Abstract

Description
본 발명은 유전자 요법 및 종양 치료 분야에 관한 것이다. 본 발명은 좀더 구체적으로 말하면 종양 세포에서 요오드의 축적을 촉진하기 위해 아데노바이러스 벡터를 이용하여 종양 세포 안으로 특정 요오드 트랜스포터(Na+/I- 심포터) NIS를 암호화 하는 유전자의 도입에 관한 것이다. 본 발명은 또한 nis 유전자를 포함하는 복제-결손 재조합 아데노바이러스 및 종양 세포 안으로 nis 유전자의 전달과 요오드-131을 이용한 대사 방사선 요법을 병행한 암 치료방법에서 이들 벡터의 용도에 관한 것이다.
요오드-131은 분화된 갑상선암의 치료에 50년 이상 사용되어왔다. 이의 치료 효능은 종양 조직으로 전달되는 방사선 조사량과 연관된다. 예를 들어, 전이의 경우, 종양 조직으로 전달되는 조사량이 80 그레이 이상일 때 요오드-131을 이용한 처리 후 종양 반응이 관찰되지만 35 그레이 이하의 조사량의 경우 종양 반응 수준은 낮거나 0이다[Maxon, NEJM, 309:937-941, 1983]. 일반적으로 종양 분량을 처리하거나 감소시키는 데 필요한 총 조사량은 방사선 감도에 따라 40 내지 60 그레이 범위에서 달라지는 것으로 판단된다. 이러한 총 조사량은 분화된 갑상선암의 고정된 전이의 치료도중 수행될 경우 요오드-131을 이용한 수회 처리에 의해 전달될 수 있다.
종양 조직으로 전달되는 방사선 조사량은 두가지 생물학적 인자에 좌우된다: 종양 조직에서 요오드-131의 유효 반감기(Teff) 및 방사능 농도.
- 종양 조직에서 요오드-131의 유효 반감기(Teff)는 상관관계 I/Teff = I/Tbiol
+ I/Tphys에 따라 생물학적 반감기(Tbiol)와 물리적 반감기(Tphys)에 좌우된다. 고정된 세포에서 요오드-131의 생물학적 반감기(Tbiol)는 요오드의 유기화(organification) 능력에 좌우된다. 유기화 부재시, 예를 들어 비갑상선 세포에서처럼, 생물학적 반감기는 수 시간 내지 수십 시간 정도로 짧다. 요오드-131의 물리적 반감기(Tphys)는 8.02일이다. 이에 따라, 기껏해야, 종양 조직에서 요오드-131의 유효 반감기는 수시간 내지 수십 시간으로 추정할 수 있다.
- 방사능 농도는 종양 조직에 의한 요오드-131의 전체 고정화 및 이러한 조직의 매스간의 비율이다.
예를 들어, 조직 1 g당 투여되는 방사능의 0.1%의 요오드 고정화 및 1.5일의 유효 반감기의 경우, 3.7 기가베크렐 (또는 100 mCi)의 투여가 종양 조직으로 30 그레이의 조사량을 전달하는 것으로 추정할 수 있다.
요오드-131의 전체 고정은 한편으론 환자에게 투여되는 요오드-131의 방사능 에 좌우되고, 다른 한편으론 요오드-131을 농축시키는 종양 조직의 능력에 좌우되는 파라미터이다.
- 환자에게 투여될 수 있는 요오드-131의 최대 방사능은 건강한 조직, 특히 골수에 전달되는 방사선 조사량으로 인한 요오드-131의 요오드 독성에 의해 제한된다. 이에 따라, 갑상선암의 경우, 환자가 갑상선 기능 저하증을 앓고 있는 동안 투여되는 치료 방사능은 일반적으로 3.7 내지 10 GBq(100 내지 300 mCi)이다. 갑상선 기능 정상 환자에게 투여될 수 있는 최대 방사능은 이보다 높은데 그 이유는 요오드 보유력이 갑상선 기능 저하증의 경우에 관찰되는 것보다 두배 정도 낮기 때문이며; 이는 8.5 내지 22.2 GBq (500 내지 600 mCi)의 값에 이른다.
- 환자에게 투여되는 정해진 방사능의 경우, 요오드-131을 농축시키는 정상 조직의 능력은 요오드 심포터의 발현 수준 및 이의 생물학적 활성에 좌우된다. 이는 정상인과 병상인의 갑상선 조직 연구 및 NIS 활성의 시험관내 연구에 의해 입증되었다. 요오드-131의 고유 방사능이 높아, 투여되는 요오드-131 방사능의 증가에 따른 포화 현상이 관찰되지 않았다.
환자에게 투여될 수 있는 요오드-131의 최대 방사능의 한계로 인해, 요오드-131의 전체 고정을 증가시키기 위해서는 바라건데 요오드-131을 농축시키는 종양 조직의 능력을 증가시킬 수 있어야 한다.
또한 요오드 고정의 증가가 종양 조직에서 균일하게 일어나는 것이 중요하다. 실제로, 고정화 조직에서 요오드의 고정화 정도는 세포간에, 또한 종양 영역간에 매우 다를 수 있다. 종양 조직에서 방사능 조사량은 본질적으로 요오드-131의 β방출에 의해 전달된다. 그러나, 생물학적 조직에서 이의 통로는 겨우 2 내지 3 mm이다. 또한, 국지화된 출처 주변에서, 방사능 조사량은 거리에 따라 지수함수적으로 감소한다. 이러한 요소들은 고정화 종양 영역에 높은 방사능 농도를 달성하는 중요성과, 다른 한편으로, 가능한 균일한 종양 고정화를 달성할 필요성을 강화시킨다.
인간 갑상선에 의한 요오드-131의 고정화, 및 이에 따른 이의 방사능은 3주간 3일분씩 나눠 L-트리이오도티로닌 1 ㎍/kg/일의 투여에 의해 쉽게 억제될 수 있음을 주목해야 한다(요오드-131의 무한한 고정화).
NIS 트랜스포터는 갑상선 세포에 의해 유동 요오드(요오드화물 I- 형태)의 농축을 담당한다[참조: P. 쥐 트랜스포터를 암호화 하는 유전자[Dai et al. Nature 379:458-460 (1996)] 및 NIS 요오드의 인간 트랜스포터[Smanik P.A. et al. Biochem. Biophys. Res. 제 1 인자는 종양 세포에서 nis 유전자의 낮은 발현 수준이다. 예를 들어, 앞서 언급된 연구에서, 시험관내에서 관찰된 결과는 비갑상선 종양 세포에서의 요오드 농도가 갑상선 세포에서 축적된 요오드 농도보다 대략 2배 낮음을 보여준다.

또다른 제한 인자는 비갑상선 세포내에서 요오드의 유기화의 부재인데; 이러한 세포내에 요오드를 유지하기 위해서는 요오드의 이러한 유기화가 필수 사항이다. 이에 따라, 앞서 보고된 연구에서, 시험관내에 축적된 요오드의 매우 빠른 방출(30 내지 60분내)이 비갑상선 세포의 경우에 관찰된다.
마지막으로, 종양 세포의 비균일성과 막 수준의 이들 세포의 엄청난 변형을 고려할 때, 종양 세포 막에서 NIS 트랜스포터를 일정하게 분배하기란 어려울 것 같다.
이에 따라, 지금껏 공지된 방법 중에는 비갑상선 종양내 요오드의 축적을 비갑상선 종양의 치료를 위해 요오드-131을 사용할 정도로 충분한 수준의 발현에 이 르게끔 할 수 있는 방법이 없다.
본 발명은 유전자 요법과 요오드-131을 이용한 방사선 요법을 병행한 개선된 종양 치료법을 제공한다.
본 발명은 특히 비갑상선 종양 수준에서 요오드의 고정화 효능을 증가시킬 수 있고 갑상선 암의 치료를 위해 성공적으로 개발된 방사선 요법의 원리를 비갑상선 종양에 적용시킬 수 있는 방법에 관해 기재하고 있다.
본 발명은 결손 재조합 아데노바이러스에 의한 특정 요오드 트랜스포터(Na+/I- 심포터) NIS를 암호화 하는 유전자의 전달이 비갑상선 종양 세포내에 요오드의 매우 높은 축적에 이르게끔 할 수 있음을 증명한 결과이다. 특정 양태에서, nis 유전자를 발현시키는 결손 아데노바이러스를 공급하여 갑상선 세포내에서 관찰된 것보다 약 5배 정도 높은 농도로 요오드를 축적할 수 있다. 요오드-125를 축적하는 비갑상선 종양 세포의 이러한 놀라운 능력으로 인해 요오드-131을 이용한 대사 방사선 요법에 기초한 신규 치료법을 이러한 종류의 종양에 적용할 수 있으며, 이러한 접근법은 지금까지는 특정 갑상선 종양의 치료에 한정되었다.
대사 방사선 요법의 주된 이점은 주변 조직을 그다지 방사선 조사하지 않고도 방사선 동위원소를 고정하는 조직으로 다량의 방사선 조사량을 전달하는 데 있다.
일반적으로 이러한 요오드 요소를 축적할 수 없는 종양 세포에서 실질적으로 요오드를 효과적으로 축적할 수 있다는 발견으로 인해 이러한 방법을 비갑상선 기원의 종양 치료를 위한 다수의 징후에 적용할 수 있다. 좀더 구체적으로 말하면 이러한 징후에는 두가지 종류가 있다: 지리적으로 외부 방사선 요법에 영향받기 어려운 종양; 예를 들면, 수술불능의 전립선암 또는 뇌종양이 언급될 수 있다; 이미 방사선 조사된 종양의 경우, 부가적인 외부 방사선 요법이 불가능한데 그 이유는 최대 조사량이 이미 투여되었기 때문이다: 이는 조사된 영역내에 일어나거나 재발하는 모든 암과 관련된다. 이러한 방법은 또한 요오드를 포획하는 능력을 이미 상실하여 대사 요법에 반응하지 않는 갑상선 종양의 치료에도 적용될 수 있다.
분화된 갑상선암의 전이의 경우 수행되는 대사 방사선 요법과 외부 방사선 요법을 병행할 수도 있다. 이러한 병행법에 의해 독성 증가없이 방사선 요법의 치료 효능을 증가시킬 수 있다. 이러한 병행법은 방사선 요법에 그다지 민감하지 않은 종양의 경우에 특히 유리한 것 같다. 또한, 과도하게 큰 크기로 인해, 대사 방사선 요법만으로 접근이 어려운 종양에 이러한 기술을 적용할 수 있다.
본 발명의 제 1 주제는 특정 요오드 트랜스포터(Na+/I- 심포터) NIS 또는 이의 유도체 중 하나를 암호화 하는 적어도 하나의 이종 DNA 서열을 포함하는 결손 재조합 아데노바이러스에 있다.

본 발명의 문맥에 이용된 아데노바이러스는 재조합 아데노바이러스, 즉, 이종 DNA 서열을 포함하는 재조합 아데노바이러스이다. 유리하게는, 이는 결손 재조합 아데노바이러스, 즉, 표적 세포에서 자가 복제할 수 없는 아데노바이러스이다.
본 발명에 따른 아데노바이러스의 작제를 위해, 다양한 혈청타입이 이용될 수 있다. 실제로 구조 및 성질면에서 다소 차이가 있지만, 대등한 유전 체계를 나타내는 다수의 혈청타입이 존재한다. 좀더 구체적으로 말하면, 재조합 아데노바이러스는 인간 또는 동물 기원일 수 있다. 인간 기원의 아데노바이러스의 경우, 바람직하게는 그룹 C로 분류된 아데노바이러스, 특히 아데노바이러스 타입 2(Ad2), 5(Ad5), 7(Ad7) 또는 12(Ad12)이 언급될 수 있다. 동물 기원의 다양한 아데노바이러스 중에서, 바람직하게는 개 기원의 아데노바이러스, 특히 모든 CAV2 아데노바이러스 균주[예를 들면 manhattan 또는 A26/61 균주(ATCC VR-800)]가 언급될 수 있다. 동물 기원의 기타 아데노바이러스는 특히 출원 WO 94/26914에서 언급된다.
아데노바이러스의 게놈은 특히 각 말단에 역 말단 반복 서열(ITR), 캡시드화 서열(Psi), 초기 유전자 및 후기 유전자를 포함한다. 주된 초기 유전자는 E1, E2, E3 및 E4 영역에 함유된다. 이들 중, E1 영역에 함유된 유전자가 특히 바이러스 증폭에 필요하다. 주된 후기 유전자는 L1-L5 영역에 함유된다. Virol. 1975, 15(2) 348-354). 기타 벡터는 바이러스 복제 및/또는 증폭에 필수적인 또다른 영역, E4 영역의 결실을 포함한다. E4 영역은 실제로 후기 유전자의 발현 조절, 후기 핵 RNA의 안정성, 숙주 세포의 단백질의 발현 소멸 및 바이러스 DNA의 복제 효율에 관여한다. E1과 E4 영역이 결실된 아데노바이러스 벡터는 전사 백그라운드 노이즈와 굉장히 감소된 바이러스 유전자의 발현을 보유한다. 이러한 벡터는 예를 들면 출원 WO 94/28152, WO 95/02697, WO 96/22378에 기재되어 있다. 부가적으로, IVa2 유전자에 변형을 운반하는 벡터도 공지되어 있다(WO 96/10088).
본 발명의 바람직한 양태에서, 재조합 아데노바이러스는 그룹 C 인간 아데노 바이러스이다. 좀더 바람직하게는, 이는 Ad2 또는 Ad5 아데노바이러스이다.

또다른 양태에 따르면, 본 발명의 문맥상 사용되는 재조합 아데노바이러스는 E1 영역에서 결실 이외에 이의 게놈의 E4 영역의 전부 또는 일부에 영향을 미치는 결실을 포함한다. 좀더 구체적으로 말하면, E4 영역에서 결실은 모든 개방 위상에 영향을 미친다. 예를 들면 출원 WO 95/02697 및 WO 96/22378에 공지된 바와 같이, E4 영역(예, ORF6 또는 ORF3)의 결실 33466-35535 또는 33093-35535 또는 이의 일부가 언급될 수 있다.
이는 예를 들면 E1과 E4 영역에서 전체적 또는 부분적으로 결손이 존재하고 임의로는 E3 영역의 경우에 결손이 존재하는 소위 3세대 재조합 아데노바이러스일 수 있다. 본 발명의 특정 양태는 하기 기능 영역의 전부 또는 일부에 영향을 미치는 결실을 운반하는 아데노바이러스의 사용에 있다:
- E1, E4 및 E3,
- E1, E4 및 E2,

이는 또한 출원 WO 99/60144에 기재되고 아데노바이러스가 또다른 바이러스에 대한 벡터, 특히 레트로바이러스 벡터로서 이용되는 경우와 같이 하이브리드 아데노바이러스 벡터를 포함할 수 있다.
요오드 트랜스포터(NIS)를 암호화 하는 핵산을 함유한 발현 카세트는 재조합 게놈의 여러 부위에 삽입될 수 있다. 이는 결실된 서열에 대한 대체물로서 또는 부가적으로 E1, E3, 또는 E4 영역에 삽입될 수 있다. 이는 또한 바이러스 생성을 위해 cis에서 필요한 서열(ITR 서열 및 캡시드화 서열) 밖의 임의 기타 부위에 삽입될 수 있다.

재조합 아데노바이러스는 캡시드화 계통, 즉, 재조합 아데노바이러스 게놈에 결핍된 1 이상의 기능을 trans로 상보할 수 있는 세포 계통에서 생성된다. 당업자에게 공지된 캡시드화 계통 중에서, 예를 들면 아데노바이러스 게놈의 일부가 통합된 293 계통이 언급될 수 있다. 좀더 정확히 말하면, 293 계통은 좌측 ITR, 캡시드화 영역, (E1a와 E1b를 포함한) E1 영역, pIX 단백질을 암호화 하는 영역 및 pIVa2 단백질을 암호화 하는 영역의 일부를 포함하는 혈청타입 5 아데노바이러스(Ad5)의 게놈의 좌측 말단(약 11-12%)을 함유한 인간 배 신장 세포 계통이다. 이러한 계통은 E1 영역에 결손이 존재하는, 즉 E1 영역 전부 또는 일부가 결핍된 재조합 아데노바이러스를 트랜스상보할 수 있고 높은 역가를 지닌 바이러스 스톡을 생산할 수 있다. 이러한 계통은 또한 허용 온도(32℃)에서 열-민감성 E2 돌연변이를 포함하는 바이러스 스톡을 생산할 수도 있다. 특히 인간 폐 육종 세포 A549(WO 94/28152) 또는 인간 망막아세포(Hum. Ther. 2(1995) 322; Krougliak et al., Hum. Gen. Ther. 6(1995) 1575) 및 E1과 E2 영역을 상보하는 계통(WO 94/28152, WO 95/02697, WO 95/27071) 또는 최소 아데노바이러스를 생성하는 데 이용될 수 있는 계통이 언급될 수 있는데, 그 이유는 특히 이들이 부가적으로 이러한 바이러스의 작제에 관여된 부위-특이성 재조합효소 활성을 발현시키기 때문이다.
재조합 아데노바이러스는 또한 종양 수준에서 감염 효율을 증가시키기 위해 캡시드의 구조에서 변형될 수 있다. 예를 들어, 캡시드는 아데노바이러스를 종양 세포에 표적화시킬 수 있는 uPAR 리간드 또는 RGD 모티브를 함유할 수 있으며, 이러한 벡터 및 표적화 서열은 특히 출원 WO 00/12738에 공지되어 있다. 표적화 서열은 헥손 단백질 또는 섬유 단백질 안으로 삽입될 수 있다. 바람직하게는, 표적화 서열은 섬유 또는 헥손 단백질의 결실 수준에서 삽입된다. 유리하게는 Ad5의 헥손의 폴리펩타이드 서열의 위치 279-292에 상응하는 헥손 13개 아미노산의 폴리펩타이드 서열에서 결실된다. 유리하게는 Ad5의 섬유(HI 루프)의 폴리펩타이드 서열의 위치 539-547에 상응하는 섬유 11개 아미노산의 폴리펩타이드 서열에서 결실된다.
재조합 아데노바이러스는 보통 바이러스 DNA를 캡시드화 계통에 도입한 다음 약 2 또는 3일 후에 세포를 용해시켜 생성된다(아데노바이러스 사이클의 동역학은 24 내지 36시간임). 방법 실행을 위해, 도입되는 바이러스 DNA는 임의로 박테리아(WO 96/25506) 또는 효모(WO 95/03400)에서 작제되어 세포 안으로 감염되는 완전한 재조합 바이러스 게놈일 수 있다. 이는 또한 캡시드화 계통을 감염시키는 데 사용된 재조합 바이러스일 수도 있다. 바이러스 DNA는 또한 단편 형태로 도입될 수 있으며 각각은 재조합 바이러스 게놈, 및 캡시드화 세포 안으로 도입 후 각종 단편간의 상동 재조합에 의해 재조합 바이러스 게놈을 재구성할 수 있는 상동성 영역의 일부를 운반한다.

발현 조절 서열. 특정 요오드 트랜스포터(Na+/I- 심포터, NIS)를 암호화 하는 유전자는 기능적이고 숙주 종양 세포에서 발현하는 프로모터 또는 프로모터/인핸서와 같은 발현 조절 서열의 통제하에 놓일 수 있다.
발현 조절 서열은 프로모터 영역 이외에 특정 요오드 트랜스포터를 암호화 하는 유전자의 3' 위치에 위치되고 전사 종결을 위한 시그널과 폴리아데닐화 부위를 제공하는 영역을 포함한다. 이들 모든 요소가 발현 카세트를 구성한다.

용어 "유효량"은 요오드 트랜스포터(NIS)를 암호화 하는 핵산을 포함하는 아데노바이러스의 투여가 요오드-135를 이용한 대사 방사선 요법 치료와 병행될 때 종양 용적을 적어도 대략 15%, 바람직하게는 적어도 50%, 좀더 바람직하게는 적어도 90% 정도 감소시키기에 충분한 양, 더욱더 바람직하게는 종양을 제거하기에 충분한 양을 말한다.

플라스미드 DNA의 예비 추출, 세슘 클로라이드 구배에서 플라스미드 DNA의 원심분리, 아가로스 또는 아크릴아미드 겔 전기영동, 전기용출에 의한 DNA 단편의 정제, 페놀 또는 페놀-클로로포름을 이용한 단백질의 추출, 에탄올 또는 이소프로판올을 이용하여 식염수 매질에서 DNA의 침전, 에쉐리키아 콜라이(Escherichia coli)에서 형질전환 등과 같은 분자 생물학에서 통상적으로 사용되는 방법은 당업자에게 익히 공지되어 있고 문헌[참조: Maniatis T. et al., "Molecular Cloning, a Laboratory Manual", Cold Spring Harbor Laboratory, Cold Spring Harbor, N.Y., 1982; Ausubel F.M. et al. 연결을 위해, DNA 단편을 아가로스 또는 아크릴아미드 겔 전기영동에 의해 크기에 따라 분리하고, 페놀 또는 페놀/클로로포름 혼합물로 추출하며, 에탄올로 침전시킨 다음 공급자의 추천에 따라 T4 파아지 DNA 리가제(Biolabs)의 존재하에 배양할 수 있다.

실시예 1: 특정 요오드 트랜스포터(Na

심포터) NIS를 암호화 하는 유전자를 발현시키는 결손 재조합 아데노바이러스의 작제.
본 실시예는 CMV 프로모터와 작동가능하게 연결된 특정 요오드 트랜스포터(Na+/I- 심포터) NIS(Genbank U60282 참조)를 암호화 하는 유전자를 운반하는 결손 아데노바이러스 벡터의 작제에 관해 기재하고 있다.
래트 nis 유전자 서열의 뉴클레오타이드 74-2046을 포함하는 단편(Dai et al., 1996, Nature 379: 458-460)을 AatII-HindIII 단편 형태로 플라스미드 pCEP-4의 PvuII-HindIII 부위 안으로 클로닝시켰다(Invitrogen). AatII-HindIII 단편의 AatII 부위를 플라스미드 pCEP-4에 삽입하기 이전에 멍 빈(Mung Bean) 뉴클레아제(Biolabs)로 처리하여 미리 블런트하게 만들었다. 생성된 플라스미드를 pAB1이라 부른다.
플라스미드 pAB1(도 1)은 발현이 사이토메갈로바이러스 CMV 인핸서/프로모터(뉴클레오타이드 -522에서 +72)(Boshart et al. 1985, cell, 41: 521-530)의 통제하에 놓인 nis 유전자를 포함하고, 그 뒤에 SV40 바이러스 폴리아데닐화 부위(SV40 게놈에 따르면 뉴클레오타이드 2538-2759, Genbank 좌위 SV4CG)가 따른다.
(i) 사이토메갈로바이러스 인핸서/프로모터, (ii) nis 유전자의 cDNA 및 (iii) SV40 바이러스 폴리아데닐화 부위에 의해 형성된 조합물은 이후 NIS 발현 카세트라 불린다.
앞서 정의된 NIS 발현 카세트를 포함하는 플라스미드 pAB1의 SspI-EcoRV 단편(3711 bp)을 미리 EcoRV로 선형화된 셔틀 벡터 pXL3048 안으로 클로닝시킨다. 생성된 벡터를 pAB2라 부른다(도 2). 플라스미드 pXL3048은 타입 5 아데노바이러스 게놈의 좌측 말단(뉴클레오타이드 1-382), 세가지 독특한 클로닝 부위를 포함하는 폴리링커 및 pIX 유전자의 일부(뉴클레오타이드 3446-4296)을 포함하는 플라스미드 Kan-SacB(Crouzet et al., PNAS vol. 94, p. 1414-1419, 1997)에서 유도된 플라스미드이다.
플라스미드 pAB2와 플라스미드 pXL3215간의 상동 재조합에 의해 Crouzet 등이 기술한 방법(PNAS vol. 94, p. 1414-1419, 1997)에 따라 아데노바이러스 Ad-NIS를 작제했다.
플라스미드 pXL3215는 E1 영역 안으로 삽입된 RSV-LacZ 카세트(ㅿE1 386-3446)와 E3 영역에서 결실(ㅿE3 28592-30470)을 함유한 아데노바이러스의 게놈을 함유한다.
작제의 기본 원리는 도 2에 설명되어 있다. 이러한 이중 재조합에 의해 생성된 플라스미드 pXL3215는 E1과 E3 영역에서 결실이 존재하고 CMV-NIS-poly A SV40 발현 카세트를 함유한 타입 5 아데노바이러스 게놈을 함유한다. 이러한 작제물은 NIS 발현 카세트를 서열분석하여 확인된다.
Ad-NIS 아데노바이러스는 PacI로 절단된 pXL3215의 DNA를 293 계통(ATCC CRL-1573) 안으로 감염시켜 생성된다. 얻어진 바이러스 입자를 동일 계통에서 증폭시키고 생성된 바이러스 스톡을 이중 CsCl 구배에 의해 정제한다.
바이러스 입자를 이용하여 각종 종양 계통에서 NIS 유전자의 발현을 연구한다.
실시예 2: Ad-NIS 벡터로 감염된 종양 세포에 의해 시험관내 요오드의 축적
Ad-NIS 벡터로 감염된 세포에서 요오드를 축적하는 능력을 다양한 계통에서 시험했다: 내피 종양 세포 SiHa(ATCC HTB-35), 유방암 세포 MCF7(ATC HTB-22) 및 T-47D(ATCC HTB-133) 및 전립선 기원 DU-145의 종양 세포(ATCC HTB-81) 및 PC-3(ATCC CRL-1435).
세포에 감염 중복도 10으로 Ad-NIS 벡터를 감염시킨다. 요오드를 포획하는 세포의 능력을 Weiss 등의 방법[Endocrinology (1984) 114: 1090-1098]에 따라 감염 후 28 내지 29시간째 시험한다. 감염된 세포를 HBSS 완충액으로 세척하고 요오 드-125 0.1 μCi를 함유한 HBSS 완충액 0.5 ml와 도 3에 나타낸 시간동안 접촉시킨다. 접촉 시간 마지막에, 세포를 콜드 HBSS 완충액으로 1회 세척하고 콜드 에탄올로 20분간 배양한다. 감마 카운터를 이용하여 에탄올의 방사능을 분석하여 세포에 의해 포획된 요오드-125의 양을 측정한다.
SiHa 계통을 이용하여 얻어진 결과는 도 3에 주어지며 이를 FRTL-5 세포(요오드를 자연스레 포획하고 대조표준으로서 작용하는 래트 갑상선 세포)를 이용하여 얻어진 결과와 비교할 수 있다.
NIS에 의한 요오드 전달의 특이적 억제를 퍼클로레이트의 존재하에 시험했다. NaClO4를 요오드-125를 함유한 HBSS 완충액에 첨가하여 최종 농도 30, 300 및 3000 μM을 얻었다. 세포와 요오드-125간의 접촉 시간은 15분이다. 결과는 도 4에 주어진다. 결과를 동일한 세포 수에 대한 카운트 수로서 표현한다.
FRTL-5 세포(양성 대조표준)는 특정 양의 요오드를 고정시키고, 이러한 현상이 매우 빠름이 관찰된다(접촉 15분 후 거의 최대에 이름). 얻어진 결과는 감염되지 않은 SiHa 세포(인간 내피 종양 세포)가 요오드를 포획하지 않음을 보여준다. 놀랍게도, Ad-NIS 벡터로 감염된 SiHa 세포가 FRTL-5 세포보다 5배 많은 요오드를 포획함이 관찰되었다. 또한 이러한 현상의 초기 동역학이 FRTL-5 세포의 경우에 관찰된 것에 필적할 만함도 주목할 만하다. nis 유전자를 함유하지 않은 대조표준 아데노바이러스로 감염시켜 전달의 특이성을 확인했다(결과는 제시되지 않음).
억제 실험의 결과(도 4)는 FRTL-5 세포내에서 요오드의 고정이 30 μM의 NaClO4 농도에서 90% 이상(30 μM의 경우 93%, 고농도에서 95%) 억제됨을 보여준다. 다른 한편으론, Ad-NIS로 감염된 SiHa 세포의 경우, 동일한 수준의 억제를 얻기 위해 필요한 억제제의 농도가 이보다 높은 것도 주목할 사항이다(30 μM NaClO4의 경우 단지 40% 억제, 300 μM NaClO4의 경우 92% 억제, 및 3 mM NaClO4의 경우 98% 억제).
억제 실험은 요오드를 포획하기 위해 Ad-NIS로 감염된 SiHa 세포의 능력이 실제로 이들 세포내에서 발현 NIS에 기인한다고 결론지을 수 있는데, 그 이유는 이러한 현상이 경쟁적이고 NIS에 의한 요오드화물 이온의 수송의 고유한 억제제인 NaClO4에 의해 억제되기 때문이다. 나머지 인간 종양 세포 계통의 경우에 유사한 결과가 얻어졌다(유방암 세포: MCF-7 및 T47-D, 및 전립선 기원의 종양 세포: DU-145 및 PC-3).
NaClO4에 의한 억제와 관련한 결과는 Ad-NIS에 의한 종양 세포의 감염이 특히 높은 수준의 NIS 발현을 야기함을 확인시켜 주는데, 그 이유는 고농도 NaClO4가 이러한 현상을 억제시키는 데 필요하기 때문이다(SiHa 세포의 경우에, FRTL-5 세포와 비교하여, 동일 수준의 억제를 얻는데 10배 높은 억제제 농도를 요구함).
이들 모든 결과는 아데노바이러스 벡터가 nis 유전자의 수송에 특히 유리한 벡터이고 이러한 벡터를 사용하여 요오드-125를 자연적으로 포획하는 세포에서 관찰되는 것보다 5배 높은 비갑상선 종양 세포내 요오드의 축적 수준을 얻을 수 있음 을 입증해 준다.
실시예 3: Ad-NIS 벡터로 감염된 종양에 의한 생체내 요오드의 축적
MCF-7 세포(유방암의 인간 종양 세포)를 누드 마우스에 피하 주사했다(5 x 106 세포). 15일 후(종양 출현 개시), 15일간 매일 티록신을 복막내 주사하여(2 ㎍/동물/일), 동물의 갑상선을 휴지시켰다. Ad-NIS 벡터를 종양내 경로에 의해 특정 동물 안으로 주사하고(2 x 109 pfu/종양); 주사시 종양의 직경은 3 내지 6 mm였다. 주사 후 3일째, 125I 6 μCi를 복막내 경로에 의해 마우스에 주사하고, 동물상의 다양한 부위로 이동될 수 있는 프로브에 의해 10초에 걸쳐 카운트를 수행했다. 프로브를 갑상선 위, 종양 위, 및 종양 바로 근처에 두어 일정한 시간 간격으로 카운트를 수행했다. 결과는 도 5에 주어진다. 결과를 시간에 따라 검출된 카운트의 수로서 표현한다(10초에 걸쳐 측정).
대조표준 동물(Ad-NIS의 투여없이)에서, 갑상선은 소량의 요오드를 고정시키지만, 티록신 처리에 의해, 시간에 따라 갑상선에서 요오드의 실질적인 축적이 관찰되지 않는다. 대조표준 동물내 종양에서 요오드의 고정화가 관찰되지 않는다(도 5a 참조). NIS가 본래 위에 존재한다는 사실과, 요오드가 소변으로 제거된다는 사실이 관찰된 비교적 높은 값을 설명해 준다(비교적 높은 백그라운드 노이즈).
종양내 경로에 의해 투여된 Ad-NIS로 처리된 동물(도 5b)은 갑상선에서 요오드의 실질적인 축적을 나타내지 않는다(티록신 처리 효과). 이들 동물 중, 종양상에 프로브를 두어 검출된 요오드의 양은 종양 근처에서 관찰된 요오드의 양보다 체 계적으로 높은데, 이는 Ad-NIS 벡터를 처리한 후 종양내에서 실제로 요오드가 고정됨을 나타낸다.
종양에서 요오드의 고정화를 확인하기 위해, 신티그래피에 의한 방사능 측정을 Ad-NIS 벡터로 처리된 동물에서 수행했다. 얻어진 결과는 도 6에 나타나 있다. 사용된 동물을 앞서 기재된 프로토콜에 따라 제조하되, 123I 50 μ/Ci를 복막내 경로에 의해 주사하고; 요오드를 주사한 후 1시간째에 이미지를 얻었다.
이미지는 동물의 복부도이다. 4군데 요오드 고정화 영역을 눈으로 확인할 수 있다: 갑상선, 방광(요오드 제거를 위한 경로), 위(NIS가 자연적으로 발현됨) 및 종양. 이러한 신티그래피 결과는 카운팅에 의해 얻어진 상기 결과를 확인시켜주고 본 발명에 따른 벡터가 종양에서 요오드의 실질적인 고정화를 얻을 수 있음을 보여준다.
실시예 4: Ad-NIS 벡터로 감염된 종양에 의한 생체내 요오드의 축적의 정량 평가
SiHa 세포(5 x 106 세포)를 누드 마우스의 두 옆구리 상으로 피하 주사했다(n = 12). 15일 후(종양 출현 개시), 15일간 매일 티록신을 복막내 주사하여(2 ㎍/동물/일), 동물의 갑상선을 휴지시켰다. 각 동물의 경우, 종양내 경로에 의해 Ad-NIS 벡터를 종양 안으로 주사하고(2 x 109 pfu/종양); 나머지 종양을 대조표준으로서 제공하며; 주사 시점의 종양의 직경은 5 내지 8 mm였다. 감염 3일 후, 125I 6 μCi를 복막내 경로에 의해 마우스에 주사했다. 125I의 투여 후 90분째에 동물을 죽이고, 125I의 양을 측정했다. 결과는 표 1에 주어진다. 결과를 조직 1 그램당 투여되는 125I의 퍼센티지로서 표현한다.

얻어진 결과는 Ad-NIS 벡터로 처리된 종양이 처리되지 않은 종양보다 요오드 125I를 매우 효율적으로 25배 많게 축적할 수 있음을 보여준다. 이러한 결과는 상기 결과를 확인시켜주며 본 발명에 따른 벡터에 의해 종양에서 요오드의 실질적인 고정화를 얻을 수 있음을 보여준다.
Claims (21)
- 요오드 트랜스포터(Na+/I- 심포터) NIS 또는 이의 유도체를 암호화하는 하나 이상의 DNA 서열을 포함함을 특징으로 하는 복제 결손 재조합 아데노바이러스.
- 제 1 항에 있어서, DNA 서열이 cDNA 서열임을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, DNA 서열이 gDNA 서열임을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, DNA 서열이 쥐 요오드 트랜스포터(Na+/I- 심포터) NIS를 암호화함을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, DNA 서열이 인간 요오드 트랜스포터(Na+/I- 심포터) NIS를 암호화함을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, DNA 서열이 전사 프로모터의 통제하에 놓여 종양 세포에서 발현됨을 특징으로 하는 아데노바이러스.
- 제 6 항에 있어서, 전사 프로모터가 바이러스 프로모터 중에서 선택됨을 특징으로 하는 아데노바이러스.
- CMV 프로모터의 통제하에 인간 요오드 트랜스포터 NIS를 암호화 하는 cDNA 서열을 포함하는 복제 결손 재조합 아데노바이러스.
- 종양 세포에서 발현시키는 프로모터의 통제하에 요오드 트랜스포터 NIS 또는 이의 유도체를 암호화하는 DNA 서열을 포함하는 복제 결손 재조합 아데노바이러스.
- 제 9 항에 있어서, 프로모터가 엘라스타제 I 유전자, 인슐린 유전자, 면역글로불린 유전자, 마우스 유방암 바이러스, PSA 유전자, 알파-페토프로테인 유전자, 알파 1-안티트립신 유전자, β-글로빈 유전자, 기본 마이엘린 유전자, 마이오신 경쇄 2 유전자 및 고나도트로핀-방출 호르몬 유전자의 조절 서열 중에서 선택됨을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, E1 영역 전부 또는 일부의 결실 및 E4 영역 전부 또는 일부의 결실을 포함함을 특징으로 하는 아데노바이러스.
- 제 11 항에 있어서, E4 영역 전부 또는 일부의 결실을 부가적으로 포함함을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, 인간 아데노바이러스 타입 Ad 2 또는 Ad5 또는 개 아데노바이러스 타입 CAV-2임을 특징으로 하는 아데노바이러스.
- 제 1 항에 있어서, 퍼옥시다제 시스템에 수반된 폴리펩타이드를 암호화하는 하나 이상의 유전자를 부가적으로 포함함을 특징으로 하는 아데노바이러스.
- 삭제
- 제 1 항에 따른 하나 이상의 아데노바이러스를 포함하는, 종양 성장의 치료 또는 억제용 약학 조성물.
