Abstract
Description
本願は、2014年8月28日に出願された米国特許仮出願62/043,149号に基づき優先権を主張し、その内容をここに援用する。
フリーフロー電気泳動法は、分離槽内で試料を緩衝液とともに上流から下流に層流状に流しながら、分離槽の両脇に設けられた電極に電圧を加えることにより、分層槽内の緩衝液の流れに垂直方向に電界を印加して,自由ゾーン電気泳動を行い、試料中の成分の電気泳動度の違いにより分離する技術である。
なお、以下の実施の実施形態は、本発明の一態様を示すものであり、この発明を限定するものではなく、本発明の技術的思想の範囲内で任意に変更可能である。また、以下の図面においては、各構成をわかりやすくするために、実際の構造と各構造における縮尺や数等を異ならせている。
エクソソーム(エキソソーム)は、直径30〜100nm程度の脂質小胞であり、エンドソームと細胞膜との融合体として、腫瘍細胞、樹状細胞、T細胞、B細胞等、種々の細胞から、血液、尿、唾液等の体液中に分泌される。
生体内に存在する癌細胞等の異常細胞は、その細胞膜に特有のタンパク質を発現している。エクソソームは細胞の分泌物であり、その表面に分泌源の細胞由来のタンパク質を発現している。
電気泳動デバイスを用いたエクソソームの分析は、一例として次のようにして行うことができる。まず、検出対象のエクソソームを分離・精製する。次に、エクソソームと特異的結合物質とを接触させる。ここで、特異的結合物質とは、エクソソームの表面に存在する分子に特異的に結合することができる物質を意味する。次に、電気泳動デバイスを用いて、エクソソームのゼータ電位を計測し、分析を行う。本分析は、エクソソームに限らず、広く細胞外小胞体一般の分析にも適用できる。
特異的結合物質としては、例えば、抗体、改変抗体、アプタマー、リガンド分子等が挙げられる。抗体としては、IgG、IgA、IgD、IgE、IgM等が挙げられる。IgGとしては、IgG1、IgG2、IgG3、IgG4等が挙げられる。IgAとしては、IgA1、IgA2等が挙げられる。IgMとしては、IgM1、IgM2等が挙げられる。改変抗体としては、Fab、F(ab’)2、scFv等が挙げられる。アプタマーとしては、ペプチドアプタマー、核酸アプタマー等が挙げられる。リガンド分子としては、エクソソームの表面に存在する検出対象分子が、レセプタータンパク質である場合の、当該レセプタータンパク質のリガンド等が挙げられる。例えば、エクソソームの表面に存在する分子がインターロイキンである場合、リガンド分子としてはGタンパク質等が挙げられる。
本分析の各工程について説明する。まず、エクソソームを含有する試料から該エクソソームを精製する。試料としては、目的に応じて、血液、尿、母乳、気管支肺胞洗浄液、羊水、悪性滲出液、唾液、細胞培養液等が挙げられる。中でも、血液及び尿からは、エクソソームを精製しやすい。
次に、エクソソームと特異的結合物質(抗体、アプタマー等)とを接触させる。エクソソームの表面に検出対象の分子が存在した場合、特異的結合物質−エクソソーム複合体が形成される。特異的結合物質を適切に選択することにより、例えば、癌、肥満、糖尿病、神経変性疾患等の疾患に関連する異常を検出することができる。詳細については後述する。
一例として、特異的結合物質として抗体を使用した場合について説明する。エクソソームと抗体とを反応させた後、抗体と反応させたエクソソームのゼータ電位を計測する。ゼータ電位とは、溶液中の微粒子の表面電荷である。例えば、エクソソームが負に帯電しているのに対し、抗体は正に帯電している。このため、抗体−エクソソーム複合体のゼータ電位は、エクソソーム単独のゼータ電位と比較して正にシフトしている。したがって、抗体と反応させたエクソソームのゼータ電位を測定することによって、エクソソームの膜表面における抗原の発現を検出することができる。これは、抗体に限らず、正に帯電した特異的結合物質でも同様である。

式(1)中、Uは測定対象のエクソソームの電気泳動移動度、ε及びηは、それぞれ、サンプル溶液の誘電率及び粘性係数である。また、電気泳動移動度Uは、電気泳動速度Sをマイクロ流路内の電界強度で除して算出することができる。
図1は、細胞外小胞体分離装置100の概略的な構成図である。
細胞外小胞体分離装置100は、電気泳動デバイスDV、電圧調整部(調整部)40、顕微鏡50、試料供給系61、緩衝液供給系62、試料回収系70、第2緩衝液供給系80、第2流路回収系90および制御部CONTを備えている。
図2は、電気泳動デバイスDVの外観斜視図である。図3は、図2におけるA−A線視断面図である。なお、図2においては、理解を容易にするために、電気泳動デバイスDVの構成要素を実線で示している。
図4に示されるように、分離槽8の−X側端部には、緩衝液導入流路6および試料導入流路7が接続されている。試料導入流路7は、X方向に延び、一端が分離槽8のY方向の中央部に接続されている。試料導入流路7の深さは、分離槽8の深さと同一である。試料導入流路7の他端は、試料導入口5に接続されている。図7は、第1基板1を下面1a側から視た下面図である。試料導入口5は、上下方向に延びており、第1基板1および第2基板2に亘って貫通して形成されている。試料導入口5は、第1基板1に形成された貫通孔5aと、第2基板2に貫通孔5aとXY平面において同一位置に形成された貫通孔5bとを含む。試料導入口5は、上述した試料を試料導入流路7に導入するための部位である。
また、ゲル高分子の末端に基材と化学結合ができる官能基を有することが望ましい。末端の官能基としては、アミノ基、イソチオシアネート基、イソシアネート基、アシルアジド基、N-ヒドロキシスクシンイミドエステル基、スルフォニルクロライド基、アルデヒド基、エポキシド基、オキシラン基、カーボネート基、アリール基、イミドエステル基、カルボジイミド基、アンハイドライド基、チオール基、ハロアセチル基、アルキルハライド基、マレイミド基、アジリジン基、アクリロイル基、ジスルフィド基、ジアゾアルカン基、ジアゾアセチル基、カルボニルジイミダゾール基、、N, N'-ジスクシニミジルカーボネート基、N-ヒドロキシスクインイミジルクロロフォメート基、ヒドラジン基、ヒドラジド基などが挙げられる。
中でも、官能基の数が4以上のものが好ましく、4つ以上のポリエチレングリコール骨格の分岐鎖であって、末端に官能基を有するものを有するポリマー種がより好ましい。係るポリマー種としては、Tetra-PEGゲルが挙げられる。
次に、上述した電気泳動デバイスDVの製造方法について、図11および図12を参照して説明する。
電気泳動デバイスDVの製造方法は、第2基板2を準備する工程、第2基板2の下面2aにマスク材を設ける工程、マスク材を設けた第2基板2に対して第2流路30(電極槽9)にゲル化前のゲル材の溶液を導入する工程、ゲル材のゲル化後にマスク材を除去する工程、第2基板2に第1基板1および第3基板3を積層する工程を含む。
第2基板2を準備する工程は、第2基材2に対して分離槽8、電極槽9、緩衝液導入流路6、試料導入流路7、試料回収流路11、電極挿入口14および貫通孔4b、5b、10bを形成する工程を含む。第2基材2に対して分離槽8、電極槽9、緩衝液導入流路6、試料導入流路7、試料回収流路11、電極挿入口14および貫通孔4b、5b、10bを形成する方法は、切削加工や射出成形を採ることができる。
なお、第1基板1〜第3基板3を構成するポリメタクリルスチレンは、(アクリエース(登録商標)MS、日本アクリエース, Japan)を使用した。
また、Tetra-PEG ゲルは生体適合性を有するため、試料として生体物質の分離時に、生体物質のゲルへの吸着の抑制が期待される。
さらに、二液混合により容易に作製可能なため、デバイスの作製工程が複雑化されないという効果を奏する。
また、TN-PEGは、SUNBRIGHT(登録商標)ITE-100HS (日本油脂)を、最終濃度100mg/mLになるように0.2Mクエン酸リン酸ナトリウムバッファー pH=5.8に溶解した。
調整したTA-PEG溶液と、TN-PEG溶液とを1:1に混合すると、10分以内に混合液の粘度が上昇し、ゲル化反応が確認される。
次に、上記の細胞外小胞体分離装置100を用いて、細胞外小胞体を分離する方法について説明する。
ここでは、試料として、電荷を持たない蛍光色素であるローダミンB、負電荷を有するスルホローダミンBの分離実験を行った。
試料供給系61から試料導入口5に1mMローダミンBと1mMスルホローダミンBの混合液を導入した。その際、緩衝液供給系62から緩衝液導入口4に10mMリン酸緩衝液を導入し、ローダミンBとスルホローダミンBの混合液の流れを整えた。
電極15に対しては、0〜80Vの電圧を印加した。
第2流路30に対しては、第2緩衝液導入口12からリン酸緩衝液を導入するとともに、第2緩衝液回収口13からリン酸緩衝液を回収した。電極15に電圧を印加した際には電気分解によって、図9に示すように、気泡18が生じるが、ポンプ91の駆動により第2流路30内を吸引することにより、図10に示されるように、リン酸緩衝液とともに気泡18を回収・排除することができ、気泡18が分離槽8に混入して、試料の分離に悪影響を及ぼすことを防止できる。
特に、本実施形態では、隔壁17を構成するゲル材が第2基板2に化学的に強固に結合しているため、リン酸緩衝液の導入・流動により第2流路30内の液圧が加わった場合でも、分離槽8と第2流路30との間の物体(緩衝液および気泡18)の移動を遮断可能である。
顕微鏡50は、試料導入口5から+X側へ40mmの位置を蛍光観察し、顕微鏡用デジタルカメラにて撮像した。
顕微鏡50における励起波長は540〜580nm、蛍光波長は600〜660nmとした。
蛍光強度分布が正規分布に従うと仮定してピーク分離を行い、双方のピーク位置から泳動距離を算出し、以下の式から分離度Rを求めた。
図16および図17に示されるように、50V以下の印加電圧では、泳動距離および分離度の双方が線形関係になっていることが確認された。
この結果より、分離槽8に対して乱れのない電界が印加でき、50Vまでの電圧範囲に対して電気泳動デバイスDVが良好な分離動作が行えていることが確認できた。
この電気泳動度から、以下のスモルコフスキーの式からゼータ電位ξを算出した。
ξ=η×μ/(ε0 ×εr)
ここで、ξはゼータ電位、ηは粘性率、ε0は真空の誘電率、εrは比誘電率である。リン酸緩衝液の粘性率、比誘電率が25℃の水と同じ値であると仮定して、粘性率は、η=0.89× 10−3kg/m2s、比誘電率は、εr=78.5、真空の誘電率はε0=8.854×10−12m−3kg−1s4A2であるため、スルホローダミンBのゼータ電位はξ=20.5mV と算出された。
次に、上述した電気泳動デバイスDVの製造方法の変形例に ついて、図18乃至図21を参照して説明する。なお、上述した電気泳動デバイスDVの製造方法と同一の手順については、その説明を簡略化または省略する。

- 第1方向に延び、試料および緩衝液が流動する第1流路と、
前記第1流路の端部に設けられ前記試料が回収される試料回収部と、
前記第1流路の前記第1方向と直交する第2方向の両側にそれぞれ配置され前記第1流路に前記第2方向に沿って電圧を印加する電極と、
前記第1流路の前記第2方向の両側にそれぞれ連通し前記電極を収容するとともに、第2緩衝液が流動する第2流路と、
前記第1流路と前記第2流路との連通部に所定の接合強度で固定され前記第1流路と前記第2流路との間の物体の移動を遮断する隔壁と、
