테스트균들은 실험전까지 글리세롤과 LB 배양액(Luria-Bertani; 트립톤 1g, 효모 추출물 0.5g, NaCl 0.5g, 글루코스 0.1g, 증류수 100㎖)의 혼합액이 든 용기에 넣어 냉동기(deep freezer)에 보관하였다. 상기 보관액을 적당량 취하여 새로운 LB 배양액에 넣어 균의 최대 생장량이 될 때까지 배양하였다. 이 배양액으로부터 포자를 취하여 10㎖의 LB 배지에서 포자 농도가 106이 되도록 첨가한 다음, 상기 실시예 1 ∼ 3에서 얻은 각각의 추출물 분획을 농도가 200 ppm, 400 ppm, 600 ppm, 1000 ppm이 되도록 첨가하여 24시간 배양한 후 각각의 최대 흡광도를 610 ㎚에서 측정하였다. 무처리구에는 소나무류 수피 추출물을 넣지 않고 배양하였으며, 균생장 억제율은 무처리구와 비교하여 다음 수학식 1에 의해 계산하였다.The test bacteria were stored in a deep freezer in a container containing a mixture of glycerol and LB broth (Luria-Bertani; tryptone 1g, yeast extract 0.5g, NaCl 0.5g, glucose 0.1g, distilled water 100ml). .
