Abstract
Description
理論に縛られるわけではないが、糖鎖特異的な糖鎖切断酵素によって処理された糖鎖は、動物生体内で極めて稀な糖鎖構造、例えばフコシル化糖ペプチド構造を生み出すため、抗体産生動物内での抗体産生が妨げられない点が理由として考えられる。

糖タンパク質を抗原として用いる際の糖タンパク質の処理方法であって、
糖鎖特異的な糖鎖切断酵素によって糖鎖を切断することを含む、
糖タンパク質の処理方法。

前記糖鎖を切断する工程が、

上記の方法により得られた、生体内に存在しないか存在しても極めて稀な糖鎖構造を有する本発明の糖タンパク質を抗原として用いることにより、当該糖鎖構造と特異的に反応する抗体を作成することができる。本発明の糖タンパク質の糖鎖と特異的に反応する抗体を作成する方法の概要を図1に示す。
本発明の糖タンパク質の処理方法によって得られた糖タンパク質は、抗体のほか、当該糖タンパク質を特異的に選択する他の選択的結合性物質、例えば、ペプチドアプタマー、DNAアプタマー、RNAアプタマー等のアプタマー、レクチン、受容体、これらの改変物、およびこれらの組み合わせを製造するために用いることができる。この目的においても、糖タンパク質として、糖鎖特異的な糖鎖切断酵素によって処理する方法によって得られたもののほか、化学合成や遺伝子組み換え技術等公知の方法により得られた同構造を持つ化合物、またはこれらの改変物を使用することができる。
本発明の方法により得られた抗体等の選択的結合性物質は、バイオマーカーなどとして機能する糖タンパク質を生体試料において検出するために使用することができる。
本発明による試料中の糖タンパク質を特異的に検出する方法を行う際には、糖タンパク質を含む可能性のある試料は糖鎖特異的な糖鎖切断酵素で処理され得る。当該糖鎖特異的な糖鎖切断酵素は、抗体等の選択的結合性物質を作成する際に用いた糖鎖特異的な糖鎖切断酵素と同じであってもよいし、異なっていてもよい。一態様において、同じ酵素または酵素の組み合わせを用いることが好ましい。
肝癌細胞株HepG2細胞の培養上清からアフィニティ精製した糖タンパク質AFPを下記条件で糖鎖切断酵素:Endo−F3(38.8kDa、New England BioLabs社製)により処理した。前記精製AFPには、AFP−L3が含まれている。
[材料および条件]
(1)AFP:8μgに対して、
(2)Endo−F3:16U(0.1M MES緩衝液(pH5.5)20μL)を混合し、
処理(酵素反応)時間:10分または30分
処理(酵素反応)温度:37℃
で酵素処理し、酵素処理したAFP液を得た。
前記酵素処理したAFP液とトリスSDSサンプル処理液(コスモバイオ社製、423420)を等量加え、10分間煮沸した。当該煮沸した試料をマルチゲル(登録商標)IIミニ 4/20(13W)(コスモバイオ社製、414879)により、30mAで70分間電気泳動を行った後、ゲルを定法のCBB染色により染色した。
上記切断された糖鎖を有するAFPを抗原(免疫原)として、公知の方法でマウス又はラットに免疫し、当該マウス又はラットの脾臓より得たB細胞からハイブリドーマを作成する。

1.試験方法
HepG2の培養上清からアフィニティ精製したAFP2μL(5〜7μg)にEndo−F2(39.8kD、New England BioLabs社製)を2μL(16U)またはEndo−F3を2μL(2μg)加え、更に0.1M MES緩衝液(pH5.5)を16μL加えた。それらの試料を37℃で10分間酵素反応させた後に、トリスSDSサンプル処理液を等量加え、10分間煮沸した。それらの試料を、マルチゲル(登録商標)IIミニ4/20(13W)に1レーン10μL添加し、30mAで70分間電気泳動を行った。その後、ゲルを定法のCBB染色により染色した。
結果を図4に示す。糖鎖切断酵素を加えていないAFPと、糖鎖切断酵素であるEndo−F2及びEndo−F3を加えたAFPでは、検出されるバンドのパターンが異なっており、酵素で処理したAFPでは、特に低分子量側のバンドの濃度が濃くなっていた(図4)。これは、AFPに修飾された一部の糖鎖が糖鎖切断酵素によって消化切断され、分子量が小さくなっていることを示している。この結果は、糖鎖切断酵素の種類によらずAFPの糖鎖を消化可能であることを示している。
1.免疫原の調製
AFPを糖鎖切断酵素処理した際に生じるアミノ酸及び糖鎖配列を模した2糖ペプチドを、KLH(Thermo社77600)等の種々のタンパク質と市販のリンカー試薬を用いて架橋し、免疫原溶液を調製した。
(1)動物への免疫
前記免疫原溶液(0.2〜2mg/mL)とフロインド完全アジュバンドを等容量ずつ混合して調製したエマルジョンを用い、Balb/cAJclマウスまたはF344ラットに1匹あたり免疫原として10〜40μgを注射した。さらに、1週間の間隔で3〜8回、当該エマルジョンの注射を繰り返した。尾部静脈より採血して得た抗血清中の抗体価を、後述する抗原固相化ELISA法にて測定した。
(2)1次スクリーニング(抗原固相化ELISA法)
前記免疫した動物の抗血清中の糖鎖ペプチドに対する抗体の存在を、免疫原と同様の方法で作製した2糖ペプチドコンジュゲートを固相化したELISA法(抗原固相化ELISA法)で確認した。抗原固相化ELISA法の詳細は以下である。
2糖ペプチドと免疫原の調製に用いたタンパク質とは異なるタンパク質とのコンジュゲート液を1μg/mLになるよう、150mM塩化ナトリウムを含む20mMリン酸緩衝液(pH7.2;以下、PBSという)に溶解し、該溶解液50μLを96穴マイクロプレートの各ウエルに分注して、室温で2時間静置した。
前記各ウエルを0.05%Tween(登録商標)20を含むPBS(以下、PBSTという)400μLで3回洗浄した後、1%牛血清アルブミンを含むPBST(以下、BSA−PBSTという)100μLを加え、室温で1時間ブロッキングを行った。これをELISA用プレートとした。
前記ELISA用プレートの各ウエルをPBST400μLで3回洗浄した後、BSA−PBSTで段階希釈した免疫動物の抗血清及び非免疫動物の血清50μLを前記各ウエルに添加し、室温で1時間静置した。前記各ウエルをPBST400μLで3回洗浄した後、BSA−PBSTで10000倍希釈したHRP標識抗マウスIgG(H&L)または5000倍希釈したHRP標識ラットIgG(H&L)を50μL前記各ウエルに分注し、室温で1時間静置した。前記各ウエルをPBST400μLで3回洗浄した後、0.2%オルトフェニレンジアミン及び0.02%過酸化水素を含むクエン酸緩衝液(pH5.0)50μLを加え、室温で10分間放置後、1.5N硫酸50μLを加えて酵素反応を停止させ、波長492nmにおける吸光度を測定した。測定の結果、抗体価の高かったマウス又はラットから、脾臓及びリンパ節を摘出して、脾臓及びリンパ節由来細胞を調製し、細胞融合に用いた。
結果の一例としてマウスに免疫した場合を図5に示す。非免疫動物の血清と比較して免疫動物の抗血清は希釈倍率が低いほど、2糖の糖ペプチドを固相化して用いた場合のみ高い吸光度を示した。この測定系では、測定される吸光度は採血した動物の血清中に含まれる2糖ペプチドに対する抗体濃度に依存する(図5)。従って、前述の結果は、免疫動物の抗血清には非免疫動物の血清と比較して、より高い濃度の2糖ペプチドに対する抗体を含んでいるため抗体価が高い、ということを示している。
前記脾臓由来細胞もしくはリンパ節由来細胞のいずれかとミエローマ細胞を細胞数で1対1の割合で混合し、電気パルス法により細胞融合した。該融合させた細胞をHAT培地に懸濁し、CO2インキュベータ内で37℃、5%CO2にて8日間培養して、融合細胞(ハイブリドーマ)を得た。
(4−1)方法
上述の抗原固相化ELISA法において、免疫動物の抗血清の代わりに融合細胞の培養上清を用いたこと、固相化抗原として、2糖ペプチドコンジュゲートに加え、単糖ペプチド、酵素未処理のAFP及びEndo−F3処理したAFP、それぞれのコンジュゲートも用いた以外は、同様の方法を行った。測定の結果、単糖ペプチドよりも2糖ペプチドに対する吸光度の高いウエルまたは酵素未処理のAFPよりもEndo−F3処理したAFPに対する吸光度の高いウエルを、抗2糖ペプチドまたは抗酵素処理AFP抗体産生ハイブリドーマの存在するウエル(陽性ウエル)として選択した。
結果の一例として2糖ペプチド又は単糖ペプチドを固相化した試験の場合を図6に示す。2糖ペプチドを免疫した動物からは2糖ペプチドに反応し、単糖ペプチドに反応しない抗体産生株を複数確認することができた(図6)。この結果は、2糖ペプチドを免疫原とすることで、酵素処理AFPの2糖の糖鎖修飾部位近傍領域への抗体産生ハイブリドーマを取得できることを示している。
1.試験方法
HepG2の培養上清から精製したAFPを試験例1と同様にEndo−F3で処理し、酵素を除去したものも免疫原とし〔試験例2〕と同様に実施した。ただし、抗原固相化ELISA法には抗原としてEndo−F3で処理したAFPを用いた。
結果の一例としてラットに免疫した場合を図7、8に示す。非免疫動物の血清と比較して免疫動物の抗血清は希釈倍率が低いほど、固相化した酵素処理AFPに高い吸光度を示した(図7)。この測定系では、測定される吸光度は採血した動物の血清中に含まれる酵素処理AFPに対する抗体濃度に依存する。つまり、前述の結果は、免疫動物の抗血清には非免疫動物の血清と比較して、より高い濃度の酵素処理AFPに対する抗体を含み、抗体価が高い、ということを示している。
上述の1次スクリーニング及び2次スクリーニングで選択された抗体産生株ハイブリドーマを用いて、ハイブリドーマの単クローン化とモノクローナル抗体の精製を行った。クローン化は定法(限界希釈法、例えば「実験医学別冊 タンパク質実験ハンドブック」ISBN4−89706−369−8を参照)で行い、上述の抗原固相化ELISA法と同様の方法で陽性ウエルを選別し、最終的に2種のモノクローナル抗体産生ハイブリドーマ(S20205R及びS20206R)を得た。
S20205Rが産生する抗体、及びS20206Rが産生する抗体は酵素処理AFPにどのような特異性で反応しているかを先ず、免疫沈降とウェスタンブロッティングで確認した。
HepG2細胞の培養上清400μLに0.1M MES緩衝液を400μL、更にEndo−F3を7.5μg加えて、37℃、30分間反応させた後に、800μLのBSA−PBSTを加え試料を調製した。次いで、S20205R抗体、S20206R抗体、既存の抗AFP抗体(自社製02211)及びAFPと反応しない抗体の4種類をそれぞれ定法により固定化したセファロースゲル100μL(GEヘルスケア社:CNBr activated Sepharose 4 Fast Flow(17098101))に上記の試料をそれぞれ400μL加え、室温で2時間攪拌した。それらを10000g、2分間遠心し、上清を除去してPBST1mLでよく懸濁し、再び10000g、2分間遠心した。この操作を2回繰り返した後に上清を除去し、トリスSDSサンプル処理液を60μL加え、10分間煮沸した。その後、10000g、2分間遠心し、1レーン15μLの量を添加し、SDS−PAGEを〔試験例1〕と同様に行った。ここまでの操作を免疫沈降とする。当該電気泳動後のゲルをPVDF膜へ定法で転写し、1%BSA−PBST内で上記PVDF膜を室温1時間で振とうした。その後、検出抗体としてビオチン標識した既存抗AFP抗体またはAFPに反応しない抗体を1μg/mLに希釈した1%BSA−PBST内で上記PVDF膜を室温1時間で振とうした。次いで、PBST内で上記PVDF膜を室温5分間振とうし、上清を除去した。この操作を3回繰り返した後に、HRP標識ストレプトアビジンを0.33μg/mLに希釈した1%BSA−PBST内で上記PVDF膜を室温30分間で振とうした。次いで、PBST内で上記PVDF膜を室温5分間振とうし、上清を除去した。この操作を3回繰り返した後に、0.2mg/mLのジアミノベンジジン及び0.0012%の過酸化水素を含む50mM Tris塩酸緩衝液(pH7.4)に浸し、バンドが目視で十分確認できた後にPVDF膜をRO水で洗浄した。
結果を図9に示す。既存の抗AFP抗体で免疫沈降を行い、抗AFP抗体で検出した場合(左レーン)は、酵素処理の有無に関わらずバンドが検出された(左レーン中の−及び+レーン)。更に、酵素処理をした場合はバンドが高分子量側と低分子量側で二重になっていることが確認された(左レーン中の+レーン)。一方で、S20205R抗体(中央左レーン)またはS20206R抗体(中央右レーン)で免疫沈降を行い、抗AFP抗体で検出した場合は、酵素処理した場合(中央左、中央右レーン中それぞれの+レーン)でさらに低分子量側のバンドのみが検出された(図9)。この測定系では、免疫沈降に用いた抗体が結合するAFPのバンドが検出される。つまり、〔試験例1〕に示す通り、酵素処理によって低分子量側のバンドが増大していることと合わせて、S20205R抗体またはS20206R抗体は酵素処理によって生じる糖分解AFPを認識していることを示している。なお、AFPと反応しない抗体(右レーン)では、実質的なバンドは検出されなかった。
1.試験方法
S20205R抗体、S20206R抗体の特異性を、上述の抗原固相化ELISA法と同様の方法で確認した。なお、プレートに固相した抗原はAFP、Endo−F3で処理したAFP、酵素PNGaseF(図3の配列中、アミノ基とGlcNAcβ1間の結合を切断する酵素)で処理したAFPである。同時に各AFPのSDS−PAGEも〔試験例1〕と同様に行った。
結果を図10、図11に示す。SDS−PAGEでは、酵素未処理のAFP、Endo−F3処理AFP、PNGaseF処理AFPの何れも異なるバンドパターンを示し、Endo−F3処理したAFPが高分子量側と低分子量側の二重のバンドとなるのに対し、PNGaseFしたAFPでは低分子量側のバンドが確認された(図10)。Endo−F3はAFPの一部の糖鎖を2糖まで切断したのに対して、PNGaseFはあらゆるN型糖鎖を消化切断し、糖鎖の無いAFPが産生されたことを示している。一方図11に示すように、既存のAFP抗体が酵素処理の有無に関わらず吸光度が増大したのに対し、S20205R抗体、S20206R抗体では吸光度がEndo−F3で処理したAFPに対してのみ増大した。この測定系ではプレートに固相化されたAFPに対する抗体の反応性が強いほど吸光度が増大する。つまり、S20205R抗体、S20206R抗体は、Endo−F3によって生じる糖鎖を認識部位の一部として含むことを示している。
これまではプレート上に固相化した抗原に対する反応性を評価していたが、液相中に存在する抗原に対する反応性の評価も行った。試験方法は以下の通りである。
(1)サンドウィッチELISA用プレートの作製
精製AFPを免疫原として作製し、酵素処理の有無に関わらず反応する既存の抗AFPモノクローナル抗体(社内品)含有液を5μg/mLになるよう、150mM塩化ナトリウムを含む20mMリン酸緩衝液(pH7.2;以下、PBSという)に溶解し、該溶解液50μLをそれぞれ96穴マイクロプレートの各ウエルに分注して、室温で2時間静置した。
前記各ウエルを0.05%Tween(登録商標)20を含むPBS(以下、PBSTという)400μLで3回洗浄した後、1%牛血清アルブミンを含むPBST(以下、BSA−PBSTという)100μLを加え、室温で1時間ブロッキングを行った。これをサンドウィッチELISA用プレートとした。
〔試験例1〕と同様にEndo−F3処理したAFPをBSA−PBSTで段階希釈した。これを抗原含有液とする。前記サンドウィッチELISA用プレートの各ウエルから液を除去した後に、抗原含有液を前記各ウエルに50μL添加し、室温で15分間静置した。前記各ウエルをPBST400μLで3回洗浄した後、BSA−PBSTで1500倍希釈したHRP標識S20205R抗体又はHRP標識S20206R抗体を前記プレートに50μL各ウエルに分注し、室温で15分間静置した。前記各ウエルをPBST400μLで3回洗浄した後、0.2%オルトフェニレンジアミン及び0.02%過酸化水素を含むクエン酸緩衝液(pH5.0)50μLを加え、室温で10分間放置後、1.5N硫酸50μLを加えて酵素反応を停止させ、波長492nmにおける吸光度を測定した。
結果を図12a,bに示す。酵素処理したAFPに対して濃度依存的に吸光度が上昇している(図12a,b)。この測定系ではプレートに固相化された既存抗AFP抗体と結合した液相中酵素処理AFPに、HRP標識S20205R抗体及びHRP標識S20206R抗体が反応することで吸光度が増大する。つまり、これらの抗体が液相中の酵素処理AFPと特異的に反応することを示す。
これまでの結果よりS20205R抗体とS20206R抗体の特性はほとんど同一であったため、以降の評価ではS20206R抗体のみを用いている。
1.試験方法
試験方法は〔試験例7〕と同様である。ただし抗原は〔試験例1〕と同様に調製したEndo−F2あるいはEndo−F3で処理したAFPを用いた。また、〔試験例7〕でHRP標識抗体を添加する工程において、BSA−PBSTで6000倍希釈したHRP標識した既存抗AFPポリクローナル抗体も同時に試験に用いた。
結果を図13a,bに示す。既存のAFP抗体同士で測定した場合には、酵素処理の有無に関わらず、AFP濃度依存的に吸光度が増大した(図13a)。一方で、S20206R抗体を用いた場合にはEndo−F2及びEndo−F3で処理したAFPのみ、濃度依存的に吸光度が増大した(図13b)。つまり、Endo−F2を用いてもEndo−F3によって生じるものと同様の糖分解AFPが生じており、Endo−F3以外の酵素を用いた場合でもこれまでの試験例と同様の測定が可能であることを示している。
これまでは精製したAFPに酵素や緩衝液を添加して、酵素処理を行っていた。しかし、実際に臨床検査で想定される測定用検体は血清であり、血清にはAFP以外に非常に高濃度のN型糖鎖タンパク質が混在していることが予想される。そこで、血清中でも酵素がAFPに作用するかどうかを試験した。
試験方法は〔試験例7〕と同様である。ただし、用いた抗原はHepG2由来のAFP200ngに3例の健常者血清を25μL加え、更に0.1M MES緩衝液(pH5.5)25μL及び2μgのEndo−F3を添加した。これらを37℃で10分間反応させた後に、BSA−PBSTを1mLmL加え、AFP濃度を200ng/mLとしたものを抗原含有液とした。
結果を図14に示す。AFPを健常者血清に添加した場合も、緩衝液に添加した場合と同様に、本発明によるサンドウィッチELISAで、酵素処理AFP特異的に吸光度が増大した(図14)。つまり、血清中においてもAFPに対してEndo−F3が作用していることを示している。
1.試験方法
これまでは緩衝液として0.1M MES緩衝液(pH5.5)を主に用いていたが、他の緩衝液(pH)でも使用可能かを検討した。試験方法は〔試験例7〕と同様である。ただし、酵素処理時に用いた緩衝液を表1に示す。
酵素処理後はAFP濃度が100ng/mLとなるようにBSA−PBSTで希釈し、サンドウィッチELISAに用いた。
結果を図15に示す。pH0.5程度からpH12.5程度の領域で吸光度が0.1以上となっており、反応していると言える(図15)。これは分子生物学で用いる通常の緩衝液のpH領域を十分に満たしており、本発明に用いる酵素はpHによらず使用できることを示している。
これまではAFPを酵素処理後にBSA−PBSTで希釈した後に、サンドウィッチELISAで測定を行っていた。そこで、サンドウィッチELISAにおける抗原抗体反応と同時にAFPの酵素処理をすることが可能か検討した。
プレートの調製までは〔試験例7〕と同様である。
(1)オンプレート酵素処理
Endo−F3を0.1M MES緩衝液(pH5.5)により0.33mg/mLに希釈し、これを酵素含有液とした。またAFPを含むHepG2培養上清を0.1M MES緩衝液(pH5.5)により段階希釈し、これを抗原含有液とした。
前記ELISA用プレートの各ウエルから液を除去した後に、酵素含有液を前記各ウエルに30μL添加した。次いで、前記各ウエルに抗原含有液を30μL添加し、37℃で20分間反応させた。前記各ウエルをPBST400μLで3回洗浄した後、BSA−PBSTで1500倍希釈したHRP標識S20206R抗体を前記プレートに50μL各ウエルに分注し、室温で15分間静置した。前記各ウエルをPBST400μLで3回洗浄した後、0.2%オルトフェニレンジアミン及び0.02%過酸化水素を含むクエン酸緩衝液(pH5.0)50μLを加え、室温で10分間放置後、1.5N硫酸50μLを加えて酵素反応を停止させ、波長492nmにおける吸光度を測定した。
結果を図16に示す。これまでの結果と同様に酵素を添加した場合で、濃度依存的に吸光度が増大した(図16)。本測定系においては、固相化した抗体と抗原が反応するのと同時に、抗原と酵素が反応している。つまり、抗原抗体反応と抗原の酵素処理反応は同時に行うことができると示している。
Claims (17)
- 糖タンパク質を抗原として用いる際の糖タンパク質の処理方法であって、
糖鎖特異的な糖鎖切断酵素によって糖鎖を切断することを含む、
糖タンパク質の処理方法。 - 前記糖鎖を切断する工程が、
当該切断によって糖鎖に内在しているエピトープを選択的結合性物質によって認識可能な状態にするものである、
請求項1に記載の糖タンパク質の処理方法。 - 前記糖鎖特異的な糖鎖切断酵素が、Endo−A、Endo−M、Endo−H、Endo−D、Endo−F1、Endo−F2、Endo−F3から選択される、請求項1または2に記載の糖タンパク質の処理方法。
- 前記糖タンパク質がAFPである、請求項1〜3のいずれか一項に記載の抗原の処理方法。
- 請求項1〜4のいずれか一項に記載の糖タンパク質の処理方法を使用する、選択的結合性物質の製造方法。
- 請求項1〜4のいずれか一項に記載の糖タンパク質の処理方法を使用する、生体試料中の糖タンパク質の検出方法。
- 請求項1〜4のいずれか一項に記載の糖タンパク質の処理方法で処理された糖タンパク質。
- 請求項1〜4のいずれか一項に記載の抗原の処理方法により処理された糖タンパク質に結合し、当該処理を受けていない同一の糖タンパク質に結合しない選択的結合性物質。
- 抗体または抗体の抗原結合フラグメントである、請求項8に記載の選択的結合性物質。
- 請求項1〜4のいずれか一項に記載の抗原の処理方法により処理された糖タンパク質を抗原として用いることを含む、請求項8または9に記載の選択的結合性物質の製造方法。

該糖タンパク質を含む可能性のある試料を糖鎖切断酵素によって処理する工程、
前記糖鎖切断酵素によって処理された糖タンパク質に、請求項8に記載の選択的結合性物質を作用させて、該糖タンパク質と該選択的結合性物質との反応物を得る工程、及び、
前記反応物を検出する工程
を含む、前記糖タンパク質の検出方法。 - 請求項12に記載の方法であって、糖タンパク質を、糖鎖特異的な糖鎖切断酵素によって処理する工程と、前記糖タンパク質と選択的結合性物質との反応物を得る工程を、同時に行う前記方法。
- 請求項12または13に記載の方法に使用するための試薬キットであって、糖タンパク質を、糖鎖特異的な糖鎖切断酵素によって処理するための試薬と、前記糖タンパク質と選択的結合性物質との反応物を得るための試薬を、同一の組成物に含有する、前記試薬キット。
- 生体内に存在しないか存在しても極めて稀な糖鎖構造を有する抗原を用いた抗体の製造方法。
- 前記抗原が、糖鎖特異的な糖鎖切断酵素によって処理された糖タンパク質である請求項15に記載の方法。
