Abstract
본 발명은 계피로부터 분리한 시남 알데하이드 및 그 유도체의 면역기능 강화제로서의 용도에 관한 것으로, 계피로부터 분리정제한 2-하이드록시 시남 알데하이드 및 그 유도체인 2-O-벤조일 시남 알데하이드는 T 세포를 보조 T 세포로 분화 촉진하고 분화된 보조 T 세포는 감마인터페론 생성을 유도하여 면역기능을 강화하며 급성독성이 없는 뛰어난 효과가 있다.
Description
본 발명은 계피로부터 분리정제한 시남 알데하이드 유도체의 신규한 면역기능 강화제로서의 용도에 관한 것으로, 보다 상세하게는 T 세포 분화 촉진과 감마인터페론 생성함으로써 면역기능을 증강시키는 계피의 주요성분중 하나인 시남알데히드의 유도체, 2-하이드록시 시남 알데하이드 또는 2-0-벤조일 시남 알데하이드을 유효성분으로 함유하는 면역증강용 조성물에 관한 것이다.
면역기능이란 동물 체내에 존재하는 자기 반응체계로서, 외부로부터 침입해 오는 생명체와 그들의 독성 생성물을 자기 자신과 구별해 내어 이 침입자를 제거하는 복잡한 생물학적 기능이다. 이러한 면역계는 외부로부터 들어온 세포, 바이러스 또는 고분자물질을 찾아내고 감염인자들을 중화시키며 그들을 파괴시켜 체내에서 몰아내는 것까지 여러 가지 기능을 포함한다. 실제로 인체는 수없이 많은 독성물질이나 미생물 등에 노출되어 있는 싱황에서 스스로를 보호하는 것이 개체의 생명을 유지해 나감에 있어서 가장 중요한 일이라 할 수 있다. 이러한 사실은 AIDS 등 각종 면역결핍증의 경우와 같이 일단 면역체계가 파괴되면 그 면역체계를 보완해 주지 않는 이상 생존하기가 어려워진다는 것을 생각해 보면 분명한 사실이 된다. 더구나 암세포와 같이 개체 내에서 생기는 이상현상을 제거하는 기능도 면역체계가 제공하고 있다. 이러한 자기방어를 위한 감시기능을 크게 두 가지 기전에 의해 이루어지는데 하나는 체액성 면역이고 다른 하나는 세포성 면역이다. 체액성 면역은 혈청 내에 존재하는 항체에 의해 일어나게 되는데 그 항체가 외부물질과 결합함으로써 그 물질을 제거한다. 항체는 B 세포가 분화된 형질세포 (plasma cel1)에서 생산되며 각 세포는 세포표면에 항원수용체로 존재하는 유일한 종류의 항체만을 만들도록 프로그램되어 있다. 따라서 침입해 온 세포나 조직을 직접 파괴하는 기능을 가지고 있으며 각종 기생충, 조직, 세포내 감염, 암세포 등을 제거하는데 그 기능을 나타낸다. 항원은 그 항원에 대응하는 표면항체를 가진 B 세포와 결합하여 그 세포를 활성화함으로써 클론 증식을 일으키고 최종적으로 항체를 생산하는 형질세포 및 면역기억세포(memory cel1)로 성숙한다. 이러한 체액성면역은 주로 세포밖에 있는 박테리아나 바이러스 그리고 단백질이나 복합탄수화물과 같은 외부물질에 대해 효과적이다. 반면 세포성 면역반응에 관여하는 T 세포는 B 세포와 마찬가지로 각기 고유한 항원 수용체 (T cell antigen receptor)를 가지고 있으며 이를 통해 MHC 분자와 복합체를 이룬 세포 표면의 항원을 인식하여 활성세포(effector cel1)와 특이 획득면역을 제공하는 기억세포로 되는 클론으로 각각 증식을 일으킨다. T 세포는 그 기능에 따라 분류할 수 있는데 그 중 보조 T 세포(Th : helper T cel1)는 대식세포의 표면에 있는 MHC class II 분자와 동반한 항원을 인식하며 사이토카인을 분비하여 대식세포를 활성화하고 세포내 미생물을 살해할 뿐 아니라 B 세포가 항체를 생산하도록 분화유도작용도 한다. 세포독성 T 세포(CTL : cytotoxic T lymphocyte)는 바이러스 감염세포 표면의 MHC class I 분자와 동반한 항원을 인식하여 그 세포내 바이러스가 증식하기 전에 감염세포를 살해하는데 이와같이 침입해 온 세포나 조직을 직접 파괴하며 각종 기생충, 조직, 세포내 감염, 암세포 등을 제거하는 CTL을 성숙 및 증식시킨다·
면역기능은 주로 B 세포, T 세포의 두 종류 림프세포에 의해 수행되는데 B 세포는 항체를 생산하고, T 세포는 세포성 면역에 직접적인 관련이 있다. T 세포는 혈액과 림프선, 비장에 주로 분포되어 있으며, 전체적으로 볼 때 B 세포보다 3-4배 많이 존재한다. 사람의 경우, 혈액에 비교적 많은 T 세포가 존재하여 전체 혈액세포의 약 15%, 혈액중 림프세포의 70-80%를 차지한다. T 세포는 골수에서 생성되어 흉선 (thymus)으로 이동하며 흉선 속에서 복잡한 분화과정을 거친다. 이 분화과정에서는 분화단계에 따라 세포의 크기도 다소 변하며 특히 세포 표면 항원의 변화가 특이적으로 일어나 세포 표면 항원을 추적함으로써 분화과정을 연구할 수 있다. 흉선 속에서 성숙된 T 세포는 혈액, 림파선, 비장 등으로 이동하여 기능을 발휘하는데 외부로부터 자극이 오면 급격히 활성화 되어 세포분열을 일으키며 활성화 과정에서 세포 표면 항원이 변화한다. 뿐만 아니라 B 세포나 다른 T 세포 혹은 대식세포와 같은 다른 면역세포에 자극을 줄 수 있는 여러 가지 인자들을 분비하기도 한다. 이러한 인자들을 생체반응 조절물질(Bio1ogical Response Modifier ; BRM)이라고 할 수 있는데 이들은 인체의 생리반응에 관여하는 물질들로 생체반응을 조절하던가 아니면 직접적으로 종양세포 등에 작용하여 이들 세포들의 성장정지, 용해, 또는 분화성숙 효과를 초래함으로써 임상에 사용할 수 있는 물질들이다. 암을 비롯한 여러 면역결핍성 질환들의 치료제로 널리 알려져 있는 BRM으로는 BCG, 베스타틴 (Bestatin), OK432와 같은 면역조걸 내지 증강제들과 인터페론 등을 포함한 사이토카인들, 인터페론 유도물질들(Tilorones), T 세포에 대한 단일클론 항체 등을 들 수 있다.
본 발명자들은 계피로부터 분리한 시남 알데하이드 유도체들이 T세포 분화를 촉진하고 감마인터페론 생성을 유도한다는 사실을 알아냈다.
따라서 본 발명의 목적은 계피로부터 시남 알데하이드 유도체들을 분리정제하여 신규한 면역기능 강화제로 제공함에 있다. 본 발명의 다른 목적은 계피로부터 분리정제한 시남 알데하이드로부터 이종의 시남 알데하이드 유도체를 합성하여 신규한 면역기능 강화제로 제공함에 있다. 본 발명의 또다른 목적은 시남 알데하이드 유도체를 유효성분으로 함유하는 면역기능 강화약품 또는 식품을 제공함에 있다.
본 발명의 상기 목적은 계피로부터 시남 알데하이드 유도체를 분리 및 치환 합성하여 이화학적 성질을 조사한 후 이들 유도체의 T세포 분화 효과와 생체활성물질의 하나인 감마인터페론의 생성 유도 효과를 조사하고 급성 독성 유무를 조사하므로써 달성하였다.
이하 본 발명의 구성과 작용을 상세히 설명하고자 한다.
본 발명은 계피로부터 시남 알데하이드 유도체인 2-하이드록시 시남 알데하이드를 분리 및 정제하는 단계; 상기 분리정제한 2-하이드록시 시남 알데하이드로부터 2-0-벤조일 시남 알데하이드를 합성하고 상기 2-하이드록시 시남 알데하이드와 이화학적 성질을 비교하는 단계; 상기 계피로부터 분리정제한 2-하이드록시 시남 알데하이드와 이로부터 합성한 2-0-벤조일 시남 알데하이드를 생쥐의 비장세포에 가한 후 T 세포의 분화 정도를 플로우사이토미터로 측정하여 시남 알데하이드 유도체의 T 세포 분화 효과를 조사하는 단계; 생쥐의 비장세포에 2종의 시남 알데하이드 유도체인 2-하이드록시 시남 알데하이드와 2-0-벤조일 시남 알데하이드를 처리한 후 엘리사 정량 킷트로 감마인터페론의 양을 측정하여 감마인터페론의 생성유도 효과를 조사하는 단계 및; 역시 상기와 동일한 2종의 시남 알데하이드 유도체를 생쥐에 주사하고 이상증상을 관찰하여 급성독성 유무를 조사하는 단계로 구성된다.
이하 본 발명의 상세한 설명을 실시예와 실험예를 통하여 설명하고자 하지만 본 발명의 권리범위를 이들 실시예와 실험예에만 한정하는 것은 아니다.
[실시예 1]
계피로부터 시탐알데하이드 유도체 분리정제 및 합성
[실험예 1]
계피로부터 2-하이드록시 시남 알데하이드 분리 및 정제
본 발명에 사용한 계피(Cinnamomum cassia Blume)는 대한민국 충청남도 금산에서 구입하여 사용하였다. 20Og의 계피를 잘게 분쇄하여 2L의 클로로포름과 아세톤을 넣어 상온에서 24시간 방치한후 교반하고 여과지를 이용하여 액상과 고체부분을 분리하였다. 액상을 모아 감압하에서 농축한 후 1L의 에칠아세테이트와 1L의 2% 수산화 나트륨용액을 가하여 물층과 유기용매층으로 분리하고, 물층을 1L의 에칠아세테이트로 3번 추출하여 유기용매에 녹는 부분은 제거하고 물층은 10% 초산용액으로 산성 용액(pH 5)을 만든후 1L의 메칠렌클로라이드로 3번 추출하고 분액 깔대기로 유기용매층과 물층을 분리하여 활성물질을 함유하고 있는 유기용매층을 모은 후 감압하에 농축 건조하여 황갈색의 고체물질을 얻었다. 메칠렌클로라이드에 녹인 활성물질을 실리카겔 (Merck, Art No.9385)에 흡착시킨 후 헥산과 에칠아세테이트의 비율을 변화시키면서 2번의 실리카겔 컬럼 크로마토 그래프를 실시해 활성분획을 분리한 후 최종적으로 순수한 2-하이드록시 시남 알데하이드를 HPLC로 정제 하였다. 이때 HPLC 컬럼으로는 페노메넥스(phenomenex)사의 울트라카브(ultracarb) 10 ODS (250 x 21.2 mm)를 사용하였으며 활성물질들은 80% 메탄올과 20% 물을 용매로 사용하여 용출시켰다. 최종적으로 분리된 순수한 2-하이드록시 시남 알데하이드는 하기 구조식(I)으로 표시되며 계피 1OOg 당 1lmg의 수율로 얻어졌다.
[실험예 2]
계피로부터 분리정제한 2-하이드록시 시남 알데하이드로부터 2-0-벤조일 시남 알데하이드 합성
계피로 부터 분리정제한 20mg의 2-하이드록시 시남 알데하이드를 50mL 둥근 바닥 플라스크에 넣고 20mL의 테트라하이드로 퓨란 용매를 가하여 녹인 후 질소가 충진된 상태에서 얼음 중탕 용기에 넣어 0℃로 만든 후 트리에칠 아민(1mL)과 벤조일 클로라이드(0.5mL)를 가하고 3시간동안 교반하였다. 반응이 종결되면 감압하에 농축하고 농축여액에 50mL의 에칠아세테이트를 가한 후 분액 깔대기에 옮겨 20mL의 물로 2번 씻고 유기용매층은 무수 황산염을 사용하여 건조시킨 후 감압하에 농축하였다. 메칠렌클로라이드에 녹인 농축액을 실리카겔 (Merck, Art No.9385)에 흡착시킨 후 헥산과 에칠아세테이트의 비율을 변화시키면서 실리카 크로마토그래피법으로 정제하여 하기 구조식(Ⅱ)로 표시되는 2-0-벤조일 시남 알데하이드를 72% 수율로 얻었다.
상기 실험예 1에서 얻은 2-하이드록시 시남 알데하이드와 실험예 2에서 얻은 2-0-벤조일 시남 알데하이드의 이화학적 성질을 비교하여 하기 표 1에 정리하였다.

시남 알데하이드 유도체들의 T세포분하 효과 조사
면역세포 중 흉선을 거쳐 성숙되는 T 세포는 그 기능에 따라 보조 T 세포(Th)와 세포독성 T 세포(cytotoxic T세포; Tc)로 분화하는데 이러한 T 세포의 아형은 그들이 발현하는 세포표면항원에 의해 구분된다. 따라서 Th 세포는 CD4 항원을, Tc 세포는 CD8 항원을 표면에 발현하는 특성에 근거하여 CD4+및 CD8세포의 비율을 분석하였다. 즉, 먼저 세포에 CD4 혹은 CD8 분자에 결합할 수 있는 항체를 결합시키고 이 항체의 존재 여부를 추척함으로써 세포표면에 CD4 혹은 CD8 분자를 갖고 있는 T 세포의 비율을 분석하였다. 이때 사용하는 항체는 미리 플루오로신 (fluoroscein)이나 파이코에리스린(phycoerythrin) 등과 같은 형광물질을 부착시켜 각 세포가 나타내는 형광 강도를 플로우사이토미터로 측정하였다
생쥐의 비장을 외과적 수술방법으로 적출하여 분리하였다. 얻어진 비장세포 (1 ×108개)를 배양배지 [3차 증류수 1ℓ에 RPMI 1640 (10.4g) NaHCO3(3.7g), N-(2-하이드록시에틸)피페라진-N'-(2-에탄술폰산) (HEPES; 2.83g), 항생제-항진균제 (Antibiotic-Antimycotic, Gibco; 10㎖)가 함유된 액에 소태아혈청을 1O%가 되도록 첨가하였다]에 혼합한 후 배양플라스크에 나누어 넣었다. 두 종류의 시남 알데하이드 유도체인, 실시예 1의 실험에 1에서 얻은 2-하이드록시 시남 알데하이드와 실험예 2에서 얻은 2-0-벤조일 시남 알데하이드 10㎛를 디메칠술폭사이드(DMSO)에 녹여 비장세포 혼합액에 가한 후 37℃의 CO2배양기에서 이틀동안 배양하였다. 배양된 비장세포를 각각 플로우사이토미터 전용 시험관 (Falcon 2052)에 넣어 400×g에서 10분간 원심분리하여 상층액을 제거하고 세포침전물에 플루오르신이 결합된 CD4 항체 (anti-CD4 FITC)와 파이코에리스린이 결합된 CD8 항체 (anti-CD8 RE) 희석액을 100㎕씩 첨가하였다. 결합된 CD4 항체(anti-CD4 FITC)를 인산완층생리식염수 (phosphate-buffered saline;PBS)로 3차례 세척한 후 1㎖의 PBS에 현탁시킨 세포부유액을 플로우사이토미터에 적용시켜 자료를 히스토그램으로 컴퓨터 상에서 출력하여 CD4, CD8 세포표면 분자에 대한 분석비율을 산출하였다. 실험결과, 표 2에 나타낸 바와 같이 두 종류의 시남 알데하이드 유도체를 넣고 배양한 세포군의 경우, 아무것도 첨가하지 않은 군과 DMSO만 넣고 배양한 대조군에 비하여 성숙되지 않은 T 세포 (CD4-CD8-) 분화의 최종 단계인 보조 T 세포로 분화된 상태를 나타내는 CD4+CD8-비율이 높았다.

시남 알데하이드 유도체들의 T 세포로부터 감마인터페로 생성 유도효과 조사
시남 알데하이드 유도체들이 보조 T 세포로 분화된 T 세포로부터 생체활성물질의 하나인 감마인터페론 생성유도 유무를 측정하였다. 즉, 생쥐 비장세포 배양 (2 x 106 cel1s/mL)에 시남 알데하이드를 처리한 후 48시간 배양한 후 이 배양액의 감마인터페론 양을 엘리사 정량 킷트 (Genzyme Corp., Cambrige, MA)로 측정하였으며 이때 처리그룹당 3번씩 측정하였다. 첫 번째로 얻어진 배양액 시료 50㎕를 항체가 미리 코팅된 마이크로타이터(microtiter) 구멍에 37℃에서 1시간 동안 넣어둔후 구멍들은 인산완충액으로 2번 세척하고 바이오틴(biotin)으로 표지된 항감마인터페론 항체 50㎕를 37℃에서 한시간 동안 반응시켰다. 다시 인산완충액으로 구멍들을 3번 세척하고 호세-라디쉬 퍼옥시다아제(horse-radish peroxidase)로 표지된 스트렙토아비딘(streptoavidin) 용액 50 ㎕를 각 구멍에 37℃에서 40분간 넣어 두었다. 구멍들을 인산완충액으로 3번 세척한 후 100㎕의 기질용액을 넣고 10분간 기다린 후 450nm에서 그 흡광도를 엘리사 측정기로 측정하였다. 표 3에 나타낸 바와 같이 두 종류의 시남 알데하이드 유도체를 넣고 배양한 세포군의 경우, 아무것도 첨가하지 않은 군과 DMSO만 넣고 배양한 대조군에 비해 많은 양의 감마인터페론이 생성되었다.

[실시예 4]
시남 알데하이드 유도체들의 급성독성조사
두 종류의 시남 알데하이드 유도체인 2-하이드록시 시남 알데하이드와 2-0-벤조일 시남 알데하이드 각각을 체중 23 내지 25g의 생쥐에 100 mg/kg 용량으로 피하주사한 결과, 특별한 이상증상은 관찰되지 않았고 투여 20일까지 사망하지 않았으며 최중의 감소도 관찰되지 않았다. 그러므로 이들 물질에 대한 급성독성효과는 없는 것으로 판단되었다.
상기 실시예를 통하여 설명한 바와 같이, 계피로부터 분리정제한 시남알데하이드 유도체인 2-하이드록시 시남 알데하이드 및 이로부터 화학합성된 2-0-벤조일시남 알데하이드는 T 세포 분화를 촉진시켜 감마인터페론의 생성을 유도하여 면역 기능을 강화시키는 효과와 급성독성이 없는 뛰어난 효과가 있으므로 생물의약 산업상 매우 유용한 발명인 것이다.

- 하기와 같은 구조식(Ⅱ)을 갖는 2-0-벤조일 시남 알데하이드를 유효성분으로 포함하는 면역증강용 조성물.
