Claims (13)
- 피검 시료 중의 소맥 내재성 DNA를 PCR에 의해 검출 또는 정량하는 방법이며, 상기 피검 시료 중의 핵산 또는 상기 피검 시료로부터 추출한 핵산을 주형으로 하고, 서열 7에 기재된 염기 서열의 적어도 80 % 이상의 연속된 영역을 증폭할 수 있는 프라이머쌍을 이용하여 상기 영역의 핵산을 증폭하는 공정, 및 상기 증폭된 핵산을 검출 또는 정량하는 공정을 포함하는 방법.
- 제1항에 있어서, 상기 프라이머쌍이
(i) 서열 20에 기재된 염기 서열을 포함하는 핵산과 서열 21에 기재된 염기 서열을 포함하는 핵산으로 이루어지는 프라이머쌍, 및
(ii) 상기 (i)의 프라이머쌍에서, 각 핵산이 갖는 염기 서열의 적어도 80 % 연속된 염기 서열을 포함하는 핵산의 쌍을 포함하는 프라이머쌍으로 이루어지는 군으로부터 선택되는 프라이머쌍인 방법. - 제1항 또는 제2항에 있어서, 상기 프라이머쌍에 포함되는 프라이머가 염기 15개 내지 40개 길이의 핵산인 방법.
- 피검 시료 중의 소맥을 PCR에 의해 검출 또는 정량하기 위한 프라이머쌍이며, 서열 7에 기재된 염기 서열의 적어도 80 % 이상의 연속된 영역을 증폭할 수 있는 것을 특징으로 하는 프라이머쌍.
- 제4항에 있어서,
(i) 서열 20에 기재된 염기 서열을 포함하는 핵산과 서열 21에 기재된 염기 서열을 포함하는 핵산으로 이루어지는 프라이머쌍, 및
(ii) 상기 (i)의 프라이머쌍에서 각 핵산이 갖는 염기 서열의 적어도 80 % 연속된 염기 서열을 포함하는 핵산의 쌍을 포함하는 프라이머쌍으로 이루어지는 군으로부터 선택되는 프라이머쌍. - 제4항 또는 제5항에 기재된 프라이머쌍을 포함하는, 피검 시료로부터 PCR에 의해 소맥 내재성 DNA 서열을 검출 또는 정량하기 위한 키트.
- 유전자 재조합 소맥 및 비유전자 재조합 소맥이 공통으로 갖는 서열 7에 기재된 염기 서열의 적어도 80% 연속된 염기서열을 포함하는 DNA인 내재성 DNA와, 유전자 재조합 소맥의 각 계통에 특이적인 서열을 포함하는 하나 이상의 유전자 재조합 소맥 특이적 DNA를 포함하는 환상 DNA.
- 서열 7에 기재된 염기 서열의 적어도 80 % 연속된 염기 서열을 포함하는 DNA를 포함하는 환상 DNA.
- 제4항 또는 제5항에 기재된 프라이머쌍을 이용하여 PCR에 의해 증폭 가능한 영역을 포함하는 환상 DNA.
- 제8항에 있어서, 유전자 재조합 소맥의 각 계통에 특이적인 서열을 포함하는 하나 이상의 DNA를 추가로 포함하는 환상 DNA.
- 피검 시료에서의 유전자 재조합 소맥의 혼입률을 결정하는 방법이며,
제7항, 제8항 및 제10항 중 어느 한 항에 기재된 환상 DNA와 피검 시료로부터 추출한 DNA를 주형으로서 정량 PCR을 행하고,
환상 DNA에 대한 상기 정량 PCR의 결과를 이용하여 주형 DNA의 분자수를 구하기 위한 검량선을 작성하고,
피검 시료에 대한 상기 정량 PCR의 결과와 상기 검량선을 이용하여 상기 피검 시료 중에 포함되는 소맥 내재성 DNA 서열 부분 영역의 분자수와 1종 이상의 유전자 재조합 소맥에 특이적인 DNA 서열 부분 영역의 분자수를 구하고,
상기 유전자 재조합 소맥에 특이적인 DNA 서열 부분 영역의 분자수를 상기 소맥 내재성 DNA 서열 부분 영역의 분자수로 나누어 얻어지는 비 A를 구하는 것을 포함하는 방법. - 제11항에 있어서, 상기 비 A와,
유전자 재조합 소맥의 표준 종자로부터 추출한 DNA를 주형으로서 정량 PCR을 행하여 구해지는, 유전자 재조합 소맥의 각 계통에 특이적인 DNA 서열 부분 영역의 분자수를 소맥 내재성 DNA 서열 부분 영역의 분자수로 나누어 얻어지는 비 B를 이용하여 수학식 100×A/B를 계산함으로써 피검 시료 중의 유전자 재조합 소맥의 혼입률을 결정하는 공정을 추가로 포함하는 방법. - 제11항에 있어서, 상기 정량 PCR에서 피검 시료 중의 소맥을 PCR에 의해 검출 또는 정량하기 위한 프라이머쌍이며, 서열 7에 기재된 염기 서열의 적어도 80 % 이상의 연속된 영역을 증폭할 수 있는 것을 특징으로 하는 프라이머쌍으로부터 선택되는 1개 이상의 프라이머쌍을 이용하는 방법.
