Abstract
Description
う蝕とは、葉の表面に付着した口腔内の原因菌の代謝産物によって歯の組織(エナメル質、象牙質、セメント質など)が破壊される感染性疾患である。ヒトの口腔内には約700種類の細菌が、総数100億個を超えて生息しており、う蝕の主な原因菌として、Streptococcus mutans、Streptococcus sobrinusなどが知られている。これらの原因菌が産生する不溶性の多糖に口腔内細菌が付着することによってバイオフィルムが歯面に形成され、形成されたバイオフィルム内の細菌が産生する酸によって歯面のpHが局所的に低下し、その結果、う蝕が形成されやすくなる。
植物乳酸菌は、植物由来の乳酸菌をいい、例えば穀物、野菜、果物、あるいはこれらを原材料に含む醗酵食品やナム等の醗酵食品の製造に関係するバナナ等の熱帯植物の葉等から分離されたものが意図される。本発明者らはこれまでに、種々の植物乳酸菌を分離しているが、他にも多くの植物乳酸菌が知られている。
(1)組成物
本発明にかかる組成物は、バイオフィルムの形成を阻害するために上記の植物乳酸菌を含有していることを特徴としている。本発明にかかる組成物は、S. mutansによるバイオフィルムの形成を阻害するのに用いることができる。また、本発明にかかる組成物は、う蝕を処置するために用いられることがより好ましい。なお、本発明の組成物は、医薬部外品であっても、食品であっても、医薬であってもよい。
本発明は、バイオフィルムの形成を阻害するための植物乳酸菌を備えているキットを提供する。本発明のキットは、う蝕を処置するために用いられることが好ましい。本明細書中にて使用される場合、用語「キット」は、特定の材料を内包する容器(例えば、ボトル、プレート、チューブ、ディッシュなど)を備えた包装が意図される。好ましくは、上記材料を使用するための指示書を備えている。指示書には、本発明のキットを、バイオフィルムの形成を阻害するために使用する際の手順が記載されていることが好ましく、う蝕を処置するために使用する際の手順が記載されていることがより好ましい。本明細書中にてキットの局面において使用される場合、「備えた(備えている)」は、キットを構成する個々の容器のいずれかの中に内包されている状態が意図される。また、本発明のキットは、複数の異なる組成物を1つに梱包した包装であってもよく、容器中に内包された溶液形態の組成物を梱包していてもよい。本発明のキットは、異なる2つ以上の物質を同一の容器に混合して備えても別々の容器に備えてもよい。「指示書」は、紙またはその他の媒体に書かれていても印刷されていてもよく、あるいは磁気テープ、コンピューター読み取り可能ディスクまたはテープ、CD-ROMなどのような電子媒体に付されてもよい。本発明のキットは、上述した組成物を構成するためにもちいられてもよく、上述した組成物に含まれる物質を別々に備えていても、上述した組成物とさらなる成分とを別々に備えていてもよい。
本発明はさらに、う蝕を処置するための方法を提供する。本発明の方法は、本発明の植物乳酸菌(または該植物乳酸菌を含有する組成物)と、う蝕の原因菌とを接触させる工程を包含する。本発明の植物乳酸菌とう蝕の原因菌とを接触させる工程は、歯磨き、指でのペーストの塗布、パッチの貼付であっても、トローチ剤またはチュアブル剤等を口に含むことであってもよく、含嗽剤を用いてうがいをする工程であってもよい。なお、本工程において、本発明の植物乳酸菌とう蝕の原因菌とを接触させる時間は、採用される手段に応じて適宜設計されるが、10秒間〜10分間の範囲内が好ましく、30秒間〜10分間の範囲内がより好ましく、1分間〜5分間の範囲内がさらに好ましい。
Claims (10)
- 籾殻または玄米から単離された、Enterococcus属の植物乳酸菌。
- E. mundtii MG3株(受託番号:NITE BP−983)である、請求項1に記載の植物乳酸菌。
- 請求項1または2に記載の植物乳酸菌を含有している、バイオフィルムの形成を阻害するための組成物。
- 前記バイオフィルムがStreptococcus mutansによって形成されるものである、請求項3に記載の組成物。
- う蝕を予防するために用いられる、請求項3または4に記載の組成物。
- 医薬部外品である、請求項3〜5のいずれか1項に記載の組成物。
- 食品である、請求項3〜5のいずれか1項に記載の組成物。
- 医薬である、請求項3〜5のいずれか1項に記載の組成物。
- 請求項1または2に記載の植物乳酸菌あるいは請求項3〜8のいずれか1項に記載の組成物を、被験体の口腔内にてStreptococcus mutansと接触させる工程を包含する、う蝕を処置する方法。
- 請求項1または2に記載の植物乳酸菌あるいは請求項3〜8のいずれか1項に記載の組成物を、被験体の口腔内へ供給する工程をさらに包含する、請求項9に記載の方法。
