자가면역 질환은 자기조직으로부터 기원한 항원에 대한 자가항체 및 세포독성 T-세포를 형성함으로써 다양한 장애를 유발하는 면역 질환에 대한 일반적인 용어를 말한다. 자가면역 질환에는 전신적 홍반성 낭창, 만성 류마치스 관절염, 전신성 경화증, 피부근염/다발성 근염, 쇼그렌 증후군(Sjogren's syndrome) 등과 같은 비-기관 특이적인 질환; 당뇨병, 하시모도 갑상선종, 그레이비스병(Graves' diease), 다발성 경화증, 특발성 아디슨 빈혈증, 중증 근무력증, 악성 빈혈 등과 같은 기관 특이적인 질환; 및 1차 담즙 경변, 만성 활성 간염 등과 같은 복합 질환이 포함될 수 있으며, 잠재적인 환자를 포함하여 이와 같은 질환으로 고생하는 환자의 총수는 매우 많다.
이와 같은 자가면역 질환을 치료하기 위해 지금까지 실질적으로 적용한 치료 요법은 면역억제 활성이 기대되는 코르티코스테로이드, 사이클로스포린 A, 사이클로포스파미드, 아자티오푸린, 메토트렉세이트와 같은 치료제의 투여; 또는 X-선 조사, 흉선절제술 등; 또는 면역조절 활성이 기대되는 인터페론, γ-글로불린, 임파구 세포 표면 마커에 대한 항체, 골드 제제(gold preparation), D-페니실라민 등의 투여를 포함한다.
다수의 상기 면역억제제는 면역경쟁성 세포를 포함하는 다양한 세포내 DNA 또는 RNA에 손상을 입힐 수 있으므로, 이들을 잘못 사용할 경우 심각한 부작용이 야기될 수 있다. 더욱이, 임산부에 대한 투여는 기형아의 출산을 야기시킬 수 있으며 또한 면역억제가 광범위하게 유도되기 때문에 기회적 감염의 위험성이 증가될 수 있다는 것이 지적되어 왔다. 한편, 면역 조절 인자에 있어서 작용의 상세한 메카니즘은 현재까지 알려져 있지 않으므로 치료학적 효과를 정확히 예견하기는 어렵다. 또한 광범위한 세포 그룹이 영향을 받음으로 인해, 부작용의 위험성이 높다는 것도 지적되어 왔다.
상술한 바와 같이, 자가면역 질환에 대한 현재의 치료에 있어서 만족스럽지 않은 많은 점들이 발견되었다. 또한 자가면역 질환의 개시 메카니즘에 있어서 해결하지 못한 많은 점들이 발견되었다. 단지 자가면역 질환을 유도하는 면역 시스템에 있어서, 비정상적 부위에 특이적으로 작용할 수 있는 치료제의 개발을 억제하는 것이 강력하게 지지되어 왔다.
발명의 기술
면역 질환에 있어서 특이적인 효과를 발휘할 수 있는 치료제가 개발될 수 있다면, 타겟화될 면역 시스템내 비정상 부위에 따라서, 부작용이 없는 완성된 치료요법을 확립할 수 있을 것으로 크게 기대된다. 즉, 특이적인 치료요법을 확립할 목적에서, 본 발명자들은 면역 시스템의 기형적인 가속화를 국소적으로 방지할 수 있는 T-세포 기원의 억제 인자(이후 TsF로 약술)로서 가용성 T-세포 수용체(이후 TCR로 약술) α-쇄를 제조함으로써 자가면역 질환의 치료 방법을 확립하기 위한 시도가 있어 왔다.
본원에서 사용된 억제 인자 T-세포(이후 Ts로 약술)는 면역 억제를 유도하는 초기 단계에서 항원-특이적인 작용을 나타내고 억제 활성을 발휘하는 최종 단계에서 항원 비-특이적인 작용을 나타내는, 면역억제 활성을 나타낼 수 있는 일련의 세포 그룹을 의미한다. 즉, 억제 인자 T-세포는 면역 반응을 항원 특이적으로 증진시킬 수 있는 헬퍼 T-세포(이후 Th로 약술)의 작용에 대해 역작용을 지니고 있는 T-세포의 아그룹이다.
자가 면역 질환이 있는 사람 및 기타 동물에 있어서, Ts의 몇가지 정량적 또는 정성적 비정상성이 보고되었다[참조: McIntosh and Crachan, Science, 232, 401(1986); Isenberg et al., Br. Med. Bull., 10, 262(1984)]. Ts 기능의 비정상성이 유도된 경우에, TsF의 생성은 감소되는 반면, Th 활성은 상대적으로 증가될 수 있다. 따라서, 세포독성 T-세포-유도성 T-세포에 이은 자가-독성 T-세포가 유도되고 활성화될 수 있을 것으로 여겨진다. 자가항체-생성 B-세포도 유사하게 활성화될 수 있다. 따라서, 조직 세포 또는 생물학적 활성 인자가 자가면역 질환에 있어서 면역 시스템에 의해 공격당하는 경우, 이와 같은 면역 질환의 비정상성에 기여하는 부위는 타겟화된 항원에 대해 특이적인 Ts에 의해서 생성된 TsF를 상기 자가면역 질환으로 고생하는 환자에게 투여함으로써 복구될 수 있는 것으로 기대되며, 이는 질환의 완전한 회복을 가져올 수 있다.
KLH[키홀 림페트 헤모시아닌(Keyhole limpet Hemocyanin)]에 의해 항원으로서 면역화된 마우스로부터 기원할 수 있는, 본 발명의 KLH-특이적인 Ts는 다니구치(Taniguch) 등에 의해 Ts 하이브리도마로서 분리되었다[참조: Taniguchi et al., Nature, 278, 555(1979)]. Ts 하이브리도마는 표준으로서 KLH-특이적인 항체의 방지를 사용하여 선별한다. 다니구치 등은 Ts 하이브리 도마에 의한 가용성 TsF의 생산을 확립하였다. TsF는 KLH로 피복된 면역흡착제에 의해 흡착되며 시험관내에서 KLH에 대한 항체 생성을 특이적으로 방지할 수 있다. 이마이(Imai) 등은 문헌[참조: Proc. Natl. Nacad. Sci. USA, 83, 8708(1986)]에서 Ts 하이브리도마 세포의 유전자 분석의 결과로서, KLH-특이적인 Ts에 의해 발현된 항원에 대한 TCR은 TCR α-쇄이며, TCR α-쇄는 억제 작용의 진행에 중요한 역할을 한다고 제안하였다.
더우기 고세키(Koseki) 등은 Ts 하이브리도마로부터 α-쇄 수용체에 대한 cDNA 및 게놈성 DNA를 클로닝함으로써 염기 서열을 측정하였다[참조: Int. Immunol., 1, 41(1989)]. 그 결과, 상기 발명자들은 당해 분야에 이미 공지된 어떠한 Vα군(family)과는 상이한, α-쇄의 가변영역(이후 Vα로 약술)에 대한 전체적으로 새로운 유전자군을 발견하고 이를 "Vα14"로 명명하였다. Vα14의 90% 이상은α-쇄의 결합 영역(이후 Jα로 약술)의 한 부류인 Jα281과의 유전자 재배열에 의해 결합한다.
최근 연구 결과는 억제 인자 T-세포로부터 기원한 TsF의 특징을 설명해 준다 [참조: Batchelor et al, Immunol. Today, 10, 37(1989)]. 더욱 상세하게는, 항원에 결합하는 TsF의 작용은 TCR α-쇄에 의한 것이며, 이것은 이후 Ts로부터 유리되어 TsF를 형성한다. 따라서, 가용성-α쇄는 면역억제를 특이적으로 유도하는 TsF로서 자가면역 질환의 치료요법에 적용될 것이다. Ts의 TCR α-쇄의 구조는 Th의 α쇄 및 세포독성 T-세포의 α쇄와는 상이한 것으로 간주될 수 있으므로[참조: Imai et al., Proc. Natle. Acad. Sci. USA, 83, 8708(1986)], 본 발명의 목적은 심지어 Th 또는 세포독성 T-세포질로부터 기원한 α-쇄를 사용하여도 달성될 수 없는 것으로 여겨진다. 따라서, 치료제를 개발하기 위한 가장 가능성 있고 타당한 시도는 Ts에 의해 생성된 α-쇄에 한정된다.
그러나, 항원 특이적인 TsF는 상기 외부 항원에 대한 항체 생성의 억제에는 적용시킬 수 있으나, 자가면역 반응의 억제, 즉 자가면역 질환의 치료요법에는 전혀 적용시킬 수 없는 것으로 예상된다.
한편, 페어칠드(Fairchild) 등은 디니트로페놀(이후 DNP로 약술) 항원 특이적인 CD(분화 집단: cluster of differentiation) 8개의 양성 Ts[참조: Fairchild et al., J. Immunol., 141, 3342(1988); Fairchild et al., ibid, 145, 2001(1990)]가 α-쇄 및 β-쇄로 이루어진 가용성 이량체 TsF를 생성시킴을 밝혀내었다. 또한, 리더(Lider)등은 실험적 알레르기성 뇌척수염(이후 EAE로 약술)의 뇌염발생성 T-세포내에서 매우 흔하게 발견되는 α-쇄 및 β-쇄로 이루어진 이량체를 TCR로 분석하고[참조: Lider et al., Science, 239, 181(1988)], 보조제 존재하에서 Vβ8 영역[참조: Vandenbark et al. Nature, 341 541(1989)] 또는 β-쇄 VDJ 영역 및 J 영역[참조: Howell et al., Science, 246, 668(1989)]의 합성 펩타이드를 사용하여 백신화하는데 성공하였다. 그러나 동일 조건하에서 수행한 확인 시험은 흔히 역결과를 나타내었다[참조: Desquenne-Clark et al., Proc. Natl. Acad. Sci. USA, 88, 7219(1991)]. 즉, 데스퀸-클락(Desquenne-Clark)등의 실험에서 뇌염발생성 T-세포로부터 기원한 상술한 합성 펩타이드 쇄를 사용한 랫트에 대한 백신화는 EAE를 억제할 수 없을 뿐 아니라, 오히려 EAE 증후는 더욱 악화됨이 나타났다.
더욱이, TCR β-쇄는 주요 조직적합성 복합체(이후 MHC로서 약술)의 제한에 관여하므로 β-쇄 뿐 아닌 α-쇄 및 β-쇄의 이량체도 치료제로서 사용하기에 적절한 것으로 고려될 수 없다. 따라서 개개의 사람은 상이한 MHC를 지니고 있으므로, 개개 환자에 대한 적합한 치료제를 제조하는 것이 불가능한 것은 아닐지라도 실용적이지 못하다. 본 발명의 TCR α-쇄는 항원만을 인지하므로, 환자에게 α-쇄를 투여함으로써 MHC의 어떠한 제한없이 치료 효과를 기대할 수 있다. 따라서, 본 발명에 따라 제조한 치료제는, 개개의 MHC에 있어서의, 어떠한 차이를 고려하지 않고서도, 특성의 자가-면역 질환으로 고생하는 환자 모두에게 적용시킬 수 있다.
상기 고려사항을 요약하면, Ts로부터 생성되어 방출된 TsF로서의 TCR α-쇄는 면역억제 활성을 발휘하므로 자가면역 질환에 대한 치료제로서의 유효 활성을 나타낼 것으로 기대된다. 항원-특이적인 면역억제 활성에 이어 최종적으로 TGF(형질전환 성장 인자)-β또는 IL(인터루킨)-10 등과 같은 사이토킨을 통해 병변 부위에서 항원 비-특이적인 면역 억제가 일어나는 것으로 여겨진다. 본 발명에서 사용된 개념은 모든 자가면역 질환에 대한 치료제 개발에 적용될 수 있다.
이제, 상기 개념의 효능성을 실험을 통해 확증하기 위하여, 본 발명자들은 인슐린-의존성 당뇨병(이후 IDDM으로 약술)의 치료 효과를 연구하였다. 본 발명자들은 TCR의 작용부위인, Vα14Jα281의 가용성 형태를 재조합 단백질 방법으로써 생성한 후, 이를 보조제 부재하에 자연발생성 IDDM을 앓고 있는 NOD(비비만성 당뇨병) 마우스에 투여하였다. 더욱 상세하게는, 마우스를 외인성 항원 KLH로 면역화하고, KLH 특이적인 Ts 하이브리도마를 시험관내에서 확립하여 TCR을 암호화하는 cDNA를 Ts 하이브리도마로부터 클로닝함으로써 본 발명에서 사용된 Vα14Jα281의 cDNA를 수득하였다. 이후, Vα14Jα281 cDNA를 마우스 Cγ3를 함유하는 발현 플라스미드내로 삽입하여 키메라성 단백질로서의 골수종 세포내에서 발현시켰다. 이 키메라성 형성에 의해서, Vα14Jα281은 구조적으로 안정하게되며 형질전환된 골수종 세포 배양물의 상층액으로부터 가용성 단백질로서 수득될 수 있다. 마우스로부터 기원한 것으로서의 Cγ3은 마우스 균주를 사용한 실험시 합텐에 대한 담체로서 작용하지 않으므로, 키메라성 단백질의 Cγ3에 대한 면역 반응은 유도되지 않는다.
Vα14에 대한 모노클로날 항체를 사용하는 간접적인 동물 시험에서는 TCR Vα14Jα281이 또한 생체내 시스템내의 항원-비특이적인 면역억제 반응에 관여할 수 있음을 나타내는 결과가 수득되었다. 또한, Vα14Jα281을 암호화하는 mRNA의 발현이 당뇨병의 발병 시기가 가까워 지고 있는 NOD 마우스내에서 억제된다는 사실은항원-비특이적인 면역 억제에 Vα14Jα281이 관여함을 강력하게 암시하고 있다. 상술한 바와 같이, IDDM 상에서 가용성 Vα14Jα281의 치료 효과 및 현저한 억제 효과가 IDDM 모델 하우스를 사용하는 동물 시험 결과로서 확증됨이 예상된다.
Vα14Jα281은 본래 KLH-특이적인 Ts의 TCR 작용부위이다. 특히 췌장내에 존재하는 다양한 자가 항원과 KLH의 상동성이 매우 높을 가능성이 있고 심지어 면역억제 시스템이 상술한 바와 같이 항원-특이적으로 유도될 수 있다해도, 자가 반응성 효과인자 세포에 대한 항원-비특이적인 억제의 최종 단계-생성을 유도시켜 IDDM내에서 이와 같은 항원-비특이적인 면역 억제 치료 효과를 설명할 수 있다.
어떤 경우이든지, Vα14Jα281의 상술한 항원-비특이적인 면역억제 효과에 따라서, 가용성 TCR은 IDDM만이 아니라 다른 모든 자가면역 질환에도 광범위한 치료 효과를 발휘할 것으로 기대할 수 있다.
TCR Vα14유전자는 야생형 마우스 균주내에서 잘 유지되어 왔다[참조: Koseki et al., Proc., Natl., Acd. Sci. USA, 87, 5248(1990)]. 마우스의 TCR Vα14의 아미노산 서열 및 사람의 상응 TCR Vα군의 아미노산 서열 사이의 상동성은 76%로 높으나, 공지된 바와 같이 Vα군 약 100중 다른 구성원의 상동성은 70% 이하이다. 이러한 높은 상동성에 비추어 볼 때, 상기 사람 TCR Vα군은 또한 유사한 면역억제 작용을 달성하는 것으로 여겨진다. 더욱이, 다수의 Vα군 이외에, 면역 억제의 특이적인 작용을 지닌 마우스 Vα14는 다양한 사람 자가면역 질환에 대한 치료제로서 유용한 것으로 여겨진다.
본 발명의 Vα14J281수용체에 의한 면역억제에 있어서의 작용 및 NOD 마우스를 사용한 IDDM 상에서의 치료효과는 하기 실시예의 방법으로써 상세히 기술될 것이다.
실시예 1
자가면역 질환 마우스에 있어서 TCR Vα14Jα281 mRNA 발현의 제한
NOD 마우스의 비장 임파구내 Vα14Jα281을 암호화하는 mRNA 발현은 RNase 보호 검정[참조: Koseki et al., Proc. Natl. Acad. Sci, USA, 87, 5248(1990)]을 사용하여 정량적으로 검정한다. 이러한 목적을 위해서, TCR α-쇄중 α-쇄의 불변 영역(이후 Cα로 약술)을 지닌 Vα14Jα281과 커플링(coupling)된 Vα14Jα281Cα를 암호화하는 cDNA를 상업적으로 시판되는 pCEM 플라스미드(Promega Biotec 제조원)내에 통합시킴으로써 Sp6 프로모터를 사용하여 방사성 동위원소로 표지시킨 Vα14Jα281Cα리보프로브를 합성한다. 임파구로부터 추출한 RNA와 프로브를 하이브리드화한다. 하이브리드화되지 않는 부위를 일본쇄-특이적인 RNase로 분해한다. 이 생성물을 아가로즈 겔 전기영동으로 분획화한다. 단지 Cα로 하이브리드화될 밴드의 농도를 100%로 정의하고 상응하는 Vα14Jα281 밴드의 농도를 비교하여 % 측면에서 계산함으로써 Vα14Jα281을 암호화하는 mRNA의 발현 비율을 측정한다. 또한 대조 그룹에서는, Jα281으 제외한 Vα14를 지닌 J영역으로 이루어진 Vα14JxCα 그룹을 암호화하는 mRNA의 발현 빈도 및 Vα14외를 제외한 Jα281을 지닌 V 영역으로 이루어진 VxJα281Cα를 암호화하는 mRNA의 발현 빈도 각각을 측정한다.
NOD 마우스내 Vα14Jα281 mRNA의 발현 빈도는 발병전에 2% 이상이었으나, 발병 이후 시간 경과에 따라 감소됨이 밝혀졌다. 한편, 시간 경과에 따른 변화는대조 그룹에서는 관측되지 않는다(참조. 제1도)
실시예 2
가용성 TCR VαJα281의 제조
마우스 IgG의 불변 영역 Cγ3를 함유하는 pSVEP 플라스미드를 우선 작제하여[참조: Kameyama et al., FEBS Letters, 244, 301(1989)], 여기에 SalI 부위에서 Vα14Jα281 cDNA를 삽입한다(참조. 제2도). 벡터(30㎍)는 전기영동에 의해서 J558L 마우스 골수종 세포(2 ×106)내로 도입한다. 도입된 세표를 마이코페놀산 20㎎/㎖를 함유하는 20% FCS-RPMI 1640 배지중에서 선별하고 상층액내에서 ELISA로써 Vα14Jα281Cγ3 키메라성 단백질의 분비를 확인한다. 상층액 내에 분비된 가용성의 Vα14Jα281Cγ3 키메라성 단백질을 단백질을 단백질 G를 사용하는 친화 크로마토그래피로써 배양 상층액으로부터 정제한다.
실시예 3
항-Vα14 모노클로날 항체의 항체 생성에 대한 억제 효과
마우스를 Vα14Jα281로 면역화시키고 Vα14에 대한 모노클로날 항체를 통상적인 방법에 따라 제조한다.
Vα14Jα281 모노클로날 항체(100㎍)를 DNP-KLH로 면역화하기 2주전부터 시작하여 매주 마우스에게 복강내 투여한다. 이후, DNP-KLH(100㎍), 백일해 백신(이후 PV로 약술; Chiba Serum Institute 제조원) 및 수산화알루미늄(Wako Pure Chemical Ind., Ltd. 제조원)의 혼합물을 제조하여 제3도 및 제4도에 나타낸 바와같이 0주에 마우스를 면역화시킨다. 마우스내 항-Vα14Jα281 모노클로날 항체를 투여하는 것을 DNP-KLH로 면역화한지 3주후에 계속 유지시킨다. 항-KLH 항체 및 항-DNP 항체의 양을 ELISA로써 측정한다.
제3도 및 제4도에서 명백해지는 바와 같이, KLH 및 DNP에 대한 항체는 항-Vα14 모노클로날 항체를 투여하지 않은 대조군 마우스 그룹에서 잘 생성된다. Vα14에 대한 모노클로날 항체를 투여한 마우스에서는, 항-KLH 항체 및 항-DNP 항체 모두의 생성이 억제된다. 이 결과는 항-Vα14 모노클로날 항체가 Ts항원-비-특이적으로 자극하여 이의 면역-억제 활성을 증진시킴을 나타낸다.
실시예 4
IDDM 동물 모델에 있어서의 치료 효과
IDDM 동물 모델은 벤델락(Bendelac) 등의 방법[참조: Bendelac et al., J. Exp. Med., 166, 823(1987); Yasunami and Bach, Eur. J. Immunol., 18, 481.(1988); Pankewycz et al., 21, 873(1991)]에 따라 제조한다. 즉, 12주된 NOD 마우스에 사이클로포스파미드 4㎎의 용량을 투여한다. 2 내지 3주 후, 마우스의 50%가 시험에서 양성 당뇨를 나타낸다. 3 ×107의 임파구를 마우스의 비장으로부터 분리하고 800㎭로 조사한 5주된 NOD 마우스에 정맥내 주사한다. 임파구-이식된 마우스는 2 내지 3주후 고비율의 양성 당뇨를 나타냄이 밝혀졌다.
Vα14Jα281Cγ3 키메라성 단백질을 투여하기 위해서, 8마리의 암컷 마우스를 준비한다. 이 마우스를 한 그룹의 구성이 4마리인 2개의 그룹으로 나누고, 음성대조군에는 아무것도 투여하지 않고, 4마리의 다른 그룹에는 Vα14Jα281Cγ3 키메라성 단백질을 투여한다. Vα14Jα281Cγ3 키메라성 단백질(100㎍)과 알루미늄 겔을 혼합하여 임파구가 이식된 NOD 마우스에 복강내 주사함으로써 투여가 수행된다. 이후, Vα14Jα281Cγ3 키메라성 단백질만을 1주에 2회 50㎍의 용량으로 복강내 투여하고, 이 투여 유형을 3과 ½주간(총 7회) 지속한다. 뇨당 시험지법(urioglucose test paper method)에 따라서 뇨중 글루코즈 양을 측정하여 IDDM의 발병을 평가한다. 이 결과를 표 1에 나타낸다.
Vα14Jα281Cγ3 키메라성 단백질 및 알루미늄 겔의 혼합물을 투여하는 실험에 있어서, Vα14Jα281Cγ3 키메라성 단백질을 투여하지 않은 4마리중 3마리의 마우스에게 임파구 이식 2주후에 발병했으며 나머지 1마리의 마우스는 2와 ½주 후에 발병했다. 이와는 대조적으로, 임파구를 이식시키고 Vα14Jα281Cγ3 키메라성 단백질을 투여한 4마리의 마우스는 4주가 지난 후에도 발병하지 않았다. Vα14Jα281Cγ3 키메라성 단백질을 불연속 투여한지 1주 후, 즉, 임파구를 이식한지 4와 ½주 후, 후자 그룹중 1마리의 마우스에서 발병했다. 7주까지 관측했으나, 이 마우스들에게서 추가의 변화는 관측되지 않았다.
더욱이, 알루미늄 겔의 영향을 제거하기 위하여, 알루미늄 겔의 부재하에서, Vα14Jα281Cγ3 키메라성 단백질만을 NOD 마우스에 투여한 경우의 실험을 수행하였다. 더욱 상세하게는, Vα14Jα281Cγ3 키메라성 단백질 50㎍을 3마리로 구성된 한 그룹의 마우스에 투여하고, 3마리로 구성된 다른 그룹에는 포스페이트 완충된 염수(PBS)를 투여하였다. 임파구 이식이 수행된 날 키메라성 단백질 또는 PBS를 1회 투여한다.
알루미늄 겔과 혼합되지 않은 Vα14Jα281Cγ3 키메라성 단백질을 마우스에게 투여한 실험 결과는 표 2에 나타낸다.
뇨 글루코즈 농도를 측정하여 평가하며 "+"는 뇨 글루코즈 농도가 200㎎/dl 이상임을 나타내고, "-"는 200㎎/dl 이하임을 나타내며, "+"는 대략 상기 2개 값의 중간값임을 나타낸다. Vα14Jα281Cγ3 키메라성 단백질을 투여한 3마리의 마우스에서는 12주의 관측기간 동안 발병이 나타나지 않았다. 그러나, PBS를 투여한 3마리의 대조군 마우스에서는, 1마리의 마우스가 2주후 전기간에 걸쳐서 발병을 나타내었고 2마리의 마우스는 상기 관측기간 동안 일시적인 발병을 나타내었으며; 궁극적으로 모든 PBS 투여한 그룹에서는 약간 중증의 발병이 관측되었다.
상술한 바와 같이, KLH-특이적인 Ts 하이브리도마로부터 기원한 가용성 Vα14Jα281은 항원-비특이적인 방식으로 TsF로써의 당뇨병 발병으로부터 NOD 마우스를 방지함이 밝혀졌다.

Claims (3)
- 예방학적 또는 치료학적 유효량의 Vα14Jα281를 함유하는 가용성 T-세포 수용체 α-쇄 및 약제학적 담체를 포함하는, 자가면역 유도된 인슐린 의존성 당뇨병의 예방 및 치료용 조성물.
- Vα14Jα281를 함유하는 가용성 T-세포 수용체 α-쇄를 약제학적 담체로서의 면역글로불린의 불변 영역과 배합함으로써 형성된 예방학적 또는 치료학적 유효량의 키메라성 단백질을 포함하는, 자가면역 유도된 인슐린 의존성 당뇨병의 예방 및 치료용 조성물.
- 제2항에 있어서, 키메라성 단백질이 Vα14Jα281Cγ3인, 자가면역 유도된 인슐린 의존형 당뇨병의 예방 및 치료용 조성물.
