Description
본 발명은 차환되지 않은 개방형 우론산부분과C(2) 또는 C(3) 위치의 0-S03기가 변화되지 않은 우론산을 갖는 황산더마탄에 관한 것이다.
헤파린, 황산헤파린, 황산더마탄의 각종 약리학적 활성은, 거의 대부분 가장긴 다당사슬에 함유된 특정구조단의 존재와 관련이 있다.
일례로, 헤파린이 항응고활성을 나타내는데 보다 중요한 요건은, D-클루쿠론산 잔기와 하나의 삼황산화 D-글루코사민 잔기를 함유하는 특이적 오당류이다.(Thunberg L. et al,. Carbohydr. Res., 100, 393, 1982: Casu B. et al., Bioch. J.,197, 599, 1981; Choay J., Biochem. Biophys. Res. Comm. 116, 492, 1983)
상기 오당류 서열은 항트롬빈Ⅲ(ATⅢ)의 실제 결합부위이다.
헤파린 코펙터 Ⅱ(HC Ⅱ)는 최소 여덟 개의 단당류로 이루어진 올리고당류에의해 활성화되며, 이 올리고당은 황산더미탄내에 함유되어 있다.(Tollefsen D.M.et al., J. Biol. Chem., 262 (19), 8854, 1986)
그러므로 좀더 거대구조인 각종 다당류 구조중에 함유된 특정 단당류 서열은 특이적 활성부위의 활성화에 필수적인 것으로 보인다.
현재 놀라운 것은 황산더마탄 다당류 사슬의 유연성을 증가시키면 혈장단백질과의 결합확률이 높아지고 따라서 황산더미탄 자체의 약리학적 활성이 증가하는 것을 발견하였다는 것이다.
과요오드산염음, 인접하는 2개의 비치환 수산기를 갖는 탄소간의 결합을 절단하는 것으로 공지되어 있다.
황산더마탄에 있어서 우론산의 C(2), C(3) 위치는 거의 대부분 2개의 비치환된 트란스-히드록시기를 가지므로 상기 조건하에 놓이게 되고 결국 과요오드산 음이온이 C(2)-C(3)결합을 절단하여 각 탄소원자를 알데히드로 산화시킨다. 그 결과, 우론산 당고리가 절단된다. C(2), C(3)위치에 0S03기를 가지는 우론산부 만이 영향을 받지 않고 남게 되는데, 이 부분은 소량이다. 그러므로 거대분자 사슬의 전부가 과요오드산 이온에 의해 산화되면 출발물질 더마탄 구조와는 다른 입체구조를 얻게된다. C(2)와 C(3)위치의 알데히드기는, 수소화붕소나트륨에 의해 제1차 알코올로 환원되거나 또는 산화될수도 있다. 어쨋든 더마탄 사슬은 산성환경에서 더 불안정하게 되어 저부자량의 올리고머(스미스분해)나 구성단위까지 분해될 수 있다(도식1 참조)
가수 분해 조건을 적당히 조절함으로써 광범위한 분자향을 가지는 올리고머를 얻을 수 있다. 얻어진 부부들은 이온 교환수지 상에서 증가하는 전하밀도에 따라 선택될 수 있다.
이런 일련의 반응은 이미 황산더마탄 분석 목적을 위해 적용되었다. (Fransson L.A. et al., Carbohydr. Res., 36, 349, 1974). 그럼에도 불구하고, 얻어진 올리고머의 구조는 밝혀지지 않았으며, 또한 출발물질 더마탄에 비해 놀랍게 향상된 올리고머의 항트롬빈 활성 역시 입증되지 않았었다. 또한 놀랍게도 C(2)-C(3) 결합이 개방된 황산더마탄은, 헤파린코펙터 Ⅱ에 대해 친화력이 약하거나 없는 것으로 판명되었다. 황산더마탄의 항혈전 작용은 헤파린 코펙터 Ⅱ의 활성화 능력에 의존할 뿐 아니라 (Tollefsen D.M. et al., J. Biol. Chem., 257, 2162, 1982, Tollefsen et al., J. Biol, Chem., 258, 6713, 1983), 아직 밝혀지지 않은 미지의 인자 및/또는 메카니즘에 의존하는 것으로 보인다.
특허문헌이나 또는 특허출원 명세서들을 보면, 황산더마탄, 황산더마탄 단편, 또는 황산더마탄 유도체들의 약리학적 활성에 대해 청구하고 있다. 그럼에도 불구하고 개방된 황산더마탄 및 개방된 황산더마탄 단편들의 약리적인 활성에 대해서는 지금까지 개시된 적 없다.
EP-A-0,097,625호에서는 항부종 활성 및 항울혈 활성을 갖는, 실질적으로 순수한 황산더마탄에 대해 청구하고 있다. EP-A-0,199,033호와 0,238,994호에서는 항혈전 활성을 갖는 순수 황산더마탄에 대해 청구하고 있다. 영국 특허 제2,098,232호는 인위적으로 다중황산화되고 화학적으로 해중합되어 약물작용을 갖는 황산콘트로이틴에 대해 포괄적으로 청구하고 있다.
PCT/EP/00291호에서는 5-6kd의 분자량을 갖도록 해중합 처리된 황산더마탄 분류물들을 청구하고 있는데, 이 분류물의 단편들은 항혈전 활성과 증가된 생체이용율을 갖는다.
EP-A-0,208,623호에서는 다당류를 포괄적으로 청구하고 있다. 따라서 결체 조직의 병상에 작용하는 저분자량(10kd이하)의 황산더마탄(특허청구범위 제6항) 역시 청구하고 있는 것이다.
일본특허 공개공보 82-40502(CA 97, 28639, 1982)호에서는, pH2이하의 DMSO중에서 해중합하고 다시 황산으로 재황산화한 황산더마탄 분류물을 청구하고 있다.
프랑스특허 2,584,728호에서는 모든 글루코사미노글리칸의 황산화에 대해 청구하고 있으며, 피리딘중에서 피리딘-SO3로 다중황산화된 황산더마탄의 황산화에 대해서도 청구하고 있다.
본 발명은 C(2) 또는 C(3)위치의 0-S03기가 변화되지 않은 우론산 및 비치화된 개방형 루온산 성분을 포함하는 황산더마탄에 관한 것이다. 본 발명은 또한 개방된 황산더마탄 단편들에 관한 것이다. 놀랍게도 상기 다당류는 항아테롬성 동맥경화 활성, 항혈전 활성, 및 혈전 분해활성을 가지며, 그 활성은 모체인 천연황산 더마탄의 활성보다 4배나 크다.
본 발명은 또한 분자량이 3,500 내지 1,000달톤인, 헤파린으로부터 얻어진 올리고당류에 관한 것이다. 이 올리고당류는 항응고활성이 없고 항트롬빈 Ⅲ(ATⅢ)에 대한 친화력도 없다. 생체내에서는 항아테롬성 동맥경화 활성을 가지며 경구투여에 의해서 혈소판 응집을 저해하고, 자발성 근수축을 차단한다. 경구투여에 의한 후자의 활성은 내피하 평활근 세포증식과 관련이 있다.
본 발명은 또한 극저분자량을 가지는 헤파린 올리고당류 단편의 제조방법에 관한 것이다.
지금까지 공지된 종래의 모든 헤파린 분류물 또는 헤파린 단편은, 높은 ATⅢ 친화력과 관련해 낮은 하응고 활성(APTT)과 낮은 항혈전 활성을 갖는 것을 특징으로 한다. 어쨋든 지금까지 기술한 모든 헤파린의 해중합 공정들은 헤파린보다 저분자량을 가지는 단편을 생성하기는 하나 생성된 단편은 헤파린과 다른 구조적 특성을 갖는다.
본 발명에 의한 헤파린 올리고당류는 놀랍게도 분자량을 빼고는 헤파린구조를 그대로 갖고 있으며, 더욱이 실험정맥혈전 모델에서 뿐 아니라 동맥혈전증 모델에서까지도 항혈전 활성을 갖고 있다.
많은 헤파린 해중합 공정들이 종래의 기술로서 공지되어 있고, 이에 따라 각공정에 대해 특이적인 서로 다른 구조적 특성을 가진 생성물을 얻을 수 있다. 상기 특성들에 의해 본 발명의 생성물들과 공지의 화합물들을 구별할 수 있다.
미국 특허 제4,303,651(EP-A-0,014,184호에 대응)호에서는 14-18개의 당단위로 된, 분자량이 약 3,600∼4,800달톤인 올리고당을 청구하고 있는데 이 올리고당은 아질산으로 탈아미노 해중합하여 얻어진다. 상기 올리고당의 주성분은 이당류 L-이두로노실-2-0-설페이트-D-글루코사민-N-6-디설페이트이고, 황산화되지 않은 이두론산은 비환원 말단으로부터 3-5개 당단위 만큼 떨어진 위치에 존재한다. 이 올리고 당류는 글루쿠론산을 함유하고 있지 않다.
미국 특허 제4,500,519호와 제4,351,938호에서는 헤파린을 아질산으로 해중합하여 얻어진 올리고당류에 대해 청구하고 있는데, 그 환원말단은 무수 만니톨로환원되거나 D-만논산으로 산화될 수 있는 무수 만노스로 되어 있다.
미국 특허 제4,401,662호와 제4,474,770호에서는 높은 항혈전 활성, ATⅢ에 대한 높은 친화력 및 매우 높은 항 Xa활성을 갖는 올리고당류에 대해 청구하고 있는데 이 올리고당류는 단지 8개의 단당류로 되어 있고 아질산이나 헤파리나제로 해중합하여 얻어진다. 그 구조에는 3-위치에 OSO3를 갖는 글루코사민이 포함되어 있다.
EP-A-0,0,048,231에서는 4∼8개의 단당류는 갖는 올리고당류에 대해 청구하고 있는데 이 올리고당은 3-0-설페이티드 글루코사민과 2,5-무수-D-만노스로 이루어진 환원 말단을 특징으로 한다.
상기 인용된 모든 특허에 기재된 해중합은 아미노당과 우론산 사이의 아세탈결합과 관련이 있다. 그러므로 얻어진 올리고당류는 항상 다수의 이당류를 갖는 화합물이다. 더욱이 분열이 일어나는 위치에서는 항상 황원자의 손실이 발생한다.
미국 특허 제4,281,108호에서는, 중간체 헤파라민을 거쳐 헤파라민의 N-탈황산화,산화제 존재하에서 산매질중의 가열에 의한 해중합 및 재황산화에 의해 얻어진 저분자량의 헤파린이 개시되어 있다. 이 공정에서는 헤파린 구조의 실질적인 변화가 일어나며 이는 재황산화에 의해서도 회복될 수 있다.
EP-A-40,144호에서는 헤파린의 알칼리 가수분해 방법을 개시하고 있다.
EP-A-44,228호에 의하면 가수분해는 항상 헤파린 카르복시기의 알킬 또는 아릴 에스테르에 대해 일어나며 평균 분자량이 2.000∼9.000달톤인 올리고당류를 생성한다. 상기 후자의 두 공정 모두 β-제거반응에 의해 말단부에 △-4.5-불포화 우론산을 갖는 올리고당류를 생성한다.
미국 특허 제4,687,765호에서는 분자량이 2.000 내지 8.000인 LMW-H의 혈전용해를 위한 약물학적 용도를 청구하고 있다.
미국 특허 제4,686,288호에서는 6개이상의 당단위(분자량이 1800달톤이상)로 이루어진 사슬과 2.5-무수 만노스 환원 말단을 갖는 뮤코다당류의 제조방법을 청구하고 있다.
본 발명의 헤파린 올리고당류는 PCT/EP/86/00291(W086-06729)에 기재된 방법과 유사한 방법에 의해 얻어졌다. 상기 방법은 2가 금속이온(예컨데, Cu++)과 과산화물(예컨데, H202)에 의해 얻어지는 HO·자유라디칼의 생성에 근거하는 바, 이라디칼은 헤파린 해중합의 개시제가 된다(도식2 참조). 본 발명은 헤파린 단편들을, 평균 분자향 3.500달톤인 경우 C 2181Ⅰ, 평균분자량 2.000인 경우 OP 381/1/1, 평균 분자향 1.700인 경우 C 2085Ⅱ, 평균 분자량 약 1.000인 경우 OP 381/1/2로 각각 분류하였다. 상기 단편들은 4-12개의 당단위로 되어 있고 다음과 같은 구조특성을 갖는데 이에 의해 종래의 생성물과 구별된다: 말단의 아노머탄소는 환원성이며 말단의 단당류는 N,6-디설페이티드 글루코사민과 2-설페이티드 우론산으로 되어 있다.
이는 각각13C NMR 스펙트라의 92.7ppm과 94.4ppm에서 나타나는 신호에 의해 입증된다.13C NMR 스펙트럼의 나머지 부분은 헤파린 구조가 그대로 보존되고 있음을 나타내고 있다. 겔침투형 크로마토 그래피는 본 발명의 공정에 의해 얻어진 헤파린 올리고당루류의 연속적 형태를 나타내는데 이는 아질산 탈아미노 해중합 공정에의해 제조된 600달톤의 올리고당류에 대해서 얻어진 비연속적 형태와는 다르다. 그러므로 이 헤파린 올리고당류의 화학 구조는 다른 특허에서 청구된 화학구조와는 완전히 다른 것이다.
본 발명의 올리고당류는 헤파린이 가지는 항응고 활성을 상실하고 동맥혈전과 정맥혈전에 대한 항아테롬성 동맥경화 활성과 항혈전 활성을 갖게 되며, 피하나경구 투여에 의해 양호환 생체이용율과 장기간의 지속효과를 갖는다. 이들은 생체외에서 콜라겐과 리스토세틴과 ADP에 의해 유도된 혈서판 응집을 억제한다. 반면에 모체 헤파린은 혈소판 응집을 유도하고 (Brace L.D., Fareed J., Seminars in Thromb. and Haemost. 11, 190, 1985; Eika C., Scand J. Haematol. 8, 248, 1972)및/또는 다른 약제에 의해 유도된 혈소판 응집을 증강시킨다. 올리고당류는 혈소판 유착을 감소시킨다. 상기 모든 활성은 EP-A-00291/86호에서 수행된 조사를 토대로는 예측할 수 없었다.
지금까지 얻어진 약리학적 결과로 본 발명의 올리고당류의 임상적 유용성이 예측되며 경구 투여량은 1일 3∼15㎎/㎏, 또는 주사가능 투여량은 1일 1∼5㎎/㎏으로 하였다.
본 발명은 또한 본 발명의 생성물을 항응고 활성이 없거나 있어도 아주 미소한 항혈전제, 항아테롬성 동맥경화제 및 섬유소 용해제로서 인간의 질병의 치료에 사용하는 모든 산업적 용도에 관한 것이다.
이 목적을 이루기 위해 본 발명은 화합물은, 종래의 기술 및 부형제를 이용해 비경구용, 외용 및 경구투여용에 적합한 약제학적 조성물로서 제공된다. 비경구투여에 적합한 제형의 예로는 바이알에 담긴 무균용액이 있다. 경구 투여에 적합한 제형의 예로는 캡슐과 정제와 시럽이 있고, 활성성분은 다시 상기 제현내에서 리포좀 이나 미셀의 형태로 담겨질 수 있다. 외용 제형의 예로는 종래의 부형제를 함유하여 만들어지는 연고가 있다.
다음의 실시예에 의해 본 발명을 상세히 설명한다. 다만, 본 발명이 하기 실시예에 의해 제한되는 것은 아니다.
순수 황산더미탄 OP 437 30g([α]D 20=-60°: S03 -/COO-의 비가 1.06(ilfarmaco Ed Pr. 43, 165, 1988), 분자량 MWp=30,000달톤, 항응고 활성 없음)을 물300㎖에 넣고 여기에 0.5M NaIO4용액 300㎖를 가한다. 용액을 하룻밤 교반한다. pH를 6으로 맞추고 NaBH420g을 3시간 동안 소량씩 가한다. pH는 아세트산을 일정 간격으로 첨가하여 7로 일정하게 유지한다. 반응온도는 25℃로 조절한다. 그런 다음 그 용액을 1N HC1로 pH를 3으로 맞춰 주고 실온에서 0.5시간 교반한 후, 1N NaOH로 pH를 5로 맞추고 용액의 부피가 반이 되게 농축한다. 3배양의 메탄올을 첨가한다: 회수된 침전물을 재용해하고 메탄올로 재침전시킨 후 건조시켜 OP616 24.7g을 얻는다(수율 82.3%).
[실시예2]
실시예1에서 얻어진 생성물 6g을 물 110㎖에 첨가하여 180㎖의 IRA 93SP수지(Rohm and Haas사제)에 침몰시킨다. 침출된 용액을 소량부피로 농축하고 3배양의 에탄올로 침전시킨다. 0.25M, 0.5M, 3M의 NaCl용액을 이용하여 칼럼으로부터 분류물을 용출시킨다.
침전 및 건조후 다음과 같은 화합물들이 얻어진다: 2078 0M:2.83g(수율47.2%), 2078 0.25M:0.71g(수율 11.8%), 2078 0.5M:0.62g(수율 10%), 2078 3M:0.19g(수율 3%)
출발물질에 대해 얻어진 화합물들의 특성이 표1에 나타나 있다. 20MHz에서 작동하는 10mm 탐침이 있는 배리안 CFT20상에 기록된 제1도의13C NMR 스펙트라를 보면, 화합물 2078 0M(제1B도)과 2078 0.5M(제1C도)은 황산더마탄(제1A도)의 전체 사슬과 비교할 때 이두론산의 C(2), C(3)에 해당하는 신호와, 동일 잔기의 C(1)에 해당하는 신호가 위쪽으로 이동한 것을 알 수 있다. 부신호들(minor signal)은 더 많이 황산화된 서열에 해당한다. 실제로, 음이온 교환수지로부터 용출된 개방된황산더마탄 분류물에 대한 부신호의 강도는 NaCl농도가 증가함에 따라 커진다.(제1C도)
얻어진 화합물은 출발 황산더마탄(OP437)에 비해 향상된 혈전증 활성을 나타내고 있다.
[실시예3]
화합물 2078 0M 1g을 100㎖의 0.1M HC1에 가하여 3시간 동안 60℃로 가열한다. 용액을 NaOH로 중화하고 동결 건조하여 생성물 OP602를 얻는다. 50MHz에서 작동하는 5mm 탐침이 있는 브루커 AC200(Bruker AC200)상에 기록된 이 화합물의13C NMR 스펙트럼 (제2도)은 분해 생성물의 특징적인 12개 신호외에도 별표로 표시한 2개의 다른 부신호를 나타내는 데, 이들중 하나인 91ppm에 있는 신호는 글루쿠론산의 잔유물에 기안하는 것 같다. A-1, A-2, A-3, A-4, A-5 및 A-6로 표시된 신호들은, 갈락토사민핵의 각 탄소에 해당된다고 본다. 신호(1-4)는, 원래 이두론산의 4-위치에 있다가 지금은 잔유물내에 있는 탄소에 해당된다. 별표로 표시한 신호(G-4)는, 본래 황산더마탄 구조내의 글루쿠론산 4-위치에 있던 탄소에 해당된다. 분해 생성물의 구조로부터 실시예2에서 얻어진 화합물의 구조를 확정할 수 있다.
[실시예4]
실시예 1에 기재된 방법을 따르되, 실시예 2에 기재된 IRA 93SP 칼럼(C1 형)상에서 분류하여 다음의 분류물을 얻는다.
상기 분류물은 아래 표 2에 기재된 특성을 갖는다.
[실시예5](OP 381/1/1)
300g의 헤파린을 2ℓ의 물에 용해하고 여기에 12g의 아세트산구리 일수화물을 가하여 반응기에 넣는다. 16% 과산화수소수 1000㎖ 3.6시간동안 반응물질에 가하여 1N 수산화나트륨용액으로 pH를 7.5로 유지하고, 제1반응시간동안 외열에 의해 44℃ 내지 66℃의 온도로 상승시킨다.
30% 아세트산 120㎖로 pH를 5.5로 맞추고 아스코르빈산 6g을 첨가한다. 올리고당류를 냉각하여 3배양의 메탄올로 침전시킨다.
여과 및 건조후 평균 분자량이 2.900 달톤인 생성물 192.7g를 얻는다. 여액A를 모아 이 중간생성물 191g을 68℃로 가열된 물 1,300㎖에 용해한다. 수산화나트륨으로 pH를 7.5로 유지하면서 16% 과산화 수소용액 400㎖를 2.3시간동안 가하여 냉각후 4배양의 메탄올을 가한다.침전물을 여과 회수하여 건조한다. 120g의 생성물을 얻는다. 고체물질 전부를 물에 다시 용해하고 컬렉스 100 (chelex100(Bio-Rad))수지칼럼(직경4cm, 높이35cm)상에 침출시킨다. 메탄올 3부피를 침출물에 가한다.
침전물을 모아 건조하면 OP 381/1/1인 생성물 99.51g을 얻는다.
화합물 OP 381/1/1의 C NMR 스펙트럼은, 헤파린 구조가 본질적인 변화없이 그대로 유지되는 것을 나타내며, 또한 이두론산 및 아미노당의 환원성 아노머 탄소원자의 C신호를 나타내고 있다. 화합물 OP 381/1/1에 대해 행한 생체내 및 시험관 내 실험에 따른 생물학적 평가(표 Ⅲ)에 의하면 본 발명의 화합물은 동맥혈전증(artr. thromb)과 정맥혈전증(ven. thromb)실험모델에 있어서 뛰어난 항혈전 활성을 보이며 양호한 생체 이용율과 작용시간의 지속성(피하투여시 투여후 8시간 이상)을 보인다.
본 발명에 의한 올리고당류는 s.c. ED/ i.v.ED비에 있어서 헤파린 보다 휠씬 더 유리하다. 카올린으로 유도된 혈전증 모델 쥐(Hladovec J., Physiol.Bohemoslovaca, 24, 551-554, 1974)에 있어서, i.v.ED, s.c.ED및 oral ED은 각각 0.5mg/kg, 3.6mg/kg 및 9.4mg/kg임이 밝혀졌다. 이 값들의 비는 1/7/19이며, 이는 본 발명의 올리고당의 생체 이용율에 대한 지표가 된다.
70mcg/㎖의 농도에서 OP 381/1/1은, 같은 조건하에서 20%까지 ADP 촉진응집을 증가시키는 헤파린에 비해, 콜라겐 2mcg에 의해 유도된 혈소판 응집, 리스토세틴에 의해 유도된 혈서판 응집 및 ADP 16mcg/㎖에 의해 유도된 혈소판 응집을 억제한다.
본 발명의 생성물은 혈소판 감소증을 일으키지 않는 생리 식염수(Mascia-Brunelli, Hellem A.J., Scand. J. Haematol. 7,374,1970의 Adelplates)에 비해 혈소판 응집을 49% 감소시킨다.
상기 특성들은 동맥 혈전증 실험 모델에 있어 381/1/1의 활성이 현저하다는 것을 설명하고 있다.
OP 381/1/2
실시예5의 전반부에서 모아진 여과액 A를 진공하에서 10ℓ로 농축시킨다. 3배양의 아세톤을 가한다: 얻어진 침전물을 여과하여 회수하고 물에 재용해하여(10%) 컬렉스(chelex 100(Bio-Rad)) 수지 100㎖에 침출시킨다. 구리가 전혀없는 침출액을 동결 건조하여 OP 318/1/2 15g을 얻는다. OP 318/1/2 화합물의 C NMR 스펙트럼은 극저분자량의 생성물에 대한, 전형적인 신호분산 형태를 보인다. 더욱이 아미노당과 우로산의 환원성 아노머탄소에 해당하는 신호가 명백히 나타나 있다. 일반적인 헤파린구조(설포아미노와 C6-황산염 신호를 특징으로 함)는 유지되었다.
β-아노머탄소에 해당하는 신호는 아노머탄소의 영역에서 명백히 보이는데, 일반적으로는 분자량이 큰 헤파린에 가려져 있다.
[실시예6](C 2085)
겔-여과 칼럼상에서의 분류
실시예5에서 얻어진 생성물 381/1/1 5g을 0.1M NaCl 용액 50㎖에 가하고 울트로겔 에이씨. 에이 202(Ultrogel Ac. A 202(LKB)) 칼럼(직경5cm, 높이 100cm)에서 2㎖/min 유속으로 침출시킨다. 초기의 사용적(dead volume)420㎖를 버리고 각분획을 10㎖씩 회수한다. 처음 30개의 분획물은 버리고 31번째 분획물에서 104번 분획물까지를 조합한다(Ⅰ). 112개의 다른 분획물들은 따로 모은다(Ⅱ).
상기(Ⅰ),(Ⅱ) 두 그룹의 분류물이 소량이 될 때가지 농축하고 트리스아크릴 지에프(GF) 0.5M로 염석시켜 동결 건조한다. 생성물 C 2085 Ⅰ(3.1g)과 C 2085 Ⅱ(0.47g)를 얻는다. 제3도는 분자향 프로필이다. EP-A-14,184호의 실시예1에 따라 아질산으로 해중합하여 얻어진 생성물의 프로필(제3a도)과 미국 특허 제4,500,519호에 따라 아질산으로 해중합하여 얻어진 생성물의 프로필(제3b도)이 보고되어 있다.
정수는 이당휴 단위를 나타낸다.
[실시예7] (C 2081 Ⅰ)
초기의 81개의 분획물만을 모아서 실시예6의 방법과 유사한 방법에 의해 화합물 C 2081 Ⅰ(1.04g)을 얻는다.
Claims (10)
- 황산화되지 않은 우론산의 C(2)-C(3) 졀합을 산화개열하는 조건하에서 과요오드산 이온과의 반응에 의해 얻어지며, C(2)-C(3) 위치의 O-SO3기가 변화되지 않은채로 보존되어 있으며 하기 구조식으로 표시되는 것을 특징으로 하는 개방사슬 황산더마탄 분류물:여기서 m은 40 내지 60이고 n 및 p는 각각 10이하이고 R은 H 또는 SO3 -이다.
- 제1항에 있어서, 황산화되지 않은 우론산의 C(2)~C(3) 결합의 개열반응에 의해 얻어진 알데히드기가 히드록시 메틸기로 환원된 것을 특징으로 하는 개방사슬 황산더마탄 분류물.
- 제1항 또는 제2항에 있어서, 상기 분류물이 전해질 용액에 의해 음이온 교환 수지상에서 더 분류되어 SO3 -/COO-비가 0.9 내지 1.4인 것을 특징으로 하는 개방사슬 황산더마탄 분류물.
- 제3항에 있어서, 상기 분확이 산 또는 알칼리 매질 중에서 분자량이 2,000 달톤인 단편으로 가수분해 되는 것을 특징으로 하는 개방사슬 황산더마탄 분류물.
- 더마탄 또는 그 분류물을 과요도드산 이온 등의 산화제와 반응시키는 단계, C(2)-C(3) 위치의 알데히드기를 히드록시메틸기로 환원시키는 단계, 전하밀도가 상이한 단편들을 분리하기 위해 음이온 교환 수지상에서 얻어진 생성물을 분류하는 단계, 및 원하는 분자량을 갖는 올리고당류를 생성하기 위해 산 가수분해하는 단계를 포함하는 것을 특징으로 하는, 제1항 내지 4항 중 어느 한항 기재의 개방사슬 황산더마탄 분류물의 제조방법.
- 헤파린의 라디칼 해중합에 의해 얻어질 수 있으며 말단에, 환원성 아노머탄소를 갖는, 2-0-설페이티드 이두론산기 또는 환원성 아노머탄소를 갖는 N, 6-디설페이티드 글루코사민기를 함유하며 평균 4 내지 12개의 단당류를 함유하며, 하기 구조식으로 표시되는 헤파린 올리고당류.또는여기서, a, b, c 및 d의 합은 2-5이다.
- 제6항에 있어서, 상기 올리고당류는 분자량이 3,500 내지 1,000달톤이고 출발 헤파린의 것과 본질적으로 동일한 SO3 -/COO-비를 갖는 것을 특징으로 하는 헤파린 올리고당류.
- 제6항에 있어서, 상기 올리고당류는 헤파린의 활성보다 60배까지 낮은 항 Xa 활성을 가지며, 동맥 혈전증에 대해서도 항혈전증 활성이 크고, 항혈소판 응집 활성이 크며, 경피투여 및 경구투여에 의한 생체 이용율이 높은 것을 특징으로 하는 헤파린 올리고당류.
- 항혈전증활성, 혈전붕괴활성, 항아테롬성 동맥경화활성이 크고, 제1항 내지 제4항 중 어느 한 항 기재의 황산더마탄 분류물과 그 단편을 활성성분으로서 함유하는 것을 특징으로 하는 약제학적 조성물.
- 항혈전증활성, 혈전분해활성 및 항아테롬성 동맥경화 활성을 가지며, 제6항 내지 제8항 중 어느한 항 기재의 헤파린 올리고당류를 활성성분으로서 함유하는 것을 특징으로 하는 약제학적 조성물.
