Abstract
Description
1.以下の工程を含む、多能性幹細胞から小腸上皮様細胞への分化誘導方法;
1)多能性幹細胞を内胚葉細胞に分化誘導する工程;
2)前記分化誘導により得られた内胚葉細胞をALK5阻害物質、Wnt3a及びEGFから選択されるいずれか一種又は複数種の物質を含む系で培養する工程。
2.前記工程1)又は2)の工程のいずれかにおいて、さらにCDX2遺伝子を導入する工程を含む、前項1に記載の多能性幹細胞から小腸上皮様細胞への分化誘導方法。
3.前記工程1)又は2)の工程のいずれかにおいて、さらにFOXA2遺伝子を導入する工程を含む、前項1又は2に記載の多能性幹細胞から小腸上皮様細胞への分化誘導方法。
4.前記工程2)において、分化誘導により得られた内胚葉細胞を、BIO及び/又はDAPTを含む系で培養する、前項1〜3のいずれかに記載の分化誘導方法。
5.前記工程2)のBIO及び/又はDAPTを含む系で培養する工程の後、基底膜マトリックスを培養細胞に重層する処理工程を含む、前項4に記載の分化誘導方法。
6.前項1〜5のいずれかに記載の分化誘導方法により得られた小腸上皮様細胞。
7.多能性幹細胞から人為的に分化誘導処理を行うことによって得られた小腸上皮様細胞であって、当該小腸上皮様細胞が、薬物代謝酵素及び/又は薬物トランスポーターを発現していることを特徴とする、小腸上皮様細胞。
8.前項1〜5のいずれかに記載の分化誘導方法が施され、培養された培養物。
9.前項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物毒性評価方法及び/又は薬物動態評価方法。
10.前項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物-薬物間相互作用の検査方法。
11.前項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物代謝酵素誘導試験方法。
1)多能性幹細胞を内胚葉細胞に分化誘導する工程;
2)前記分化誘導により得られた内胚葉細胞をALK5阻害物質(SB431542)、Wnt3a及びEGFから選択されるいずれか一種又は複数種の物質を含む系で培養する工程。
3)また、前記1)又は2)の工程のいずれかにおいて、CDX2遺伝子及び/又はFOXA2遺伝子を導入する工程を含んでいても良い。これにより、80%以上の効率で小腸上皮様細胞を作製することができる。
4)さらに、前記2)の工程において、基底膜マトリックスを重層する工程を含んでいても良い。これにより、小腸上皮マーカーの遺伝子発現量を向上させることができる。
(A)ヒトES/iPS細胞未分化維持培地としては、ReproStem(商品名)、iPSellon(商品名)、Essential 8(商品名)、TeSR-E8(商品名)などの各種幹細胞維持培地にbFGFなどを添加して使用することができる。
(B)分化誘導用培地として、ES細胞培養用基本培地であるhESF-GRO(商品名、Cell Science & Technology Institute社)に、インスリン(10μg/ml) 、トランスフェリン(5μg/ml)、2-メルカプトエタノール(10μM)、2-エタノールアミン(10μM) 、亜セレン酸ナトリウム(20 nM)及びウシ血清アルブミン(BSA: 1 mg/ml)を含む培地を使用することができる。分化誘導用培地の他の態様として、ES細胞分化誘導用基本培地であるhESF-DIF(商品名、Cell Science & Technology Institute社)に、インスリン(10μg/ml)、トランスフェリン(5μg/ml)、2-メルカプトエタノール(10μM)、2-エタノールアミン(10μM)、亜セレン酸ナトリウム(20 nM)を含む培地 (FASEB J. 23:114-22 (2009))を使用することができる。
(C)分化誘導用培地の他の態様として、RPMI1640培地(Sigma社)に4 mM L-Glutamine、B27 Supplement(Invitrogen社)、ペニシリン/ストレプトマイシンを含む培地も使用することができる。内胚葉細胞を分化誘導する際に使用する培地は同等の機能を発揮しうるものであれば、上記に限定されない。
(D)内胚葉細胞以降の分化誘導にはdifferentiation DMEM-high Glucose培地、10% Knock Serum Replacement (Invitrogen)、1 % Non Essential Amino Acid Solution (Invitrogen)、ペニシリン/ストレプトマイシン、2 mM L-Glutamine、100 μmol/l β-メルカプトエタノールを含むdifferentiation DMEM-high Glucose培地 (Invitrogen))を使用することができる。
本実施例で示す培養方法では、ヒトiPS細胞又はヒトES細胞に対する各種培地が必要である。本参考例では、各種培養に使用可能な培養液の組成について説明する。
(A)ヒトES/iPS細胞未分化維持培地としては、ReproStem(商品名)、iPSellon(商品名)、Essential 8(商品名)、TeSR-E8(商品名)などの各種幹細胞維持培地にbFGFを添加して使用することができる。以後、当該培地を「培地1」という。
(B)分化誘導用培地として、ES細胞培養用基本培地であるhESF-GRO(商品名、Cell Science & Technology Institute社)に、インスリン(10μg/ml) 、トランスフェリン(5μg/ml)、2-メルカプトエタノール(10μM)、2-エタノールアミン(10μM) 、亜セレン酸ナトリウム(20 nM)及びBSA(1 mg/ml)を含む培地を使用することができる。分化誘導用培地の他の態様として、ES細胞分化誘導用基本培地であるhESF-DIF(商品名、Cell Science & Technology Institute社)に、インスリン(10μg/ml)、トランスフェリン(5μg/ml)、2-メルカプトエタノール(10μM)、2-エタノールアミン(10μM)、亜セレン酸ナトリウム(20 nM)を含む培地 (FASEB J. 23:114-22 (2009))も使用することができる。

(D)内胚葉細胞以降の分化誘導にはdifferentiation DMEM-high Glucose 培地(10% Knock Serum Replacement(Invitrogen)、1 % Non Essential Amino Acid Solution(Invitrogen)、ペニシリン/ストレプトマイシン、2 mM L-Glutamine、100 μmol/l β-メルカプトエタノールを含むdifferentiation DMEM-high Glucose培地 (Invitrogen))を使用することができる。以後、当該「differentiation DMEM-high Glucose 培地」を「培地3 」という。
本実施例では、以降の比較例及び各実施例で使用される内胚葉細胞の作製について説明する。内胚葉細胞は、多能性幹細胞であるヒトiPS細胞又はヒトES細胞を用いて作製した。本実施例では、ヒトiPS細胞株としてTic(JCRB1331)を用いた。ヒトES細胞株(KhES3)は、文部科学省のガイドラインに従って扱い、実験は倫理委員会の承認を経て行った。上記多能性幹細胞は、フィーダー細胞とし、Tiss. Cult. Res. Commun., 27: 139-147 (2008) に記載の方法に従い、上記培地1を用いて培養した。
本比較例では、ヒトiPS細胞株(Tic)又はヒトES細胞株(K3)から上記実施例1に示す方法で作製した内胚葉細胞に、BIO(5μM)及びDAPT(10μM)を培養系に加え、培地3を用いて20日間培養した(図1A)。本比較例の方法は、非特許文献3に示す方法に準じる。
本実施例では、小腸上皮細胞マーカーであるANPEP及びVILLINを指標とし、比較例の分化誘導方法に改良を加えた分化誘導方法を試みた。
本実施例では、小腸上皮細胞マーカーであるANPEPを指標とし、培養期間を変えた場合の分化誘導効果を確認した。
本実施例では、小腸上皮細胞マーカーであるVILLINを指標とし、SB431542、EGF及びWnt3aの作用の有無、培養期間の変更が分化誘導効率に及ぼす影響を確認した。
本実施例では、HiPS-ELCについてトランスポーター(PEPT1)の遺伝子発現を確認し、HiPS-ELCの薬物代謝能を確認した。特にPEPT1、P-gp、BCRPは小腸の薬物輸送において重要な役割を果たす輸送タンパクである。
小腸上皮細胞は細胞同士で強固に結びつき、タイトジャンクションを形成することが知られている。本実施例では、HiPS-ELCのタイトジャンクション機能を確認した。本実施例では、実施例5と同様に、ヒトiPS細胞から作製した内胚葉細胞を、BIO(5μM)及びDAPT(10μM)を含む系で培地3を用いて15日間培養し、さらにBIO(5μM)、DAPT(10μM)並びにSB431542(10μM)、EGF(250 ng/ml)及びWnt3aを培養系に加えた。そして、さらに15日間(培養開始から19〜34日目)培養し、分化誘導により本実施例のHiPS-ELCを作製した(図7A)。
本実施例では、HiPS-ELCのCYP3A4遺伝子の発現を確認した。ヒトiPS細胞を実施例6−1と同手法により処理した(図8A)。
(A)本実施例では、HiPS-ELCのCYP3A4遺伝子の発現を確認した。ヒトiPS細胞を実施例6−1と同手法により処理した(図9A)。
以上のことから、HiPS-ELCはVD3やRIF等の薬物によるCYP3A4誘導能を有していることが示唆された。
本実施例ではヒトiPS細胞から小腸上皮様細胞への分化を促進するために、分化途中の細胞に対して分化に関与する転写因子を遺伝子導入した。
Adベクターの作製はin vitro ライゲーション法により行った。シャトルプラスミドpHMEF5のマルチクローニング部位に、FOXA2遺伝子又はCDX2遺伝子を挿入した、FOXA2遺伝子又はCDX2遺伝子発現シャトルプラスミドを作製した。導入する遺伝子は、FOXA2遺伝子では例えばGenBank Accession No. NM_021784に登録されているものを、CDX2遺伝子では例えばGenBank Accession No. NM_001265に登録されているものを用いた。
本実施例では、ヒトiPS細胞から小腸上皮様細胞への分化効率を解析した。
(A)ヒトiPS細胞を実施例7(A)と同手法により処理した(図11A)。
(B)小腸上皮様細胞への分化効率を評価するために、分化誘導35日目にVILLIN陽性率をFACSを用いて測定した。FOXA2遺伝子、CDX2遺伝子を導入することによって、VILLIN陽性率は38%から96%に上昇した。以上のことから、FOXA2遺伝子、CDX2遺伝子を導入することによって、ヒトiPS細胞から小腸上皮様細胞への分化効率が上昇したことが確かめられた(図11B)。
本実施例では遺伝子導入して作製した小腸上皮様細胞におけるCYP3A4、P-gp誘導能の評価を行った。
(A)ヒトiPS細胞を実施例7(A)と同手法により処理した(図12A)。
(B)CYP3A4、P-gp誘導剤としてVD3を使用した。VD3を作用することにより、HiPS-ELC-LacZでは約100倍CYP3A4が上昇したが、HiPS-ELC-TFsでは約10倍程度の上昇であった。一方、HiPS-ELC-TFsにおけるP-gp誘導能はHiPS-ELC-LacZよりも高かった(図12B)。(FOXA2遺伝子、CDX2遺伝子導入細胞=HiPS-ELC-TFs、コントロール細胞=HiPS-ELC-LacZ)
(C)CYP3A4、P-gp誘導剤としてRIFを使用した。RIFを作用することにより、HiPS-ELC-LacZでは約7倍CYP3A4が上昇したが、HiPS-ELC-TFsでは約3倍程度の上昇であった。一方、HiPS-ELC-TFsにおけるP-gp誘導能はHiPS-ELC-LacZよりも高かった(図12C)。(FOXA2遺伝子、CDX2遺伝子導入細胞=HiPS-ELC-TFs、コントロール細胞=HiPS-ELC-LacZ)
以上のことから、FOXA2遺伝子、CDX2遺伝子を導入することによって、CYP3A4誘導能は低下するものの、P-gp誘導能は上昇することが示唆された。
本実施例では、本発明の小腸上皮様細胞(HiPS-ELC-TFs:FOXA2, CDX2遺伝子導入済みのヒトiPS細胞由来小腸上皮様細胞)について、RIF(20μM/ 0.1% DMSO)を48時間作用させたときのCYP3A4遺伝子発現量を測定した。小腸上皮様細胞(HiPS-ELC-TFs)は、実施例7と同手法により作製した。コントロール細胞としてCaco-2細胞について同様に測定した。遺伝子の発現量はリアルタイムRT-PCR法により測定した。
本実施例では、本発明の小腸上皮様細胞(HiPS-ELC-TFs)の薬物刺激により誘導されたCYP3A4について、薬物代謝に及ぼす影響を確認した。CYP3A4によって代謝される薬物として、代謝により肝毒性を示すAMを用いた。実施例10と同様に、HiPS-ELC-TFsは、実施例7と同手法により作製し、コントロールとしてCaco-2細胞を用いた。HiPS-ELC-TFs又はCaco-2細胞に、RIF(20μM/ 0.1% DMSO)を48時間作用させたのち、AM(25μM、50μM、又は100μM)を培地に添加した。AMの添加と同時に、HiPS-ELC-TFs又はCaco-2細胞と、ヒトiPS細胞由来肝細胞(HiPS-HLC)の共培養を開始した。48時間後、HiPS-HLCの細胞生存率を測定した(図14)。細胞生存率はWST-8 assay kit(Dojindo)を用いた。細胞生存率はDMSO作用群を100%とした。なお、DMSOの使用濃度は0.1%以下にした。
本実施例では、本発明の小腸上皮様細胞(HiPS-ELC-TFs)について、CYP3A4阻害する物質と薬物代謝に及ぼす影響を確認した。薬剤を服用する際に、例えば食物含有成分によって当該薬剤の薬物動態に変化が生じることがある。そこで、本実施例では、CYP3A4活性を阻害することが知られているGFJの成分の一つであるクマリン類(6',7'-dihydroxybergamottinとbergamottin)を含む系でのAMの薬物代謝に及ぼす影響を確認した。
本実施例では、本発明の小腸上皮様細胞(HiPS-ELC-TFs)のタイトジャンクション機能を確認した。小腸上皮細胞は細胞同士で強固に結びつき、タイトジャンクションを形成することが知られている。タイトジャンクション機能は、実施例6と同手法により、TEERを測定することで確認した。また、FD4(Fluorescein isothiocyanate-dextran:平均分子量4,000 )透過試験も実施した。小腸上皮様細胞(HiPS-ELC-TFs)の TEER は約430 Ω・cm2であった(図18A)。タイトジャンクションの開口剤であるカプリン酸(C10)を作用させることにより、HiPS-ELC-TFsのTEER値はほぼ0 Ω・cm2になった(図18A)。また、FD4のPapp (透過係数)は、約 0.4 × 10-6 (cm/sec) であった(図18B)。FD4のPapp値はC10の作用により大幅に上昇した(図18B)。以上の結果から、HiPS-ELC-TFsはタイトジャンクション機能を有し、バリア能を有していることが示唆された。

P-gpは、消化管粘膜、腎尿細管上皮細胞、脳血管内皮細胞(血液脳関門)などで、異物、薬物などを細胞外へ排出するABCトランスポータファミリーの一つである。薬物の生物学的利用能や薬物の標的組織への分布に影響を与える。本実施例では、本発明の小腸上皮様細胞(HiPS-ELC-TFs)のP-gpの機能を評価するため、Rhodamine 123(作用濃度は10 μM)を用いた薬物透過試験を実施した。Rhodamine 123はP-gpの基質であることが知られている。なお、P-gpは頂側膜(apical)側に発現するトランスポーターであるため、P-gpが機能していればapical側におけるRhodamine 123の蓄積が確認できるようになる。実際に、HiPS-ELC-TFsにおける基底側(basolateral)(A to B)へのRhodamine 123の透過係数はapical側(B to A)への透過係数よりも有意に小さいことが分かった(ER値は2.1)。したがって、HiPS-ELC-TFsにおいてP-gpが機能していることが示唆された。さらに、P-gpをcyclosporin A (CysA、作用濃度は10 μM)により阻害することによって、B to Aへの有意なRhodamine 123の透過が確認できなくなった(ER値は0.9)。この結果より、HiPS-ELC-TFsにおいて確認されていたB to AへのRhodamine 123の輸送はP-gpを介したものであることが示唆された(図19)。
本実施例では、マトリゲルを重層した系での小腸上皮様細胞への分化誘導法について説明する。本実施例では、ヒトiPS細胞株としてTic(JCRB1331)を用い、実施例1に示す方法で培地1を用いて培養した。ヒトiPS細胞株(Tic)の培養系に、Activin Aを100 ng/ml加えて4日間培養し、分化誘導処理を行い内胚葉細胞を作製した。作製した内胚葉細胞を、さらに培地3を用いて培養した。分化誘導開始4日目から8日目の間に、BIO(5μM)及びDAPT(10μM)を培養系に加え、その後BIO(1μM)及びDAPT(2.5μM)を培養系に加えた。さらにその後25日目まで、BIO(0.1μM)、DAPT(1μM)SB431542(2μM)、EGF(250 ng/mL)及びWnt3aを加えた培地3を用いて更に5日間培養した。その後、EGF(250 ng/ml)を含む培地3を用い、50μg/cm2のマトリゲルを重層したのちに、5日間培養した(図20)。以降の実施例において、本実施例においてマトリゲル重層処理をせずに作製した小腸上皮様細胞はHiPS-ELC-Cといい、マトリゲルを含む系で作製した小腸上皮様細胞をHiPS-ELC-MGということとする。
上記実施例15で作製した小腸上皮様細胞(HiPS-ELC-C、HiPS-ELC-MG)及び各種細胞について、小腸関連遺伝子発現の解析を行った。各遺伝子の発現は、iPSCs(未分化ヒトiPS細胞)、HiPS-ELC-C、HiPS-ELC-MG、LS180細胞(ヒト結腸腺癌由来細胞)、Caco-2細胞(ヒト結腸癌由来細胞)、及びAIについて解析した。各遺伝子の発現は、各々SYBR Green gene expression assays(Applied Biosystems)を用いた定量的リアルタイムRT-PCR法により測定した。
上記実施例15で作製したHiPS-ELC-MGについて、小腸関連遺伝子であるSI、VILLIN及びCDX2各遺伝子発現割合をFACS解析により測定した。その結果、HiPS-ELC-MGではSI陽性細胞が25.3%、VILLIN陽性細胞が55.6%及びCDX2陽性細胞が90.1%であった。これによりHiPS-ELC-MGは小腸上皮様細胞であることが確認された(図22)。
上記実施例15で作製したHiPS-ELC-MGについてCYP3A4発現量を測定し、各種CYP3A4誘導剤によるCYP3A4誘導能を評価した。CYP3A4発現量は実施例10と同手法により測定した。CYP3A4誘導剤として、デキサメタゾン(dexamethasone:DEX)、フェノバルビタール(phenobarbital:PB)、RIF、VD3を用いた。比較細胞として、Caco-2細胞やLS180細胞についても同様に検討した。HiPS-ELC-MGでは、いずれのCYP3A4誘導剤を用いた場合においても、有意なCYP3A4の発現誘導が確認された。一方、Caco-2細胞ではVD3を用いた場合のみCYP3A4誘導が確認され、LS180細胞ではDEXによるCYP3A4誘導が確認されなかった。以上のことから、ヒトiPS細胞由来小腸上皮様細胞はCaco-2細胞やLS180細胞よりもCYP3A4誘導の評価に適したモデルであることが示唆された(図23)。
CYP3A4誘導において核内受容体が重要な役割を担うことが知られている。上記実施例15で作製した小腸上皮様細胞(HiPS-ELC-MG)及び各種細胞について、小腸核内受容体の遺伝子発現解析を行った。小腸核内受容体として、フェノバルビタール(PB)やRIFによるCYP3A4発現誘導のために必須のプレグナンX受容体(PXR)、DEXによるCYP3A4発現誘導のために必須の糖質コルチコイド受容体(GR)及びVD3によるCYP3A4発現誘導のために必須のビタミンD受容体(VDR)について確認した。各遺伝子の発現は、iPSCs(未分化ヒトiPS細胞)、HiPS-ELC、LS180細胞、Caco-2細胞及びAIについて解析した。各遺伝子の発現は、各々SYBR Green gene expression assays(Applied Biosystems)を用いた定量的リアルタイムRT-PCR法により測定した。上記の結果、PXRはHiPS-ELC-MGとLS180細胞においてSmall Intestineの約半分程度発現していた。GRはHiPS-ELC-MGのみで強く発現していた。VDRはHiPS-ELC-MG、LS180細胞、Caco-2細胞のいずれにおいても弱いながらも発現していることが確認できた(図24)。このことから、HiPS-ELC-MGはLS180細胞やCaco-2細胞よりもAI組織に近い核内受容体の発現パターンを有していることが示唆される。
Claims (11)
- 以下の工程を含む、多能性幹細胞から小腸上皮様細胞への分化誘導方法;
1)多能性幹細胞を内胚葉細胞に分化誘導する工程;
2)前記分化誘導により得られた内胚葉細胞をALK5阻害物質、Wnt3a及びEGFから選択されるいずれか一種又は複数種の物質を含む系で培養する工程。 - 前記工程1)又は2)の工程のいずれかにおいて、さらにCDX2遺伝子を導入する工程を含む、請求項1に記載の多能性幹細胞から小腸上皮様細胞への分化誘導方法。
- 前記工程1)又は2)の工程のいずれかにおいて、さらにFOXA2遺伝子を導入する工程を含む、請求項1又は2に記載の多能性幹細胞から小腸上皮様細胞への分化誘導方法。
- 前記工程2)において、分化誘導により得られた内胚葉細胞を、BIO(GSK-3 Inhibitor IX)及び/又はDAPT(γ-secretase inhibitor)を含む系で培養する、請求項1〜3のいずれかに記載の分化誘導方法。
- 前記工程2)のBIO(GSK-3 Inhibitor IX)及び/又はDAPT(γ-secretase inhibitor)を含む系で培養する工程の後、基底膜マトリックスを培養細胞に重層する処理工程を含む、請求項4に記載の分化誘導方法。
- 請求項1〜5のいずれかに記載の分化誘導方法により得られた小腸上皮様細胞。
- 多能性幹細胞から人為的に分化誘導処理を行うことによって得られた小腸上皮様細胞であって、当該小腸上皮様細胞が、薬物代謝酵素及び/又は薬物トランスポーターを発現していることを特徴とする、小腸上皮様細胞。
- 請求項1〜5のいずれかに記載の分化誘導方法が施され、培養された培養物。
- 請求項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物毒性評価方法及び/又は薬物動態評価方法。
- 請求項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物-薬物間相互作用の検査方法。
- 請求項6又は7に記載の小腸上皮様細胞を用いることを特徴とする、薬物代謝酵素誘導試験方法。
