Abstract

PURPOSE: A perhydroazepin derivative is used as a starting material to effect its dehydrative cyclization reaction to easily produce the titled substance having important pharmacological activities such as analgesic, antitussive and so on with no need of many steps such as demethylation and lessened loss.
CONSTITUTION: A perhydroazepin derivative of formula I (R, R1, R2, R3 are H, lower alkyl) is used as a starting material to effect the dehydrative cyclization reaction using a Lewis acid to produce the objective compound of formula II. The preferable example of the Lewis acid is hydrobromic acid or polyphosphoric acid and the amount is 5W15 times the weight of the substance to be dehydrated. The dehydration reaction temperature is 0W120°C. In case of using a solvent such as water or acetic acid, the reaction progresses smoothly, but the reaction time becomes longer than in case of using no solvent.
