Abstract

PURPOSE: To prepare the titled compound useful as an intermediate of antiphlogistic agent and analgesic agent, easily, economically, in high yield, by cyclizing a compound derived from o-phthalaldehydic acid and p-aminoacetophenone in a low-cost solvent with a reducing agent.
CONSTITUTION: The compound of formula I (R is H or lower alkyl) is used as a raw material, and is cyclized with a reducing agent (e.g. sodium borohydride, potassium borohydride, etc.) in a solvent such as methanol at room temperature or under heating to obtain 4-(1-oxo-2-isoindolinyl)acetophenone of formula II. As an alternative method, the compound of formula II is obtained by the one-bath reaction comprising the reaction of 4-aminoacetophenone with o-phthalaldehydic acid and the cyclization of the resultant compound of formula I without separation from the reaction mixture. The apparatus, steps, and operation of the reaction are simple. The compound of formula II can be converted easily to indoprofen (antiphlogistic agent, analgesic agent) by hydrolysis.
