Abstract
Description
ただし、式1において、Io(λ)は照明光源部21の出力を示し、τ1(λ)は波長制限フィルタ24の影響を示し、τ2(λ)は培地(培地に添加されたフェノールレッド)の影響を示し、Aは、以下の式2で表される試料の周囲媒体による光吸収特性を示す。
ただし、式2において、α(λ)は吸光係数を示し、Lは培地の光路長を示し、Cは培地中のフェノールレッドの濃度を示す。
本実施形態では、波長制限フィルタ24を照明光源部21とリング絞り22との間に配置する例を示したが、照明光源部21と撮像部8との間で、照明光源部21の光軸上であれば、どの位置に配置しても良い。波長制限フィルタ24の配置位置は、波長制限フィルタ24により制限される波長や顕微鏡1の構成に応じて決めると良い。図12に、ステージ5と撮像部8との間に波長制限フィルタ24を配置した場合の、照明光源部21から照射された光が撮像素子8Aに到達し、画像が生成されるまでの流れ図を示す。図12に示すように、波長制限フィルタ24を配置する位置を変更しても、入射光Iin(λ)は図6の場合と同様である。
Claims (6)
- 培養容器で培養される試料を観察する観察装置であって、
照明光学系を備え、前記試料を照明する照明部と、
撮像素子を備え、前記照明部により照明された前記試料の像を撮像して画像を生成する撮像部と、
前記照明光学系の光軸上で、かつ、前記照明部と前記撮像部との間に配置され、前記試料の培養に用いる培地に含まれる添加物の光吸収特性に応じて、前記照明光学系からの照明光の一部の波長を制限する波長制限フィルタと
を備えたことを特徴とする観察装置。 - 請求項1に記載の観察装置において、
前記添加物の光吸収特性は、前記培地のpH変化に伴う光吸収特性と前記培地の濃度変化に伴う光吸収特性との少なくとも一方である
ことを特徴とする観察装置。 - 請求項1または請求項2に記載の観察装置において、
前記波長制限フィルタは、前記添加物の光吸収特性に加えて、前記撮像素子の感度特性に応じて、前記照明光の一部の波長を制限する
ことを特徴とする観察装置。 - 請求項1から請求項3の何れか1項に記載の観察装置において、
制限波長の異なる複数の前記波長制限フィルタを備え、
前記添加物の光吸収特性に応じて、前記複数の波長制限フィルタのうち何れかの波長制限フィルタを前記照明光学系の光軸上で、かつ、前記照明部と前記撮像部との間に配置するフィルタ制御部を備える
ことを特徴とする観察装置。 - 請求項1から請求項3の何れか1項に記載の観察装置において、
前記波長制限フィルタは、制限波長が可変であるフィルタであり、
前記添加物の光吸収特性に応じて、前記波長制限フィルタによる制限波長を変更する制限波長制御部を備える
ことを特徴とする観察装置。 - 照明光学系を備え、試料を照明する照明部と、撮像素子を備え、前記照明部により照明された前記試料の像を撮像して画像を生成する撮像部とを備えるとともに、培養容器で培養される前記試料を観察する観察装置用の波長制限フィルタであって、
前記照明光学系の光軸上で、かつ、前記照明部と前記撮像部との間に配置され、前記試料の培養に用いる培地に含まれる添加物の光吸収特性に応じて、前記照明光学系からの照明光の一部の波長を制限する
ことを特徴とする波長制限フィルタ。
