신-내성 엔테레코커스 파에칼리스(Enterococcus faecalis)(심장 내막염, 비뇨기 관 감염 및 상처 감염의 원인 생물), 메티실린-내성 스타필로코커스 아우레우스(MRSA; 다양한 피부 감염, 눈 감염 및 상처 감염 등) 및 살모넬라 티피(Salmonella typhi, 장티푸스 원인 생물) 등을 포함한다. 다양한 표면으로 상기 갈륨 화합물의 적용은 비용 효과적이고, 항생제-내성 미생물의 진전의 증가없이 주위에서 미생물성 오염을 현저하게 감소시킨다.
5.7 동물 먹이
동물 성장을 후원하고, 감염을 방지하기 위한 동물 먹이 내에 항생제의 사용은 약품-내성 박테리아의 진전을 크게 초래하였다. 가금류 및 쇠고기 산업에서 상기한 항생제의 사용은 살모넬라, 캄필로박터(Campylobacter ), 쉬겔라(Shigella) 및 브루셀라(Brucella) 속 등 중에 항생제-내성 병원균의 출현의 결과를 가져왔다. 따라서, 일부 국가는 동물 먹이에 항생제의 사용을 금지하고 있는 중이다.
상기 논의한 바와 같이, 갈륨 화합물은 통상의 항생제 보다 약품-내성 병원균의 발생할 위험이 더 낮고, 따라서 실용적인 종래 항생제의 대안이다. 따라서, 본 발명은 유효량의 갈륨 화합물을 포함하는 동물 먹이를 제공한다. 동물 성장을 촉진시키고 감염을 방지하기에 충분한, 동물 먹이 중의 갈륨 화합물의 유효량은 상기 부분(5.2 부분 참조)에 기재된 갈륨 화합물의 치료적 또는 예방적인 투여량에 기초하여 즉시 결정될 수 있고, 상기 결정은 기술 분야의 당업자의 능력 내에 있다.
바람직한 구현예에서, 본 발명의 동물 먹이는 추가로 프로바이오틱을 포함할 수 있다. 프로바이오틱의 건강상 잇점은 3.1 및 5.3 부분에 논의되어 있다. 동물의 위장관 내의 프로바이오틱 또는 상주 미생물의 수를 증가시키기 위한 전형적인 프로바이오틱 조성물 및 방법은, 예를 들어 미국등록특허 제6,221,350호에 개시되어 있다.
5.8 기타 적용

피부 감염의 원인 생물로는, 한정하는 것은 아니지만, S. aureus 및 S. epidermidis와 같은 스타릴로코커스(Staphylococcus); 스트렙토코커스 피오제네스(Streptococcus pyogenes)와 같은 그룹 A 스트렙토코커스(Group A streptococci); 슈도모나스 아에루지노사(Pseudomonas aeruginosa ) 등의 종이 포함된다. 피부 감염을 초래하는 전형적인 효모 또는 진균류로는 칸디다 알비칸스(Candida albicans ), M. auduini 및 M. canis와 같은 미크로스포론(Microsporon) 속의 종; T. metagrophyte 및 T. tonsurans 등과 같은 트리초피톤(Trichophyton) 종이 포함된다. 안구 감염의 원인 생물로는 스트렙토코커스 아우레우스(Staphylococcus aureus ), 스트렙토코커스 피오제네스(Streptococcus pyogenes ), 해모필러스 인플루엔자( Haemophilus influenzae ), 클라미디아 트라초마티스(Chlamydia trachomatis ), 넬세리아 고노르호애(Neisseria gonorrhoeae ), 프로피오니박테리움 속의 종(Propionibacterium spp.), 노카디아 속의 종(Nocardia spp .), 박테로이데스 속의 종(Bacteroides spp.) 및 푸사리움 속의 종(Fusarium spp .) 등이 포함된다. 스트렙토코커스 뉴모니아(Streptococcus penumoniae)에 의해 초래되는 비강 감염은 귀 감염, 부비강염, 기관지염 및 폐렴 등을 야기할 수 있다. 이러한 생물에 의해 초래되는 감염은 메티실린-내성 스타필로코커스 아우레우스 (MRSA)와 같은 약품-내성 미생물 뿐만 아니라, HIV 감염 또는 AIDS, 기관 이식, 자기면역성 질환용 치료 등으로 인한 면역능력이 감소된 개인의 수가 증가하는 것 때문에 더욱 일상화되고 있다. 따라서, 수의사 용도뿐만 아니라 사람 용도를 위한 갈륨 화합물의 적용은 낮은 독성 및 내성 미생물을 생성하는 위험이 낮다는 점에서 특히 매우 유익하다. 식품 산업 분야, 농업 및 어업 등의 분야에서 다른 적용이 또한 가능하다.
본 발명은 갈륨 화합물을 감염된 영역에 적용함으로써, 박테리아, 진균류 및 기생충과 같은 철-의존성 병원성 미생물에 의해 초래되는 질환 및 장애를 치료 또는 예방하는 방법을 제공한다. 구체적으로, 본 발명은 충치, 질 감염 및 피부 감염 등을 치료 또는 예방하는 방법을 제공한다. 갈륨 화합물은 치약, 구강 세정제, 크림, 연고, 젤, 용액, 눈물액 및 좌약 등으로 제형화될 수 있다. 또한 본 발명은 칫솔, 의치, 치아 교정기, 콘택트 렌즈, 카테터 및 식료품 등의 표면을 포함하여, 주위의 표면상에 미생물 성장을 제어하는 방법을 제공한다. 추가적으로, 본 발명은 동물 성장을 촉진시키고, 동물을 감염으로부터 보호할 뿐만 아니라 소비자를 후행하는 감염으로부터 보호하는 갈륨 화합물을 포함하는 동물 먹이를 제공한다.
도 1은 미생물이 철 또는 갈륨을 흡입하는 메카니즘을 나타내는 모식도이다.
갈륨에 대한 미생물의 감수성
갈륨 나이트레이트를 사용하여 각 미생물에 대한 최소 저해 농도(MIC) 및 최소 살세균 농도(MBC)를 결정함으로써, 갈륨에 대한 다양한 미생물의 감수성을 테스트하였다. 일반적으로, MIC는 (i) 각각 표준 수의 미생물을 함유하는 일련의 브로스(broth)를, 순차적으로 희석한 갈륨 화합물 용액과 믹싱하고, (ii) 인큐베이트한 후에, 미생물의 성장을 저해하는 갈륨 화합물의 가장 낮은 농도인 MIC를 결정하여 결정된다. MIC가 낮을 수록, 미생물은 더욱 감수성이 있는 것이다. 갈륨 화합물을 포함하지 않는 적당한 아가 플레이트 상에 MIC 테스트로부터 각각의 분취한 샘플을 2차배양(subculture)하여 MBC를 결정하였다. 인큐베이트한 후, MBC는 성장이 발견되지 않는 갈륨 화합물의 최소 농도인 것으로 결정되었다.
구체적으로, 본 실험에서는, 2 g의 갈륨 나이트레이트 분말을 10 ㎖의 필터-멸균한 탈이온수에 용해시키고 생성되는 20%(w/v)(즉, 200 ㎎/㎖) 용액을 다시 한번 필터-멸균하였다. 10%, 1%, 0.5%, 0.1%, 0.05%, 0.01%, 0.005% 및 0.001%의 갈륨 나이트레이트 용액이 준비된 칸디다 알비칸스(Candida albicans ) 및 스트렙토코커스 뮤탄스(Streptococcus mutans)의 테스트를 제외하고는 대부분 생물을 테스트 하기 위해 멸균된 탈이온수 내에서 0.156%(즉, 1.56 ㎎/㎖)으로 2배 연속 희석액(2-fold serial dilution)을 준비하였다.
표 1은 MIC 및 MBC를 테스트한 미생물의 목록을 나타낸다. 모든 생물은 버지니아 매나사스(Manassas)의 미국 표준 균주(Amrican Type Culture Collection, ATCC)로부터 입수하였다. 시드 배양으로부터 각각의 미생물을 선택하고(표 1 참조), 적당한 유형의 브로스 내에 접종하여 0.5 McFarland 탁도 표준을 얻었다. 그리고 나서, 미생물의 표준 현탁액을 브로스와 1:100으로 희석하고 테스트에 사용하였다.
96-웰 플레이트 내에서 미량희석(microdilution) 브로스법(즉, 0.1 ㎖의 미생물 현탁액과 혼합된 0.1 ㎖의 갈륨 나이트레이트 용액) 또는 테스트 튜브 내에서 다량희석(macrodilution) 브로스법(즉, 1 ㎖의 미생물 현탁액과 혼합된 1 ㎖의 갈륨 나이트레이트 용액)에 의해 두개로 각각의 미생물을 다음과 같이 테스트하였다;
미량희석 브로스법: 칸디다 알비칸스(Candida albicans ); 스트렙토코커스 뮤탄스(Streptococcus mutans ); 대장균(Escherichia coli) O157:H7; 및 캠필로박터 제주니(Campylobacter jejuni ).
대량희석 브로스법: 메티실린-내성 스타필로코커스 아우레우스(MRSA); 반코마이신-내성 엔테로코커스 파에칼리스(VRE); 살모넬라 티피(Salmonella typhi ); 슈도모나스 애루지노사(Pseudomonas aeruginosa ); 포피로모나스 진지아발리스(Porphyromonas gingivalis ).
미생물의 성장은 샘플에서 탁도의 육안 관찰에 의해 결정되었다.
하기의 대조군을 테스트 샘플과 함께 인큐베이트하였다:
생존성 대조군: 동일 부피의 탈이온수와, 테스트 미생물로 접종되지만 갈륨 나이트레이트가 없는 적절한 브로스와의 혼합물; 및
불임성 대조군: 동일 부피의 탈이온수와, 미생물 또는 갈륨 나이트레이트가 없는 적절한 브로스와의 혼합물.
각 미생물의 순도는 적절한 아가 플레이트 상에 적절하게 희석된 미생물 현탁액을 스트리킹(streaking)하여 단리된 콜로니를 얻고, 콜로니 형태를 관찰하여 확인하였다.
MIC 테스트에 사용된 현탁액 중의 미생물의 농도는 일련의 현탁액의 희석액을 적절한 아가 플레이트 상에 접종하고, 콜로니의 수를 세어서 결정되었다.
MBC를 결정하기 위해, MIC에 사용된 각각의 샘플 10㎕를 적절한 아가 플레이트 상에 접종하고, 인큐베이트하였다. 성장을 보이지 않는 갈륨 나이트레이트의 최저 농도를 MBC인 것으로 결정하였다.
결과는 하기 표 2에 나타내었다.
하기 실시예는, 한정하는 것은 아니고, 세균성 및 진균성 감염의 방지 및 치료를 위한 실용적인 양식에 적용되듯이 본 발명의 바람직한 구현예를 예시하고자 하는 것이다.
실시예 1. 치약 제형
치아 및 치주 질환의 치료와 예방에 있어 활성성분으로서 50 μM 갈륨 나이트레이트 및 0.2 중량%의 불화나트륨을 포함하도록 치약을 제조한다. 상기 제형은 또한 물, 소르비톨, 글리세린, 수화 실리카, 폴리에틸렌글리콜, 크산탄(xanthan) 검, 사카린나트륨, 메틸파라벤 및 프로필파라벤을 포함하는 통상적인 불활성 성분을 포함한다. 이런 것들은 적절한 밀도의 치약을 얻기 위해 기술 분야의 당업자에게 알려진 양으로 혼합된다. 상기 제형은 통상적인 치약 튜브에 포장되어 보통의 방법으로 사용될 수 있다.
실시예 2. 질 효모 감염의 치료
본 발명은 내부 및 외부 질 효모 감염의 치료를 위하여 좌약 및 크림의 제형으로 각각 다음과 같이 적용된다. 좌약의 경우에는 젤라틴, 글리세린, 레시틴, 미네랄 오일, 산화티타늄 및 백색 페트롤레이텀(petrolatum)을 포함하거나, 외용 크림의 경우에는 벤조산, 이소프로필미리스테이트, 폴리소르베이트 60, 수산화칼륨, 프로필렌글리콜, 증류수 및 스테아릴알코올을 포함하는 불활성 성분과 200 μM 갈륨 시트레이트를 조합한다. 이 제형들은 통상적인 방법으로 사용하기에 적합하게 포장될 수 있다.
실시예 3. 무생물 물체의 소독
5 중량%의 갈륨포스페이트 및 5 중량%의 디메틸벤질암모늄클로라이드의 수용액 형태로 액상 제형을 제조하고, 병원성 생물의 성장 지점으로 작용하기 쉬운 표면을 갖는 물체를 소독하기 위한 표면 처리제로서 가정, 병원, 이발소 및 미장원에서 사용되도록 포장한다. 상기 제형은 통상적인 방법으로 상기 표면에 적용하여 사용될 수 있다.
실시예 4. 동물 먹이
가축에게 공급되는 곡물에 다양한 영양제 및 보조제가 첨가될 수 있는 동물 먹이의 제형 형태로서, 200㎎/먹이㎏의 갈륨 타르트레이트를 단독으로 또는 상기한 성분과 최적의 혼합이 되도록 조합한 상태로 상기 먹이에 첨가된다. 먹이 전체에 갈륨 타르트레이트의 분산되도록 하기 위한 통상적인 장치를 사용하여 수용액으로서 상기 갈륨 타르트레이트가 적용될 수 있다.
7. 동등물
본 발명이 관련된 분야에 당업자는 단지 일상적인 실험을 사용하여 본 명세서에 기재된 발명의 특정 구현예에 대한 많은 동등물을 확인할 수 있거나 인지할 것이다. 상기 동등물은 하기 청구범위에 의해 포함될 것이다.
