Claims (15)
- 비평면 형상의 고체 지지체와, 미생물 및 소듐 폴리아네톨 술포네이트를 포함하는 혈액 배양물을 pH 3.0 내지 6.0의 범위에서 접촉시켜 상기 미생물을 상기 고체 지지체에 부착시키는 단계,상기 고체 지지체에 부착되지 않은 물질을 세척하여 제거하는 단계, 및상기 고체 지지체에 부착된 상기 미생물에 대하여 PCR을 수행하여 핵산을 증폭하는 단계를 포함하는, 미생물로부터 핵산을 증폭하는 방법으로서,상기 접촉단계, 세척 단계, 및 증폭 단계는 동일한 용기 내에서 수행되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 미생물은 박테리아, 곰팡이 또는 바이러스인 것인 방법.
- 삭제
- 제1항에 있어서, 상기 시료는 포스페이트 버퍼 또는 아세테이트 버퍼로 희석된 것인 방법.
- 제4항에 있어서, 상기 희석은 1:1 내지 1:10의 비율로 수행되는 것을 특징으 로 하는 방법.
- 제4항에 있어서, 상기 시료는 10 mM 내지 500 mM의 염 농도를 갖는 것인 방법.
- 제6항에 있어서, 상기 시료는 50 mM 내지 300 mM의 염 농도를 갖는 것인 방법.
- 제1항에 있어서, 상기 비평면 형상의 고체 지지체는 표면에 복수 개의 기둥 (pillar)이 형성되어 있는 기둥 구조를 갖는 고체 지지체, 비드 형상을 갖는 고체 지지체 및 표면에 복수 개의 공극이 형성되어 있는 체 (sieve) 구조를 갖는 고체 지지체로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제8항에 있어서, 상기 기둥은 어스팩 비가 1:1 내지 20:1인 것인 방법.
- 제8항에 있어서, 상기 기둥 구조는 기둥 높이 대 기둥 사이의 거리의 비율이 1:1 내지 25:1인 것인 방법.
- 제8항에 있어서, 상기 기둥 구조는 기둥 사이의 거리가 5 ㎛ 내지 100 ㎛의 범위를 갖는 것인 방법.
- 제1항에 있어서, 상기 비평면 형상의 고체 지지체는 물 접촉각이 70°내지 95°인 소수성을 갖는 것을 특징으로 하는 방법.
- 제12항에 있어서, 상기 소수성은 상기 고체 지지체의 표면을 옥타데실디메틸(3-트리메톡시실릴 프로필)암모늄 (OTC) 또는 트리데카플루오로테트라히드로옥틸트리메톡시실란 (DFS)의 화합물로 코팅하여 부여되는 성질인 것을 특징으로 하는 방법.
