Abstract
Description
これらの問題を解決するため、トランスポゾン法を用いたCAR-T細胞の作製工程において、CAR発現細胞の増大を目的とした種々の検討がなされている。
[1] 以下のステップ:
a)哺乳動物由来のT細胞を含む細胞集団にキメラ抗原受容体(chimeric antigen receptor:CAR)タンパク質をコードするポリヌクレオチドをトランスポゾン法により導入して遺伝子改変細胞集団を得るステップ;
b)前記CARに結合するタンパク質を発現する該哺乳動物由来の内因性細胞集団を準備するステップ;及び
c)ステップa)の遺伝子改変細胞集団とステップb)の内因性細胞集団とを共培養するステップ
を含む、哺乳動物遺伝子改変細胞の作製方法。
[2] 上記ステップb)において、内因性細胞集団を1種または複数種のサイトカインの存在下で培養する、上記[1]に記載の作製方法。
[3] 上記ステップb)におけるサイトカインがGM-CSF及び/又はIL-4である上記[2]に記載の作製方法。
[4] 上記ステップc)を、1種または複数種のサイトカインの存在下で行う、上記[1]〜[3]のいずれかに記載の作製方法。
[5] 上記ステップc)におけるサイトカインがIL-7及び/又はIL-15である上記[4]に記載の作製方法。
[6] 上記内因性細胞集団が、不活化処理を受けていないPBMCsに由来する細胞集団である上記[1]〜[5]のいずれかに記載の作製方法。
[7] トランスポゾン法がpiggyBac法である上記[1]〜[6]のいずれかに記載の作製方法。
[8] 遺伝子改変細胞集団と、内因性細胞集団が同一の個体に由来する、上記[1]〜[7]のいずれかに記載の作製方法。
[9] 上記CARタンパク質がヒト顆粒球-マクロファージコロニー刺激因子(GM-CSF)受容体に特異的な結合ドメインを含むタンパク質であり、上記内因性細胞集団がGM-CSF受容体発現細胞である、上記[1]〜[8]のいずれかに記載の作製方法。
本明細書は本願の優先権の基礎となる日本国特許出願番号2018-202336号の開示内容を包含する。
本発明は、以下のステップ:
a)哺乳動物由来のT細胞を含む細胞集団にキメラ抗原受容体(chimeric antigen receptor:CAR)タンパク質をコードするポリヌクレオチドをトランスポゾン法により導入して遺伝子改変細胞集団を得るステップ;
b)前記CARに結合するタンパク質を発現する該哺乳動物由来の内因性細胞集団を準備するステップ;及び
c)ステップa)の遺伝子改変細胞集団とステップb)の内因性細胞集団とを共培養するステップ
を含む、遺伝子改変細胞の作製方法を提供する。
本発明における「T細胞を含む細胞集団」としては、例えば、血液(末梢血、臍帯血など)、骨髄などの体液、組織又は器官により採取、単離、精製、誘導された細胞集団を使用することができる。好適には末梢血単核細胞(PBMC)を使用することができる。
細胞外スペーサードメインは、標的結合ドメインと、膜貫通ドメインとの間に存在し得るように、それぞれのドメインのアミノ酸配列をコードする塩基配列を連結してベクターに挿入し、宿主細胞において発現させることができる。あるいはまた、細胞外スペーサードメインは、予め作製したプラスミドCARタンパク質をコードするポリヌクレオチドを鋳型として改変することもできる。
細胞外スペーサードメインの改変は、例えば、CARをコードするポリヌクレオチドを導入したCAR-T細胞の宿主細胞におけるCAR遺伝子発現率の向上、シグナル伝達、細胞の老化、腫瘍への分布、抗原認識又はin vivo活性への影響を考慮した場合に有用である。
上記の方法により、1×106〜2×107個の範囲の細胞に遺伝子導入することが可能である。
本発明の方法では、ステップa)で調製する遺伝子改変細胞集団が発現するCARに結合するタンパク質を発現する内因性細胞集団を準備することを特徴とする。従って、本発明の方法は、内因性細胞集団にCARタンパク質に結合するタンパク質が発現していることを必要とするが、そのような内因性細胞集団を取得できる限り、CARタンパク質は特に限定されない。
ステップb)における培養条件は、特に限定するものではないが、例えば37℃で0〜10日間の範囲内とすることが好適である。
本発明の方法は、ステップa)で得られた遺伝子改変細胞集団と、ステップb)で得られた内因性細胞集団とを共培養するステップを含む。
上記ステップc)を、1種または複数種のサイトカインの存在下で行うことができ、例えばサイトカインはIL-7及び/又はIL-15であり得る。IL-7は、例えば5〜20 ng/mL、IL-15は、例えば1〜10 ng/mLの範囲の濃度で使用することが好ましい。
ステップc)における培養条件は、特に限定するものではないが、例えば37℃で1〜14日間とすることが好適である。
ヒトGM-CSF(NCBI Accession No.: NM_000758の翻訳領域(配列番号16、33〜464塩基:終止コドン前)の5’側にEcoRIサイト、3’側にIgG1のヒンジをコードする塩基配列及びBclIサイトをつけたPCR産物をpTA2クローニングベクター(TOYOBO)にサブクローニングした。ヒトGM-CSFとIgG1のヒンジ配列をサブクローニングしたpTA2クローニングベクターをEcoRIとBclIでダブルダイジェストし、ヒトGM-CSFとIgG1のヒンジ配列を切り出した。
実施例1で作製したCAR 001を鋳型にしたinverse-PCR法により、CH2CH3を部分欠失させ、(G4S)3を挿入したスペーサー改変体を作製した。inverse-PCRに用いるPCRプライマーは、以下に示すようにデザイン(配列番号17及び18)し、合成した(ユーロフィンジェノミクス株式会社に委託)。

実施例2で作製したCAR 002を鋳型にしたinverse-PCR法により、GM-CSFポリペプチド(配列番号1)の21番目のEをKに置換した変異型スペーサー改変体を作製した。inverse-PCRに用いるPCRプライマーは、以下に示すようにデザイン(配列番号19及び20)し、合成した(ユーロフィンジェノミクス株式会社に委託)。
フォワードプライマー:

リバースプライマー:

<4-1 PBMCの精製>
健康成人ドナーから末梢血を採取し、D-PBS(富士フィルム和光純薬株式会社)で2倍に希釈した後、Ficoll-Paque PLUS(GE Healthcare)に希釈末梢血をSepMate-50(STEMCELL Technologies)を用いて重層し、1200 x gで15分の遠心分離を行い、PBMCを分取した。分取したPBMCをD-PBSで2回洗浄し、遠心分離により精製した。精製したPBMCを、ACEフィーダー細胞(4-2)、ベクターを電気的導入する細胞(4-3)、OKT3 blast作製用の細胞(4-4)としてそれぞれ使用した。
上記4-1で精製したPBMCにpeptivatorペプチドプール(登録商標、ミルテニーバイオテック)によるウイルスペプチドパルスを行った。具体的には、100 μLのD-PBSに0.05 μg/μLのAdV5 Hexon、CMV pp65、EBV BZLF1及びEBV EBNA-1(4種類のペプチドを総称してACEペプチドと呼ぶ)を2 μLずつ添加したペプチドプールに、PBMCを12 x 106個懸濁し、CO2インキュベーター内で37℃、30分間ウイルスペプチドパルスを施した。30分後、ウイルスペプチドパルスされたPBMCに5 mLのD-PBSを加えて懸濁し、その後4分間のUV照射を行った。UV照射されたPBMC(PBMC ACEフィーダー細胞)を回収し、遠心分離した。遠心分離後、2 x 106個/ウェル/2 mLとなるように10 ng/mL human IL-7(ミルテニーバイオテック)、5 ng/mL human IL-15(ミルテニーバイオテック)及びArtificial serum Animal-free(株式会社細胞科学研究所)2%含有ALyS705(株式会社細胞科学研究所)中に懸濁し、24ウェル細胞培養用平底マルチウェルプレート(FALCON)に移した。
上記4-1で精製した15 x 106個のPBMCへの遺伝子導入のために、実施例3で作製したCAR 004プラスミド及びpCMV-piggyBac (kan)プラスミドをそれぞれ7.5 μg添加した上で、Artificial serum Animal-free 2%含有ALyS705中に懸濁した。懸濁した細胞をNEPAキュベット電極2 mmギャップ(ネッパジーン株式会社)へセットし、遺伝子導入装置であるNEPA21(ネッパジーン株式会社)を用いて懸濁液の抵抗値を測定した。その際、メーカー推奨の適正範囲内の抵抗値(0.038〜0.046 kΩ)になるようにArtificial serum Animal-free 2%含有ALyS705を用いて液量調節した。調節後、NEPA21で設定可能な範囲内で電気的導入を実施した。
Day 0にD-PBSで1 μg/mLに調製したAnti-human CD3抗体(ミルテニーバイオテック)及びAnti-human CD28抗体(ミルテニーバイオテック)を24ウェルノントリートメントプレート平底(FALCON)に500 μL添加し、37℃、2時間CO2インキュベーターへ入れることで、抗体をプレートに固相した。2時間後、上清を除くことで抗体固相したプレートを作製した。
一方、Day 0から刺激をしていたOKT3 blastは抗体を固相していない24ウェル細胞培養用平底マルチウェルプレートに移すことで、一時的に刺激を行わなかった。
上記4-4で作製したOKT3 blastに対して、上記4-2と同様のpeptivatorペプチドプールによるウイルスペプチドパルス及びUV照射を行ってOKT3 ACEフィーダー細胞とした。このOKT3 ACEフィーダー細胞を回収し、5 x 106個を30 mLの10 ng/mL human IL-7、5 ng/mL human IL-15及びArtificial serum Animal-free 2%含有ALyS705中に懸濁し、G-Rex 細胞培養プレート 6well(wilson wolf manufacturing corporation)に移した。
上記4-3で電気的導入した細胞全量を、上記4-5でOKT3 ACEフィーダー細胞が播種されたG-Rex 細胞培養プレート 6well に移し、Day 14までGMR.CAR-T細胞の培養・増幅を行ったが、その期間中、適宜培地交換、IL-7及びIL-15の添加を行った。発現プラスミドCAR 004を用いてPBMCへの電気的導入操作及びT細胞の培養・増幅によって得られるGMR.CAR-T細胞を、CAR-T 004とした。
Day 14まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率をフローサイトメトリー解析により評価した。上記4-6の細胞培養懸濁液100 μL を採取し、遠心分離した細胞にPE Anti-human GM-CSF抗体(ミルテニーバイオテック)5 μL及びAPC Anti-human CD3抗体(ミルテニーバイオテック)5 μLを加えて懸濁し、4℃、遮光にて20分間抗体標識反応を行った。20分後、D-PBSで細胞を洗浄し、遠心分離により細胞を沈殿させ上清を除去した後、D-PBS適量に再懸濁したサンプルについてFACSCantoII(ベクトン・ディッキンソン)及びFLOWJO(ベクトン・ディッキンソン)を用いて解析し、GM-CSF/CD3陽性を指標としてGMR.CARの発現率を測定した。
CAR-T 004のday 14における発現解析結果を図5に示す。GMR.CAR発現率は11.1%であった。
<5-1 PBMCの精製(Day 0)>
実施例4と同一の健康成人ドナーから末梢血を採取し、D-PBS(富士フィルム和光純薬株式会社)で2倍に希釈した後、Ficoll-Paque PLUS(GE Healthcare)に希釈末梢血をSepMate-50(STEMCELL Technologies)を用いて重層し、1200 x gで15分の遠心分離を行い、PBMCを分取した。分取したPBMCをD-PBSで2回洗浄し、遠心分離により精製した。精製したPBMCを、ベクターを電気的導入する細胞(5-3)及び内因性のCAR刺激用細胞(5-4及び5-5)としてそれぞれ使用した。
上記5-1で精製したPBMCを24ウェル細胞培養用平底マルチウェルプレート(FALCON)に20 x 106個/ウェル/2 mLとなるようにD-PBSで懸濁し、播種した。播種した細胞を37℃、2時間CO2インキュベーター内で静置させた後、培養上清とともに浮遊細胞を回収し、下記5-3の電気的導入を実施した。
一方で、プレートに付着した細胞は、2mLの10 ng/mL human IL-7(ミルテニーバイオテック)、5 ng/mL human IL-15(ミルテニーバイオテック)及びArtificial serum Animal-free(株式会社細胞科学研究所)2%含有ALyS705(株式会社細胞科学研究所)で培養し、内因性のCAR刺激用細胞として下記5-4の操作に使用した。
上記5-2で回収した浮遊細胞への遺伝子導入のために、CAR 004プラスミド及びpCMV-piggyBac (kan)プラスミドをそれぞれ7.5 μg添加した上で、Artificial serum Animal-free 2%含有ALyS705中に懸濁した。懸濁した細胞をNEPAキュベット電極2 mmギャップ(ネッパジーン株式会社)へセットし、遺伝子導入装置であるNEPA21(ネッパジーン株式会社)を用いて懸濁液の抵抗値を測定した。その際、メーカー推奨の適正範囲内の抵抗値(0.038〜0.046 kΩ)になるようにArtificial serum Animal-free 2%含有ALyS705を用いて液量調節した。調節後、実施例4と同様の設定値で電気的導入を実施した。
上記5-3で電気的導入した細胞全量を、上記5-2で用意した内因性のCAR刺激用細胞が予め付着した24ウェル細胞培養用平底マルチウェルプレートへ移し、37℃のCO2インキュベーター内で培養を開始した。プラスミドを添加せず、電気的刺激を施したMock-T細胞を同様に培養した。7日間の培養期間中、適宜培地交換、IL-7及びIL-15の添加を行った。
上記5-1で精製したPBMCを、5 x 106個/ウェル/15 mLとなるように10 ng/mL human GM-CSF(ミルテニーバイオテック)及び10 ng/mL human IL-4(ミルテニーバイオテック)含有ALyS705で懸濁し、G-Rex 細胞培養プレート 6well(wilson wolf manufacturing corporation)に播種し、37℃のCO2インキュベーター内で培養を開始した。7日間の培養期間中、適宜GM-CSF及びIL-4の添加を行った。
上記5-5で7日間培養したG-Rex 細胞培養プレート 6wellから上清を除き、30mLの10 ng/mL human IL-7、5 ng/mL human IL-15及びArtificial serum Animal-free 2%含有ALyS705を添加した。さらに、7日間24ウェル細胞培養用平底マルチウェルプレートで培養していた細胞全量をG-Rex 細胞培養プレート 6wellへ移すことで、遺伝子導入した細胞への2回目のCAR刺激を実施した。その後さらに6日間培養・増幅し、その期間中、適宜培地交換、IL-7及びIL-15の添加を行った。Day 13まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率を以下の方法にて測定した。本実施例において内因性の細胞を用いてCAR刺激を施したGMR.CAR-T細胞の培養・増幅によって得られる、発現プラスミドCAR 004を用いたGMR.CAR-T細胞をCAR-T 004-Aとした。
Day 13まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率をフローサイトメトリー解析により評価した。上記5-6の細胞培養懸濁液100 μL を採取し、遠心分離した細胞にPE Anti-human GM-CSF抗体(ミルテニーバイオテック)5μL及びAPC anti-human CD3抗体(ミルテニーバイオテック)5μLを加え懸濁し、4℃、遮光にて20分間抗体標識反応を行った。20分後、D-PBS適量で細胞を洗浄し、遠心分離により細胞を沈殿させ上清を除去した後、D-PBS適量に再懸濁したサンプルについてFACSCantoII(ベクトン・ディッキンソン)及びFLOWJO(ベクトン・ディッキンソン)を用いて解析し、GM-CSF/CD3陽性を指標としてGMR.CARの発現率を測定した。
<6-1 PBMCの精製(Day 0)>
実施例4及び5と同一の健康成人ドナーから末梢血を採取し、D-PBS(富士フィルム和光純薬株式会社)で2倍に希釈した後、Ficoll-Paque PLUS(GE Healthcare)に希釈末梢血をSepMate-50(STEMCELL Technologies)を用いて重層し、1200 x gで15分の遠心分離を行い、PBMCを分取した。分取したPBMCをD-PBSで2回洗浄し、遠心分離により精製した。精製したPBMCを、ベクターを電気的導入する細胞(6-2)及び内因性のCAR刺激用細胞(6-3)としてそれぞれ使用した。
上記6-1で精製した15 x 106個のPBMCへの遺伝子導入のために、CAR 004プラスミド及びpCMV-piggyBac (kan)プラスミドをそれぞれ7.5 μg添加した上で、Artificial serum Animal-free(株式会社細胞科学研究所)2%含有ALyS705(株式会社細胞科学研究所)中に懸濁した。懸濁した細胞をNEPAキュベット電極2 mmギャップ(ネッパジーン株式会社)へセットし、遺伝子導入装置であるNEPA21(ネッパジーン株式会社)を用いて懸濁液の抵抗値を測定した。その際、メーカー推奨の適正範囲内の抵抗値(0.038〜0.046 kΩ)になるようにArtificial serum Animal-free 2%含有ALyS705を用いて液量調節した。調節後、実施例4及び5と同様の設定値で電気的導入を実施した。
上記6-1で精製したPBMCを、5 x 106個/ウェル/15 mLとなるように10 ng/mL human GM-CSF(ミルテニーバイオテック)及び10 ng/mL human IL-4(ミルテニーバイオテック)含有ALyS705で懸濁し、G-Rex 細胞培養プレート 6well(wilson wolf manufacturing corporation)に播種し、37℃のCO2インキュベーター内で3日間培養した。
上記6-3で3日間培養したG-Rex 細胞培養プレート 6wellから上清を除き、30mLの10 ng/mL human IL-7、5 ng/mL human IL-15及びArtificial serum Animal-free 2%含有ALyS705を添加した。さらに、3日間24ウェル細胞培養用平底マルチウェルプレートで培養していた上記6-2の遺伝子導入細胞全量をG-Rex 細胞培養プレート 6wellへ移すことで、遺伝子導入した細胞へのCAR刺激を実施した。その後さらに7日間培養し、その期間中、適宜培地交換、IL-7及びIL-15の添加を行った。Day 10まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率を以下の方法にて測定した。内因性の細胞を用い、かつ実施例5より簡便な方法でCAR刺激を施したGMR.CAR-T細胞の培養・増幅によって得られる、発現プラスミドCAR 004を用いたGMR.CAR-T細胞をCAR-T 004-Bとした。
Day 10まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率をフローサイトメトリー解析により評価した。上記6-4の細胞培養懸濁液100 μL を採取し、遠心分離した細胞にPE Anti-human GM-CSF抗体(ミルテニーバイオテック)5μL及びAPC Anti-human CD3抗体(ミルテニーバイオテック)5μLを加え懸濁し、4℃、遮光にて20分間抗体標識反応を行った。20分後、D-PBS適量で細胞を洗浄し、遠心分離により細胞を沈殿させ上清を除去した後、D-PBS適量に再懸濁したサンプルについてFACSCantoII(ベクトン・ディッキンソン)及びFLOWJO(ベクトン・ディッキンソン)を用いて解析し、GM-CSF/CD3陽性を指標としてGMR.CARの発現率を測定した。
養子免疫療法において効果が期待されるCAR-T細胞は、持続的な細胞傷害活性が要求される。したがって、実施例6で作製したCAR-T 004-Bを用いて抗腫瘍細胞活性の持続性を評価した。
<8-1 PBMCの精製(Day 0)>
実施例4、5及び6と同一の健康成人ドナーから末梢血を採取し、D-PBS(富士フィルム和光純薬株式会社)で2倍に希釈した後、Ficoll-Paque PLUS(GE Healthcare)に希釈末梢血をSepMate-50(STEMCELL Technologies)を用いて重層し、1200 x gで15分の遠心分離を行い、PBMCを分取した。分取したPBMCをD-PBSで2回洗浄し、遠心分離により精製した。精製したPBMCを、ベクターを電気的導入する細胞(8-2)及び内因性のCAR刺激用細胞(8-3)としてそれぞれ使用した。
上記8-1で精製した20 x 106個のPBMCへの遺伝子導入のために、CAR 004プラスミド及びpCMV-piggyBac (kan)プラスミドをそれぞれ7.5 μg添加した上で、Artificial serum Animal-free(株式会社細胞科学研究所)5%含有ALyS705(株式会社細胞科学研究所)中に懸濁した。懸濁した細胞をNEPAキュベット電極2 mmギャップ(ネッパジーン株式会社)へセットし、遺伝子導入装置であるNEPA21(ネッパジーン株式会社)を用いて懸濁液の抵抗値を測定した。その際、メーカー推奨の適正範囲内の抵抗値(0.038〜0.046 kΩ)になるようにArtificial serum Animal-free 5%含有ALyS705を用いて液量調節した。調節後、実施例4、5及び6と同様の設定値で電気的導入を実施した。
上記8-1で精製したPBMCを、5 x 106個/ウェル/15 mLとなるようにALyS705で懸濁し、G-Rex 細胞培養プレート 6well(wilson wolf manufacturing corporation)に播種し、37℃のCO2インキュベーター内で3日間培養した。
上記8-3で3日間培養したG-Rex 細胞培養プレート 6wellから上清を除き、30mLの10 ng/mL human IL-7、5 ng/mL human IL-15及びArtificial serum Animal-free 5%含有ALyS705を添加した。さらに、3日間24ウェル細胞培養用平底マルチウェルプレートで培養していた上記8-2の遺伝子導入細胞全量をG-Rex 細胞培養プレート 6wellへ移すことで、遺伝子導入した細胞へのCAR刺激を実施した。その後さらに8日間培養し、その期間中、適宜培地交換、IL-7及びIL-15の添加を行った。Day 11まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率を以下の方法にて測定した。実施例6とは異なり、IL-4やGM-CSFといったサイトカインを添加せずに内因性の細胞を培養し、かつ実施例5より簡便な方法でCAR刺激を施したGMR.CAR-T細胞の培養・増幅によって得られる、発現プラスミドCAR 004を用いたGMR.CAR-T細胞をCAR-T 004-Cとした。
Day 11まで培養・増幅したGMR.CAR-T細胞におけるGMR.CARの発現率をフローサイトメトリー解析により評価した。上記8-4の細胞培養懸濁液100 μL を採取し、遠心分離した細胞にPE Anti-human GM-CSF抗体(ミルテニーバイオテック)5μL及びAPC Anti-human CD3抗体(ミルテニーバイオテック)5μLを加え懸濁し、4℃、遮光にて20分間抗体標識反応を行った。20分後、D-PBS適量で細胞を洗浄し、遠心分離により細胞を沈殿させ上清を除去した後、D-PBS適量に再懸濁したサンプルについてFACSCantoII(ベクトン・ディッキンソン)及びFLOWJO(ベクトン・ディッキンソン)を用いて解析し、GM-CSF/CD3陽性を指標としてGMR.CARの発現率を測定した。
実施例5で作製したCAR-T 004-A及び実施例6で作製したCAR-T 004-BをDay 14まで培養してGMR.CAR-T細胞のフェノタイプを解析した。フェノタイプ解析することで、PD-1陽性率を指標にした疲弊率と、CD45RA/CCR7陽性率を指標にしたステムセルメモリー分画の割合を算出した。
上記と同様の要領で1x 106個の細胞をPE-Cy7 Isotype control抗体(ベクトン・ディッキンソン)、AF647 Isotype control抗体(ベクトン・ディッキンソン)、BV421 Isotype control抗体(ベクトン・ディッキンソン)と反応させたものをアイソタイプコントロールとした。
その結果、図10に示すように、CAR-T 004-A及びCAR-T 004-BのPD-1陽性率はそれぞれ4.2%、1.82%であり、CAR刺激しても疲弊した細胞の割合は少なかった。また,図11に示すように、CAR-T 004-A及びCAR-T 004-BのCD45RA/CCR7陽性率はそれぞれ34.0%及び30.7%であった。

Claims (9)
- 以下のステップ:
a)哺乳動物由来のT細胞を含む細胞集団にキメラ抗原受容体(chimeric antigen receptor:CAR)タンパク質をコードするポリヌクレオチドをトランスポゾン法により導入して遺伝子改変細胞集団を得るステップ;
b)前記CARに結合するタンパク質を発現する該哺乳動物由来の内因性細胞集団を準備するステップ;及び
c)ステップa)の遺伝子改変細胞集団とステップb)の内因性細胞集団とを共培養するステップ
を含む、哺乳動物遺伝子改変細胞の作製方法。 - 上記ステップb)において、内因性細胞集団を1種または複数種のサイトカインの存在下で培養する、請求項1に記載の作製方法。
- 上記ステップb)におけるサイトカインがGM-CSF及び/又はIL-4である請求項2に記載の作製方法。
- 上記ステップc)を、1種または複数種のサイトカインの存在下で行う、請求項1〜3のいずれか一項に記載の作製方法。
- 上記ステップc)におけるサイトカインがIL-7及び/又はIL-15である請求項4に記載の作製方法。
- 上記内因性細胞集団が、不活化処理を受けていないPBMCに由来する細胞集団である請求項1〜5のいずれか一項に記載の作製方法。
- トランスポゾン法がpiggyBac法である請求項1〜6のいずれか一項に記載の作製方法。
- 遺伝子改変細胞集団と、内因性細胞集団が同一の個体に由来する、請求項1〜7のいずれか一項に記載の作製方法。
- 上記CARタンパク質がヒト顆粒球-マクロファージコロニー刺激因子(GM-CSF)受容体に特異的な結合ドメインを含むタンパク質であり、上記内因性細胞集団がGM-CSF受容体発現細胞である、請求項1〜8のいずれか一項に記載の作製方法。
