Abstract

PURPOSE: To obtain high purity acyl-coenzyme A oxidase, by contacting an acyl- coenzyme A oxidase-containing liquid with a compound bonded to a water-soluble carrier, and eluting the adsorbed oxidase.
CONSTITUTION: A solution containing acyl-coenzyme A oxidase is brought into contact with a water-soluble carrier bonded with a group of formula CH3-(CH2)n- (n is 1W3) or NH2-(CH2)m-(m is 2W8), and the adsorbed acyl-coenzyme A oxidase is eluted to obtain the purified objective product. The acyl-coenzyme A oxidase- containing liquid can be prepared by culturing various microorganisms in an enriched medium. The medium pref. contains 0.5% of oleic acid, 0.5% of palmitic acid, 0.3% of yeast extract, 0.5% of (NH4)H2PO4, 0.25% of KH2PO4, and 0.1% of MgSO4.7H2O, and the culturing is pref. carried out aerobically, at 20W40°C for 1W5 days.
