Description
アミノ酸系分子抱合量=(アミノ酸系分子添加量)−(フィルター通過液中のアミノ酸系分子の量)
アミノ酸系分子抱合率(%)=アミノ酸系分子抱合量÷アミノ酸系分子添加量×100
塩基性アミノ酸(アルギニン、ヒスチジン、リジン)、酸性アミノ酸(アスパラギン酸、グルタミン酸)及び中性アミノ酸(アラニン、グリシン、ロイシン、バリン、イソロイシン、セリン、スレオニン、フェニルアラニン、トリプトファン、チロシン、シスチン、メチオニン、グルタミン、アスパラギン、プロリン)を用いて、各アミノ酸を抱合するシアノアクリレートポリマー粒子を製造した。

2.1 Dex-Amino acid抱合系粒子の抗菌活性(その1)
製造例1のDex-Amino acid抱合系で製造した粒子の各種菌株に対する抗菌活性(最小発育阻止濃度(MIC))を調べた。ポジティブコントロールとしてアンピシリン(ABPC)及びバンコマイシン(VCM)を使用した。測定方法は微量液体希釈法(NCCLS)に準拠した。すなわち、APBC及びVCMについては256μg/mlを原液(1倍希釈)とし、粒子については6.4mg/mlを原液(1倍希釈)として、それぞれ2倍希釈〜2048倍希釈までの12段階の希釈系列を調製して抗菌活性を評価した。96ウェルプレートにこれらの希釈系列を添加して、各ウェルに菌株を105 CFU/mlとなるように加えて35℃で18時間インキュベートし、目視にて濁りが観察された場合を菌株の発育ありとして、発育が認められなかった最低濃度をMICとした。結果を表3に示す。また、表3の結果を図1(黄色ブドウ球菌)及び図2(腸球菌)にグラフで示す。
製造例1のDex-Amino acid抱合系で製造した粒子の各種菌株に対する抗菌活性(最小発育阻止濃度(MIC))を調べた。ポジティブコントロールとしてアンピシリン(ABPC)及びレボフロキサシン(LVFX)を使用した。測定方法は上記2.1と同様に微量液体希釈法(NCCLS)に準拠した。結果を表4に示す。
(Dex-Amino acid抱合系とAmino acid単独抱合系の比較)
デキストランを使用しない製造例2のナノ粒子4種(Gly, Ala, His, Pro)について、上記と同様の方法で抗菌活性を評価し、製造例1のナノ粒子と活性を比較した。その結果を表5に示す。また、表5の結果を図3(黄色ブドウ球菌)及び図4(腸球菌)にグラフで示す。
製造例1で合成したアミノ酸抱合ナノ粒子(0.01HCl-Gly+Dex70)を処理したグラム陽性細菌の形態変化を電子顕微鏡により観察した。ナノ粒子による処理は以下の通りに行なった。
VREを感染させたマウス(マウス1匹に対し108cfu/0.5mlのVREを経口接種)において、アミノ酸抱合ナノ粒子の抗菌活性を確認した。VRE接種の3日前又は1時間後に、グリシン抱合ナノ粒子を1mg/mlの濃度でマウスに経口投与した。マウス体内でのVRE量は、糞便1gあたりのVRE菌数を計測することで確認した。その結果、感染前投与と感染後投与のいずれにおいても、コントロールの生理食塩水投与群マウスと比較して菌数が顕著に低下していた。これにより、アミノ酸抱合粒子がin vivoでも感染の予防効果及び治療効果があることが確認された。
(Dex-Amino acid抱合系とAmino acid単独抱合系の比較)
製造例1及び製造例2のアミノ酸抱合粒子で各種細胞株を処理し、MTT法による細胞増殖アッセイを行なうことにより、各種アミノ酸抱合粒子の細胞障害活性を評価した。アッセイにはMTTアッセイキット(ロシュ)を用いた。細胞株としては、H9(ヒト悪性リンパ腫)、HCT116(ヒト大腸癌)、MIAPaCa(ヒト膵臓癌)及びHepG2(ヒト肝臓癌)を用いた。
製造例1で合成したアミノ酸抱合ナノ粒子(0.01HCl-Gly+Dex70)を処理したH9細胞を電子顕微鏡により観察した。ナノ粒子による処理は上記6と同様に行なった。図7はナノ粒子添加2時間後のH9細胞のSEM像である。細胞は球形を保っており、細胞の周囲に遊離のナノ粒子と、細胞表面に接合したナノ粒子が確認された(図7A)。細胞表面を拡大して観察すると、ひだは退化し滑面化が見られ(図7B)、滑面化した部位には複数のナノ粒子の接合が認められた(図7C)。
(1) 正常マウス及びヌードマウスに対し、アミノ酸抱合ナノ粒子を1mg/mlの濃度で投与(経口投与、静注投与、腹腔投与、皮下投与)した。投与後6ヶ月まで観察しても、マウスに異常は認められず正常に生存していた。アミノ酸抱合ナノ粒子は正常細胞を障害しないことが確認された。
(2) ICR系6週齢雌マウス(体重20g〜24g)にグリシン抱合ナノ粒子50mg/bodyを経口投与、又は10mg/bodyを腹腔内投与、尾静脈内投与、皮下投与、若しくは皮膚塗布により単回投与し(各投与群5匹ずつ)、投与後8日間毎日体重変化及び排泄・行動の観察を行なった。対象として生理食塩水投与群と比較したところ、異常を認めなかった。
Claims (26)
- アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種が共存し、かつ、糖類及びポリソルベートのいずれも実質的に共存しない条件下において、シアノアクリレートモノマーをアニオン重合することを含む、シアノアクリレートポリマー粒子の製造方法。
- アミノ酸及びその誘導体並びにそれらのオリゴマーから成る群より選択される少なくとも1種が共存する条件下において前記アニオン重合を行なう請求項1記載の方法。
- アミノ酸が共存する条件下において前記アニオン重合を行なう請求項2記載の方法。
- 前記アミノ酸が、アルギニン、ヒスチジン、リジン、アスパラギン酸、グルタミン酸、アラニン、グリシン、ロイシン、バリン、イソロイシン、セリン、スレオニン、フェニルアラニン、トリプトファン、チロシン、シスチン又はシステイン、グルタミン、アスパラギン、プロリン、メチオニン、β−アラニン、γ−アミノ酪酸、カルニチン、γ−アミノレブリン酸、及びγ−アミノ吉草酸から成る群より選択される少なくとも1種である請求項1ないし3のいずれか1項に記載の方法。
- 前記誘導体が、クレアチン、オルニチン、サイロキシン、デスモシン、ヒドロキシプロリン、ヒドロキシリジン、ホスホセリン、テアニン、カイニン酸、トリコロミン酸、及びサルコシンから成る群より選択される少なくとも1種である請求項1又は2記載の方法。
- アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種を抱合し、平均粒径が1000nm未満であるシアノアクリレートポリマー粒子を有効成分として含有する細胞障害剤。
- アミノ酸及びその誘導体並びにそれらのオリゴマーから成る群より選択される少なくとも1種を抱合する前記粒子を有効成分とする請求項6記載の細胞障害剤。
- アミノ酸を抱合する前記粒子を有効成分とする請求項7記載の細胞障害剤。
- 前記アミノ酸が、アルギニン、ヒスチジン、リジン、アスパラギン酸、グルタミン酸、アラニン、グリシン、ロイシン、バリン、イソロイシン、セリン、スレオニン、フェニルアラニン、トリプトファン、チロシン、シスチン又はシステイン、グルタミン、アスパラギン、プロリン、メチオニン、β−アラニン、γ−アミノ酪酸、カルニチン、γ−アミノレブリン酸、及びγ−アミノ吉草酸から成る群より選択される少なくとも1種である請求項6ないし8のいずれか1項に記載の細胞障害剤。
- 前記アミノ酸誘導体が、クレアチン、オルニチン、サイロキシン、デスモシン、ヒドロキシプロリン、ヒドロキシリジン、ホスホセリン、テアニン、カイニン酸、トリコロミン酸、及びサルコシンから成る群より選択される少なくとも1種である請求項6又は7記載の細胞障害剤。
- グリシン、アスパラギン及びヒスチジンから成る群より選択される少なくとも1種を抱合するシアノアクリレートポリマー粒子を有効成分とする請求項8記載の細胞障害剤。
- 前記細胞が細菌である請求項6ないし11のいずれか1項に記載の細胞障害剤。
- 前記細胞がグラム陽性細菌である請求項12記載の細胞障害剤。
- 前記粒子が、アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種が共存し、かつ、糖類及びポリソルベートのいずれも実質的に共存しない条件下において、シアノアクリレートモノマーをアニオン重合させることにより製造された粒子である請求項6ないし13のいずれか1項に記載の細胞障害剤。
- 前記粒子が、アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種と、糖類及びポリソルベートから成る群より選択される少なくとも1種とが共存する条件下において、シアノアクリレートモノマーをアニオン重合させることにより製造された粒子である請求項6ないし13のいずれか1項に記載の細胞障害剤。
- 前記糖類が、水酸基を有する単糖類、水酸基を有する二糖類及び水酸基を有する多糖類から成る群より選択される少なくとも1種である請求項15記載の細胞障害剤。
- 前記粒子の平均粒径が20nm〜700nmである請求項6ないし16のいずれか1項に記載の細胞障害剤。
- シアノアクリレートポリマー粒子に抱合させるアミノ酸若しくはその誘導体又はそれらのオリゴマー若しくはポリマーの種類の選択、並びにシアノアクリレートポリマー粒子の重合方法の選択の少なくともいずれかにより、シアノアクリレートポリマー粒子の細胞障害活性を調整することを特徴とする、請求項6記載の細胞障害剤の細胞障害活性の調整方法。
- 前記重合方法が下記のいずれかから選択される請求項18記載の方法。
(1) アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種が共存し、かつ、糖類及びポリソルベートのいずれも実質的に共存しない条件下において、シアノアクリレートモノマーをアニオン重合する方法。
(2) アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種と、糖類及びポリソルベートから成る群より選択される少なくとも1種とが共存する条件下において、シアノアクリレートモノマーをアニオン重合する方法。 - アミノ酸及びその誘導体並びにそれらのオリゴマー及びポリマーから成る群より選択される少なくとも1種を抱合し、抗菌活性成分を実質的に含まず、かつ、平均粒径が1000nm未満であるシアノアクリレートポリマー粒子を有効成分として含有する、抗菌剤。
- グラム陽性細菌に対する抗菌活性成分を実質的に含まない前記粒子を有効成分とするグラム陽性細菌用抗菌剤である請求項20記載の抗菌剤。
- アミノ酸及びその誘導体並びにそれらのオリゴマーから成る群より選択される少なくとも1種を抱合する前記粒子を有効成分とする請求項20又は21記載の抗菌剤。
- アミノ酸を抱合する前記粒子を有効成分とする請求項22記載の抗菌剤。
- アラニン、グリシン、バリン、イソロイシン、セリン、スレオニン、フェニルアラニン、チロシン、トリプトファン、アスパラギン酸、グルタミン酸、アスパラギン、グルタミン、ヒスチジン及びプロリンから成る群より選択される少なくとも1種を抱合する粒子を有効成分とする請求項23記載の抗菌剤。
- 前記粒子が請求項1ないし5のいずれか1項に記載の方法で製造された粒子である請求項20ないし24のいずれか1項に記載の抗菌剤。
- 請求項1ないし5のいずれか1項に記載の方法で製造された粒子を有効成分として含有するがんの治療及び/又は予防剤。
