Claims (5)
- 에바네센트파 형광 현미경을 이용하여, 생체재료로부터 유래한 DNA가 칩상에 고정된 DNA칩 자체를 검출하는 방법에 있어서,Au가 증착되어 있는 담체를 칩상에 고정화하는 단계;표적 DNA의 말단에 형광 물질을 수식시키는 단계;표면 플라스몬 공명(Surface plasmon resonance, SPR)에 의해 발생하는 에바네센트파를 이용하여 DNA에 수식된 형광 물질을 여기시키는 단계;상기 여기된 형광 물질을 에바네센트파 형광 현미경으로 검사하는 단계;를 포함하는 것을 특징으로 하는 에바네센트파 형광 현미경을 이용한 DNA칩 검출방법.
- 제1항에 있어서, 상기 형광 물질은 로다민(Rhodamin)인 것을 특징으로 하는 에바네센트파 형광 현미경을 이용한 DNA칩 검출방법.

- 제1항에 있어서, 상기 DNA는 매치 또는 미스매치 DNA인 것을 특징으로 하는 에바네센트파 형광 현미경을 이용한 DNA칩 검출방법.
- 제1항에 있어서, 상기 담체(particles)에 생체재료를 고정화하는 방법은3,3`-dithiodipropionic acid(3,3'-디티오디프로피온산) 수용액에 담체를 담그는 단계;NHS(엔-하이드록시서시니마이드, N-hydroxy succinimide)와 EDC(에틸디메틸아미노프로필카르보디이미드, 1-Ethy1-3-(3-dimethylaminopropyl) carbodiimide hydrochloride)의 혼합액에서 상기 수용액을 카르복실산과 반응시킨 후 건조시키는 단계;상기 건조된 담체를 아비딘 완충액에 담구어 놓는 단계;상기 아비딘 완충액의 담체를 에탄올아민 수용액에 담그고 카르복실기를 불활성화시키는 단계;아비딘을 수식한 담체를 완충액에 비오틴화 프로브 DNA를 용액에 담그는 단계;를 포함하는 것을 특징으로 하는 에바네센트파 형광 현미경을 이용한 DNA칩 검출방법.
