Claims (18)
- 숙신산 생성능을 가지는 루멘 박테리아의 젖산 탈수소화효소(lactate dehydrogenase)를 코딩하는 유전자(ldhA)와 피루브산-개미산 분해효소(pyruvate-formate lyase)를 코딩하는 유전자(pfl)가 결실되어 있고, 혐기적 조건에서 다른 유기산은 거의 생성하지 않으면서 숙신산을 고농도로 생성하는 특성을 가지는 루멘 박테리아 변이균주.
- 제1항에 있어서, 상기 루멘 박테리아는 만헤이미아 속(Mannheimia sp.), 액티노바실러스 속(Actinobacillus sp.) 및 언에어로바이오스피리륨 속(Anaerobiospirillum sp.)으로 구성된 군에서 선택되는 것을 특징으로 하는 루멘 박테리아 변이균주.
- 제2항에 있어서, 상기 루멘 박테리아는 만헤이미아 속(Mannheimia sp.) 균주인 것을 특징으로 하는 루멘 박테리아 변이균주.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 상기 루멘 박테리아는 다른 유기산은 생성하지 않으면서 숙신산만을 생성하는 동형발효 균주인 것을 특징으로 하는 루멘 박테리아 변이균주.
- 만헤이미아 속(Mannheimia sp.), 액티노바실러스 속(Actinobacillus sp.) 및 언에어로바이오스피리륨 속(Anaerobiospirillum sp.)으로 구성된 군에서 선택되는 숙신산 생성능을 가지는 루멘 박테리아 균주로부터 젖산 탈수소화효소(lactate dehydrogenase)를 코딩하는 유전자(ldhA)와 피루브산-개미산 분해효소(pyruvate-formate lyase)를 코딩하는 유전자(pfl)를 결실시켜 수득되고, 혐기적 조건에서 다른 유기산은 거의 생성하지 않으면서 숙신산을 고농도로 생성하는 특성을 가지는 루멘 박테리아 변이균주의 제조방법.
- 제5항에 있어서, 상기 ldhA 유전자와 pfl 유전자의 결실은 상동성 재조합 (homologous recombination)에 의해 수행되는 것을 특징으로 하는 루멘 박테리아 변이균주의 제조방법.
- 제6항에 있어서, 상동성 재조합은 결실된 ldhA 유전자를 포함하는 유전자 교환벡터 및 결실된 pfl 유전자를 포함하는 유전자 교환벡터를 사용하여 수행되는 것을 특징으로 하는 루멘 박테리아 변이균주의 제조방법.
- 제7항에 있어서, 상기 결실된 ldhA 유전자를 포함하는 유전자 교환벡터는 pMLKO-sacB이고, 상기 결실된 pfl 유전자를 포함하는 유전자 교환벡터는 pMPKO-sacB인 것을 특징으로 하는 루멘 박테리아 변이균주의 제조방법.
- 도 1과 같은 개열지도를 가지며, 결실된 ldhA 유전자를 포함하는 유전자 교환벡터 pMLKO-sacB.
- 도 2와 같은 개열지도를 가지며, 결실된 pfl 유전자를 포함하는 유전자 교환벡터 pMPKO-sacB.
- 만헤이미아 속 55E(Mannheimia succiniciproducens 55E, KCTC 0769BP)의 ldhA 유전자와 pfl 유전자가 결실된 만헤이미아 속 LPK(Mannheimia sp. LPK, KCTC 10558BP).
- 제1항 내지 제3항 중 어느 한 항의 루멘 박테리아 변이균주를 혐기적 조건에서 배양하는 단계와 배양액으로부터 숙신산을 회수하는 단계를 포함하는 숙신산의 제조방법.
- 제12항에 있어서, 혐기적 조건은 이산화탄소 또는 질소를 이용하여 조성하는 것을 특징으로 하는 숙신산의 제조방법.
- 제12항에 있어서, 상기 배양은 다른 유기산은 생성되지 않으면서 숙신산만이 생성되는 동형발효인 것을 특징으로 하는 숙신산의 제조방법.
- 제12항에 있어서, 배양은 35~45℃의 온도와 6.0~7.5의 pH에서 이산화탄소 또는 이산화탄소를 포함하는 공기를 0.1~0.4vvm의 유속으로 공급하면서 수행하는 것을 특징으로 하는 숙신산의 제조방법.
- 제11항의 만헤이미아 속 LPK(Mannheimia sp. LPK, KCTC 10558BP) 균주를 혐기적 조건에서 배양하는 것을 특징으로 하는 숙신산의 제조방법.
- 제16항에 있어서, 혐기적 조건은 이산화탄소 또는 질소를 이용하여 조성하는 것을 특징으로 하는 숙신산의 제조방법.
- 제16항에 있어서, 상기 배양은 다른 유기산은 생성되지 않으면서 숙신산만이 생성되는 동형발효인 것을 특징으로 하는 숙신산의 제조방법.
