Abstract

Description
본 발명은 약품 및 치료 약제를 환자에게 전달하기 위한 전기운반장치에 관한 것이다. 또한, 본 발명은 전기운반장치용의 속도 제어막에 관한 것이다.
전기운반장치 및 경피투여 패치 (transdermal patch) 는 건강보호산업의 직면한 많은 요구를 충족시킬수 있는 의약품 전달장치이다. 예를 들어, 전달속도를 제어하고 증가시키고, 약품과 약제의 품질저하를 감소시키며, 오염의 위험을 줄이기 위한 시도가 이루어져 왔다. 그러나, 전달될 약품과 치료제의 분자크기가 증가함에 따라, 그러한 목적은 더욱더 달성하기 어려워졌다. 그러한 약품과 약제로는 펩티드, 폴리펩티드, 및 프로틴이 있다.
전기운반장치는 일반적으로 적어도 두 개의 전극 및 하나의 전력원을 포함한다. 각 전극은 피부, 손톱, 점막 또는 다른 신체의 막표면과 전기 전달 및 물리적 접촉한다. 일반적으로 하나의 전극은 액티브 또는 도너 전극이라 하며, 다른 하나의 전극은 카운터 또는 리턴 전극이라 한다. 도너 전극은 또한 전달될 약품 또는 약제를 담고 있는 도너 저장기를 포함하며, 카운터 전극은 일반적으로 전해질을 담고 있는 카운터 저장기를 포함한다. 조합하여, 전극은 환자 신 체표면을 통해 전기적 회로를 폐쇄한다.
약품 또는 치료 약제는 인가된 전류에 의해 신체표면을 통해 전달되는 전하를 띤 분자 또는 이온이다. 약품 또는 약제의 전하는 도너 전극이 캐소드 인지 애노드 인지를 결정한다. 예를 들어, 약품이 양의 전하를 띨 때, 도너 전극은 애노드이며, 카운터 전극은 캐소드 이다. 반대로, 약품이 음의 전하를 띨 때, 도너 전극은 캐소드 이며, 카운터 전극은 애노드이다.

그러나, 수동성 약제/약품 유속을 더 정확하고 미세하게 제어하기 위하여 반투과성막이 필요할 수 있다. 기공의 크기가 작고 (직경이 약 0.09 미크론), 기공밀도가 낮은 (cm2 당 4 ×105 경로, 또는 체적당 0.003 %) 종래의 막들은 수동적인 유속을 막았지만, 적층 및 어셈블리 후에 충분한 흐름을 얻을 수 없는 단점이 있었다. 종래의 장치는 또한 장치 동작시 약품 저장기 또는 피부의 오일 및 기타 오염물질에 의해 오염 또는 손상될 수 있었다. 오염은 약품 전달속도에 영향을 주는 막의 폐색을 유발할 수 있다. 예를 들어, 막이 저항성을 잃으면, 약품은 신체표면으로 과다한 수준으로 녹아 들어갈 수 있다. 또한, 막이 과도한 저항을 가져, 약품전달을 저해하고 효능을 감소시키고 전원을 소모시킬수 있다. 그러한 약품전달 속도제어의 손실은 약품이 마취제 (opiod) 같은 진통제인 경우 매우 해롭다.
막의 기공은 또한 막이 신체표면과 접촉할 때 유해한 영향을 받을 수 있다. 피부상의 바디 오일, 더러운 미립자 및 다른 오염물질들은 반투과성막으로 전달되어 막의 기공을 막는다. 도너 및/또는 카운터 저장기의 소수성 (hydrophobic) 성분은 또한 막의 기공을 막아 폐색을 일으킬 수 있다. 그러한 반투과성막의 손상은 약품 전달속도의 제어, 정확성과 정밀도를 손상시킨다.
종래의 장치는 또한 막의 기공이 도너 및/또는 카운터 저장기로 막이 부착될 때 변경될 수 있기 때문에 불리하다. 반투과성막은 접착제로 부착되거나 및/또는 열압착으로 부착되므로 막의 기공의 변형 및 폐색을 일으킬 수 있다 그러한 손상은 약품/약제의 수동적인 및/또는 이온영동 (iontophoretic) 유속에 심각한 영향을 준다.
발명의 요약
본 발명은, 다른 이유 가운데서도, 반투과성막 어셈블리가 채용되기 때문에 종래의 기술에 대하여 장점을 갖는다. 본 발명의 전기운반장치 전용의 막 어셈 블리는 피부 오일 및 다른 오염물질 또는 저장기의 소수성 성분에 의해 실질적으로 영향을 받지 않는다. 본 발명은 또한 막을 저장기로 접착하는 방법에 있어서 접착제로 부착하고, 열압착 되는 것과 관련한 문제점이 없는 방법을 포함한다.
본 발명의 다른 장점으로는, 바람직한 실시예에 예시되는 바와 같이, 저 유공성막이 하나 이상의, 바람직하게는, 두개의 고 유공성 보호층에 의해 구조적으로 지지된다. 또한, 본 발명의 막 어셈블리는 종래의 전기운반장치에 비하여 빠른 정상상태의 약품전달을 얻을 수 있다. 예를 들어, 본 발명의 막 어셈블리는 일반적으로 동일한 두께와 구조적 내구성을 갖는 하나의 저 유공성층에 비하여 더 작은 유속저항을 갖는다. 따라서, 본 발명은 정상상태 약품전달/유속을 얻는데 필요한 시간에 실질적으로 영향을 주지 않으면서도 구조적 내구성을 증가시킨다.
본 발명의 또 다른 태양으로는 바디 오일, 접착제, 미립자의 오염물질 및/또는 소수성 성분에 의한 속도 제어막의 폐색효과를 줄인 방법이다. 그러한 폐색은 전기운반장치의 막의 수동적 유속에 나쁜 영향을 준다.
본 발명의 또 다른 태양은, 도너 저장기 및 신체 표면 사이에 위치하는 속도제어막 어셈블리를 포함하는 전기운반장치이다. 상기 어셈블리는 상기 저장기 부근의 저 유공성층 및 상기 저 유공성층과 신체 표면 사이에 위치하는 고 유공성층을 포함한다. 본 발명의 또 다른 태양은, 고 유공성층이 도너 저장기 부근에 위치하고, 상기 고 유공성층 및 신체 표면 사이에 저 유공성층이 위치한다.
본 발명의 또 다른 태양은, 저 유공성층이 고 유공성층 사이에 샌드위치 된, 제 2 고 유공성 보호층을 포함하는 전기운반장치이다. 저 유공성 속도제어층의 유공성을 실질적으로 변경시키지 않고, 장치 및/또는 저 유공성막으로 고 유공성 막이 적층에 의해 부착될 수 있다. 그렇게 함으로서, 막 어셈블리가 도너 저장기로 부착될 때 많은 량의 기공 폐색이 억제될 수 있다. 그 결과, 저 유공성층의 정확도 및 정밀도는 심각하게 손상되지 않는다. 고 유공성층은 저 유공성 층이 폐색 및 기타 손상을 일으키는 바디 오일 및 미세 오염물질로 노출되는 것으로부터 보호한다.

바람직하게는, 본 발명의 저 유공성의 속도제어막은 치료 약품 전달속도와 실질적으로 같은 약품 전달속도를 허용하도록 충분히 낮은 유공성을 갖는다. 더욱 바람직하게는, 유공성은 1 % 이하이며, 더더욱 바람직하게는, 0.1 % 이하이다.
본 발명의 또 다른 태양은 막 어셈블리 조립에 사용되는 저 유공성 및 고 유공성 형성 방법이다. 예를 들어, 예비적으로 형성된 저 유공성 기판상에 필름을 리소그라피로 디포지션 (depostion) 시킴으로서 막이 형성될 수 있다. 바람직하게는, 예비 형성된 해당 기판의 주 대향표면상에 리소그라피로 저 유공성 또는 고 유공성 막이 형성된다. 고 유공성막은 접착제를 사용하거나 열압착 밀봉을 적용하여 저 유공성막으로 부착될 수 있다. 바람직하게는, 실질적으로 고 유공성막의 주변부에만 접착 또는 압착된다. 바람직하게는, 저 유공성막이 고 유공 성층 사이에 들어가 저유공성층의 손상을 보호하게 된다.

본 발명의 바람직한 일실시예는 전기운반장치용의 속도-제어막 어셈블리에 관한 것이다. 전기운반장치에서 막 어셈블리는 약품 및/또는 약제 유속의 정확도 및 정밀도를 개선되도록 설계 및 조립된다. 유속의 정확도 및 정밀도는 상기 어셈블리가 고 유공성층 및 저 유공성층을 포함함으로서 개선된다. 일실시예에서, 고 유공성층이 도너 저장기와 신체 표면과 인접한 저 고유공성층 사이에 위치한다.
고 유공성층은, 조립 또는 동작전 충분히 높은 유공성을 가지며, 요구되는 기공 폐색에 따라, 고 유공성층은 저 유공성층보다 더 다공성을 갖는다. 상기 폐색은 접착제 또는 적층물로 인해 조립중 발생할 수도 있고, 조립 또는 동작중 오염물질에 노출되어 발생될 수도 있다. 그 결과, 약품/약제 전달속도는 전기운반장치의 조립 또는 동작중 고 유공성막에는 실질적으로 영향을 받지 않는다.
본 발명의 다른 바람직한 실시예에서, 속도-제어막 어셈블리는 저 유공성층 및 신체 표면 사이에 위치되는 고 유공성막을 포함한다. 상기 고 유공성층은 조립 또는 동작전에 충분히 높은 유공성을 갖지며, 장치의 조립 또는 동작중 기공 폐색 또는 손상을 입은 상태에서, 고 유공성층은 저 유공성층보다 더 유공성이 높다. 고 유공성층은 저 유공성층이 손상되는 것을 막는다. 예를 들어, 고 유공성층은 신체 표면의 오일 또는 미세 오염물에 의해 손상될 수 있다. 폐색이 및/또는 기공이 폐색 또는 차단될 수 있다. 상기 손상은 막이 부착되거나 조립될 때 발생할 수 있다.

본 발명의 바람직한 실시예에서, 도 1 에 도시된 바와 같이, 전기운반장치 (10) 은 도너 전극 (12) 및 카운터 전극 (14) 을 포함한다. 도너 및 카운터 전극 (12, 14) 은 도너 및 카운터 저장기 (16, 18) 에 각각 인접하게 위치한다. 도너 저장기 (16) 는 전달될 약품 또는 약제를 보관하며, 카운터 저장기 (18) 는 생체용 전해 염류를 보관한다. 반대로, 약품 및/또는 약제가 도너에서 카운터 전극으로 동시에 또는 교대로 전달될 수 있다. 그러한 경우, 속도 제어막 어셈블리는 카운터 전극 및 저장기와 연계하여 채용된다.

바람직하게는, 본 발명의 막 어셈블리는 유공성과 기공분포, 균일성, 크기, 형태, 및 치수의 측면에서 높은 정확도와 정밀도를 갖는 리소그라피 공정으로 만들어진다. 저 유공성막을 만드는 한가지 바람직한 방법은 저항성 또는 에너지 감지 물질로 저 유공성 기판을 코팅하는 것이다. 고 유공성 기판은 고 유공성막을 형성하는데 사용될 수 있다. 다른 방법으로, 레이져 드릴링으로 기공이 형성될 수 있다.
